The present study was to assess the in vitro viability and sexing rate of bovine embryos. Blastocysts were harvested on day 7~9 day after insemination(in vitro and in vivo), and the sex of the embryos was examined using the LAMP method. Embryo cell biopsy was carried out in a $80{\mu}l$ drop $Ca^{2+}$, $Mg^{2+}$ free D-PBS and, biopsied embryos viability were evaluated after more 12 h culture in IVMD culture medium. The formation of recovered embryo to expanded and hatching stages had ensued in higher of sexed embryo in vivo than in vitro (100% vs. 89%, p<0.05), and in vitro, the rates of degeneration after sexing were significantly (p<0.05) higher in vitro than in vivo(11% vs. 0.0%). The rates of the predicted sex were female 61% vs. 56%, and male 39% vs. 44% in vivo and in vitro, respectively. The rates of survival following different biopsy methods were seen between punching and bisection method in vivo and in vitro (100% vs. 89% and 100% vs, 78% respectively). Biopsy method by punching was significantly (p<0.05) higher than bisection between produced embryos in vivo and in vitro. The present data indicate that with microblade after punching for embryo sexing results in high incidence of survivability on development after embryo biopsy. It is also suggested that LAMP-based embryo sexing suitable for field applications.

Persimmons on the trees are often exposed to weather condition of cold temperature before harvest, which affects the fruit quality at harvest time and later storage. This study investigated the effect of the harvest season temperature on the storage quality of 'Fuyu' sweet persimmons. Monitoring the temperature changes in the orchard showed five times of cold temperature below the freezing temperature of persimmon, $-2.1^{\circ}C$, in the harvesting period between mid October and mid November with the lowest being as low as $-5.4^{\circ}C$. Among three different harvestings, the tertiary one at mid November showed decreased 'L' value and consistently high 'a' value of color index and high reduction in hardness during 150 days storage of fruits at $0^{\circ}C$ showing freeze-injured symptoms, compared to the first one at mid October and the second one at early November. The fruits of first harvest maintained consistently hard texture with increased 'a' color value probably due to postharvest ripening. The tertiary harvest exhibited increase in fructose and glucose contents and decrease in sucrose of the fruits, being very different from the first and secondary harvests which maintained constant level of all three free sugars. Mid November with possible sudden temperature drop is unsuitable period for harvesting sweet persimmons in the quality at harvest and later storage, and thus the harvest should be designed to be done before that time.
This study performs to evaluate the role of filter material at alluvial well for intake of riverbank filtration and the applicability and improvement effect of dual filter well. To achieve this objective, dual filter intake well and single filter intake well were installed with different filter conditions at riverbank free surface aquifer in soil layer then we evaluated filter material condition, permeability, optimum yield and well efficiency according to yield in drawdown test. As a results, we assumed forming dual filter layer minimizes sudden speed changes at boundary between aquifer and filter layer by cushioning of groundwater flow. This suppresses warm current then intake groundwater efficiently, therefore it seems decreasing peripheral groundwater level changes in spite of higher intake water amount than single filter intake well. Furthermore, we confirmed by test, installing dual filter improves permeability, optimum yield and well efficiency. The result will be used by combining with former study to set up standard of design/construction of dual filter intake well at alluvial aquifer layer. Furthermore, we expect this result will be used to prove application effect of dual filter intake well compared to single filter one and radial collector well which are mainly applied on riverbank filtration.
The object of this work is to improve the maintenance of bioluminescence from stored Photobacterium phosphoreum in a view of developing continuous monitoring system for pollutants. The long-term experiments were performed to determine the effect of storage temperature and immobilization on the maintenance of bioluminescence and viability of P. phosphoreum. A naturally luminescent bacterium, P. phosphoreum was starved in 2.5% Nael solution at $20^{\circ}C$, $4^{\circ}C$, -$20^{\circ}C$ and -$70^{\circ}C$ for 30 days. In vivo luminescence was measured by luminometry, and total cell concentrations and concentrations of culturable and viable cells were determined by acridine orange staining, dilution plate counting, and direct viable counting, respectively. The bioluminescence emission from cells stored at 4De was maintained up to 10 days while those with starved cells at other temperature ranges decreased to background level within 3 days. In terms of viability of cells, concentrations of cells stored at $20^{\circ}C$ were rapidly decreased as a result of cell lysis, leading to a drop in culturable and viable counts while cells stored at $4^{\circ}C$ was shown viable but nonculturable state during starvation. With immobilized cells on strontium alginate, the bioluminescence showed higher maintenance than free cells and decreased with count number of nonculturable cells.
