Free Radical Scavenging Activity of NokJoongTang Prepared from Antler and Various Oriental Medicinal Materials (녹용과 한약재료로 조제된 녹중탕의 유리기 소거 활성)

  • Two different NokJoongTangs were prepared by boiling antler and various oriental medicinal materials. The phenolic contents and free radical scavenging effects against DPPH(1,1-diphenyl-2-picrylhydrazyl) radical and hydroxyl radical of the NokJoongTangs which are used as the health supplement foods, were investigated. NokJoongTang-II prepared dried antler with various oriental medicinal materials, had higher phenolic content than that of NokJoongTang-I prepared 1mm raw antler with the same materials. In addition, NokJoongTang-II exhibited higher scavenging activities than these of NokJoongTang-I against DPPH radical and hydroxyl radical. These result indicate that free radical scavenging activity of NokJoongTang showed a difference by using raw and dry antler for NokJoongTang preparation, and that the findings also show a positive correlation between radical scavenging activity and total phenolic contents.

  • This study was performed to investigate the effect of kimchi intake on antiaging characteristics in liver of senescence-accelerated mouse (SAM) in terms of free radical production and anti-oxidative enzyme activities. Two hundred twenty SAM were divided into four groups and fed kimchi diet for 12 months. Experimental groups were kimchi free AIN-76 diet (control) group, Korean cabbage kimchi diet (KCK) group, mustard leaf added (30%) Korean cabbage kimchi diet (MKCK) group, and mustard leaf kimchi diet (MLK) group. Amount of freez-dried kimchi added to the diet was 5% that is equivalent to 50 g of fresh kimchi. Concentrations of total free radical, OH radical, $H_2O$$_2$in the liver significantly increased as aged (p<0.05). But those free radical concentrations from kimchi diet groups were lower than those of control (p<0.05). Among kimchi groups, MKCK and MLK groues showed greater inhibiting effect than KCK. Protective Effect of Oenanthe javanica Extract on the Carbon Tetrachloride-Induced Hepatotoxicity in Mice (미나리추출물이 사염화탄소에 의한 마우스 간손상에 미치는 영향)

  • The present work was undertaken to investigate the protective mechanism of Oenanthe iavanicu n-butanol extract on the carbon tetrachloride-induced hepatotoxicity in mice. It was observed that a striking enhancement of serum alanine aminotransferase and hepatic lipid peroxide content after carbon tetrachloride administration were markedly decreased by the presentment of Oenanthe javanica extract for 5 days. It was also observed that the hepatic aniline hydroxylase, catalase, glutathione S-transferase activity and glutathione content were not changed by the injection of Oenanthe javanica extract for 5 days. Whereas, hepatic xanthine oxidase activity was inhibited by the treatment of Oenanthe javanica extract for 5 days. After treatment with Oenanthe javanica extract, xanthine oxidase activity was decreased with dose and time-dependent manner as compared to control group. However, hepatic xanthine oxidase activity was not affected by the addition of Oenanthe javanica extract in vitro. These results suggest that the inhibition of hepatic xanthine oxidase activity by the injection of Oenanthe javanica extract is believed to be a possible protective mechanism for the carbon tetrachloride-indured hepatotoxicity in mice.

Comparative Study of Antioxidant Activities at Different Cultivation Area and Harvest Date of the Gomchwi 'Sammany' Variety (곰취 '쌈마니' 품종의 재배지역 및 수확기별 항산화 활성 비교)

