Evaluation on the Durability of High Performance Concrete Used Expansive Additive and Shrinkage Reducing Agent (팽창재와 수축저감제 사용 고성능 콘크리트의 내구성 평가)

  • Generally, the high performance concrete of drying cracking and autogenous shrinkage are tend to be increased. In the previous study, it was found that the using method in combination with expansive additive and shrinkage reducing agent was more effective than the separtely using method of that. This study is to investigated the durability of high performance concrete using expansive additive and shrinkage reducing agent. Test results showed that the high performance concrete using expansive additive and shrinkage reducing agent had very good not only the durability performance such as salt injury, carbonation, resistance to freezing-thawing and permeability but also the resistance to shrinkage.

  • In this study, the repair method using PVA fiber reinforced mortar evaluated on durability performance. Test of salt injury, carbonation, freezing-thawing, chemical attack, permeability was performed As for the test results, it was found that durability performance of the repair method using PVA fiber mortar showed more better than the existing repair method. Therefore, appling on the repair method using PVA fiber reinforced mortar, the repaired concrete structures can be increased to service life.

  • The freezing susceptibilities of two larval stages (D-shaped and umbo) of the pearl oyster (Pinctada fucata martensii) were evaluated by the electron microscopy (light, transmission electron and scanning electron). The morphological shapes were examined from each pre-frozen or frozen-thawed stage of the cryopreserved larvae in liquid nitrogen by using the cryoprotectant, dimethyl sulfoxide ($Me_2SO$) mixed with sucrose. Although a portion of the shell was damaged, the hinge and prodissoconch were intact and clearly visible after preservation in liquid nitrogen. In addition, the cytoplasm of the frozen-thawed larvae maintained the normal organelle integrities, e.g., endoplasmic reticula, lipid droplets, mitochondria, nucleus and microvilli. However, some of the frozen-thawed larvae showed irregularly arranged cilia, rough shell surfaces and round-lumped cilium heads. These results indicate that P. fucata martensii larvae are susceptible to freezing, at least at those two critical developmental stages (D-shaped and umbo), and suggest a new industrial investigation including reduction method of cell injury for preserving microbial starter cultures need to be developed.

  • Grapevines will experience various types of winter damage. Some winter damages are caused by mechanical injury, freezing temperatures or poor vine vigor. This research was conducted to find out the appropriate control methods through selection of resistant rootstocks and improvement of overwintering methods for the control of crown gall disease on 'Kyoho' grape. The crown gall symptoms were not found when three stock plants of grapevine SO4, 5BB and 3306 were inoculated with $10^4cfu/ml$ of Agrobacterium vitis strains (YK2823, YK3312, LMG259, HKA234). But when they were inoculated with higher concentration $(10^6 cfu/ml)$ of A. vitis, irrespective of stocks plants, crown galls were formed all of them and the gall size was much smaller than that of kyoho. Three stock plants were selected as resistant based on above mentioned. Covering trunks and branches with rice straw and insulating coverlet was the most effective method for prevention of crown gall disease. This treatment minimized the ambient temperature changes on grapevine trees during winter season to $9.6^{\circ}C$ and the normal plant growth was due to the absence of freezing injury.

  • Despite frequent freezing injury to tea trees due low temperature, drought, and strong wind during wintertime, no comprehensive measurements have been taken. We selected and examined 9 locations in Hwagae-myeon and 4 places in Agyang-myeon, Hadong-gun, Gyeonsanggnam-do where low temperature damage had occurred between December 2010 and February 2011. Our objective is to examine the effect of frost damage on the morphological symptom and harvest of a tea tree exposed to a constant low temperature environment during wintertime. The results of our analyses on meteorological environment, tea leaf chromaticity, water content and trypan blue are as follows: (1) the number of days with temperature of $-10^{\circ}C$ or less, which were subject to frost damage to a tea tree were 8 and 13.6% during the winterization period in 2011; (2) the accumulated time was 1,308 minutes, and the longest duration at $-10^{\circ}C$ was 588 minutes from 21:08 p.m. 15 January to 7:30 a.m. $16^{th}$ January. The rainfall was only 104 mm which was 306 mm less than the previous year; (3) the lightness L values in 2011 were higher than in 2012 due to dehydration and necrosis by blue discoloration and red discoloration at all areas in chromaticity measurement; (4) the water content in a tea leaf in 2011 was higher than in 2012 due to low rainfall and strong wind, and almost no cell death phenomenon was observed from normal tea leaves subject to no low temperature stress in a trypan blue analysis; and (5) partial coloration due to cell death, however, took place in the leaves damaged by blue discoloration subject to low temperature stress, and most coloration due to cell death took place in the leaves damaged by red discoloration.

