Characterization of Photobacterium sp. YW2207 isolated from rainbow trout (Oncorhynchus mykiss) raised in a fresh water farm in South Korea (국내 양식 무지개송어(Oncorhynchus mykiss)에서 분리된 Photobacterium sp. YW2207의 특성)

  • Photobacterium sp. YW2207 was isolated from rainbow trout raised in a fish farm located in Yeongwol-gun, Gangwon Province, South Korea. Based on 16S rRNA sequence analysis and phylogenetic analysis, it was confirmed that Photobacterium sp. YW2207 showed 100% similarity with Photobacterium piscicola and Photobacterium phosphoreum, and 94.6% similarity with P. damselae subsp. damselae. Biochemical analysis revealed that Photobacterium sp. YW2207 is a Gram-negative, motile bacterium with a cell size of 1.5~3×3~5 ㎛. The bacteria were cultured on nutrient agar, brain heart infusion agar, Muller-Hinton agar, tryptic soy agar, and thiosulfate citrate bile sucrose agar with NaCl concentrations ranging from 0 to 2.5%. The API50CHE and API20E tests indicated lower utilization capabilities compared to the P. damselae strains provided in the API database. Furthermore, unlike most Photobacterium species, Photobacterium sp. YW2207 presented negative for catalase test. Results from the flow cytometric measurement indicated that Photobacterium sp. YW2207 exhibited a more diverse distribution of cell sizes and had larger cell sizes compared with P. damselae subsp. damselae. Minimum inhibitory concentration tests showed that Photobacterium sp. YW2207 had low susceptibility to β-Lactam and aminoglycoside antibiotics, while having high susceptibility to tetracycline, doxycycline, and quinolone antibiotics. Pathogenicity on rainbow trout revealed that an immersion of 1×105 CFU/ml did not cause mortality or clinical symptoms.

  • This study aims to find the taste compounds of wild loach, Misgurnis mizolepis, which is important fresh-water fish in this country. In order to elucidate the origin of the taste of wild loach meat, free amino acids, organic bases, nucleotides and their related compounds, organic acids, sugars and minerals were analyzed, and then followed by organoleptic test of synthetic extract prepared on the basis of the analytical data. Sensory evaluations of synthetic extracts prepared by omitting each extractive components were carried out by a triangle difference test, and changes in taste profile were assessed. In free amino acid composition, histidine, threonine, glycine, lysine, alanine were abundant. IMP and AMP were dominant in the wild loach in content. Total creatinine and butyric acid were abundant. As for the minerals, $Na^+,\;K^+,\;Cl^-\;and\;PO_4^{3-}$ were found to be the major ions. From the results of omission test of synthetic extracts the major components which contribute to produce the taste were arginine, glutamic acid, valine, aspartic acid, IMP, succinic acid, $Na^+,\;K^+,\;Ca^{2+}\;Mg^{2+}\;Cl^-\;and\;PO_4^{3-}$.

Metarercariae of Echinochusmus japonicus Encysted in a Fresh Water Fibh, Pseudorasboru purva, and Their Development in Experimental Mice (참붕어(Pseudorasbora parva)에서 분리한 Echinochasmus japonicus 피낭유충 및 마우스 실험감염)

