Conservation for Pocket of Wooden Dagger Excavated from Sinchang-dong, Gwangju (광주 신창동 출토 칠기칼집 보존처리)

  • The immersed ancient wood relics excavated from the low swampy area of historic site in Shinchangdong, Gwangju are extremely weak. Since they are quickly dried inducing shrinkage when exposed to air, special preservative treatment is required to exhibit and preserve such items. In the case of the heavily varnished lacquerware such as the lacquered sheaths, in particular, it is extremely difficult to preserve them because chemicals are not easily penetrated and the coating might swell or come off during the process of coating. The lacquer sheaths have been preserved by carrying out freeze-drying after impregnating them in aqueous solution with low concentration of PEG#4000 40%.

  • We have fabricated gelatin-immobilized polyurethane foams (PUFG) by dipping polyurethane foam (PUF) in an aqueous solution containing gelatin and by subsequent reaction with glutaraldehyde after freeze-drying. Gelatin aqueous solutions of different concentrations were used as the dipping solutions to control the amount of immobilized gelatin. The average pore size of PUF decreased with an increase in gelatin concentration. It was found from the hepatocyte adhesion experiment that the amount of hepatocytes seeded on PUFG1, prepared by using a 1% aqueous gelatin solution, was higher than that on other PUFGs. The hepatocytes inoculated in PUFG1, were slightly aggregated as the incubation time increased. The cells inoculated in PUFG1 showed higher ammonia removal ability than those monolayer-cultured on a gelatin-immobilized polystyrene dish (PSG) after 1 and 4 days of incubation time. The inoculated cells exhibited higher albumin secretion relative to monolayer-cultured hepatocytes on PSG. Albumin secretion by hepatocytes seeded on PUFG1 was increased by the presence of serum and was further increased by both the presence of serum and cytokines. The results obtained from a 3-(3,4-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide (MTT) assay indicated that PUFG can provide a better microenvironment for hepatocyte culture along with nutrition and metabolite transfer through the high porosity of PUF.

  • An inclusion complex of muscone with ${\beta}-cyclodextrin$ (CD), as a solid form of muscone, was prepared to increase the solubility of muscone. The molar ratio of muscone to ${\beta}-CD$ in complex was in the range of $1:1{\sim}1:5$ when prepared by freeze-drying method. The interaction of muscone with ${\beta}-CD$ in solid state was investigated by Infrared (IR) spectroscopy and differential scanning calorimetry (DSC). IR and DSC studies between $muscone-{\beta}-CD$ inclusion complex and physical mixture showed that $muscone-{\beta}-CD$ inclusion complex was prepared stably. From the amount of muscone incorporated in the inclusion complex, it was found that the molar ratio of muscone : ${\beta}-CD$ was 1:1. Relative spatial position of muscone and ${\beta}-CD$ was observed by Hyperchem molecular modelling program.

Effect of Sublimable Vehicle Compositions in the Camphor-Naphthalene System on the Pore Structure of Porous Cu-Ni (Camphor-Naphthalene 동결제 조성이 Cu-Ni 다공체의 기공구조에 미치는 영향)

  • The effect of sublimable vehicle composition in the camphor-naphthalene system on the pore structure of porous Cu-Ni alloy is investigated. The CuO-NiO mixed slurries with hypoeutectic, eutectic and hypereutectic compositions are frozen into a mold at $-25^{\circ}C$. Pores are generated by sublimation of the vehicles at room temperature. After hydrogen reduction at $300^{\circ}C$ and sintering at $850^{\circ}C$ for 1 h, the green body of CuO-NiO is completely converted to porous Cu-Ni alloy with various pore structures. The sintered samples show large pores which are aligned parallel to the sublimable vehicle growth direction. The pore size and porosity decrease with increase in powder content due to the degree of powder rearrangement in slurry. In the hypoeutectic composition slurry, small pores with dendritic morphology are observed in the sintered Cu-Ni, whereas the specimen of hypereutectic composition shows pore structure of plate shape. The change of pore structure is explained by growth behavior of primary camphor and naphthalene crystals during solidification of camphor-naphthalene alloys.

Comparative Estrogenic Effects of Yak-Kong and Soy Bean on the Proliferation of Human Osteoblastic Cell Line, MG-63 (MG 63 조골세포에서 약콩과 대두의 천연 에스트로겐 효과 비교)