  • The objective of this study was to optimize the freezing/thawing method of in vitro produced Hanwoo blastocysts. Day 7 blastocysts after IVF were vitrified using EFS40 (40% ethylene glycol, 18% ficoll, 0.3 M sucrose and 10% FBS added m-DPBS) as a freezing solution and electron microscope (EM) grid (V-G) or straw (V-S) as an embryo container. In both method, freezing/thawing were treated by 2-step, treatment time was required in V-G method and V-S method, for 2 min / 3 min and 3.5 min / 10 min, respectively. Embryo survival was assessed as re-expanded and hatched rates at 24 h and 48 h after warming, respectively. The results obtained in these experiments were summarized as follows: when the effect of exposure in vitrification solution and chilling injury from freezing procedure on in vitro produced expanded blastocysts were examined, at 24 h after warming, embryo survival in exposure group (100.0%) was not different compared to that in control group (100.0%), although those results were significantly different with two vitrified groups (V-G: 87.8, V-S: 77.8%) (P<0.001). However, at 48 h after warming, hatched rates of V-G group (67.8%) were significantly higher than those of V-S group (53.3%) (P<0.05). In addition, this hatched rate in V-G group was not different with that in exposure group (73.3%). When the effects of embryo developmental stage (early, expanded and early hatching blastocysts) and embryo container (EM grid and straw) to the in vitro survival of vitrified-warmed day 7 Hanwoo blastocysts were simultaneously examined, fast developed embryos were indicated the better resistance to freezing than delayed developed one, irrespective of embryo containers (early; 57.1 & 24.4%, expanded; 84.7 & 60.6%, early hatching; 91.7 & 80.0%) (P<0.001). Especially, in expanded and early hatching blastocysts, embryo survival of V-G group (67.8, 95.0%) was significantly higher than those of V-S group (53.0, 65.0%) at 48 h post warming, respectively (P<0.05, P<0.001). Therefore, this study indicates that Hanwoo blastocysts can be cryopreserved more simple, efficient and successful by vitrification method using EM grid.

Plant Hardiness Zone Mapping Based on a Combined Risk Analysis Using Dormancy Depth Index and Low Temperature Extremes - A Case Study with "Campbell Early" Grapevine - (최저기온과 휴면심도 기반의 동해위험도를 활용한 'Campbell Early' 포도의 내동성 지도 제작)

  • This study was conducted to delineate temporal and spatial patterns of potential risk of cold injury by combining the short-term cold hardiness of Campbell Early grapevine and the IPCC projected climate winter season minimum temperature at a landscape scale. Gridded data sets of daily maximum and minimum temperature with a 270m cell spacing ("High Definition Digital Temperature Map", HD-DTM) were prepared for the current climatological normal year (1971-2000) based on observations at the 56 Korea Meteorological Administration (KMA) stations using a geospatial interpolation scheme for correcting land surface effects (e.g., land use, topography, and elevation). The same procedure was applied to the official temperature projection dataset covering South Korea (under the auspices of the IPCC-SRES A2 and A1B scenarios) for 2071-2100. The dormancy depth model was run with the gridded datasets to estimate the geographical pattern of any changes in the short-term cold hardiness of Campbell Early across South Korea for the current and future normal years (1971-2000 and 2071-2100). We combined this result with the projected mean annual minimum temperature for each period to obtain the potential risk of cold injury. Results showed that both the land areas with the normal cold-hardiness (-150 and below for dormancy depth) and those with the sub-threshold temperature for freezing damage ($-15^{\circ}C$ and below) will decrease in 2071-2100, reducing the freezing risk. Although more land area will encounter less risk in the future, the land area with higher risk (>70%) will expand from 14% at the current normal year to 23 (A1B) ${\sim}5%$ (A2) in the future. Our method can be applied to other deciduous fruit trees for delineating geographical shift of cold-hardiness zone under the projected climate change in the future, thereby providing valuable information for adaptation strategy in fruit industry.

