Santalum album Linn. [Family: Santalaceae] is commonly known as white sandalwood, sandal safaid and safed chandan. It is one of the most valuable trees and second costliest wood in the world. Sandalwood and its oil is extensively used in the Unani and other traditional systems of medicine as it has blood purifier, anti-inflammatory, analgesic, exhilarant, cardiotonic, antiseptic, nervine tonic and expectorant properties. It is used in skin, cardiac, liver, gastrointestinal, respiratory, integument and urogenital disorders. These uses are supported and proven by many in vitro or in vivo studies. The proven pharmacological activities of S. album are antimicrobial, anti-oxidant, anti-inflammatory, antimutagenic and anti-fatigue. The research has proven that sandal oil or its constituents have anti-microbial activity. Sandalwood oil showed skin cancer preventive effect in mice and its constituent alpha santalol showed the anticancer property. The methanolic extract of wood was confirmed for antioxidant, free radical scavenging, analgesic and anti-inflammatory activities. ${\alpha}$ and ${\beta}$ santalols present in sandal oil showed sedative effects. Sandalwood tea had a significant effect on heart muscles of frog and showed increased myocardial contractility. Its oil showed significant changes in hepatic xenobiotic metabolizing enzymes. Sandalwood oil and its major constituents showed less acute oral and dermal toxicity in laboratory animals. Hence, the aforementioned studies justify the uses of sandalwood and its oil mentioned in the classical Unani literature. However, further clinical trials are suggested to confirm its efficacy and safety in humans.
Previously, we have isolated authentic bombesin and another bombesin like peptide named bombesin like immunoreactivity (BLI)-K2 from the skin of Korean fire-bellied toad, Bombina orientalis. In the present study, we have newly purified three heterogeneous forms of BLI named BLI-K3, BLI-K4, and BLI-K5 from side fractions obtained in previous isolation of bombesin like peptide. The BLIs were separated into five peaks on a column of $C_{18}$ preparative HPLC. Among them, three minor peaks containing BLI-K3, K4, and K5 were purified by means of sequential chromatography on the columns of SP cation exchange HPLC and $C_{18}$ reverse phase HPLC. The purified BLI-K3 and K4 showed high binding affinity to an anti-bombesin serum (LBE 2G-2) with binding potency of 72 and 95%, respectively, relative to that of bombesin. However, they did not possess any distinctive biological activity of bombesin like peptide. On the contrary, the biological activity of BLI-K5 was similar to that of bombesin but its binding affinity to an anti-bombesin serum was low. The results indicate that three heterogeneous forms of BLI were coexpressed with bombesin and BLI-K2 in the skin of B. orientalis. All forms of the purified BLI in the present study were immunologically active but only BLI-K5 possessed the distinctive biological activity of bombesin like peptide.
김치의 숙성과정 중 일어날 수 있는 식품위생상 이물질의 오염방지를 위하여 혼입시험을 고안하여 이를 모니터링 하였다. 먼저, 시험군은 소금으로 절이고 양념으로 버무린 김치였고, 대조구는 소금에 절이지 않았거나 양념을 하지 않은 배추를 선택하였다. 오염원으로는 배추와 색깔이 유사한 청개구리를 선택하였다. 그 결과, 염분이 첨가되지 않은 대조구에서는 오염원이 10일 이상 생존하였으나 염분이 첨가된 시험구의 이물질은 5일 이상 생존할 수 없었다. 김치제조 중에 혼입된 생물학적 이물질은 대개 5일 정도 생존하므로 소비자에 의하여 살아있는 상태로 발견된 사실은 제조 후에 혼입되었다고 추정할 수 있다.
To know if small changes in rearing water temperature and density affect the number of bacterial and fungal colonies in metamorphosed frogs, Dybowski's frog tadpoles were reared from Gosner 25-26 stages at either low ($1^{\circ}C$ low to ambient water temperature), ambient, or high ($1^{\circ}C$ high) water temperature (each 15 tadpoles in 20 L water) condition and at either low (10 tadpoles/20 L water), medium (20 tadpoles), or high (30 tadpoles) density condition. Immediately after metamorphosis, we sampled bacteria and fungi from skin, liver, and heart of six metamorphosed frogs, randomly selected for each treatment group. After separate incubation of bacteria and fungi on 3M Petrifilm plates, we counted the number of bacterial and fungal colonies appeared on the plates and compared the numbers among the temperature and density treatment groups. For temperature treatment, high-temperature group had fewer bacterial colonies, while low-temperature group had more fungal colonies than the other two groups. For density treatment, low-density group had fewer bacterial colonies than the other two groups, but the number of fungal colonies were not different among the groups. Our results suggest that small increased rearing water temperature and lowered rearing density could potentially reduce pathogens in farming frogs.
