Quantitative analysis of coumarins in Artemisia keiskeana and A. stolonifera using HPLC/PDA

  • Artemisia keiskeana and A. stolonifera are plants of the genus Artemisia, distributed in various regions, especially China and Korea. They are renowned as medicinal plants with biological and pharmacological activities. Fraxidin, isofraxidin, and daphnoretin are coumarins present in Artemisia spp.; however, research on them is limited. Therefore, this study was carried out to quantify the content of these compounds in the aerial parts of A. keiskeana and A. stolonifera in different regions in Korea. High-performance liquid chromatography was performed with a photodiode array detector and a reverse-phase INNO column. A. stolonifera only contained fraxidin with the highest amount found in Yongmun commune. A. keiskeana cultivation in Soyang commune gave the highest fraxidin and daphnoretin content. However, isofraxidin was not present in all samples. The findings suggest that the concentrations of the three compounds may differ depending on the growth site and provide a foundation for future studies.

  • Five constituents were isolated from the stem of Acanthopanax senticosus. Their structures were elucidated as (-)-sesamin (1), iso-fraxidin (2), 5-hydroxymethylfurfural (3), syringin (4) and acanthoside D (5) by spectral analysis. Among these compounds, 5-hydroxymethylfurfural (3) was isolated for the first time from this plant.

  • The radical scavenging effect of the MeOH extract of Fraxini Cortex on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was examined. The $CH_2Cl_2$-and n-BuOH-soluble fractions of MeOH extract showed the promising DPPH radical scavenging effects, and further purified by silica gel, Sephadex LH-20 column chromatography, and reversed-phase C-18 MPLC to yield five coumarins, esculetin (1), fraxidin (2), fraxetin (3), fraxidin $8-O-{\beta}-D-glucopyranoside$ (fraxin methyl ether) (5), esculin (6), and a secoiridoid oleuropein (4), and a coumarin-secoiridoid escuside (7). Compounds 1, 3, and 4 showed potent DPPH radical scavenging effects, exhibiting $IC_{50}$ values of 14.68, 9.64, and $22.03\;{\mu}M$, respectively. Compounds 6 and 7 also showed moderate effects with $IC_{50}$ values of 147.79 and $72.73\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.31\;{\mu}M$.

Inhibition of Interleukin-4 and β-Hexosaminidase Release in RBL-2H3 Cells by Compounds Isolated from Lobelia chinensis

  • Lobelia chinensis Lour. has commonly been used in traditional Chinese medicine for the treatment of antidote, diuretic, diarrhea, and inflammation. This study aimed to identify the active compounds in an aqueous extract of L. chinensis responsible for its anti-atopic effect in vitro using RBL-2H3 cells. A chemical investigation of secondary metabolites in an aqueous extract of L. chinensis led to the isolation of nine chemical constituents, which included the four marker compounds, and these were evaluated for their inhibitory effects on IL-4 mRNA expression and the release of β-hexosaminidase in propidium iodide-induced RBL-2H3 cells. We found diosmetin and fraxidin inhibited cellular IL-4 mRNA expression, and that diosmetin and 6,8-dimethoxycoumarin inhibited DNP-specific IgE-induced degranulation in these cells. Our study suggests that diosmetin, fraxidin, and 6,8-dimethoxycoumarin are potential candidates for the treatment of atopic diseases.

  • Seven compounds were isolated from the stem of Acanthopanax senticosus by repeated col-umn chromatography. Their structures were elucidated as isovanillin (1), (-)-sesamin (2), iso-fraxidin (3), (+)-syringaresinol (4), 5-hydroxymethylfurfural (5), eleutheroside B (6), and eleuth-eroside E (7) by spectral analysis. Among them, isovanillin (1) was isolated for the first time from the family Araliaceae.

Their structures were elucidated as iso-fraxidin, H-sesamin, 5-hydroxymethylfurfural, eleutheroside B, eleutheroside E and an unknown compound by spectral analysis. Of them, 5-hydroxymethylfurfural was isolated for the first time from A. senticosus.
