아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐의 혈청 지질 성분에 미치는 영향 (Effects of Feeding the Mixture of Linseed and Sunflower Seed Oil on the Lipid Components of Serum in Dietary Hyperlipidemic Rats)

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks, then they were fed with the diet containing lard 3.0 % and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil and antihyperlipidemic drugs for 2 weeks. Analysis of the lipid component of the serum showed following results. Concentration of total cholesterol in serum was significantly lower in the other groups except group 2 (lard 3.0 % + olive oil 12.0%) than in the control group , especially in groups 3 (cholestyramine 2.0%) and 5 (lard 3.0% + linseed oil 12.0%). HDL-C and the ratio of HDL-C to total cholesterol concentration were higher in groups 1, 3, 5 and 9 (lard 3.0% + sunflower seed oil 12.0%) , while the atherosclerotic index was low in groups3, 5. Concentrations of free cholesterol and choesteryl ester in serum were significantly lower in the other groups except group 2 than in the control group, especially lower in grop 5. Concentration of LDL was significantly higher in group 2 while it was remarkably lower in groups 3 and 5, while those of triglyceride in groups in 3 and 4 (lipraroid) were significantly higher than in the control groups. From the data on concentration of total choelsterol , HDL-cholesterol, LDL, phospholipid and triglyceride in serum, the results suggested that the feeding mixed with 3.0% lard and 12.0% linseed oil or 2.0% cholestyramine were most effective for the improvement of the serum lipids.

Apo E 유전자 변이형이 혈청지질 및 유리아미노산 농도에 미치는 영향 (Effects of Apo E Polymorphism on the Plasma Lipid Profiles and Free Amino Acids in Korean Women)

  • Apo E polymorphism(e2, e3, e4) was among the first reported genetic polymorphism that explained part of the normal variation in plasma cholesterol concentrations. Among 62 normolipidemic healthy females, aged 19 up to 22 years, the relative frequencies of E3/3 was 0.806(n=50), E3/2 was 0.081(n=5), E3/4 allele was 0.113(n=7), and no E2/2, E2/4 and E4/4 were found. Based on the five samples of E2 allele, five subjects were randomly selected by E3 and E4 groups for the study of effects of apo E polymorphism on the distribution of serum lipid and amino acids profiles. No differences in the anthro pometric data among apo E isomers were found, otherwise the pulsation was higher in E4 than that in the others. There were no differences in plasma total HDL , HDL3 , HDL2 & LDL cholesterol, and apo A I concentrations. However, phenotype means significantly rank E2>E3>E4 allele in average TG levels(p=0.014), and rank E4>E3>E2 in total cholesterol levels(p=0.011). Atherogenic index(AI) such as lipoproteins was significantly increased in E2 & E4 than that in E3(p=0.045). Subjects with E3/2 allele had significantly higher concentrations of glutamine, phosphoserine and taurine, while subjects with E3/4 allele showed significantly lower concentrations of arginine and am inobutyrate and elevated level of phosphoserine in plasma com pared to those of E3/3 allele. Higher level of plasma taurine in subjects with E3/2 or E3/4 allele appears to be related to the elevated level of plasma total and LDL cholesterol concentrations compared to those of E3/3 allele.

만금탕(萬金湯) 및 가미만금탕(加味萬金湯)이 고지혈증(高脂血症)에 미치는 영향(影響) (Experimental Study on the Effects of Mangumtang and Gamimangumtang on Hyperlipidemia)

  • In order to investigate the effects of Mangumtang(MT) and Gamimangumtang(GMT) experimental studies were performed in Ex.1(treated with cholesterol and administration of sample at a time) and Ex.2(administration of sample after the treatment of cholesterol). The results were summerized as follows : 1. The content of Total cholesterol in the serum was not significantly decreased in MT group, whereas in GMT group significantly decreased compared with control group in Ex.1 and all significantly decreased in MT group and GMT group in Ex.2. 2. The content of Triglyseride in the serum was all significantly decreased in MT group and GMT group in all case. 3. The content of Phospholipid in the serum was not significantly decreased in MT group, whereas in GMT group was significantly decreased compared with control group in Ex.1 or Ex.2. 4. The content of Free Fatty Acid in the serum was all significantly decreased in MT group and GMT group in all case. 5. The content of HDL-Cholesterol in the serum was tended to decreased compared with control group but did not show a significance in Ex.1 or Ex.2. 6. The content of LDL-Cholesterol in the serum was not significantly decreased in MT group and GMT group in Ex.1, whereas in Ex.2 not significantly decreased in MT group but significantly decreased in GMT group. According to the above results, it is assumed that Mangumtang and Gamimangumtang have a valid effect on Hyperlipidemia, especially Gamimangumtang have a more valid effect on prevention and cure of Hyperlipidemia.

