Effect of the Ethanol Extract of Lycium chinense on the Oxygen Free Radical and Alcohol Metabolizing Enzyme Activities in Rats (흰쥐에 있어서 구기자 알콜 추출물이 Oxygen Free Radical 및 Alcohol 대사효소 활성에 미치는 영향)

  • To investigate an effect of the ethanol extract of Lycium chinense(EELC) on the activities of enzymes scavenging oxygen free radicals or detoxicating alcohol. The ground Lycium chinense was extracted with 30% edible ethanol and then diluted with 6% ethanol to contain 2% EELC(w/v). Three different groups of male Sprague-Dawley rats had taken a drink EELC, ethanol(ETH) or water(control), respectively for 2 months. At the end of experimental period, the animals were sacrificed and obtained the following findings. The EELC-treated animals showed the highest activity of hepatic glucose-6-phosphatase among three groups. The activities of xanthine oxidase and cytochrome p-450 from EELC treatment group were lower than those from ETH-treated group. However, the activity of superoxide dismutase was higher in the EELC-treated group than the ETH-treated(p<0.005). Furthermore, hepatic alcohol or aldehyde dehydrogenase activity, and glutathione content and glutathione peroxidase were significantly higher in EELC-treated animals than in ETH-treated those. The activity of glutathione S-transferase in liver was appeared the orderly higher value in EELC, ETH and control-treated group. As the result, EELC may affect the reduction of oxygen free radical production and help the detoxication of ethanol.

Analysis of Total Phenolic Content and Antioxidant Activity from Fruits of Vaccinium oldhamii Miq. (정금나무 열매의 총 페놀 함량 및 항산화활성 분석)

  • Vaccinium oldhamii Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. To develop superior cultivars for edible or medicinal purposes, we evaluated total phenolic content and antioxidant activity. Total phenolic content was measured by folin-denis method and antioxidant activity was measured by DPPH free radical scavenging activity (DPPH method) and reducing power (potassium ferricyanide method). Total phenolic content, DPPH free radical scavenging activity (at 400 ppm) and reducing power (at 700 ppm) were 15.8~19.9 mg/g, 69.1~91.4% and 0.56~0.96, respectively. A linear correlation was shown between DPPH-radical scavenging activity and reducing power (r=0969).

Antioxidant Constituents from the Stem of Sorghum bicolor

  • The EtOAc soluble fraction from the stem of Sorghum bicolor showed a strong free radical scavenging activity. Five major compounds were isolated from this fraction. They were identified by spectral data as methyl ferulate (1), methyl p-hydroxycinnamate (2), p-hydroxybenzaldehyde (3), tricin (4), and quercetin 3,4 -dimethyl ether (5). Among these compounds, 1 exhibited a strong, free radical scavenging activity on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) with an $IC_50$ value of 0.7 $\mu$M. We further studied the effects of these isolated compounds on the lipid peroxidation in rat liver microsomes induced by non-enzymatic method. All five compounds showed anti-lipid peroxidation activity ($IC_50$ values of 0.5, 0.4, 0.3 and 0.3 $\mu$ M, respectively).

  • Free radical-mediated cell damage and free radical attack on polyunsaturated fatty acids result in the formation of lipid radicals. These lipid radicals react readily with molecular oxygen to produce peroxy radicals responsible for initiating lipid peroxidation. The peroxidation of cellular membrane lipid can lead to cell necrosis and considered to be implicated in a number of pathophysiological conditions as well as in the toxicity of many xenobiotics. DPPH is known to abstract labile hydrogen and the ability to scavenge the DPPH radical is related to the inhibition of lipid peroxidation. (omitted)

  • Oxygen free radical have recently been found to mediate cell injury after ischemia in the kidney. The purpose of our study was to determine whether selenium had an effect on damge mediated by oxygen free radical in inorganic mercury induced renal failure, toxic model of renal failure. Toxic renal failure model was produced by subcutaneous injection of mercuric chloride (4mg/kg) once a day for 7 consecutive days. In additionally, coadministration of sodium selenite (1mg/kg) was performed by the same condition. As a consequence of this study, we were able to detect partially unequivocal role of selenium as below dipicted. The combination of sodium selenite showed that markedly inhibited production of superoxide radical in mercuric chloride alone. On the other hand, combined sodium selenite was unable to enhance against significantly lowered superoxide dismutase activity after mercuric chloride insult. However, simultaneous administration of sodium selenite was inclined to induce mitochondrial superoxide dismutase and catalase.

  • The EtOAc and n-BuOH soluble fractions from the aerial part of Setaria viridis showed a strong free radical scavenging activity. Six major compounds were isolated from these fractions. They were identified by spectral data as tricin (1), p-hydroxycinnamic acid (2), vitexin 2"-Ο-xyloside (3), orientin 2"-Ο-xyloside (4), $tricin-7-Ο-{\beta}-D-glucoside$ (5) and vitexin 2"-Ο-glucoside (6). Among these compounds, 4 and 5 exhibited strong free radical scavenging activities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH). We further studied the effects of these isolated compounds on the lipid peroxidation in rat liver microsomes induced by non-enzymatic method. As expected, 4 and 5 exhibited significant inhibition on $ascorbic/Fe^{2-}$ induced lipid peroxidation in rat liver microsomes.ver microsomes.

Free radical Scavenging Activities and Inhibitory Effects on Xanthine oxidase of Buckwheat (Suwon No. 5) (수원 5호 메밀의 유리라디칼 소거 및 Xanthine oxidase 활성 저해)

  • To examine the free radical scavenging activities from buckwheat, Suwon No. 5 was extracted with various solvents. The seed of Suwon No. 5 had a high contents of carbohydrate, but the vegetable of that had a 31.4% of protein, 28.6% of ash and 25.7% carbohydrate as moisture free basis. Phenols contents of vegetable extracts had higher than those of seed extracts. Acetone extract of vegetable showed the greatest electron donating ability. Butanol and acetone extracts of vegetable showed high inhibitory effect on lipid peroxidation. Acetone extract of vegetable has also excellent activity in the superoxide radical scavenging activity by xanthine/xanthine oxidase-cytochrome c reduction system. The inhibitory effects of extracts on xanthine oxidase were measured. Acetone extract had the strongest inhibitory effect on xanthine oxidase and $IC_{50}$ was 2.2$\mu\textrm{g}$.

  • Assay-guided fractionation of the methanol extract of the leaves of Albizzia julibrissin has furnished quercitrin (1) and afzelin (2) as the scavengers of DPPH radical. Quercitrin and afzelin showed scavenging activity of DPPH free radical with the $IC_{50}$ values of 17.3 and 71.5 ${\mu}M$, respectively.

Antioxidative Activity of Lichen Thamnolia vermicularis in vitro

  • This study was aimed at evaluating the antioxidant activities of methanol extract of Thamnolia vermicularis. The antioxidant activity, reducing power, superoxide anion radical scavenging and free radical scavenging activities were studied. The antioxidant activity of the extract correlated with its concentration ($0.2{\sim}2\;mg/ml$) in the reaction mixtures containing linoleic acid. Upto 67% of lipid peroxidation was inhibited by 2 mg/ml of the lichen extract. The extract showed strong free radical scavenging activity similar to that of BRA (positive control) in a manlier of concentration dependent. The lichen extract also showed moderate effects on superoxide anoin scavenging activity and reducing power, which was not so effective as that of Quercetin and BHA used as positive controls. This study suggests that T. vermicularis lichen can be used as a novel source of natural antioxidant.
