본 연구는 거봉종 포도의 종자와 과피로부터 에탄올용매의 농도와 추출 온도변화에 의한 추출물의 수율의 차이와 그에 따른 free radical 소거 활성을 비교하였다. 각 분획물은 에탄올 추출물로부터 hexane 분획물, chloroform 분획물, ethyl acetate 분획물, butanol 분획물 및 물 분획물로 나누어 free radical 소거 활성을 $RC_{50}$으로 타나내었다. 추출 수율은 종자 및 과피 모두에서 추출 온도 $78^{\circ}C$, 에탄올 농도 100%에서 36.0%와 49.5%로 다른 온도와 농도 조건에 비하여 높은 수율을 나타내었다. 각 분획물 중에서는 물 분획물이 종자에서는 72.1%인데 비하여 과피에서는 92.7%로 다른 분획층에 비하여 많이 차지하였다. DPPH법에 의한 free radical 소거 활성에서는 종자의 경우, $78^{\circ}C$ 에탄올 추출물이 $RC_{50}\;=\;409.7\;{\mu}g/mL$이며, 분획물에서는 ethyl acetate층이 $RC_{50}\;=\;136.7{\mu}g/mL$로 가장 높은 활성을 나타내었다. 과피의 경우, $78^{\circ}C$ 에탄올 추출물이 $RC_{50}\;=\;6528.8\;{\mu}g/mL$로, 분획물에서는 ethyl acetate층이 $RC_{50}\;=\;694.7\;{\mu}g/mL$로 가장 높은 활성을 나타내었으나, 물 분획물은 free radical 소거 활성을 보이지 않았다. 이상의 결과를 통하여 추출온도와 용매 온도에 따른 수율의 차이가 나타났으며 추출온도와 free radical 소거 활성의 효능은 별다른 차이가 없는 것으로 나타났다. 전반적으로, 본 연구 결과 종자 에탄올 추출물과 분획물 모두 과피 추출물보다 현저히 높은 활성을 나타내었다.
Green tea catechins (GTC) and its major component, (-)-epigallocatechin gallate (EGCG) were studied for their protective effects against reactive oxygen species (ROS)-induced oxidative stress. GTC and EGCG skewed the strong antioxidative effects on the lipid peroxidation of ethyl linolate with Fenton's reagent and free radical scavenging effect to DPPH radical generation. They also protected $H_2O$$_2$- or KO$_2$-induced cytotoxicity in CHL cells or mouse splenocytes. These results indicate that GTC and EGCG are capable of protecting the lipid peroxidation, flee radical generation and cytotoxicity induced by ROS. The mechanism of inhibition in ROS-induced cytotoxicity may be due to their antiofidative and free radical scavenging properties. Therefore, GTC and EGCG may be useful chemopreventive agents by protecting the free radical generation which are involved in cancer and aging.
In this study, the Rhus chinensis Mill was divided into bark and inner bark. The antimicrobial activity and the antioxidative activity of the extracts were investigated by using the organic solvent fractions after the extraction and concentration with ethanol. This study showed the possibility of functional materials such as raw materials for cosmetics and food supplements. This study was the antimicrobial activity and antioxidant activity of five microorganisms (S. aureus, E. coli, B. subtilis, P. aeruginosa, and P. ovale). The following conclusions were obtained. First, the antimicrobial activity of B. subtilis was found to be high in the ethanol extract of. Second, DPPH scavenging activity was 86.4% free radical scavenging activity at 2.5 mg/mL bark part and 61.9% free radical scavenging activity at 2.5 mg/mL in inner Rhus chinensis Mill bark part. The ABTS scavenging activity was 79.2% free radical scavenging at 1 mg/mL bark fraction and 63% free radical scavenging activity at 1 mg/mL in inner bark, and bark showed higher antioxidant activity than inner bark. These results suggested that the antimicrobial activity and antioxidant activity of Rhus chinensis Mill extract can be used as a natural material. Specific and diverse physiological activity studies are expected in the future.
The potential antioxidative activity of water-soluble enzymatic hydrolyzates from a kelp, Ecklonia cava was evaluated by free radical scavenging and lipid peroxidation assays. To prepare water-soluble hydrolyzates from E. cava the seaweed was enzymatically hydrolyzed by five carbohydrases (Viscozyme, Celluclast, AMG, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase). Among all the hydrolyzates, Celluclast hydrolyzate effectively scavenged free radicals released from DPPH (1,1-diphenyl-2- pricrylhydrazyl) and recorded around 73% scavenging activity at the concentration of 4 mg ${\cdot}ml^{-1}$. This hydrolyzate was thermally stable and DPPH radical scavenging activity remained 80% or higher at heating temperatures of 40 and 60$^{\circ}C$ up to 12 h and around 80% at 100$^{\circ}C$ up to 8 h. AMG and Ultraflo hydrolyzate inhibited the lipid peroxidation of fish oil as that of $\alpha$-tocopherol. These results suggested that an enzymatic extraction will be an effective way for the production of a potential antioxidant from seaweeds.

