Effects of Defatted Safflower and Perilla Seed Powders on Lipid Metabolism in Ovariectomized Female Rats Fed High Cholesterol Diets (고콜레스테롤식에서 홍화박과 들깨박이 난소절제 흰쥐의 지질대사에 미치는 영향)

  • To study effect of non-fat components present in plant seeds on lipid metabolism, defatted safflower and perilla seed powders were used in high cholesterol diets for ovariectomized (ovx) female Sprague-Dawley rats weighing 227$\pm$15g. Experimental groups were six as follows; normal group without ovariectomy and cholesterol-free diet, sham and ovx-control groups with high cholesterol and cellulose for dietary fiber, ovx-est group with the same diet as ovx-control but with eight subcutaneous injections of 50$\mu\textrm{g}$ 17$\beta$-estradiol. ovx-safflower and ovx-perilla with 29% and 16% (w/w) of each defatted powder in high cholesterol diets at the expense of cellulose. Weight gains were lower in normal and sham groups and food efficiencies were lower in normal,ovx-est and ovx-safflower groups compared with ovx-control. Uterus weights were dramatically reduced by ovariectomy but restored completely by 17$\beta$-estradiol and partially (~5%) by defatted safflower. Plasma levels of total cholesterol were not different among ovx-control, sham, vx-est and ovx-safflower groups (90.8~95.1 mg/dL) but that was lower in ovx-perilla (80.4$\pm$6.2 mg/dL). Plasma triglyceride (TG) levels were lower in sham (76.6$\pm$7.0 mg/dL) and ovx-perilla (79.2$\pm$5.8 mg/dL) groups. Liver cholesterol levels were lower in sham, ovx-est, ovx-safflower and ovx-perilla groups (26.6~29.8 mg/g) than ovx-control (36.5$\pm$3.2 mg/g). But liver TG levels were reduced only sham and ovx-est groups compared to control group. Fecal excretions of bile acid and cholesterol were highest in ovx-safflower group (30.8$\pm$5. and 32.1$\pm$5.7 mg/g) compared with other ovx groups (20.8~23.1 and 12.1~19.5 mg/g). These results suggest that both perilla and safflower seeds contain groups (20.8~23.1 and 12.1~19.5mg/g). These results suggest that both perilla and safflower seeds contain non-fat and non-fiber components having lipid lowering effects.

Calcium-Independent Acrosome Reaetion by Methyl Beta Cyclodextrin in Mouse Epididymal Sperm In Vitro (생쥐 부정소 정자의 첨체반응 유도의 Calcium 비의존성)

  • Sperm capacitation and acrosome reaction (AR) have been known to be Ca$^{2+}$-dependent events. Sperm capacitation accompanies with cholesterol efflux fiom plasma membrane, that eventually stimulates AR. However, whether the AR mediated by cholesterol efflux is Ca$^{2+}$ dependent has not been verified yet. Recently, methyl beta cyclodextrin (MBCD) was found to evoke AR by stimulating the cholesterol efflux fiom sperm membrane. In the present study, we examined the requirement of Ca$^{2+}$ in the MBCD-induced AR. During incubation of sperm in the bicarbonate buffered media MBCD increased AR in a dose-dependent manner regardless of the Ca$^{2+}$ presence. In the presence of low molar concentration of Ca$^{2+}$ (100 ${\mu}$M), MBCD-induced AR was slightly increased compared to Ca$^{2+}$-free condition. In the absence of Ca$^{2+}$ supplement, spontaneous AR was slightly increased during the incubation but inhibited by 100 ${\mu}$M EGTA. MBCD potentiated AR even the presence of EGTA. However, EGTA attenuated MBCD-induced AR, suagesting the functional involvement of intracellular Ca$^{2+}$ in the MBCD-induced AR. Taken together, it was suggested that cholesterol efflux from the sperm plasma membrane was sufficient for induction of AR even in the absence of extracellular Ca$^{2+}$and that a condition permissive for mobilization of intracellular Ca$^{2+}$ is important for MBCD-induced AR.

