Occurrence of Syzygites Megalocarpus on Wild Mushrooms and Its Cultural Characteristics (야생(野生)버섯의 기생균(寄生菌)인 이분지털곰팡이의 출현(出現)과 배양(培養) 특성(特性))

  • Syzygites megalocarpus occurred on the fruit bodies of Tricholoma matsutake, Leccinum rugosiceps, Pulveroboletus ravenelii, Russula emetica, and Amanita pseudoporphyria in the field. We could select Malt Extract Agar, Mueller Hinton Medium and Potato Dextrose Agar as optimal media among eight media. Syzygites megalocarpus showed an optimal temperature around $23^{\circ}C$ with optimal pH 6.0. Growth of S. megalocarpus on PDA was inhibited by 36.5% at 5% NaCl compared with without NaCl and did not grow at more than 8% NaCl in 2 days after inoculation. However, it grew 1.1 cm in 10% NaCl in 5 days after inoculation. Growth of Tricholoma matsutake on PDA was inhibited by increasing contration of NaCl and did not nearly grow at 2.5% NaCl in 60 days after inoculation. Because S. megalocarpus grew at high concentration of NaCl, we concluded that NaCl should not use for controlling S. megalocarpus on the fruit body of T. matsutake.

Growth Characteristics of Variety of Oyster Mushroom (Pleurotus ostreatus) as Affected by Number of Air Exchanges (느타리버섯의 품종별 환기횟수에 따른 생육특성)

  • In this study, we investigated the effects of the number of air exchanges (NAE) on shape fruit body in two oyster mushrooms, Chunchu 2# and Suhan 2#. The suitable NAEs of Chunchu 2# were $1/10h^{-1}$ at primordial induction, $1/6h^{-1}$ at early stage of growth, $1/4h^{-1}$ at middle stage of growth, $1/4h^{-1}$ at late stage of growth and those of Suhan 2# were $1/6h^{-1}$ at primordial induction, $1/6h^{-1}$ at early stage of growth, $1/4h^{-1}$ at middle stage of growth, and $1/2h^{-1}$ at late stage of growth. In those conditions, the fruit bodies grew well. $CO_2$ concentration hardly affected the primordial formation of both mushrooms. However there were ventilation disturbances over 1500ppm. For instance, the end of pileus rolled up etc. As a result, Suhan 2# required higher ventilation compared with Chunchu 2# and the lower NAE was favorable for growth.

Studies on Improvement of Cultural Practice for Lyopyllum ulmarium (만가닥버섯 병재배법 개선 연구)

  • Substrate improvement, maturation period and method of pinheading promotion were investigated to establish the culture method of Lyopyllum ulmarium which could obtained high yield in the short culture period. Rice bran+wheat bran (10%+10%) combination was selected from various additives, which showed vigorous growth of fruit body and high yield (133.8 g/850 cc). Oyster shells powder substance increased yield to 155.5 g per 850 cc capacity bottle. The suitable method for pinheading promotion was to stand the bottle reversely. The number of fruit bodies per 850 cc capacity bottle in this method was 27.2 stipes and bunch formation was good. When the maturation period of mycelium culture was 15 days to 30 days, pinheading period was 10 days. But when the maturing mycelium was 30 to 45 days, yield per 850 cc bottle was 148.2 g.

The effect of RGB LED lights on oyster mushroom (Pleurotus spp.) fruit-body characteristics (RGB LED 광원이 느타리류의 자실체 특성에 미치는 효과)

