포도주스의 보충섭취가 흡연성인의 혈압, 혈장지질 및 자유 라디칼 생성에 미치는 영향 (The Effects of Purple Grape Juice Supplementation on Blood Pressure, Plasma Lipid Profile and Free Radical Levels in Korean Smokers)

  • Flavonoids contained in grapes are potent antioxidants that may protect against oxidative stress and reduce the risk of chronic diseases related with free radical damage. In this study we investigated the effect of daily grape juice supplementation on blood pressure (BP), plasma lipid profiles and the generation of free radicals in 67 healthy volunteers (29 smoker, 38 nonsmokers). The daily 480 ml of grape juice supplementation for 8 weeks resulted in a significant decrease in diastolic BP by 6.5% in smokers and systolic and diastolic BP by 11.2 and 3.7% in non-smokers. Plasma total cholesterol, HDL- and LDL-cholesterollevels in smokers and total cholesterol in non-smokers were significantly increased after the intervention. Plasma triglycerides and conjugated dienes were not affected by grape juice supplementation. Levels of free radical determined by reading the lucigenin-perborate ROS generating sources, decreased significantly by 18% compared to the beginning of the study. The results indicated that the consumption of grape juice may reduce BP and free radical generation in smokers, which was possibly exerted by flavonoids. Our findings suggested that the grape juice has protective effect on chronic disease due to the overproduction of free radical in smokers.

빈도에 따른 복합운동프로그램이 노인들의 체내 총항산능 및 활성산소 유리기에 미치는 영향 (The Analysis of Old Person's Total Antioxidant Capacity and Free Oxygen Radical According to Exercise Frequency of Combination Exercise)

  • Purpose:The purpose of this study was analyze the effect combination exercise program on plasma total antioxidant capacity and free oxygen radical according to exercise frequency of elderly. Age range of subject is 65~70 years. Methods:Subjects divided into three groups: control group(0-time), week-3 times group(3-times), week-5 times group(5-times). Each group were performed combination exercise during 8 weeks. Results:The results of each group by ANOVA free oxygen radical have a change(p<.05). We performed comparison of total antioxidant capacity and free oxygen radical pre-test and post-test on each groups. Total antioxidant capacity was increased in the 3-times and 5-times groups but there have not significant. Total antioxidant capacity is improve better than to 5-times. Free oxygen radical is significant increased in the 0-time (p<.05). But 5-times but 3-times is no significant decrease. Free oxygen radical is improve better than to 3-times. Conclusion:In the old people, exercise frequency week 3 times doing appeared effectively in improvement of the free oxygen radical and 5 times doing appeared effectively in improvement of the total antioxidant capacity. So rightly must apply a exercise frequency well in characteristic of old person each person.

Free Radical Scavenging Activity of Enzymatic Hydrolyzates of Hot Water Extract from the Shell of Reeve's Turtle (Chinemys reevesii)

  • The shell of Reeve's turtle has been used as a traditional folk medicine in Korea. We produced a hot water extract from Reeve's turtle shell according to the traditional medical practice. To release bioactive peptides, the hot water extract was enzymatically hydrolyzed with various proteases, and the free radical scavenging activity of the hydrolysate was investigated against 1,1-diphenyl-2-picryl-hydrazyl (DPPH), hydroxyl and peroxyl radicals. The free radical scavenging activity of the enzymatic hydrolysates varied from 1 to 79% depending on the enzymes, free radical species, and concentration. The $EC_{50}$ values demonstrated that the enzymatic hydrolysates of hot water extract from the shell of Reeve's turtle are potential antioxidants.

  • Methanolic aqueous extracts of 100 plants were screened for antioxidative activity using Fenton's reagen/ethyl linoeate system and free radical scavenging activity using DPPH free radical generating system. The results suggest that at least six plants including Eugenia caryophyllata, Alpinia offiicinarum, Rhus verniciflua, Curcuma longa, Rheum palmatvm and Evodia officinalis may be the potential sources of antioxidant, But only one plant, Cornus officinalis, may be the potential source of free radical scavenger from natural plants.