  • The echinostomatid metacercariae encysted in the gill of the fresh water fish, Pseudorasbora larva were identified through obtaining adult worms after eBperimental infection to mice. In addition, a brief course of worm development and maturation was observed in this experimental host. The results were as follows: 1. The echinostomatid metacercariae were elliptical, golden yellow, 0.073∼0.078 mm long and 0.0541∼0.065 mm wide. Their head portions were characterized by the presence of a head crown armed with collar spines of total 24 in number and interrupted at the mid-dorsal side of the oral sucker. 2. The average rate of worm recovery froth 12 mice (on the 1-2lth postinfection days) was 19.4 % and the rate revealed no decrease in accordance with the increase of infection duration. The worms were collected chiefly from the lower part of the small intestine. 3. After the infection, their sexual maturation was attained in 5 days and their growth in size nearly completed in 7 days. The early growth curve of genital organs was S shape while that of nongenital organs was C form. In 5 day old worms, 1 or 2 eggs were found from their uteri and the stools of mice revealed echinostomatid eggs from the 5-6th postinfection day. 4. The 7 day old adult worms were ovoid in shape, 0.54-0.69 mm long and 0.29-0.34 mm wide, and characterized by a well developed head crown with 24 collar spines and vitelline follicles distributed from the acetabular level down to the posterior end of body. Based on these characters they were identified to be Echinochasmus japonicus Tanabe, 1926. From these results, it is verified that p. larva is one of the second intermediate hosts of 5. jatonicus in Korea.

  • Mitochondrial DNAs of two Mdh allelotypes of the dark chub, Z. temmincki inhabiting in Korean fresh water, were analysed. Samples of each type were collected from four populations, and the fragment patterns for mtdNA of each type were explained from 7 of the eleven restriction enzymes with hexanucleotide recognition site. Genome size was approximately 16.7 kilobases. The highly typical mtdNA fragments of each type were discovered in digestion profiles produced by Eco RI and Pst I enzyrnes. The comparisons of restriction fragment patterns and relative digestion maps permitted the estimation of fragment homology (F) and nucleotide sequence divergence(p). Between the two identical types, sequence divergence(p) was 0.128(MS), and 0.045(MM), ; between the two different types, 0.195 (range 0.177-0.226). These result may provide a distinct difference more than the value derived from allozyrne analysis, and a powerful new molecular approach for assessing genetic-evolutionary relationship among fishes.

BREEDING OF THE PUFFER FUGU RUBRIPES (자주복 Fugu rubripes (Temminck et Schlegel)의 종묘 생산에 관한 연구)

  • Fingerlings hatched from the eggs of the puffer, Fugu rubripes, which were spawned on May 21, 1969, and were cultivated. The results of their growth during 150 days, until October 25th, are summarized as follows: 1. The eggs began to hatch after 163 hours, at a water temperature of 15.9 to $17.4^{\circ}C$, and the hatching rate was $61.56\%$. 2. They reached the post-larval stage 6 to 7 days after hatching, and at this time a high mortality occurred. The mortality rate was 57.26 to $68.0\%$. 3. Sixteen days after hatching some of the larger fingerlings (6.7mm in total length) began attacking some of the smaller ones (4.6mm in to total length). 4. Twenty five to twenty eight days after hatching, the fish changed their food, and at this time a second high mortality occured. The total mortality rate amounted to 90.7 to $90.9\%$ of the total hatch. 5. After the fingerling stage. cannibalism occurs. The fish usually attack the caudal part of other fingerlings. It occurs regardless of body length and when the food supply is short. 6. The food coeffiicient at the age of 46 days (when body length is 53 to 68 mm) was 5.5 for short-necked clams, 8.5 for earth-worms, and 8.7 for fishes. 7. A third hish mortality occurred 53 to 63 days after hatching, the total mortality amounting to 95.76 to $97.34\%$, and the main cause of the mortality was found to be rickets resulting from nutritional deficiency 8. The growth rates were as follows: 2.68mm just after hatching, 3.84mm at the age of 10 days; 7.96mm after 25 days; 20.96mm or 130mg after 40 days; 73.68mm or 9.06g after 80 days: and 123mm or 31.8g after 150 days. 9. The water temperature during the above period was 15.7 to $28.4^{\circ}C$ with an average of $22.10^{\circ}C$ and the salinity was 25.53 to $34.50\%$ with an average of $32.07\%$, 10. The young of this species could endure well a wide range and sudden rise in salinity, and could survive easily when the salinity suddenly fell to $5\%$, but a considerable mortality occured when it fell to $3\%$. 11. When the fish were tranferred directly to fresh water from normal sea water they died out in 9 hours and 40minutes. However, when transferred from water of $5\%$ salinity at which they were reared for 54 days, they survived for 60 hours and 40 mimutes longer than in the former case.

