한국산 복어의 독성 3. 선어와 냉동어 국매리복의 독성차이 (Toxicity of Pufferfish in Korea 3. Comparison in the toxicity of fresh and frozen pufferfish Tikifugu vermicularis radiatus (Gukmeri-bok))

  • 국매리복은 근육의 독성이 약독이라고 알려져 왔다. 하지만 국내에서 국매리복의 섭식이 많은데도 불구하고 아직 식중독 사고가 발생하지 않았는데, 이는 근육의 독성이 이제까지 알려진 것과 다를 가능성을 시사한다. 따라서, 본 연구에서는 선어와 냉동어에서 근육의 독성 (유독개체출현율, 최고독성 및 평균독성)을 조사하였으며, 그 결과 선어의 근육은 무독이었지만 냉동어의 근육은 약독이라는 것을 확인할 수 있었다 냉동한 국매리복의 근육이 약독인 것은 냉동과 해동과 같은 물리적인 처리로 인해 생긴 것임을 강하게 보여주며, 아울러 앞으로 복어류의 독성을 조사할 경우에는 냉동어뿐 아니라 선어도 조사해야 할 것이다.

Mitochondrial Malate Dehydrogenase 활성을 이용한 냉장계육과 냉동계육의 판별법에 관한 연구 (Study on the Method of Differentiating between Fresh and Frozen Chicken Meat by Using Mitochondrial Malate Dehydrogenase Activity)

  • 본 연구는 미토콘드리아 malate dehydrogenase활성을 이용하여 냉장계육과 냉동계육의 판별법을 개발하기 위하여 실시되었다. 단백질의 함량은 8.5mg/mL에서 12.7mg/mL 범위로 나타났으며, 동일 저장 기간 동안의 온도 별 차이점은 모든 부위에서 1일에서 15일 저장기간이 길어짐에 따라 단백질함량이 늘어나는 경향을 보인다. 냉장육의 보수력은 저장 기간 1일 60.5%에서 15일 33.9%로 감소하였으며 유의적 차이는 없었다(p>0.05). 냉장계육과 냉동계육은 저장 기간이 길어짐에 따라 육즙손실량이 증가하였으며 유의적 차이는 없었다.(p>0.05). 냉장계육의 육즙손실량이 냉동계육의 육즙손실량보다 낮게 나타났다. 냉장계육과 냉동계육 간의 mitochondrial malate dehydrogenase의 활성은 냉동계육이 냉장계육보다 효소활성이 높게 나타났다. 따라서 본 연구결과로부터 mitochondrial malate dehydrogenase의 활성 여부로 계육의 냉장, 냉동 유무를 판별하는데 유효한 지표로 사용 가능한 것으로 사료되었다.

Production of Plasminogen Activators during In Vitro Maturation of Fresh or Frozen- Thawed Oocytes in the Pig

  • This study were examined whether plasminogen activators (PAs) are produced by porcine fresh or frozen-thawed cumulus-oocytes complexes (COCs) and cumulus cell free-oocytes. In fresh or frozen-thawed COCs and oocytes for 0 hour cultured, no activity of PAs was detected. However, at 24 hours of culture urokinase-type plasminogen activator (uPA) was detected in COCs and denuded oocytes. In the frozen-thawed COCs and cumulus cell free-oocytes cultured for 24 hours, no PAs were observed. After COCs were cultured for 48 hours, tissue-type plasminogen activator (tPA) and tPA-PAI were observed in COCs only. In the frozen-thawed COCs and cumulus cell free-oocytes cultured for 48 hours, no PAs were observed. These results suggest that uPA, tPA and tPA-PAI are produced by porcine COCs, but only uPA by oocytes during maturation for 24 hours. Only tPA, and tPA-PAI are produced by COCs cultured for 48 hours, and no PAs are produced by denuded-oocytes cultured for 48 hours. In all of the frozen-thawed groups, no PAs are observed by COCs and denuded-oocytes.