The present study was to conducted the sexing efficiency and accuracy of bovine embryo by LAMP (Loop-mediated isothermal amplification) method, the development of the biopsied embryos into re- reformation and the freezability of these blastocysts by slow-freezing and vitrification. In vivo embryos were superovaluted with gonadotropin(Antorin R-10) for 4 days combined with progestrone releasing intravaginal(CIDR) insertion in Hanwoo donors, and in vitro embryos were used blastocyst embryos at Day 7 or Day 8 after post-insemination in vitro. The biopsy of bovine embryo was carried out in a 80μl drop with Ca2+-Mg2+ free D-PBS and the viability of biopsied embryos were evaluated in IVMD medium at over 12 h culture time in 5% CO2 incubator.For embryo sexing, about five or seven blastomeres were isolated from in vitro and in vivo embryos at blastocysts with microblade. and were then subjected to LAMP. The survivability of biopsied embryos were no difference in the development rate to re-formation of blastocysts between in vivo and in vitro embryos(100% and 90% respectively). The rates of sexed embryos were compared according to two groups, the female rate was lower than that the male in the in vivo and in vitro embryos(46% vs, 54% and 40% vs, 60%, respectively). However, there were no difference in the overall sexing ratio between the two groups. The survivability of frozen-thawed sexed embryos were lower in the in vitro than in vivo embryos in the slow-freezing(Group 1) and vitrification method(Group 2), (41.7% vs. 58.8% and 57.1% vs, 77.8. respectively).

The nitrogenous compounds and minerals in the raw and cooked meat of cockle shell were analyzed, and compared with those of cooked meat extracts. In abundant free amino acids, the content of glutamic acid was $129mg\%$ in raw meet, $105mg\%$ in cooked meat, $28mg\%$ in cooked meet extracts, aspartic acid, glycine, arginine, lysine, leucine, and alanine in order. The major components were lysine, arginine and leucine, and the minor components of essential amino acids were proline, tyrosine, serine and cystine. Some of ATP, ADP, AMP, inosine and hypoxanthine were identified in raw and cooked meat, but IMP and inosine were not detected in cooked meat extracts. A slight drop in content of ATP was showed in cooked meat and those had a higher content in inosine and hypoxanthine compared with raw meat. TMA, TMAO and betaine were also checked in all meat products and TMA slightly increased during cooking. Minerals in cooked cockle shell products were phosphorous, potassium, calcium and zinc. The content of phosphorous showed the highest value($16mg\%$ in raw, $185mg\%$ in cooked meat, and $25mg\%$ in extracts).

Motor imagery based Brain-computer Interface(BCI), which has recently attracted attention, is the technique for decoding the user's voluntary motor intention using Electroencephalography(EEG). For classifying the motor imagery, event-related desynchronization(ERD), which is the phenomenon of EEG voltage drop at sensorimotor area in ${\mu}$-band(8-13Hz), has been generally used but this method are not free from the performance degradation of the BCI system because EEG has low spatial resolution and shows different ERD-appearing band according to users. Common spatial pattern(CSP) was proposed to solve the low spatial resolution problem but it has a disadvantage of being very sensitive to frequency-band selection. Discriminative filter bank common spatial pattern(DFBCSP) tried to solve the frequency-band selection problem by using the Fisher ratio of the averaged EEG signal power and establishing discriminative filter bank(DFB) which only includes the feature frequency-band. However, we found that DFB might not include the proper filters showing the spatial pattern of ERD. To solve this problem, we apply a band-selection process using CSP feature vectors and linear discriminant analysis to DFBCSP instead of the averaged EEG signal power. The filter selection results and the classification accuracies of the existing and the proposed methods show that the CSP feature is more effective than signal power feature.