Effects of Dietary Xylooligosaccharides on HMG-CoA Reductase Activity and Lipid Cmposition of Liver in Rat Fed High Cholesterol Diets (Xylooligo당이 고콜레스테를 식이 횐쥐에서 간조직 HMG-CoA Reductase 활성과 지질 초성에 미치는 영향)

  • This study was conducted to examine the effects of dietary xylooligosaccharides on HMG-CoA reductase activity and lipid composition of liver in rat fed high cholesterol diet. Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly divided into groups of one normal diet, and four high cholesterol diet containing 1% cholesterol. The high cholesterol (1%) diet groups were classified into xylooligosaccharides free diet (C group), 5% xylooligosaccharides diet (C5XO group), 10% xylooligosaccharides diet (C10XO group), and 15% xylooligosaccharides diet (C15XO grcup) according to the levels of dietary xylooligosaccharides supplementation. The experimental diets were fed ad libitum for 4 weeks. The hepatic lipid contents, cholesterol and triglycerides in xylooligosaccharides supplemented groups were significantly lower than those of C group. An antithrombGsis index, a ratio of n-3 to n-6 fatty acids of liver was significantly increased in 10% xylooligosaccharides supplemented groups compared to that of C group. The activity of hepatic HMG-CoA reductase, a rate limiting enzyme of cholesterol biosynthesis, in xylooligosaccharides supplemented groups was more significantly increased than in C group. These results suggest that dietary xylooligosaccharide may be act as potential substitute for a dietary fiber to improve lipids metabolism in rat fed high cholesterol diet.

Effects of Liquid Culture of Agaricus blazei Murill on Growth, Lipid and Protein Levels, and Enzyme Activities in Rats (신령버섯 균사체 배양액이 흰쥐의 성장률, 지질과 단백질 농도 및 효소활성에 미치는 영향)

  • The effects of liquid culture of Agaricus blazei Murill on the body weight gain, lipid metabolism, protein levels, and enzyme activities were studied in growing male rats. Sprague-Dawley rats were divided into three groups ; control group (A. blazei free water), 20 or 30% A. blazei groups (20 or 30% A. blazei in water) according to the levels of A. blazei supplementation. The rats were fed ad libitum each of the experimental beverage for 5 weeks. The body weight gain, food intake, food efficiency ratio, and organ weight of the 20 or 30% A. blazei groups were similar to those of the control group. There were no differences in the concentrations of triglyceride, total cholesterol, HDL-cholesterol, total protein and albumin, and the activities of GOT, GPT, LDH and alkaline phosphatase of the serum among the experimental groups. The calcium and phosphorus concentrations of serum in the 20 or 30% A. blazei group were significantly decreased compared with those of the control group. The concentrations of LDL-cholesterol and the atherogenic index of the 20 or 30% A. blazei groups were significantly lower than those of the control group, but the serum HDL-cholesterol/total cholesterol ratio was higher. These results showed that the 20 or 30% A. blazei feeding decreased the LDL-cholesterol and the atherogenic index, and increased HDL-cholesterol/total cholesterol ratio in serum of rats.

Effects of Cellulose and Pectin on Postprandial Blood Glucose and Plasma Lipid Concentration (셀룰로오스 및 펙틴이 식후 혈당과 혈장 지질 농도에 미치는 영향)

  • The purpose of this study was to observe the effects of cellulose and pectin on glucose and plasma lipid concentrations in healthy women. Eight female collage students were participated voluntarily. All subjects received a fiber-free control diet (CD), cellulose diet (CED), pectin diet (PTD) with each diet for a period of three days with a 4 day interval. The food intake of subjects were monitored every day and plasma level of glucose, triglyceride, total cholesterol were determined at last day. Blood glucose, plasma triglyceride, total cholesterol were measured at fasting state and 30, 60, 90, 120, and 180 minute after consuming each diet. Plasma glucose concentrations (AUC: Area Under the Curve) of pectin diet was $122.7{\pm}4.0mg/dl$ and that of cellulose diet was $147.6{\pm}8.4mg/d$, but they were significantly lower than in comparison with those of control diet ($197.1{\pm}11.6mg/dl$) (p<0.05) Plasma triglyceride concentrations of the CED ($-83.9{\pm}22.2mg/dl$) PTD ($-9.7{\pm}26.1mg/dl$) showed gradual decrease after each test diet feeding but not significantly different in each dietary fiber added diet (p<0.05). Plasma cholesterol concentrations of the CD was not significantly different in PTD, but PTD was significantly lower than the CED until 3 hours after consuming each test meal (p<0.05). In conclusion, in spite of total cholesterol was reduced only by pectin, glucose and triglyceride were lower than control diet in dietary fiber intake. But there were no significant differences each dietary fiber.

