Development of Nested-PCR Assay to Detect Acidovorax citrulli, a Causal Agent of Bacterial Fruit Blotch at Cucurbitaceae (박과 작물에 과일썩음병을 일으키는 Acidovorax citrulli 검출을 위한 nested-PCR 검사법 개발)

  • The specific and sensitive nested-PCR method to detect Acidovorax citrulli, a causal agent of bacterial fruit blotch on cucurbitaceae, was developed. PCR primers were designed from the draft genome sequence which was obtained with the Next Generation Sequencing of A. citrulli KACC10651, and the nested-PCR primer set (Ac-ORF 21F/Ac-ORF 21R) were selected by checking of specificity to A. citrulli with PCR assays. The selected nested-PCR primer amplified the 140 bp DNA only from A. citrulli strains, and detection sensitivity of the nested PCR increased 10,000 times of $1^{st}$ PCR detection limit (10 ng genomic DNA/PCR). The nested PCR detected A. citrulli from the all samples of seed surface wash (external seed detection) of the artificially inoculated watermelon seeds with $10^1cfu/ml$ and above population of A. citrulli while the nested PCR could not detected A. citrulli from the mashed seed suspension (internal seed detection) of the all artificially inoculated watermelon seeds. When the naturally infested watermelon seeds (10% seed infested rate with grow-out test) used, the nested PCR detected A. citrulli from 2 seed samples out of 10 replication samples externally and 5 seed samples out of 10 replication samples internally. We believe that the nested-PCR developed in this study will be useful method to detect A. citrulli from the Cucurbitaceae seeds.

  • Geometrical characteristics of fruit including perimeter, projected area and length of minor and major axis were calculated by computer programs to be used in fruit sorting by image processing system. The results are summerized as follows. 1. A program calculating perimeter, projected area, and length of minor and major axis by edge detection and chain code was developed. 2. Geometrical characteristics of given figures were calculated to verify the program and the discrepancies from the measured values were about 5%. 3. Regression models for estimating volums of apples were developed and regression coefficients for each variety were found. 4. Abnormal apples could be recognized by comparing the ratio of minor axis to major axis and the standard value was proposed.

Histological Detection of Phytoalexin Scoparone from Heat-Treated and UV-Illuminated Lemon Fruits After Inoculation with Penicillium digitatum

  • Phytoalexin scoparone (6,7-dimethoxycoumarin) was induced in flavedo tissue of lemon fruit inoculated with Penicillium digitatum during heat treatment for 3 days at $36^{\circ}$. The compound was also induced in the flavedo tissue after UV illuminatiion. Induction of scoparone was deteected in the flavedo tissue by histological analysis. This fluorescent scoparone accumulated only on the 4-5 layers of cells adjacent to the inoculation site. Preinoculation with P. digitatum and subsequent heat-treatment induced resistance in the lemon fruit tissues after challenge-inoculation at the site of the first infection. the data obtained in the study suggest that lemon fruit acquired resistance against P. digitatum parallel with the scoparone production at the infection site.

Studies on the Tin Content for Fruit Tinplated Can (과실 통조림의 주석 함량에 관한 연구)

  • Authors investigated on the content of tin metals in 108 samples of tinplated canned fruit commercial. 1. The highest concentration of tin in 11 kinds of samples of canned fruit that is, apricot nectar was 114ppm and lowest of that 2.5∼3ppm. 2. There was 48.6ppm of tin content in orange nectar A that of pineapple nectar 56.6ppm in higher remarkably and that of the other samples was no appearance difference. 3. There was 114ppm(number 10), 5.3∼56.6ppm(number 8) and 8.7∼48.6ppm(number 3) of tin content in minimum between maximum concentration for difference remarkably and that of others there was no difference considerably, The highest concentration in detection frequency of content range was 8∼ 14ppm in 41. 7% and 2.5"6.6ppm in 36.1% and that of concentration was lower for comparison the others. 5. There was lower contration of tin in all samples for law of food hygiene in limitation 150ppm.

Detection of Xanthomonas axonopodis pv. citri on Satsuma Mandarin Orange Fruits Using Phage Technique in Korea

  • A phage technique for detection of Xanthomonas axonopodis pv. citri, a causal bacterium of canker on Sastuma mandarin fruits was developed. Phage and ELISA techniques were compared for their sensitivity for detection of Xanthomonas axonopodis pv. citri on orange fruits. Both of techniques revealed a similar efficiency for the bacterial detection; the pathogenic bacteria were observed in pellet from the fruits with over one canker spot with below 2 mm in diameter. In field assays, the increase of phage population(120%) on surface of the fruits related to the disease development one month later indicated that the bacterial pathogens inhabit on the surface. The procedure will be effectively used for detection of only living bacterial pathogen on fruit surfaces of Satsuma mandarin and for the disease forecasting.

  • In the operation of farms, it is very important to evaluate the quality of harvested crops and to classify defective products. However, farmers have difficulty coping with the cost and time required for quality assessment due to insufficient capital and manpower. This study thus aims to detect defects by analyzing the epidermis of fruit using deep learning algorithm. We developed a model that can analyze the epidermis by applying YOLOv3 algorithm based on Region Convolutional Neural Network to video images of peach. A total of four classes were selected and trained. Through 97,600 epochs, a high performance detection model was obtained. The crop failure detection model proposed in this study can be used to automate the process of data collection, quality evaluation through analyzed data, and defect detection. In particular, we have developed an analytical model for peach, which is the most vulnerable to external wounds among crops, so it is expected to be applicable to other crops in farming.