  • Light plays an important role in fruit-body development and morphology during Pleurotus spp. cultivation. To understand the effects of light color on fruit-body properties, we evaluated the fruit-body characteristics of Pleurotus spp. Varieties cultivated under blue, red, and purple LED light sources. The main results are as follows: The overall fruit-body shape showed differences depending on the color of the LED light. The fruit-bodies of mushroom cultivated under blue and purple light were generally similar to the mushroom shapes typically produced, while those of mushroom cultivated under green light were abnormally shaped, probably due to the absence of effective light source. The average cap lightness of mushrooms cultivated under blue, green, and purple LED lights was 57.0, 57.4, and 59.4, respectively. The average cap lightness of all varieties except Wonhyeong1ho and Hwang-geumsantari cultivated under the three LED light sources were statistically significantly different (P<0.05). The cap redness varied significantly depending on the LED lighting and variety. Only Gonji7hoM, the cap color mutant of Gonji7ho, showed negative cap redness values under all three LED light sources. Among the eight varieties excluding Gonji7ho, the highest cap redness was observed when cultivated under the blue LED. The average harvest weight of the varieties cultivated under purple, blue, and green LED light were 68.0, 58.3, and 50.1 g, respectively. The yield of Gonji7ho, the mushroom variety with the highest yield, cultivated under blue, green, and purple LED light were 92.8, 77.1, and 98.6 g, respectively. The earliness when grown under the purple, blue, and green LED lights were 5.3, 5.8, and 5.8 days, respectively. Among the varieties, six, three, and two cultivars showed the shortest earliness under the purple, green, and blue LED, respectively. The fruit-body lengths were 66.4, 51.8, and 46.8 mm when cultivated under green, purple, and blue lights, respectively. These results are expected to serve as a foundation for producing mushrooms with traits demanded in the market.

Varietal characteristics of cross-bred Cordyceps militaris 'Dowonhongcho' (교배육종에 의한 밀리타리스 동충하초 '도원홍초'의 품종특성)

  • Cordyceps militaris is being studied and cultivated as a medicinal mushroom having many valuable biological and pharmaceutical activities. In the breeding of new C. militaris mushroom, single ascospores were isolated and examined their mycelial growth, mycelial density, and production of stroma and perithecia. Among them selected isolates were crossed and hybrids were produced showing high quality fruiting bodies on artificial media. Mycelial growth rate of new strain 'Dowonhongcho' was higher than that of better on 'Yedang 3' on SDAY at $10-25^{\circ}C$. The stromata of new strain were club-shaped and bright orange-red. Its height was 6.1 cm and the cordycepin content was 0.34% on average. The new strain showed 9% higher yield than 'Yedang 3' with producing firmer fruit bodies. The optimum temperature for mycelial growth was $22{\sim}25^{\circ}C$ and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. Fruiting bodies were began to produce 45 days later after inoculation. This new cultivar may serve as a valuable one for artificial cultivation and industrial-scale production of C. militaris.

Changes in the microbial community of substrate and fruit body of Pleurotus ostreatus (느타리 배지 및 자실체 미생물 군집 변화)

  • In this study, we investigated the microbial community of oyster mushrooms at different growth stages at the species level. Gram-positive bacteria were predominant in the presterilized medium. On the other hand, Gram-negative bacteria were predominant in the culture-completed medium, post-harvest medium, and fruiting bodies. In addition, Pseudomonas tolaasii, which is known to cause disease in mushrooms, was confirmed in the cultured medium, post-harvest medium, and fruiting bodies, and it was determined that the mycelium culture stage was contaminated, and the reason why no disease occurred was Sphingobacterium psychroaquaticum. It was confirmed that this was because the growth of Pseudomonas tolaasii was suppressed by producing a component called tolacin. As a result of confirming the diversity of microorganisms, it was confirmed that the presterilization medium contains a variety of microorganisms compared to other growth stages, and the diversity decreases in the order of culture completion medium, fruiting body, and post-harvest medium. showed a trend. As a result of microbial similarity analysis, it was confirmed that the cultured medium and the post-harvest medium showed similar microbial communities, and in the case of fruiting bodies, there were some similarities but overall differences.