Analysis of the electrical properties of pork to discriminate between fresh and frozen/ thawed pork

  • The thawing process is usually essential for imported pork because this product is typically distributed frozen. Consumers prefer fresh pork because discoloration, nutrient spills, and microbial contamination are high during the thawing process. The illegal act of selling frozen pork by disguising it as fresh pork through various methods can occur for the benefit of the difference in the sales price. However, there is some difficulty in securing systematic and objective data, as sensory tests are generally performed on imported pork. In the experiment conducted here, the electrical conductivity and dielectric properties of pork neck and pork belly products were measured. The amounts of change before and after freezing were compared through a statistical analysis, and a new method for determining frozen meat was proposed based on the analysis results. The weight was reduced compared to that before freezing due to the outflow of drips from the thawing process, but there was no difference in the drip loss level due to the thawing method. Vacuum packaging was found to lead to more drip loss than regular packaging, but the difference was not statistically significant. Frozen pork neck meat can be determined by measuring the electrical conductivity in the lean parts and the dielectric characteristic in the fatty parts. Frozen pork belly is determined by measuring the dielectric constant of the part closest to the outer fat layer.

동결.융해시킨 햄스터 난자에서 장자침입의 지연 (Delayed Sperm Penetration into Frozen-Thawed Zona-Free Hamster Oocytes)

  • Frozen storage of the oocytes has been used in a few mammalian species including mouse, hamster, human and cattle. However, frozen4hawed oocvtes show different sperm penetration on the levels of the zona pellucida and the plasma memhrane when compared with fresh oocytes. To elucidate biological changes occurring during freezing and thawing, we examined the kinetics of sperm penetration into frozen-thawed hamster oocytes. Oocytes obtained from superovulated female golden hamsters were frozen-thawed in an autofreezer according to an established method. Fresh and frozen4hawed oocytes were fertilized in vitro with capacitated hamster spermatozoa after removing the zona pellucida. The oocytes were examined at 1, 2, 3 and 6 h postinsemination. Sperm penetration found to be 1 h delayed in frozen-thawed oocytes. Other parameters such as degree of polyspermy and decondensing sperm heads were not affected by freezing and thawing. The results suggest that freezing and thawing may cause changes in the egg membrane surface and subsequently which leads to delay in the sperm-egg fusion.

한우에서 수정란 이식의 효율 증진에 대한 연구 I. 수정란의 조건이 이식 후 수태율에 미치는 영향 (Studies on the Improvement of Embryo Transfer Efficiency in Korean Cattle I. Effect of Embryo Conditions on Pregnancy Rate after Embryo Transfer)

  • This study was carried out to establish an effective system for embryo transfer techniques by analyzing several factors affecting in-vivo embryo transfer in Korean cattle. Embryos produced in-vivo were transferred into a total of 301 recipients. The results obtained in studies on the factors affecting pregnancy rate after embryo transfer by condition of embryos were as follow ; 1. The pregnancy rate of 301 recipients was 45.2% and higher with fresh embryos than with frozen embryos(63.5% : 21.4%, P<0.01). Embryos superovurated by FSH-P had slightly greater than by SUPER-OV in pragnancy rate, athough these were no difference between two treatments. 2. The pregnancy rates of transferred morulae and blastocysts showed no difference between fresh and frozen embryos(63.5% : 63~6% ; 20.0% : 25.8%). However, the pregnancy rates by quality of flesh and frozen embryos were significantly different(P<0~05). The pregnancy rates were outstandingly high in the grade A, B of fresh embryos(59.0~66.4%), and in the grade A of frozen embryos(43.6%). 3. The number of transferred embryos showed no difference in pregnancy rate, but when frozen embryos transferred, the pregnancy rate was slightly higher with two embryos than that with one embryo.

동결 전처리가 매실의 착즙과 건조 특성에 미치는 영향 (Effects of Freezing Pretreatment on Juice Expression and Drying Characteristics of Prunus mume Fruit)

  • 매실(Prunus mume)의 착즙과 건조 특성에 동결 전처리가 미치는 영향을 조사하기 위하여, 매실을 동결처리($-20^{\circ}C$)하거나 처리하지 않고 착즙수율과 주스의 품질특성 및 건조속도와 분말의 품질특성을 각각 측정하였다. 매실의 착즙수율과 건조속도는 동결처리구가 무처리구보다 유의적으로 높은 수준을 나타내었다. 압착주스와 건조분말의 L과 b값 및 a값은 동결처리구가 무처리구보다 유의적으로 낮고 높은 수준을 각각 보였다. 압착주스와 건조분말의 유리당과 유기산 함량은 동결 전처리에 따른 유의적인 차이를 나타나지 않았다. 주스의 가용성 고형분 함량, 적정산도 및 pH는 동결 전처리의 영향을 받지 않은 것으로 나타났다. 이로써 동결 전처리는 매실의 착즙과 건조 효율을 증대시킬 수 있는 유효한 방법이지만 착즙액과 건조분말의 갈변도가 높은 문제점이 있는 것으로 확인되었다.

