An improved mass spectrometric method for multicomponent analysis of metabolites in urine, well-suited for clinical biochemistry, is described. The method involves solvent elution of the metabolites from an adsorbent and the concentration of the eluate on a microadsorption column. This is administered by a direct inlet probe into the ionizing source of field ionization mass spectrometry (FIMS), which yield a molecular weight profile of the metabolites. The procedure provides rapidly (within one hour) reproducible profiles from a small volume of urine. The optimization of the sampling technique and the reproducibility are discussed.
Alcoholic extract of Spathodea campanulata P. aerial parts, and two of the isolated fractions from celite column showed strong antioxidant activity (92, 94 & 89% RSA, Radical Scavenging Activity). Phytochemical investigation of chloroform/EtOAc faction of this column led to the isolation of phenolic acids, caffiec acid (1), and ferulic acid (2), fraction EtOAc/MeOH on further fractionation afforded 3 Flavonoids, kampferol 3-O-glucoside (3), quercetin 3-methyl ether (4) and 8-methoxy kampferol 3-O-glucoside (5). The isolated constituents were identified by co chromatography with authentic samples, TLC, PC., UV, MS and $^1H-NMR$. Also the lipoidal matter of the plant was studied. The unsaponifiable matter was found to be mixture of hydrocarbons from $(C_{14}-C_{28})$, cholesterol, campasterol, stigmasterol, and ${\alpha}-amyrin$. Fatty acid methyl esters were found to contain 12 fatty acids. The fatty acids containing $C_{18}$ farmed ca.65% of the total mixture.
The hot-water extract of fruiting bodies in Fomitella fraxinea had potent anti-complementary activities. After fractionation of water-soluble polysaccharides by DEAE-Sephadex A-25 column chromatography, major anti-complementary activity was concentrated into the FF-AP1 among three polysaccharides (FF-NP, FF-AP1, FF-AP2). FF-AP1 was fractionated into $FF-AP1{\alpha}$ and $FF-AP1{\beta}$ obtained from the adsorbed fraction and unadsorbed fraction by affinity chromatography using a ConA-sepharose 4B column, respectively. $FF-AP1{\beta}$, which exihibited the highest anti-complementary activities had an IR absorption peak of $890cm^{-1}$, and a M.W. of about 15,000 (gel filtration). Anti-complementary activity of FF-AP1 decreased greatly by pronase treatment and periodate oxidation. $FF-AP1{\beta}$ responsible for potent anti-complemenary activities of Fomitella fraxinea was an acidic protein-containing heteroglycan consisted of 48% glucose, 13% mannose, and 12% galactose as major component sugars, 9.6% protein, 6% uronic acids.
An outer membrane-associated 2-furaldehyde dehydrogenase, catalyzing the oxidation of 2-furaldehyde to 2-furoic acid from Klebsiella pneumoniae was purified to homogeneity and characterized. The enzyme showed its highly specific dependency on $\beta$-$NAD^{+}$. Enzyme activity was monitored during purification by using substrate 2-furaldehyde and coenzyme $\beta$-$NAD^{+}$ by means of high performance liquid chromatography. The outer membrane was successfully collected by the methods of Percoll density gradient ultracentrifugation and ultracentrifugation after preferential solubilization of the membrane with $Mg^{2+}$ and Triton X-100. The enzyme was purified by the series of procedures including extraction of outer membrane protein with EDTA and lysozume, and fractionation by column chromatography on QAE-Sephades Q-50, and subsequently Sephadex G-100. The enzume showed its optimal activity at $85^{\circ}C$, pH 9.5, and in the presence of 1.5% (vol/vol) Triton X-100. The enzyme exhibited a native molecular size of 88,000 by nondenaturing polyacrylamide gel electrophoresis and had an apparent Km of 4.72mM for 2-furaldehyde.
This study was performed to purify fig pectinesterase(F-PE) and characterize its in situ activity. Three kinds of F-PE were partially separated by using ammonium sulfate fractionation, Q-Sepharose column, CM-cation exchanger column chromatography, and HPLC. One of those was anionic protein and the others were cationic proteins. All of them had approximate molecular weight of 27,000 and lost rapidly their activity during storage. Therefore alternative crude enzyme was prepared by suspending the freeze dried and milled fig powder in 0.1 M NaCl at pH 7.5. F-PE had the optimum pH of 8.5, the optimum temperature of $50^{\circ}C$ with activation energy of 7,671 cal $mol^{-1}K^{-1}$ and stability up to $55^{\circ}C$ with 10 minutes heating. Optimum activity was obtained in $0.2{\sim}0.4$ M NaCl with optimum solubility at above 0.8 M NaCl.