효소적 갈변 단백질이 흰쥐 혈청콜레스테롤 농도에 미치는 영향 (Effect of Proteins Modified by Enzymically Oxidized Caffeic Acid on the Concentration of Serum Cholesterol of Rats)

  • 카제인, 대두 단백질에 caffeic acid와 tyrosinase를 첨가한 후 온도 $30{\sim}35^{\circ}C$, pH 6.8에서 5시간 반응시켜 갈변단백질을 조제하였다. 이러한 갈변단백질 및 무처리 카제인, 대두단백질을 단백질 수준 20.0% 되도록 콜레스테롤 무첨가 사료에 혼합, 7주령의 Wistar계 흰쥐 수컷에 14일간 급여하였다. 갈변단백질의 아미노산 cystine의 손실이 눈에 띄는 것 이외 아미노산 손실은 인정되지 않았다. 단백질 소화율은 갈변에 의하여 저하되고, 갈변 카제인의 경우는 현저한 맹장의 비대가 인정되었다. 혈청 총콜레스테롤, LDL-콜레스테롤, 트리글리세라이드 농도는 갈변에 의해 유의차는 인정되지 않았으나, 상승하는 경향이었다. 단백질은 효소적 갈변에 의해 단백질 소화율이 저하되고, 그로 인한 분량(糞量)도 증가하지만, 혈청지질의 저하작용은 없었다.

양릉천 인진 약침이 흰쥐의 고지혈증에 미치는 영향 (A Study on the Effect of Herbal-acupuncture with Artemisiae Capillaris Herba at GB34 on Hyperlipidemia in Rat)

  • Objective & Methods: The purpose of this study is to investigate the effects of Artemisiae Capillaris Herba herbal-acupuncture (ACH-HA) at GB34 (Yangleungchean) on hyperlipidemia induced with alloxan injection and high fat diet in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum, liver, as well as the histological changes of liver and aorta. Results: 1. ACH-HA solution increased the DPPH radical scavenging activity in rat liver cells. 2. ACH-HA at GB34 significantly decreased the levels of serum total cholesterol, low-density lipoprotein (LDL)-cholesterol, free cholesterol and atherogenic index (AI), while significantly increased the ratios of high-density lipoprotein (HDL)/total cholesterol and phospholipid/total cholesterol in hyperlipidemic rats. 3. ACH-HA at GB34 significantly decreased serum aspartate aminotransferase (AST) level in hyperlipidemic rats. 4. ACH-HA at GB34 significantly increased hepatic glutathione (GSH) activity in hyperlipidemic rats. Conclusion: From the above results, it is suggested that ACH-HA at GB34 may have therapeutic and preventive effects on hyperlipidemia.

흑축이 3T3-L1 세포와 비만 유도 흰 쥐에 미치는 영향 (Effects of Pharbitidis Semen on 3T3-L1 Cells and Obesity-induced Rats)