A Study on the Prevalence of Clonorchis Sinensis and the Effects of Educational Program among Residents in the Basin of the Youngsan River, Korea (영산강 유역 주민의 간흡충 감염실태와 감염 예방교육 프로그램의 효과)

  • Purpose: The goal of this study was to analyze the prevalence of Clonorchis sinensis infection in people living within 5 km of the Youngsan River basin, to develop an educational program to prevent the infection, and to examine the effects of the educational program. Methods: This study employed a one group pretest-posttest design, the subjects were 384. This study was conducted from November 20, 2008 to June 16, 2009. The results were analyzed with the SPSS/WIN 18.0 program. Results: Of the residents, 5.7% were infected with Clonorchis sinensis and the knowledge level about the prevention of Clonorchis sinensis was improved significantly from $9.57{\pm}5.12$ points before the education to $15.05{\pm}2.43$ points after the education. One of the important effects was that the willingness to eat raw fresh-water fish was reduced after the education. Conclusion: Based on the results of this study, a continuous research needs to be conducted on how education and knowledge level-up change people's living attitudes for the prevention of Clonorchis sinensis infection and the reduction of the infection rate in the future.

A Study on the Fine Structure of Clonorchis sinensis, a Liver Fluke V. The Mature Spermatozoa (간흡충의 미세구조에 관한 연구 V. 정자의 미세구조)

  • An ultrastructural study on the mature spermatoBoa oi Cloptorchis sineitsis was carried out. For this study, the liver cukes were collected from the livers of rabbits and rats artificially infected with the metacercariae obtained from the fresh water fish, Pseudorasbora parve. Six-month old worms were used. The collected liver fiukes were washed with 0.85% saline solution and then immediately moved to cold 2% glutaraldehyde buffered with 0.1M Millonig's phosphate buffier (pH 7.4) . The materials were dissected into appropriate pieces in the fixative about 30 minutes after beginning of the fixation. Two hours later the materials containing the seminal receptacle were rinsed several times with the buffier and were secondarily fixed with cold, bugeyed 1% osmium tetroxide(OsO4) for 2 hours. The fully mixed tissue blocks Ivere dehydrated in a series of graded concentrations of acetone and were embedded in Epon 812 mixture. Thin sections obtained from LKB-5 ultramicrotome were stained with uranyl acetate and Reynold's lead citrate. Observations of the sections were carried out with JEM-100CX II electron microscope, In general, the mature sperm was long thread-like form with a sickle-shaped head. According to the longitudinal sectioned view of the sperm tail, the nucleus seemed to be spirally coiled and run a little far along the tail. The acrosome was not observed. The cytoplasm of the tail was biflagellated as usual in trematodes. Unlike other platyhelminth spermatozoa, the sperm tail of Clenorchis sinensis showed the ${\ulcorner}9+2{\lrcorner}$ in the microtubular arrangement. The mitochondria with poorly developed cristae were observed throughout the middle piece. The middle piece of the tail showed dull ladder or triangular shapes with the two flagella at the bottom. But, the principal piece of the tail was slightly flattened cylindrical shape with two aagella within the cytoplasm. The end piece was uniflagellated. It was not clearly identised whether the end piece was subdivided into two by aagellum or the lengths of the two aagella were different. The glycogen granules were rich in the cytoplasm throughout the lenght of the spermatozoa. These granules might be the energy source for the movement of the spermatosoa.