Effect of Antioxidant Vitamin Supplementation on Plasma Lipid Pattern in Smoking College Men (항산화성 비타민 보충 급여가 흡연자의 혈중 지질 양상에 미치는 영향)

  • Cigarette smoking is a well known risk factor for cardiovascular disease and has negative effects on blood lipid and lipoprotein . Some of the associations between smoking and chronic disease can be attributed to the less healthful lifestyles of smokers. A large body of epidemiologic evidence suggests inverse relationships between ischemic heart disease and plasma vitamin C and E concentrations . Smokers have lower plasma concentrations of these vitamins than do nonsmokers. Smokers therefore need antioxidant vitamin supplementation. The purpose of this study was to investigate the effect vitamin supplementation on plasma lipid patterns in smoking college men. 24subjects were divided into 3 groups of which were the vitamin C supplementation group (n=8), the vitamin E supplementation group(n=8) and the vitamin C+E supplementation group(n=8). The vitamin C supplementation group consumed 500mg of ascorbic acid, the vitamin E supplementation group consumed 200IU of D-$\alpha$-tocopherol, and the vitamin C+E supplementation group consumed 500mg of ascorbic acid+ 200IU of D-$\alpha$-tocopherol for 4 weeks. We examined the plasma lipid patterns before and after the vitamins were supplemented. The results obtained were as follows ; In the vitamin C supplementation group, the concentration of total cholesterol decreased significantly and HDL-cholesterol increased significantly with the supplementation of vitamin. In the vitamin E and vitamin C+E supplementation groups, however, there were no significant differences observed with the supplementation of vitamin. Concentration of plasma LDL, triglyceride, free fatty acid were not significantly affected by the supplementation of vitamin in all groups. In terms of plasma fatty acid composition, the concentrations of saturated fatty acid were not significantly affected by the supplementation of vitamin in all groups. The concentrations of palmitoleic acid, arachidonic acid, and docosahexaenoic acid, however, significantly increased in the vitamin E supplementation group(p<0.05). The concentration of plasma linoleic acid significantly increased in the vitamin C+ E supplementation group)(p<0.05). The results of this study show that antioxidant vitamin supplementation in smokers has a tendency to decrease coronary heart disease risk in view of the plasma total cholestrol and HDL-cholesterol concentrations of the vitamin C supplementation group and fatty acid concentration of the vitamin E supplementation group.

Effects of the Korean Mistletoe Hot-Water Extract on the Lipid Components and Blood Pressure Level in Spontaneously Hypertensive Rats (한국산 겨우살이 열수추출액이 본태성 고혈압쥐의 지질성분 및 혈압 저하에 미치는 영향)

  • This study was designed to observe the effects of Korean mistletoe(Viscum album L. var. coloratum Ohwi) hot-water extract administration on the improvement on the lipid components, apolipoprotein and blood pressure level in spontaneously hypertensive rats (SHR, Wistar strain, male) fed the experimental diets for 33 days. Concentrations of total cholesterol, triglyceride (TG), LDL-cholesterol, free cholesterol and atheroscieotic index in serum were significantly lower in the Korean mistletoe extract administration group [group KM(Korean mistletoe 10.0 g% hot-water extract)] than those in the control group. In the ratio of HDL-cholesterol concentration Korean mistletoe 10.0% hot-water extract administration group were higher percentage than in the control group. However, concentrations of total cholesterol and TG in liver and brain were significantly lower in the group KM than those in the control group. But the components of apolipoprotein (Apo) A-I and Apo A-II in serum were significantly higher in the KM group than in the control group. However, components of Apo C-II, Apo C-III, Apo E and ratio of Apo B to Apo A-I in serum were fairly reduced in the group KM than in the control group. The levels of systolic and diastolic blood pressure were significantly lower in KM cup than control group. From these results, Korean mistletoe hot-water extract administration were effective on the improvement of the lipid components, coronary heart disease and hypertension in spontaneously hypertensive rats. And particularly, Korean mistletoe extract administration were more effective as a therapeutic regimen for the control of blood pressure in hypertension.

An anti-Dxidative Effect of Cheongeumyeonsudan of Rats Induced Aging by D-Galactose (천금연수단(千金延壽丹)이 노화 유발된 철쥐의 항산화효능에 미치는 영향)