  • This study established a method for preserving chicken primordial germ cells (PGCs) that enables long-term storage in liquid nitrogen ($LN_2$) for preservation of the species. The purpose of this study was to compare the effects of Ethylene Glycol (EG) and Propylene Glycol (PG) on viability of cryopreserved PGCs with vitrification in Korean Native Chicken (Ogye), and to fine should be find or to the optimal protocol for PGCs freezing. One of the important components of cryopreservation process is cryopreservation medium that plays a vital role in preventing cellular injury during freeze-thawing. Cryoprotective agents have been known to improve cell viability after freeze-thawing. PGCs obtained from the germinal gonade of 5.5~6 day (stage 28) chick embryos, using the MACS method were suspended in a freezing medium containing a freezing and protecting agents. Gonads were harvested from stage 28 chick embryos and pooled in groups of 10E embryos, contributing gonads to the cell suspension. The gonadal cells, including PGCs, were then frozen in 1 of the following cryoprotectant treatments: 2.5% EG, 5% EG, 10% EG, 2.5% PG, 5% PG, 10% PG, and 0% cryoprotectant as a control. Effects of exposure to vitrification solution and vitrification, with different concentrations of the cryoprotectant solution, were examined. After freezing and thawing, survival rates of the frozen-thawed PGCs from the 0, 2.5, 5, 10 and 15% EG plus FBS treatment were 44.24%, 64.51%, 85.63%, 80.51% and 73.52% (p<0.05), respectively. The viability of PGCs after freeze-thawing was significantly higher for 10% EG plus FBS treatment than for 10% PG + FBS treatment (p<0.05)(85.63% vs 66.81%). Therefore, these systems may contribute in the improvement of cryopreservation for a scarce species in birds preservation. This study established a method for preserving chicken PGC that enables systematic storage and labeling of cryopreserved PGCs in liquid N at a germplasm repository and ease of entry into a database. In the future, the importance for this new technology is that poultry lines can be conserved while work is being conducted on improving the production of germline chimeras.

Comparison of Two Vitrification Methods of In Vitro Development Oocytes Collected from Porcine Antral Follicles Using Open Pulled Straw (OPS) Techniques

  • The advantages of the OPS techniques(Vajta G et al, Mol Reprod Dev 51: 53-58,1998) give 1) high survival rates of various types of eggs, 2) quick and simple process, 3) inexpensive equipment and reduced chilling injury. The efficiency of IVM/IVF technique in the porcine species is relatively lower than that obtained in other species such as ruminants. Two experiments were designed to investigate the effects of in-vitro fertilization of porcine oocytes matures using different OPS protocol for chilling and warming of vitrification. Porcine oocytes from ovaries collected at abattoir were matured for 44 hours in TCM199 Earle's salt supplemental with pyruvate, pff, L-cysteine, hormones and gentamycin. Oocytes were denuded and fertilized with frozen boar semen by common method. Porcine embryos produced routinely by in-vitro culture system of NCSU23 medium. The vitrification and the warming were conducted by OPS method with the glass micropipette instead of straw vessels and modified the protocol of G.Vajta(1999). In Exp 1, Chilling/Warming:Holding Medium(HM)+EG+DMSO/HM +sucrose Medium(SM) at 39$^{\circ}C$ warm stage. In Exp 2, : PBS+CS+EG+Ficoll+ Trehalose/PBS+Trehalose at 25$^{\circ}C$ stage. Filling, freezing, packing, thawing out and further culturing were performed to follow the basic protocol of G Vajta. During IVM-lVC and post-warming, fertilization parameter and developmental potential were compared to and statistically analysed. It was not significantly different from Exp 1 and Exp 2 but 25$^{\circ}C$ of stage was slightly higher on the morula/blastocyst forming rate and better atmosphere for worker than that at 39$^{\circ}C$ stage.

  • To investigate the optimal conditions for the storage of fresh garlic bulbs, garlic was stored at the different temperature, relative humidity and moisture content. From the standpoint of weight loss and sprouting ratio of garlic bulbs, the optimal temperature for storage was $-4^{\circ}C$. No significant changes in quality of garlic bulbs occurred even after 10 months of storage, when preserving the completely predried sample at $-4^{\circ}C$. However, freezing injury was observed in sample with incomplete drying or without predrying. From these results, The optimal condition for long term storage of fresh garlic was concluded to preserve at $-4^{\circ}C$ after complete predrying.