In the course of searching for hepatoprotective agents from natural products, six compounds were isolated from the MeOH extract of the leaves of Juglans sinensis, as guid ed by their DPPH free radical scavenging activity. The structures were determined as juglanoside B (1), quercetin 3-O-${\alpha}$-L-arabinofuranoside (avicularin, 2), quercetin 3-O-${\alpha}$-L-arabinopyranoside (guaijaverin,3), quercetin 3-O-${\alpha}$-L-rhamnopyranoside (quercitrin,4), (+)-catechin (5) and quercetin 3-O-${\beta}$- D-galactopyranoside (hyperin,6). Compounds 2-6 showed significant DPPH free radical scavenging effects. An evaluation for the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity was conducted, and compounds 1, 2, and 5 showed protective effects against nitrofurantoin-induced cytotoxicity, and compound 5 also exhibited a moderate protective effect on amiodarone-induced cytotoxicity in Hep G2 cells.
To develop new functional materials, Saccharina japonica (S. japonica) was cultivated with Phellinus linteus (P. linteus) mycelia by solid culture. S. japonica was fermented with P. linteus mycelia. The various radical scavenging activities of extracts of fermented S. japonica with P. linteus mycelia (FSPM) were investigated and compared to the antioxidant capabilities of unfermented S. japonica and P. linteus mycelia. The antioxidant activities of FSPM extracts were evaluated using ferric reducing antioxidant power (FRAP) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. FSPM showed stronger free radical scavenging abilities than P. linteus mycelia or S. japonica alone. These results indicate that fermentation of S. japonica changes its chemical nature and could provide beneficial antioxidant effects.
The study was aimed at evaluating the antioxidant and free radical scavenging activities of ethanolic extract and its fractions of Cleome rutidosperma. The antioxidant activity, reducing power, total phenolic content, total flavonoid content, superoxide anion scavenging activity, nitric oxide anion scavenging activity, in vitro antilipid peroxidation activity and in vitro non-enzymatic hemoglobin glycosylation were studied. The results obtained in the study indicate that Cleome rutidosperma is a potential source of natural antioxidant. All the parameters were found to be concentration dependent and increased with increasing amounts of sample. Flavonoids, phenolic compound like tannins, terpenoids may be responsible for the antioxidant activity of the plant. Variation of solubility parameters in various models may be attributed to non-linearity of activity of ethanol extract fractions models. Further investigation on the isolation and identification of antioxidant component(s) in the plant may lead to chemical entities with potential for clinical use.

In the course of searching for hepatoprotective agents from natural products, four compounds were isolated from the MeOH extract of Sanguisorbae Radix, as guided by their DPPH free radical scavenging activity. The structures were determined as 4,5-dimethoxy-3-hydroxybenzoic acid methyl ester (1), (+)-gallocatechin (2), methyl $6-O-galloyl-{\beta}-D-glucopyranoside$ (3), and pomolic acid $3-O-[{\alpha}-L-arabinopyranoside]-28-O-[{\beta}-D-glucopyranosyl]$ ester (ziyu-glycoside I) (4). Compounds 2 and 3 showed significant DPPH free radical scavenging effects, exhibiting $IC_{50}$ values of 11.4 and $13.0\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.3\;{\mu}M$. In evaluation of the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity, compound 2 showed the significant hepatoprotective effect with the $EC_{50}$ value of $91.84\;{\pm}\;11.0\;{\mu}M$ on tacrine-induced cytotoxicity in Hep G2 cells, while silybin, a positive control, exhibited $EC_{50}$ value of $122.4\;{\pm}\;12.5\;{\mu}M$.

The ethanolic extract of chestnut (Castanea crenata S. et Z., Fagaceae) inner shell (CISE) and one of its components, ellagic acid (EA), were evaluated for their protective effects against 1, 1-diphenyl-2-picryl hydrazine (DPPH) free radical generation and hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line. CISE and EA were shown to possess the free radical scavenging effect against DPPH radical generation, significantly. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from Chinese hamster lung (CHL) cell, assessed by single cell gel electrophoresis assay and 8-hydroxy -2'-deoxy guanosine (8-OH-2'dG) assay. Furthermore, topical application of CISE [$12.5\%$(w/w) cream] and ellagic acid [$1.0\%$(w/w) cream] for 14 days potently inhibited malondialdehyde (MDA) formation of mouse dorsal skin (a marker of lipid peroxidation) induced by ultraviolet B exposure. Therefore, CISE and its component, ellagic acid, may be the useful natural antioxidants by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage when topically applied.
The aim of the present study was to evaluate the antioxidant activity of red algae in Jeju Island. The algal extracts were obtained with MeOH and fresh water at 20 and 70°C, and screened for antioxidant activities using hydroxyl radical (HO·), superoxide anion (O2–), hydrogen peroxide (H2O2) and DPPH free radical scavenging assays. Among them, Gracilaria verrucosa methanolic extract at 20°C (20ME, 96.85%), G. textorii aqueous extract at 20°C (20AE, 88.01%), Grateloupia filicina 20AE (85.35%), and Polysiphonia japonica 20ME (94.92%) exhibited the highest scavenging activities against HO·, O2–, H2O2, and DPPH free radicals, respectively. Moreover, P. japonica (20ME and 70ME) is correlated between DPPH free radical scavenging activity and polyphenolic contents. These results indicate that some red algae in Jeju Island could be potential candidates for development of antioxidants.