Analysis of Membrane Integrity and Mitochondrial Activity in Fresh and Cryopreserved Boar Sperm Using Flow Cytometry

  • This study was carried out to evaluate the effects of washing medium, breed and washing temperature of fresh and frozen-thawed boar sperm on mitochondrial activity and membrane integrity by flow cytometry. More than $80\%$ of fresh sperm washed with mTLP-PVA medium at $20^{\circ}C$ exhibited an intact membrane and a functional mitochondrion. With frozen-thawed samples, a large number of sperm showed both damaged membrane $(36.4\~46.9\%)$ and nonfunctional mitochondrion $(55.1\~71.1\%)$ in the mTLP-PVA and BTS washing media at $20^{\circ}C$. There were no breed effects of fresh and frozen-thawed sperm on mitochondrial activity and membrane integrity. The percentages of damaged membrane of fresh and frozen sperm, respectively, were higher at $4^{\circ}C$ washing temperature than at $20^{\circ}C$ washing temperature in the mTLP-PVA medium. We found that washing medium and washing temperature of fresh and frozen-thawed boar sperm were important for the analyses of mitochondrial activity and membrane integrity by flow cytometry.

Ribose-induced Maillard Reaction as an Analytical Method for Detection of Adulteration and Differentiation of Chilled and Frozen-thawed Minced Veal

  • Quality control of meat products is one of the main concerns of consumers, governmental control authorities, and retailers. The purpose of this study was to employ ribose-induced Maillard reaction in detection of meat adulteration and differentiation of fresh-chilled from frozen-thawed minced veal. The browning intensity was assessed through measuring the absorbance at 420 nm with a spectrophotometer as well as the direct analysis of the color and pH. The results showed that CIE b*, CIE a*, and A420* values in the extract of fresh-chilled veal were significantly (p<0.05) higher than frozen-thawed samples. The extract of frozen meat samples stored at -18℃ became significantly darker and more yellowish compared to -4℃. The results showed that the A420* value in the frozen-thawed veal stored at -4℃ and -18℃ was reduced by approximately 17.22±3.53% and 11.68±2.49%, respectively, compared with fresh-chilled veal. The findings also showed that the storage temperature of minced veal and the heating time in this reaction had a significant effect on all tested variables (p<0.0001). The proposed method can be considered as an easy, quick, and inexpensive test for differentiating between the fresh-chilled and frozen-thawed minced veal.

신선편의 냉장·냉동 과채류에서 Listeria monocytogenes의 예측모델 개발 (Development of Predictive Models of Listeria monocytogenes in Fresh-Cut Fruits and Vegetables)

  • 본 연구에서는 농산물에서 오염 가능성이 있는 병원성식중독 균 L. monocytogenes에 대해 신선편의 샐러드, 파인애플, 냉동망고에서 예측 모델을 개발하고, 본 연구에서 개발된 예측 모델을 다른 제품에서 적용 여부를 검증하였다. 시료에 L. monocytogenes를 접종하여 각각의 저장 온도에 보관 시 샐러드는 13℃, 파인애플은 10℃ 이상에서 성장하였으며, 두 식품 중 파인애플에서 L. monocytogenes가 더 빠르게 성장하는 것으로 확인 되었다. 또한, 냉동망고에 접종한 L. monocytogenes는 -2, -10, -18℃의 저장온도에서 온도가 낮아질수록 delta 값이 커지며 생존력이 높아지는 양상을 보였다. 본 실험 검증을 통해 같은 신선편의 과일, 채소 식품 그룹에 속하더라도 식품 각각의 특성에 따라 L. monocytogenes의 성장 패턴은 일정하지 않으며 각기 다른 행동 패턴을 보이는 것으로 확인하였다. 신선편의 샐러드 및 절단된 과일류는 냉장유통 되며 추가세척 없이 소비되는 제품 특성상 공정과정에서 L. monocytogenes에 의한 오염이 일어나지 않도록 위생관리에 주의하고 유통과정에서 온도 남용이 되지 않도록 유통온도 관리에도 유의해야할 것으로 사료된다.