  • Objectives: This study was designed to evaluate the efficacy of Pharbitidis Semen (PS) on obesity using 3T3-L1 cells and high-fat-diet rats. Methods: After the administration of PS at a concentration of 0.125, 0.25, and 0.50 mg/ml in 3T3-L1 cells, the degree of differentiation of adipocytes was measured by Oil Red O staining. The experimental animals were divided into five groups in vivo. Normal group: diet-fed normal group (Normal), Control group (Control): high-fat-diet (HFD)-fed, Positive control group (PC): HFD+Contrave 200 mg/kg, PS_Low group: HFD+PS 250 mg/kg group, PS_High group: HFD+PS 500 mg/kg. Results: The total cholesterol, low density lipoprotein-cholesterol (LDL-cholesterol), and triglycerides (TG) content of the PS medication groups were significantly lower than those of the control group. In addition, high density lipoprotein cholesterol (HDL-cholesterol) tended to increase, and free fatty acid, total lipid, and phospholipid tended to decrease, but the decrease was not statistically significant. The administration of PS inhibited the differentiation of 3T3-L1 adipocytes, and a statistically significant difference was found in the group administered with 0.125 mg/ml. The mRNA expression of $PPAR-{\gamma}$ and leptin, which are involved in the differentiation of adipocytes, was observed using reverse transcription polymerase chain reaction (RT-PCR). The expressions of $PPAR-{\gamma}$ and leptin significantly decreased in comparison with the control and the positive control.

갈화(Puerariae Flos) 추출물이 고지혈 유발 흰쥐의 항산화활성 및 지질농도에 미치는 영향 (Effects of Puerariae Flos on Antioxidative Activities and Lipid Levels in Hyperlipidemic Sprague-Dawley Rats)

  • 갈화 추출물을 농축하여 동결 건조해서 실험동물인 SD계 수컷 흰쥐의 실험식이에 0.5% 수준으로 첨가하여 4주간 사육하면서 체중은 7일 간격으로 섭취는 2일 간격으로 오전9시에 측정하였으며, 활성산소 및 항산화활성 측정은 2주간격으로 측정하였다. 또한 실험이 종료된 후 실험동물을 단두 도살하여 혈청 중 총콜레스테롤, HDL-콜레스테롤, 중성지방, 혈당, 인지질을 측정하여 지질 함량에 미치는 영향을 측정하였다. 갈화 추출물을 SD계 수컷 흰쥐에 갈화 추출물이 함유된 식이를 급여 후 식이섭취량, 체중증가량을 측정한 결과 갈화 추출물을 섭취한 rat이 대조군에 비해 모두 증가하였지만 군간 유의적 차이를 보이지 않았다. 갈화 추출물 급여 후 적출된 실험동물의 장기에서 대조군과 실험군의 무게 차이는 보이지 않았다. 사육 기간 동안 2주 간격으로 측정한 혈액 중 항산화와 활성산소에서는, 항산화에서는 4주째 갈화가 대조군에 비해 유의적으로 수치가 높게 나왔으며, 활성산소는 유의적인 차이를 보이지 않았다. 실험식이 종료 후 실험동물에서 적출한 장기의 무게를 측정한 결과 심장, 신장, 장, 간 모두 차이를 보이지 않았다. 또한, 혈청중의 총콜레스테롤 함량 측정 결과 갈화 추출물 급여군이 대조군에 비해 낮은 함량을 나타내었고, HDL-콜레스테롤 함량에서는 대조군에 비해 높은 함량을 나타내었지만 유의적으로 증가하지는 않았다. HDL-cholesterol/total cholesterol의 비율에서도 갈화 추출물 급여군이 유의적으로 증가하였다. LDL-cholesterol 함량은 갈화 추출물 급여군이 대조군에 비해 낮은 함량을 나타내었으나 유의적인 차이는 아니었다. 중성지방 함량은 갈화 추출물 급여군이 대조군에 비해 낮은 수치를 보였으며, 혈당 함량과 인지질 함량은 대조군에 비해 높은 수치였으나 유의적인 차이가 없었다. 이상에서와 같이 갈화 추출물 급여한 흰쥐를 이용한 동물실험에서 항산화능 및 혈청의 지질대사 개선 효과를 확인할 수 있었다. 이는 천연물의 조추출물을 이용한 기반연구로서 향후 갈화의 항산화 효과와 지질개선에 대한 분자생물학적 조직병리학적 후속연구가 필요하다고 사료되며, 이 연구의 결과를 바탕으로 갈화의 항산화식품이나 지질 개선 기능성식품 소재로의 활용이 기대된다.