Purpose : To evaluate the treatment outcome for patients with locally advanced, unresectable esophageal cancer treated with relatively high dose radiation therapy(RT). Materials and Methods : From January 2000 to December 2008, 32 patients with locally advanced unresectable or medically inoperable esophageal cancer were treated with radiation therapy(RT) with or without concurrent chemotherapy. Ten patients were excluded from analysis because of distant metastasis and drop off. Patient distributions according to AJCC stages II, III IVa were 7(31.8%), 12(54.6%), 3(13.6%) respectively. The locations of tumor were cervical/upper thorax 3 (13.6%), mid thorax 13(59.1%), and lower thorax/abdominal 6(27.3%), respectively. Eleven patients received RT only, and 11 patients received cisplatin based concurrent chemoradiotherapy(CCRT). Median radiation dose was 65 Gy(range 57.6~72 Gy). Results : The median follow-up was 9.1 months(range 1.9~43.8 months). The response rates for complete response, Partial response, stable disease and Persistent disease were 6(27.3%), 11(50.0%), 4(18.2%) and 1(4.5%), respectively. Two patients(9.1%) suffered from esophageal stenosis and stents were inserted. Two patients(9.1%) had Grade 3 radiation pneumonitis and one of them expired due to acute respiratory distress syndrome(ARDS) at 36 days after completion of radiation therapy. The recurrence rate was 11(50.0%). The patterns of recurrence were persistent disease and local progression in 5(22.7%), local recurrence 3(13.7%) and concomitant local and distant recurrence in 3(13.7%). The overall survival(OS) rate was 32.1% at 2 years and 21.4% at 3 years(median 12.0 months). Disease free survival(DFS) rate was 17.3% at 2 and 3 years. All patients who had no dysphagia at diagnosis showed complete response after treatment and 100% OS at 3 years(p=0.0041). The OS for above 64.8 Gy group and 64.8 Gy or below group at 3 years were 60.6% and 9.1%(p=0.1341). The response to treatment was the only significant factor affecting OS(p=0.004). Conclusion : Relatively high dose radiation therapy in unresectable esophageal cancer tended to have a better outcome without increased complication rate. Further study with more patients is warranted to justify improved result.

Recently, the probability of rock joints being exposed to free faces is getting higher for the scale of rock mass structures gets larger. Also, the frequency of occurring dynamic events such as earthquakes and blasting has been increasing. Thus, the shear behavior of rock joints under different conditions needs to be investigated. In this study, a series of direct shear tests were carried out under various conditions to examine the velocity-dependent shear behavior of saw-cut rock joints. Two types of direct shear test were carried out. The first was to examine the velocity-dependent shear behavior of saw-cut rock joints at seven different shear velocities, each with three different normal stresses. The second was to examine the shear behavior of saw-cut rock joints when three different instantaneous shear velocities changed. As a result, the coefficient of friction was affected by normal stress. The breakpoint velocity, the point when the change of shear velocity starts to affect the frictional behavior, became lower as normal stress increased. Also, as the shear velocity became lower, the degree of stress-drop on stick-slip behavior became larger. As a result of examining the changes of friction coefficient, velocity weakening (decrease of friction coefficient) was observed. The decrement of friction coefficient due to the changes of shear velocity under slow shear velocity was larger than that under fast shear velocity.
Oyster is a nutritionally good food ingredient. Also, oyster is used to make source for taste and flavor. This study tried to investigate optimal condition of hydrolysis of oyster and oyster cooking drip for better amino acid content to make good taste and flavor. And then this study characterized hydrolysate of oyster and oyster cooking drip. Enzymes are Acalase, Flavourzyme, Neutrase, and Protamax. The optimal condition for the highest enzyme activity is given by the company. Under the best condition of each enzymes, they react with the homogenized oyster and oyster cooking drip for 0.5, 1.0, 1.5, 2, 4, 6 hr. The degree of oysters’ hydrolysis is 13.2±0.1%. But, in the case of using enzyme, the rate of hydrolysis sharply increased as time went on during 2 hr. After 8 hr, the rate is 36.9~40.5%. Protamax showed 27.4±0.4% of hydrolysis rate in 2 hr. And the degree of oyster cooking drop hydrolysis is 42.7±0.1%. The highest of hydrolysate concentration is 72.1±0.1% using protamax. In the case of oyster, it has a similar tendency of all enzymes. Otherwise, the hydrolysate of oyster cooking drip had a difference among the enzymes. Composition of free amino acid of hydrolysate using protamax was investigated how much time showed highest rate of hydrolysis to find best amino acid composition. Hydrolysis using Protamax during 6 hr is selected for best condition.