Gaegurin 4 (GGN4), a novel peptide isolated from the skin of a Korean frog, Rana rugosa, has broad spectrum antimicrobial activity. A number of amphipathic peptides closely related to GGN4 undergo a coil to helix transition with concomitant oligomerization in lipid membranes or membrane-mimicking environments. Despite intensive study of their secondary structures, the oligomeric states of the peptides before and after the transition are not well understood. To clarify the structural basis of its antibiotic action, we used analytical ultracentrifugation to define the aggregation state of GGN4 in water, ethyl alcohol, and 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP). The maximum size of GGN4 in 15% HFIP corresponded to a decamer, whereas it was monomeric in buffer. The oligomeric transition is accompanied by a cooperative 9 nm blue-shift of maximum fluorescence emission and a large secondary structure change from an almost random coil to an ${\alpha}$-helical structure. GGN4 induces pores in lipid membranes and, using electrophysiological methods, we estimated the diameter of the pores to be exceed $7.3{\AA}$, which suggests that the minimal oligomer structure responsible is a pentamer.
Bombesin (BBS)은 유럽에 서식하는 무당개구리(Bombina bombina)의 피부로 부터 발견되었으며 이것은 14개의 amino acid로 구성되어 있음이 잘 알려져 있다. 한국에도 유럽의 무당개구리와 종(species)이 다른 무당개구리인 Bombina orientalis가 서식함이 알려졌으나 이 무당개구리의 피부에 BBS가 존재하는 지는 확실치 않다. 그러므로 본 실험에서는 한국에 서식하는 무당개구리의 피부로부터 BBS 유사물질(BBS-LS)을 추출하여 분자적 이질성과 생물학적 활성을 측정하고자 하였다. 박리한 무당개구리 피부를 100% methanol로 처리하여 얻은 추출물의 일부를 Sephadex G-50, superfine column에 부하하고 여기에서 얻은 chromatogram을 분석한 결과 BBS-LS의 3.5%는 합성 gastrin releasing peptide (GRP-27) 보다 앞에 용출되었으며 나머지 96.5%는 GRP-27과 합성 BBS-14의 사이에 용출되었다. alkaline alumina column과 Sephadex G-10, fine column 그리고 Sephadex G-50, superfine column을 연속적으로 이용하여 methanol 추출물로 부터 BBS-LS를 정제 하였다. 정제된 BBS-LS를 Sephadex G-50, superfine column으로 분석한 결과 합성 GRP-27과 합성 BBS-14 사이에 용출되어 BBS-LS가 이미 알려진 BBS-14와 분자의 크기에 있어서 다른 물질임을 알 수 있었다. 정제된 BBS-LS를 reversed phase HPLC로 분석한 실험은 이러한 결과를 더욱 확실히 뒷받침하였다. 정제된 BBS-LS를 합성 BBS-14와 비교하여 생물학적 활성을 검토한 결과 흰쥐에서 gastrin의 분비, 췌장액의 분비, 회장의 수축을 촉진하고 guinea pig 담낭의 수축을 증가시키는 등 이들 두물질이 매우 유사한 생물학적 활성을 지니고 있음을 발견하였다. 이러한 결과로 보아 한국에 서식하는 무당개구리의 피부는 서로 다른 두가지 형태의 BBS-LS를 함유하고 있으며 그 중에서 96.5%를 차지하는 BBS-LS는 합성 BBS-14와 분자적 이질성을 지니나 유사한 생물학적 활성을 지니고 있음을 알 수 있었다.