백삼 및 홍삼추출물이 고지방 식이 급여 흰쥐의 혈청 지질 함량에 미치는 영향 (Effect of White and Red Panax ginseng Extract on Serum Lipids Level in High-fat-diet Fed Rats)

  • 본 연구는 고지방 식이를 급여한 암컷 rat(wister/ST, 8주령, 320g)에서 백삼과 홍삼이 체중증가, 식이효율, 부고환 지방 무게 및 혈청 콜레스테롤과 triglyceride 수치에 미치는 영향을 측정하기 위하여 실행하였다. rat는 기초식이(NC)와 고지방식이(HFD)군으로 분리하고 고지방식이군은 고지방식이 대조군과 고지방식이 인삼 투여군으로 분리하였다. 또한 인삼식이군은 다시 백삼(WG)과 홍삼투여군(RG)으로 나누었다. 인삼투여군은 4주 동안 인삼 에탄올 추출물 250, 500, 1000 mg/kg b.w.를 투여하였다. 전체 40마리의 rat를 8군으로 분리하였다. WG-500(p<0.05), WG1000(p<0.01), RG500(p<0.05) 투여군의 체중 증가 및 식이 효율에서 대조군에 비해 유의성있게 낮게 나타났다. WG500, WG1000, RG250 투여군의 부고환 지방무게는 HFD에 비해 매우 낮게 나타났다(p<0.05). 또한 WG250, WG500, WG1000과 RG1000군의 혈청 콜레스테롤 수치가 HFD에 비해 유의하게 낮게 나타났다(p<0.05). 전체 콜레스테롤 수치는 WG100군에서 가장 많이 감소하였다. 백삼 투여군에서 혈청 유리 콜레스테롤 수치가 감소하였으나 HFD군의 triglyceride 수치는 백삼과 홍삼 모두 관련이 없었다. 매일 1,000 mg/kg b.w.를 먹인 실험군이 유리 콜레스테롤과 triglyceride 억제활성이 가장 크게 나타났다. 위 결과를 종합해 볼 때, 인삼의 복용은 고지방 식이 동물에서 혈청 전체 혹은 유리콜레스테롤과 triglyceride수치를 감소시키며, 백삼이 홍삼보다 더 강한 효능을 나타내었다.

  • The purpose of this study is to examine the effects of Gamimahaenggamseok-tang and Typhae Pollen on Body Weight and Lipid Metabolism of Rats with Obesity induced by high fat diet. Sixty four male Sprague-Dawley rats, around 250g of B.W., were divided into normal, control, sample A and sample B groups. Normal groups were orally administrated with saline and normal animal feed, and control groups with saline and high fat diet, sample A with liquid extract of Gamimahaenggamseok-tang and high fat diet, and sample B with liquid extract of Typhae Pollen and high fat diet. After 4 and 8 weeks administration, body weight, epididymal fat pad weight, weight of liverㆍspleenㆍkidney, abdominal skin fold thickness, feed intake, and lipid level in liver tissue on morphological examination; total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, free fatty acid, and phospholipid on serum examination were measured in all of the groups.

석류 껍질 추출물이 고지방 고콜레스레롤 식이 급여 C57BL/6J 마우스의 항산화 지표 및 DNA 보호에 미치는 영향 (Effects of Pomegranate Peel (Granati pericarpium) Extracts on the Antioxidant Biomarkers in C57BL/6J Mice Fed a High Fat and Cholesterol Diet)

  • The present study evaluated the effects of pomegranate peel (Granati pericarpium) extract on the lipid profiles and antioxidant biomarkers in mice fed a high fat and cholesterol diet: the measured biomarkers included the TBARS value, GPx, GR, SOD and GST activities. Body fat depositions were significantly decreased in the group that received pomegranate peel. In addition, the activities of GPx, GST and SOD were significantly higher in the liver and plasma of the pomegranate peel group than in the control group. Also, the pomegranate peel diet decreased lipid peroxidation of the liver and kidney. Alkaline single-cell gel electrophoresis (comet assay) showed that the DNA damage in the plasma of the pomegranate peel group was decreased compared to that of control. The present results show that a diet with added pomegranate peel exerts protective effects against oxidative DNA damage and lipid peroxidation possibly via effects on the free radical levels in mice fed a high fat and cholesterol diet.