Ion chromatographic determination of chlorite and chlorate in chlorinated food using a hydroxide eluent

  • This study was conducted to develop an analytical technique for determination of chlorite and chlorate concentrations in fresh-cut food and dried fish products by an ion chromatography/conductivity detection method using a hydroxide mobile phase. Deionized water was added to homogenized samples, which were then extracted by ultrasound extraction and centrifuged at high speed (8,500 rpm). Subsequently, a Sep-Pak tC18 cartridge was used to purify the supernatant. Chlorite and chlorate ions were separated using 20 mM KOH solution as the mobile phase and Dionex IonPac AS27 column as the stationary phase. Ethylenediamine was used as sample preservative and dibromoacetate was added to adjust for the disparity in extraction efficiencies between the food samples. The method detection limit) for chlorite and chlorate were estimated to be 0.2 mg/kg and 0.1 mg/kg, respectively, and the coefficient of determination ($r^2$) that denotes the linearity of their calibration curves were correspondingly measured to be 0.9973 and 0.9987. The recovery rate for each ion was 92.1 % and 96.3 %, with relative standard deviations of 7.47 % and 6.18 %, respectively. Although neither chlorite nor chlorate was detected in the food samples, the analytical technique developed in this study may potentially be used in the analysis of disinfected food products.

Comparison of Carotenoid Pigments in Korean Bittering, Cheilognathus signifer and Bride Bittering, Rhodeus ukekii in the Subfamily Cyprinidae (잉어아과에 속하는 묵납자루와 각시붕어의 Carotenoid 색소성분의 비교)

  • Differences in carotenoid composition in the integument of Korean bittering, Cheilognathus signifer and bride bittering, Rhodeus ukekii which are Korean native fresh water fish were compared. Total ca rotenoid contents in the integument of wild Korean bittering was 2.11mg% and composed of 42.6% zeaxanthin, 12.1% diatoxanthin and 12.1% lutein epoxide which are predominant carotenoids and 10.3% cynthiaxanthin, 8.3% zeaxanthin epoxide, 6.4% lutein and 1.5% cryptoxanthin which are minor carotenoids. Total ca rotenoid contents in the integument of wild bride bittering was 4.99mg% during a spawning period but after the spawning period it was decreased to 4.17mg% and carotenoid composition of bride bittering during the spawning period was 46.7% zeaxanthin, 26.5% diatoxanthin and 12.3% lutein which are predominant carotenoids, and 6.2% zeaxanthin epoxide, 3.1% cynthiaxanthin, 2.9% cryptoxanthin and 0.7% canthax anthin which are minor carotenoids. These results indicated that the carotenoid composition of bride bittering during spawning period was very similar to that of Korean bittering and carotenoid composition of bride bittering after the spawning period was 30.5% diatoxanthin, 21.5% cynthiaxanthin and 16.8% zeaxanthin which are predominant carotenoids and 14.0% cryptoxanthin, 11.3% lutein and 3.4% can thaxanthin which are minor carotenoids, indicating that after the spawning period, the content of zeaxanthin was decreased while that of cryptoxanthin and cynthiaxanthin was increased as compared to that of the spawning period. Total carotenoid contents in Korean bittering and bride bittering was relatively higher than that in other species of cyprinidae whereas composition of the carotenoid was similar.

A case of Diphyllobothrium latum infection with a brief review of diphyllobothriasis in the Republic of Korea

  • A case of Diphyllobothrium latum infection in a 49-year old man is described, and diphyllobothriasis latum in the Republic of Korea is briefly reviewed. An incomplete strobila of a tapeworm, 95 cm in length, without scolex and neck, was spontaneously discharged in the feces of a patient. On the basis of morphologic characteristics of the worm and eggs, the worm was identified as D. latum. The patient was successfully treated with a single dose (15 mg/kg) of praziquantel. The most probable source of infection was salmon flesh according to the past history of the patient. The first case of D. latum infection was documented in 1971, and this is the 43rd recorded case in the Republic of Korea. The 43 cases were briefly reviewed. The patients' main complaints were gastrointestinal troubles, such as mild abdominal pain, indigestion, and diarrhea, and discharge of tapeworm segments in the feces. The suspected infection sources included raw or improperly cooked flesh of fresh or brackish water fish, including the perch, mullet, salmon, and trout.