  • Objectives : The purpose of this study is to find out the effect of antioxidation related to aging of Cheongeumyeonsudan which is written on Dongui-bogam experimentally. Method : 14 weeks aged SD albino rats were separated into uncontrolled group, controlled group and CGY group. As controlled and CGY groups were induced aging by subcutaneous injection of D-galactose, at the same time we administered the extract of Cheongeumyeonsudan to CGY group for 6 weeks. After then we drew blood from each group, and took measurements; the activity of SOD, GSH-px, catalase in erythrocytes, TBARS value, concentration of total lipid, tryglycende, total cholesterol, HDL-cholesterol in blood plasma. Results : The activities of SOD, GSH-px in erythrocytes were significantly increased in the CGY group compared with control group. The activity of catalase showed a tendency to increase, but it was nor remarkable. The concentration of total lipid, the values of TBARS and total cholesterol was significantly decreased in the CGY group compared with control group, and the concentration of plasma HDL-cholesterol was not remarkable. The concentration of tryglycende in plasma showed a tendency to decreased. Conclusions : it is suggested that Cheongeumyeonsudan decreased the activities of free radical, the concentration of lipid in plasma and generate enzyme which form lipid peroxide.

Effects of Feeding Dried Leftover Food on Growth and Body Composition of Broiler Chicks

  • This study was conducted to investigate the effects of feeding dried leftover food (DLF) on growth, body composition and feed conversion of broiler chicks. One hundred ninety-six of one-day old Ross broiler chicks were assigned to 7 treatments in a completely randomized design. Each treatment had four replications with seven chicks per replication. The treatments groups included control without DLF, dietary 10% level of DLF, dietary 20% level of DLF and dietary 30% level of DLF, 5% higher protein level of diet containing 10% DLF, 10% higher protein level of diet containing 20% DLF and 15% higher protein level of diet containing 30% DLF than control diet. Body weight gain was slightly higher in control group than that of DLF-fed groups. However, there were no significant differences in body weight gain among those groups fed diets containing different levels of DLF. In general, increasing dietary level of DLF resulted in decreasing feed conversion. Content of crude protein in whole broiler body was slightly higher in control group although any significant difference was not found among treatments (p>0.05). Content of crude fat in whole broiler body was lowest in groups fed diets containing 30% DLF with 15% higher protein level than control diet, showing significant difference from groups fed diets containing 20% DLF (p<0.05). Contents of total cholesterol, free cholesterol, cholesterol ester and LDL- cholesterol in blood of broilers fed DLF-containing diets generally appeared to be higher compared with control group without significant difference (p>0.05). Fatty acid contents in broiler meat were higher in the order of oleic acid, palmitic acid and linoleic acid without significant differences among treatments. Content of DHA in broiler meat was higher in groups fed diets containing DLF than that of control group although there were no significant differences among treatments (p>0.05).

A Study of Hyperlipidemia in Koreans -I. Specially Related to physical Characteristics and It's Risk Factors for Hypercholesterolemia- (한국인의 고지혈증에 관한 연구 -I. 고콜레스테롤혈증 소견자의 신체 특성과 그 위험인자를 중심으로-)

  • This study was conducted to investigate the physical characteristics and risk factors for hyperchol-esterolemia (HC) in Korean. 344 adult men who took the annual health check-ups at D or J hospitals were participated in this cross-sectional study. The subjects were grouped by plasma total cholesterol level in to three groups: normal cholesterolemic (n=139) borderline hypercholesterolemic(n=93) and hypercholesterolemic (n=112) groups. The data of height weight and plasma cholesterol level were col-lected from medical records. Body circumferences(midarm, waist, hip, and thight) skinfold thicknesses (biceps, triceps, subcostal, abdomen, and suprailic), and body composition (fat mass and fat free mass) were measured. Body mass index (BMI) height/weight ratio (HWR) waist/hip circumference ratio (WHR) waist/ thigh circumference ratio (WTR) central skinfold thickenss (CSF) and peripheral skin-fold thickness were calculated. The subjects with HC had significantly higher weight BMI waist cir-cumference skinfold thickness and body fat mass than those of the normal subjects. The relative and attributable risks on HC were 1.61 and 0.17 for obesity (BMI$\geq$25) 1,30 and 0.11 for upper body obesity (WTR$\geq$1.30) and 1.54 and 0.18 for central body obesity (CSF$\geq$95.7). Plasma total cholesterol level was positively correlated with several antropometric parameters: BMI (p<0.001) weight(p<0.001) waist circumference(p<0.001) and skinfold thickness of abdomen (p<0.001) spraillic (p<0.01) triceps(p<0.01) subcostal (p<0.01) and biceps (p<0.05) In conclusion the major influencing factors to plasma cholesterol level was BMI. Among the each physical parameters the circumference of waist the skinfol-d thickness of abdomen and the percentage of body fat were closely related to plasma cholesterol level. The important risk factor for hypercholesterolemia was obesity specially upper body obesity and central body obesity.