Efficiency of virus elimination in apple calli (cv. Hongro) derived from meristem culture of dormant buds (사과 품종 홍로의 휴면아 분열조직 배양을 통해 형성된 캘러스에서의 바이러스 제거효율)

  • Various sizes (0.2 ~ 1.2 mm) and developmental stages (referred to as Stage 1 ~ 3) of apical and lateral meristems were excised, together or separately, directly from dormant buds of apple 'Hongro'. They were mixed infected by Apple scar skin viroid (ASSVd), Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV) and Apple stem grooving virus (ASGV), which are major viruses attacking apples. A total of 31 callus lines (> 10 mm in diameter) were obtained by culturing the explants on Murashige and Skoog (MS) medium supplemented with 3% sucrose, 3.0 mg/L benzyladenine (BA) and 0.1 mg/L indole-3-butyric acid (IBA), and they were subjected to RT-PCR analysis for virus detection. A high rate of virus elimination (expressed as the percentage of calli that did not amplify during RT-PCR, i.e., RT-PCR negative calli per total number of calli obtained) was achieved for ACLSV (100%), ASSVd (93.7%), and ASPV (93.7%), whereas it was only 25.8% for ASGV. ASPV was detected in the presence of 2 ~ 3 bracts. Simultaneous virus elimination of ASSVd, ASPV, ACLSV, and ASGV occurred during the meristem culture, in which the early stages of the dormant buds (Stage 1) were used, because ASGV was mostly eliminated during that stage. The results of the present study will be valuable for the production of virus-free apple trees.

Production System of Virus-free Apple Plants Using Heat Treatment and Shoot Tip Culture (열처리와 경정배양을 이용한 바이러스 무병 사과 생산 시스템)

  • In worldwide, viral diseases of apple plants has caused the serious problems like reduced production and malformation of fruits. Also, the damages of apple plants by virus and/or viroid infection (Apple chlorotic leaf spot virus, Apple stem grooving virus, Apple mosaic virus, and Apple scar skin viroid) were reported in Korea. However there is few report about the protection approach against the infection by apple viruses. Therefore, this paper introduced the experimental protocol for the development of virus-free apple cultivars (Danhong, Hongan, Saenara, Summerdream). Apple plants were treated at $37^{\circ}C$ for 4 weeks and shoot tips were cultured in vitro. After heat treatment, the detection of apple viruses was performed by RT-PCR using virusspecific detection primers in new apple cultivars. With the heat treatments followed by in vitro shoot tip culture, the proportion of virus-free stocks of 'Danhong', 'Hongan', 'Saenara', and 'Summerdream' was 28%, 16%, 12%, and 12%, respectively. Taken together, this approach can be a good tool for production of virus-free apple stocks.

Fruit's Defective Area Detection Using Yolo V4 Deep Learning Intelligent Technology (Yolo V4 딥러닝 지능기술을 이용한 과일 불량 부위 검출)

  • It is very important to first detect and remove defective fruits with scratches or bruised areas in the automatic fruit quality screening system. This paper proposes a method of detecting defective areas in fruits using the latest artificial intelligence technology, the Yolo V4 deep learning model in order to overcome the limitations of the method of detecting fruit's defective areas using the existing image processing techniques. In this study, a total of 2,400 defective fruits, including 1,000 defective apples and 1,400 defective fruits with scratch or decayed areas, were learned using the Yolo V4 deep learning model and experiments were conducted to detect defective areas. As a result of the performance test, the precision of apples is 0.80, recall is 0.76, IoU is 69.92% and mAP is 65.27%. The precision of pears is 0.86, recall is 0.81, IoU is 70.54% and mAP is 68.75%. The method proposed in this study can dramatically improve the performance of the existing automatic fruit quality screening system by accurately selecting fruits with defective areas in real time rather than using the existing image processing techniques.

Application of time-of-flight near infrared spectroscopy to Satsuma mandarin

  • In this study, a newly constructed optical measurement system, whose main components were a parametric tunable laser and a near infrared photoelectric multiplier, was applied to detection of the information for the inside of Satsuma mandarin using time-of-flight near infrared spectroscopy (TOF-NIRS). The combined effects on the time resolved profile of sample diameter, sugar content, the wavelength of the laser beam, and the detection position of transmitted light were investigated in detail. The samples used were Satsuma mandarin (Citrus unshu $M^{ARC}$.) (location: Wakayama, Japan) having the diameters of 50-84 mm. The sugar content measured by a refractometer varied from 9.9 to 16.3 Brix%. Equator of sample was irradiated vertically with the pulsed laser, and transmitted output power was measured on the restricted position of the equator using the optical fiber cable. The sampling time and the number of averaging the output power were 100 ns and 100 times, respectively. The variation of the attenuance of peak maxima At, the time delay of peak maxima t and the variation of full width at half maximum w were strongly dependent on the detection position and the wavelength of the laser beam. At, t and w increased gradually as the sample diameter increased to be much absorbed and vigorously scattered. On the other hand, each optical parameter had a tendency to increase as the sugar content increased. Such behavior was remarkable when the transmitted light was detected at the side face of a sample. When we apply TOF-NIRS to detection of the information for the inside of fruit with high moisture content like Satsuma mandarin, it is very important to give attention to the difference in the scattered light within tissues and the semi-straightly propagated light. Furthermore, we tried to express the resulting phenomena by using a model samples composed of water, sucrose, and milk. The variation of the time resolved profile is strongly governed by the combination of the light absorption component, scattering medium, and refractive index.