Free radical scavenging activity and kinetic behavior of the Galgeuntang water extract

  • Galgeuntang water extract exhibited a strong free radical scavenging activity and reducing power determination. However, a gradual increase in the free radical scavenging activity and reducing power determination was obtained with increasing concentrations. The highest radical scavenging activity was shown by the water extract from Galgeuntang (116.93 ${\mu}g$/mL) and the water extract from Cinnamonum cassia Presl. (95.01 ${\mu}g$/mL). These results of phenolic and flavonoid contents of the extracts indicated that the strong radical scavenging activity of the Cinnamonum cassia Presl. extract might be in part due to the phenolic compounds. The correlation coefficient between TPC and DPPH ($r^2$ = 0.9312), TFC and DPPH ($r^2$ = 0.9677), showed positive correlation among total phenolic/flavonoid contents and antioxidant activity. These results suggest that Galgeuntang has a potential antioxidant activity.

  • The effects of aqueous extracts from Korean commercial teas on excessive free radicals were examined utilizing spin trapping, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation. A potent scavenging effect of green tea and oriental senna tea was dound using sipin trapping. The most effective teasagainst the DPPH radical was green tea, followed inorder by pine leak tea, Chinese gutta percha tea and orietnal senna tea. Similar to the effects of DPPH radical , green tea, pine leaf tea, Chinese gutta percha tea and oriential senna tea had an inhibitory effect on lipid peroxidation. These findings predict that Korean tea is a promising material for scavenging free radicals, and for curing diseases related to free-radical reactions.

국내산 포도 캠벨종의 종자 및 과피 추출물의 Free Radical 소거능 탐색 (Free Radical Scavenging Effect of Seed and Skin Extracts from Campbell Early Grape (Vitis labruscana B.))

  • 본 연구는 캠벨종 포도의 종자와 과피 에탄올 추출물의 추출조건과 그에 따른 DPPH법에 의한 free radical 소거 활성을 비교하여 최적의 추출 조건의 확립과 항산화 활성을 살펴보았다. 추출 조건은 추출 용매인 에탄올의 농도와 추출 온도를 달리 하여 각각의 수율을 비교하였으며, 각 분획물은 hexane 분획물, chloroform 분획물, ethyl acetate 분획물, butanol 분획물 및 물 분획물로 나누어, free radical 소거 활성을 RC$_{50}$으로 나타내었다. 추출 수율은 종자에서는 추출 온도 78$^{\circ}C$, 에탄올 농도 100%에서 11.4%로 다른 온도와 농도 조건에 비하여 높은 수율을 나타냈으며, 과피 에서도 역시 추출 온도 78$^{\circ}C$, 에탄올 농도 100%에서 44.5%로 다른 온도와 농도 조건에 비하여 높은 수율을 나타냈다. 각 분획물 중에서는 물 분획물이 종자에서는 39.2%인데 비하여 과피 에서는 85.6%로 다른 분획물에 비하여 많이 차지하였다. DPPH법에 의한free radical 소거 활성에서는 종자의 경우, 5$0^{\circ}C$ 에탄을 추출물이 RC$_{50}$=16.8 $\mu\textrm{g}$/mL로, 분획물에서는 ethyl acetate 분획물이 RC/syb 50/=15.4 $\mu\textrm{g}$/mL로 가장 높은 활성을 나타내었다. 과피의 경우, 78$^{\circ}C$ 에탄올 추출물이 RC$_{50}$=2437.5 $\mu\textrm{g}$/mL로, 분획물에서는 butanol 분획물이 RC$_{50}$=698.4 $\mu\textrm{g}$/mL로 가장 높은 활성을 나타내었다. 이상의 결과를 통하여 추출 온도와 용매 농도에 따른 수율의 차이가 있었으며 free radical 소거 활성에서는 종자 에탄을 추출물이 과피 에탄올 추출물 보다 145배 이상의 현저히 높은 활성을 나타내었다.을 나타내었다.