  • This study was conducted for the comparison of antioxidant activities in different cultivation areas, harvest date and processing statuses in the new variety 'Sammany' of Gomchwi (Fischer ligularia). The lowland was Gangneung, 20 m above sea level, and the highland was Daegwallyeong, 750 m above sea level. The total phenolic contents, flavonoids, DPPH free radical scavenging activities, and reducing power were analyzed by freezing and drying the raw and blanched of Gomchwi leaves from April 18th to July 15th, and extracting them through in the use of methanol. Total phenolic contents, flavonoids, DPPH free radical scavenging activities and reducing power were found to be more pronounced during May than they were in the period of early harvest on April 18th in the lowland, as well as to be more effective during June than in May in the highland. Total phenolic contents, flavonoids, DPPH free radical scavenging activities, and reducing power by region, harvest date, and processing status were more effective in May than the early harvest of April in flat areas, and antioxidant activity in the highlands was elevated as of June. When compared with raw and blench leaves, raw leaves exhibited higher antioxidant activities across the board. Highland cultivation also displayed higher antioxidant activities than did lowland cultivation. Combining the above results, the 'Sammany' variety of Gomchwi was found to be more active in terms of antioxidants, which were harvested in May in flat areas and June in the highlands. Also, consuming them raw showed higher antioxidant activity than when blanched.

In vitro Free Radical Scavenging and Hepatoprotective Compound from Sanguisorbae Radix

  • In the course of searching for hepatoprotective agents from natural products, four compounds were isolated from the MeOH extract of Sanguisorbae Radix, as guided by their DPPH free radical scavenging activity. The structures were determined as 4,5-dimethoxy-3-hydroxybenzoic acid methyl ester (1), (+)-gallocatechin (2), methyl $6-O-galloyl-{\beta}-D-glucopyranoside$ (3), and pomolic acid $3-O-[{\alpha}-L-arabinopyranoside]-28-O-[{\beta}-D-glucopyranosyl]$ ester (ziyu-glycoside I) (4). Compounds 2 and 3 showed significant DPPH free radical scavenging effects, exhibiting $IC_{50}$ values of 11.4 and $13.0\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.3\;{\mu}M$. In evaluation of the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity, compound 2 showed the significant hepatoprotective effect with the $EC_{50}$ value of $91.84\;{\pm}\;11.0\;{\mu}M$ on tacrine-induced cytotoxicity in Hep G2 cells, while silybin, a positive control, exhibited $EC_{50}$ value of $122.4\;{\pm}\;12.5\;{\mu}M$.

Lipid Peroxidation Induced by the Reaction of Cytochrome c with Hydrogen Peroxide

  • Lipid peroxidation induced by the reaction of cytochrome c with $H_2O_2$ was investigated. When linoleic acid micelles or phosphatidyl choline liposomes were incubated with cytochrome c and $H_2O_2$, lipid peroxidation was increased in cytochrome c and $H_2O_2$ concentrations-dependent manner. Radical scavengers, azide, formate and ethanol prevented lipid peroxidation induced by the cytochrome c/$H_2O_2$ system. Iron specific chelator, desferoxamine also prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. These results suggest that lipid peroxidation may be induced by the cytochrome c/$H_2O_2$ system via the generation of free radicals. Carnosine, homocarnosine and anserine are present in the muscle and brain of many animals and human. Previous studies show that these compounds have an antioxidant function. In the present study, carnosine, homocarnosine and anserine significantly prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. Carnosine and related compounds also inhibited the free radical-generating activity of cytochrome c. The results suggest that carnosine, homocarnosine and anserine may prevent lipid peroxidation induced by the cytochrome c/$H_2O_2$ system through a free radical scavenging.

  • The ethanolic extract of chestnut (Castanea crenata S. et Z., Fagaceae) inner shell (CISE) and one of its components, ellagic acid (EA), were evaluated for their protective effects against 1, 1-diphenyl-2-picryl hydrazine (DPPH) free radical generation and hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line. CISE and EA were shown to possess the free radical scavenging effect against DPPH radical generation, significantly. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from Chinese hamster lung (CHL) cell, assessed by single cell gel electrophoresis assay and 8-hydroxy -2'-deoxy guanosine (8-OH-2'dG) assay. Furthermore, topical application of CISE [$12.5\%$(w/w) cream] and ellagic acid [$1.0\%$(w/w) cream] for 14 days potently inhibited malondialdehyde (MDA) formation of mouse dorsal skin (a marker of lipid peroxidation) induced by ultraviolet B exposure. Therefore, CISE and its component, ellagic acid, may be the useful natural antioxidants by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage when topically applied.