  • Phytoestrogens, especially soy-derived isoflavones, are receiving great scrutiny as a food supplement for preventing hormone dependent diseases such as cardiovascular diseases, cancer, and osteoporosis. These beneficial effects of phytoestrogens are caused by functioning as partial agonists or antagonists of estrogens. In contrast to the common usage of soy bean, Yak-kong(Rhynchosia Molubilis ; ) has been used as supplements of estrogen fir preventing postmenopausal osteoporosis in Oriental medicine. To investigate estrogenic effects of Yak-kong and soy bean on the proliferation of MG-63 osteoblastic cells, each bean was extracted with 70% methanol and dried by freeze-drying. Yak-kong treatment of MG-63 cells resulted in an increase of cell proliferation to a maximum of 76% compared to 68% of soy bean treatment. Treatment of MG-63 cells with Yak-kong extract also resulted in an increase of transactivation of an ERE(estrogen response element)-luciferase reporter plasmid and IGF-I expression selectively. Despite increased effects of both bean treatments on the expression of estrogen receptor $\alpha$(ER$\alpha$) and $\beta$(ER$\beta$), soy bean treatment decreased transactivation of an ERE-luciferase reporter plasmid and did not further enhance IGF-I expression. Together, our data demonstrates that the greater estrogenic response of Yak-kong extract for MG-63 cell proliferation is mediated by ER derived transactivation of ERE and selective induction of IGF-I expression.

  • The interactions of ${\alpha}-cyclodextrin({\alpha}-CyD)$ and ${\beta}-cyclodextrin({\beta}-CyD)$ with several non-steroidal antiinflammatory drugs were studied on the effects of ${\alpha}-$ and ${\beta}-CyD$ on the solubility of the drugs in aqueous medium. Indoprofen, niflumic acid, alclofenac, and naproxen were chosen as representatives of antiinflammatory drugs. The solubility of all drugs studied increased with the addition of ${\beta}-CyD$, while not with glucose or ${\alpha}-CyD$. The increase of the solubility with ${\beta}-CyD$ was considered due mainly to the formation of inclusion complexes between ${\beta}-CyD$ and drugs. From the solubility data, the apparent stability constants K of the complex could be calculated. Ultraviolet absorption and circular dichroism confirmed the inclusion of indoprofen, niflumic acid and naproxen with ${\beta}-CyD$ in the molar ratio of 1 : 1. Inclusion complexes in solid powder form were obtained by the freeze-drying method and the inclusion formation was confirmed again by infrared, diffential thermal analysis, and X-ray diffraction measurements.

Conservation for Wooden Lacquer Ware Excavated from Isong Sansung Fortress (이성산성 출토 목제칠기 보존)

  • The wooden lacquer wares, which had excavated from the water reservoir in the Isong Sansung Fortress, were applied with vacuum freeze drying method and treated, after they had been substituted with tbutanol, and treated with PEG-pretreatment. The manufacturing technique, which had not been investigated in the past, was examined through the investigation of the wood and the lacquer used for wooden lacquer wares, and the pieces of the lacquer wares, which had been stored for a long time, were restored for the exhibition and conservation. As the result, the wooden lacquer wares were manufactured, using black alder belonging to the group of Alnus, and the lacquer was painted on the bottom-layer by mixing the powdered soil and lacquer, and it was confirmed that the lacquer mixed with the black pigment and the pure lacquer had been painted alternatively over 2 times, and that the wooden lacquer ware had been manufactured through total 5 times of being painted.

  • For thestudying and utilizing the thorn sea cucumber synthetically, the method preparing polysaccharide and polypeptide from sea cucumber was studied. The comparatively rational craft for preparing crude polysaccharide is fresh raw materials deal with oar form, $60\%$ of the ethanol precipitate after hydrolyzed by pretense and vibrated with ultrasonic wave auxiliary. The purification of polysaccharide and removal of protein fromcrude polysaccharide were made through precipitation method using the acetate. The polypeptide is obtained by concentrating in vacuum, freeze-drying after mixing precipitations of two times. Polysaccharide acute poisoning experiment indicate there is no bad reaction when LD50>5000g/kg, there strain rate of liver tumor H11 approach present generally acknowledged tumor treat medicine cyclophosphamide, and there is no side effect. The fatigue resistance function of polypeptide experiment also indicates that the fatigue resistance ability of mouse which fed on added sea cucumber polypeptide has a great improvement.

  • Nanoparticles have been developed and applied for various applications to intelligently deliver bioactive materials. Herein, lactocin was processed into nanoparticles with methacrylic acid-methyl methacrylate copolymer (1:1) (eudragit L100). The eudragit polymer can protect lactocin from the stomach acid and release lactocin in the intestines. When acetone and pH 7 buffer solution were used as non-solvent and solvent, respectively, the smallest volume-average particle size (290 nm) could be obtained. Freeze drying in presence of carrageenan (dispersant) can process the particles into dried powders with minimum aggregation. SEM observation revealed the primary particles prepared based on lactocin and eudragit were of a few tens of nanometers.

  • This research investigates the moisture and aging effects of wastepaper fiber-cement composites. Wastepaper fibers is obtained by a dry process. Wastepaper fiber-cement composites was manufactured by the hatscheck process. The effects of moisture and aging on the performance of wastepaper fiber-cement composites were investigated through accelerated laboratory tests simulating the effects of moisture sensitivity and wet-dry cycles as well as freeze-thaw cycles and long-term drying. They were shown to possess acceptable moisture and aging performance compared with virgin fiber cement composites.