Oxidative DNA 손상에 대한 Quercetin 및 그 배당체들의 유전독성억제효과와 작용기전 (Antigenotoxicity and Action Mechanism of Quercetin and its Glycosides against Oxidative DNA Damage)

  • Quercetin and its glycosides showed a strong free radical scavenging effect to DPPH radical generation. However, there were not big differences between quercetin aglycone and glycosides under experimental condition of this study. On the other hand, quercetin had pro-oxidant effect in bleomycin-dependent DNA assay. Quercetin aglycone and its glycosides, quercitrin inhibited $H_2$$O_2$- induced DNA damage in CHL cells. They also have an anticlastogenicity toward DNA breakage agent by radical generation like bleomycin. These results indicate that quercetin aglycone and its glycosides are capable of protecting the free radical generation induced by reactive oxygen species like $H_2$$O_2$. The mechanism of inhibition in hydrogen peroxide-induced genotoxicity may be due to their free radical scavenging properties. Therefore, quercetin aglycone and its glycosides may be useful chemopreventive agents by protecting of free radical generation which are involved in carcinogenesis and aging. However, quercetin and its glycosides must also carefully examined for pro-oxidant properties before being proposed for use in vivo.

Microwave Assisted Extraction, Optimization using Central Composite Design, Quantitative Estimation of Arjunic Acid and Arjunolic Acid using HPTLC and Evaluation of Radical Scavenging Potential of Stem Bark of Terminalia arjuna

  • The optimization and microwave assisted extraction of stem bark of Terminalia arjuna, quantitative estimation of the marker compounds arjunic acid and arjunolic acid using HPTLC and the evaluation of free radical scavenging activity has been performed in this study. The central composite design was used for optimization and the values of parameters for optimized batch of microwave assisted extraction were 1000 W (Power), 3 minutes (Time) and 1/120 (Solid/solvent ratio). The solvent system to carry out the HPTLC was toluene: acetic acid: ethyl acetate (5: 5: 0.5) and quantitative estimation was done using standard equations obtained from the marker compounds. The in-vitro free radical scavenging activity was performed spectrophotometrically using ascorbic acid as standard. The value of estimated percentage yield of arjunic acid and arjunolic acid was 1.42% and 1.52% which upon experimentation was obtained as 1.38% and 1.51% respectively. The DPPH assay of the different batches of microwave assisted extraction and marker compounds taken suggested that the marker compounds arjunic acid and the arjunolic acid were responsible for the free radical scavenging activity as the batch having the maximum percentage yield of the marker compounds showed best free radical scavenging effect as compared to standard ascorbic acid. The $IC_{50}$ value of the optimized batch was found to be 24.72 while that of the standard ascorbic acid was 29.83. Hence, the yield of arjunic acid and arjunolic acid has direct correlation with the free radical scavenging activity of stem bark extract of Terminalia arjuna and have potential to serve as active lead compounds for free radical scavenging activity.

참당귀(Angelica gigas)의 DPPH Radical 소거 활성과 항산화 효과 (DPPH Radical Scavenger Activity and Antioxidant Effects of Cham-Dang-Gui (Angelica gigas))

  • 본 연구의 결과를 요약하면 참당귀추출물 1 mg/mL의 농도에서 열수추출물의 free radical소거 효과는 메탄올추출 물보다 높게 나타났으며 농도 의존적으로 DPPH free radical 소거 효과가 상승하는 것을 관찰하였다. 또한 참당귀 투여에 의하여 화학요법으로 발생된 지질과산화물의 생성을 감소시켰으며, 단백질 산화를 유의적으로 억제함으로써 참당귀추출물은 CYP를 투여로 생성된 free radical을 감소시킴으로써 독성을 감소시켜, 혈액학적 빈혈지표가 더 감소되는 것을 막았다고 사료되며, 항산화제로서의 가능성이 있는 한약재로서 활용성이 높을 것으로 사료된다 항산화효소인 SOD 활성은CYP투여에 의하여 활성이 증가하였으며 참당귀추출물의 투여에 의한 효과가 나타나지 않았으나 감소하는 경향을 보였고 catalase 활성은 CYP 투여에 의하여 활성이 증가하였으나 참당귀의 효과는 차이가 없었다. 결론적으로 참당귀추출물은 free radical 소거능을 갖고, 화학요법으로 발생된 지질과산화물의 생성을 낮춰 화학요법에 의한 부작용으로 발생되는 혈액학적 수치의 감소를 막음으로써 항산화제로의 활용성을 기대해본다.