피부선이 잘 발달되어 있는 Bombina속 중에서 유일하게 국내에 서식하는 무당개구리를 재료로 하여 피부 점액선의 미세구조와 점액분비물질의 조직화학적 성질을 광학 및 전자현미경으로 관찰하였다. 무당개구리의 점액선은 진피에 위치한 단포상선으로서 선분비부는 외부의 근상피세포층과 내부의 선상피세포층으로 이루어져 있었다. 조직화학반응의 결과, 점액선에서 생성되는 물질의 주성분이 산성다당류임을 확인하였으며, 배피에 비해 복피에서 강한 양성반응을 나타내었다. 또한 주사전자현미경을 이용한 피부표면의 관찰 결과, 점액선의 개구부는 피부의 표면에 돌출된 식물의 기공과 비슷한 모양을 하고 있으며, 과립선의 개구부에 비해 분포밀도는 높았으나 크기는 소형인 것으로 관찰되었다. 점액선의 선상피세포층에는 점액분비세포와 미토콘드리아가 잘 발달되어 있는 세포의 2종류로 이루어져 있었고, 선의 대부분을 차지하고 있는 점액분비세포는 세포의 분화정도에 따라서 성숙 또는 미성숙된 세포로 구분되었으며, 세포질 내에 함유된 점액분비물질의 주변에서는 조면소포체가 잘 발달되어 있었다.
상처회복은 염증, 재상피화 그리고 기질의 재형성등이 관여하는 복잡한 과정이다. 이중 재상피화에 관여하는 각질화세포의 이동경로를 분석하기 위하여 무당개구리 피부 상처유도 후 투과전자현미경과 cytokeratin에 대한 조직면역화학법을 이용하였다. 정상조직의 cytokeratin발현은 기저층의 세포들과 선상피에서 확인되었다. 상처 유도후 3시간 조직에서, 기저세포층에서 강한 반응이 관찰되었고, 1일과 2일 사이에서는 재생되는 각질화세포에서 강한 면역반응이 확인되었다. 상처반응 중기인 7일부터 10일 사이에서도 재생된 세포의 기저층세포에서 강한 반응이 일어났다. 19일경과의 조직에서는 기저층과 유극층의 세포들에서 cytokeratin의 발현이 증가하였다. 따라서, 재상피화에 관여하는 각질화세포는 기저층의 세포로부터 시작하여 상처부위로 이동하여 과립층과 유극층으로 분화됨으로서 재상피가 진행되었다.
금개구리 (Rana nigromaculata coreana Okada) 배부피부의 색소보유세포를 전자현미경으로 관찰하였다. 3종류의 진피 색소보유세포는 서로 평행으로 위치하였으며, 그들 세포들사이의 intercellular space에 의하여 인접되었다. 각 색소보유세포의 특징은 다음과 같다. 1. 대황세포 : 대황세포들은 서로 밀접하게 접하여 있었다. 그들 세포는 pterinosomes과 carotenoid vesicles로 채워져 있었으며, 그중 pterinosomes은 5가지 형태로 구분되었고 (제1형, 제2형, 제3형, 제4형 및 제5형 pterinosomes), 특히 전형적인 제2형과 제3형 pterinosomes이 발달되었다. 2. Iridophore:들은 위에 위치한 대황세포와 인접하였고, 그들은 iridophore reflective platelets로 채워져 있었으며, 이 reflective platelets는 대개 평행하게 배열되었으나, 일부분은 이 평행면과 다른 방향으로 위치하였다. 3. 멜라닌보유세포:멜라닌보유세포는 멜라닌과립으로 채워져 있었으며, 이 과립들은 같은 전자밀도를 나타내었다. 멜라닌보유세포의 돌기는 iridophore 양 측면으로 뻗어 있었고, 이 돌기속의 멜라닌과립은 드문드문 분산되어 관찰되었다.

The structure and dynamics of a 37-residue antimicrobial peptide gaegurin 4 (GGN4) isolated from the skin of the native Korean frog, Rana rugosa, was determined in SDS micelles by NMR spectroscopy. The solution structure of the peptide in SDS micelles was determined from 352 NOE-derived distance constraints and 22 backbone torsion angle constraints. Dynamic properties for the amide backbone were characterized by $^1H-^{15}N $heteronuclear NOE experiments. The structural study revealed two amphipathic helices spanning residues 2-10 and 16-32 and that the helices were connected by a flexible loop. An intraresidue disulfide bridge was formed between residues Cys31 and Cys37 near the C-terminus. The loop region (11-15) connecting the two helices are were slightly more flexible than these helices themselves. From the fact that since there is no contact NOEs between two helices, it is implied that the GGN4 peptide shows an independent motion of both helices which has an angle of about $ 60^{\circ}-120^{\circ}$ from each other.