Some Factors Affecting Growth of Diehlomyces microsporus and Chemical Control of Truffle Disease in Cultivation of Agaricus bisporus (양송이 괴균병균(塊菌病菌)의 방제(防除) 및 생장요인(生長要因)에 관한 연구(硏究))

  • Mycelial growth and fruit body formation of Diehlomyces microsporus were best on mushroom spawn extract medium and rice bran extract medium, respectively. L-asparagine, fructose and glucose were good nutrient sources for mycelial growth. Optimum temperature for mycelial growth ranged at $25{\sim}28^{\circ}C$. Maximum mycelial growth occurred at pH 5.5 while optimum pH for ascospore germination was 6.0. Mycelial mats of D. microsporus did not survive at $60^{\circ}C$ for 60 minutes while ascospores at $80^{\circ}C$ for 120 minutes. Damages of fruit body of Agaricus bisporus caused by D. microsporus were maximum when the fruit bodies were infected at spawning and casing on the compost. The truffle disease could be controlled by basamid with $100{\sim}150 ppm$ treating on the compost after filling.

Effect on Mycelial Growth and Fruit Body Development According to Additives and Mixing Ratio in Pot Cultivation of Pleurotus ostreatus (느타리버섯 봉지재배시 첨가제 및 첨가량이 균사배양 및 자실체 생육에 미치는 영향)

  • This study was carried to investigate suitable additive materials and its mixing ratio in pot cultivation of Pleurotus ostreatus. The main substrate for Pleurotus ostreatus cultivation was poplar sawdust plus with waste cotton at the ratio 1 : 1 (v/v). Using pot diameter and substrate weight were 12 cm and 1 kg/pot, respectively. The higher amount of additives resulted in higher total nitrogen content and C/N ratio of substrate also decreased according to increasing mixing ratio of additives. The supplementation of cotton seed flour more than 20% caused the failure of devolopment of fruit body. The hardness of substrate in supplementation of rice bran after mycelial growth was highest any other additives. The ratio of harvest was highest in supplementation of beet pulp, $75{\sim}85%$. The fruit bodies yield and biological efficiency of supplemented with 20% beet pulp were highest during second flush.

Occurrence and Damage by Thrips on Greenhouse-Cultivated Fig (시설재배 무화과에서 총채벌레의 발생과 피해)

  • This study was conducted to assess the occurrence and damages by thrips on greenhouse-cultivated fig in Hwaseong, Gyeonggi Province, Korea. We identified the collected species as Thrips tabaci Lindeman, Frankliniella occidentalis Pergande, and F. intonsa Trybom. The density of thrips in the greenhouses during the summer months was monitored using yellow sticky traps; T. tabaci showed the highest density, followed by F. occidentalis and F. intonsa. The damages by thrips were characterized by stunted plant growth because of delayed discoloration of the pericarp, and development of rough fruit surface. Stereomicroscopic observation on the fruit flesh revealed the growth of gray mold at the damaged area, as well as the dead bodies and exuviae of thrips. The rates of fig fruit damages per month, were 18.2%, 9.7%, 2.9%, and 1.3% in July, August, September, and October, respectively.

Characteristics and breeding of a new variety Pleurotus eryngii, Gonji No. 8 (신품종 큰느타리버섯 '곤지8호' 육성 및 특성)

  • A new variety of Pleurotus eryngii which is named 'Gonji No. 8' was bred by mating monokaryotic strain isolated from E085D2 and a monokaryotic strain 'aerini No.3' obtained from the Mushroom Research Institute, Gyeonggi-Do A.R.E.S. The characteristics of the new variety 'Gonji No.8' is as follows. The optimum temperature for mycelium growth was from $23^{\circ}C$ to $26^{\circ}C$ on potato dextrose agar (PDA) medium. For the primodia formation and the growth of fruit bodies, the optimum temperature was from $14^{\circ}C$ to $16^{\circ}C$. The period of spawn running was around 30 days at $22^{\circ}C$ and the days taken after the removal of the spawn layer to initiate primodia was seven days. The hardness value of fruit body was $8,432{\pm}2,193$ $g/cm^2$, which was two times more than that of 'Keunneutari No.2'. The yield of 'Gonji No.8' was about 133 g per bottle(900cc) and it was same as 'Keunneutari No.2'.