As a study on the ATP-inhibited ribonuclease of Bacillus subtilis the screening work for obtaining the ATP-inhibited ribonuclease negative mutant were carried out. And mutant strain was selected by the treatment of N-methyl-N'-nitro-N-nitrosoguanidine (NTG). For the selected strain the enzyme purification and some physiological properties were examined and the results obtained were as follows. 1. Among tested 1817 strains with the treatment of NTG, 101 strain was selected as a mutant strain. 2. ATP-inhibited ribonuclease was tentatively purified by several independent column chromatography. The results with Sephadex G-75 column were 30 times purification, 99% recovery, and 20 times purification, 98% recovery, respectively. 3. ATP-inhibited ribonuclease was purified by 60 times through acid treatment, ammonium fractionation, and two successive chromatography. 4. The purified ribonuclease were shown to be effectively concentrated in robonnclease content and to have reduced numbers of protein band on Disc electrophoresis. 5. This enzyme degraded single-stranded RNA to 2',3'-cyclic AMP, 2',3'-cyclic CMP, 2',3,-cyclic GMP, 2',3'-cyclic UMP and some unknown intermediates. The enzyme could not split double-stranded RNA.

Streptomyces griseus trypsin (SGT) is an extracellular proteinase produced by S. griseus. The sprT gene, which encodes premature SGT protein, was cloned into the plasmid pWHM3, a Streptomyces-E. coli shuttle vector. When the recombinant plasmid was introduced into Streptomyces lividans TK24, two proteins with molecular weights of 28 kDa and 42 kDa were detected. The 28-kDa protein was a SGT protein while the larger 42-kDa protein is thought to have been a premature form of the SGT protein. The SGT protein was purified to homogeneity via ammonium sulfate fractionation and many column chromatographies, including CM -sepharose chromatography, Mono-S chromatography, and Superose-12 chromatography, from the culture broth of S. lividans TK24 harboring the sprT gene. The N-terminal amino acid sequence, isoelectric points, and stabilities at various conditions of the SGT proteins purified from the Pronase and transformant were almost identical. The amount of the expressed SGT in S. lividans TK 24 was determined to be 5 times more than that of S. griseus based on the enzymatic activity against artificial substrate.
The carboxymethyl cellulase (CMCase) was purified from the induced culture filtrate of hybrid TAPW15703 between Aspergillus niger and penicillium verruculosum made by nuclear transfer. The enzyme was purified 80 fold with an overall yield 17% from the culture medium by ammonium sulfate fractionation, Sephadex G-75 gel permeation chromatography, and DEAE-ion exchange column chromatography. The molecular weight of the CMCase has estimated to be 32,000 daltons on SDS-polyacrylamide gel electrophoresis and Sephadex G-150 gel permeation chromatography. The purified enzyme functions optimally at pH 4.0 and 4$0^{\circ}C$ The Km value for carbosymethyl cellulose was 68 mM. The enzyme activity was increased by the presence of $Mg^{2+}$and Mn$^{2+}$.
The properties of polygalacturonase by Ganoderma lucidum in liquid culture were investigated. The enzyme was composed of an endo- and an exo-polygalacturonase. The endo- and exo-polygalacturonase were purified approximately 56 and 9.2-fold, respectively, through ammonium sulfate fractionation, gel filtration on Biogel P-100, anion exchange chromatography on DEAE-cellulose, gel chromatography on Sephadex G-150 and re-gel chromatography on Sephadex G-150. The endo- and exo-polygalacturonase had higher affinity for apple pectin than for citrus pectin or pectic acid. The Km values of the endo- and exo-polygalacturonase for apple pectin, determined on the Lineweaver-Burk plot, were 1.44 and 10.6 mg $ml^{-1}$ for apple pectin, respectively. Purified endo-polygalacturonase was found to be homogeneous electrophoretically and had a molecular weight of 54,000 estimated on SDS polyacrylamide gel. The optimal pH for the activity of the enzymes was 4.0. The endo- and exo-polygalacturonase were stable in the pH range of 4.0 to 6.0 and 3.5 to 5.5, respectively. The optimal temperatures of the endo- and exo-polygalacturonase were 40 and $60^{\circ}C$, respectively. The exo-polygalacturonase was more resistant to heat than the endo-polygalacturonase, requiring heating for 40 min at $80^{\circ}C$ for complete inactivation. The activity of the endo-polygalacturonase was increased by $Ca^{++}$ and $Mn^{++}\;ions$, while that of the exo-polygalacturonase was increased by $Ca^{++}\;ion$ only, and was not affected by $Mn^{++}\;ion$.

Soyasaponins are phytochemicals of major interest fur their health benefits. Chemical investigation of a soybean phytochemical concentrate resulted in the isolation and identification of triterpenoid saponins. The MeOH extraction of defatted hypocotyl separated from soybeans was peformed by the automated solvent extractor (ASE). Fractionation was performed on a flash column ($150mm{\times}40mm$ i.d.) packed with a preparative $C_{18}$ reverse phase bulk packing material $(125\AA,\;55-105{\mu}m)$ and monitored at 210 nm, and collected 14 fractions. Consequent Fsat preparative column liquid chromatography (Fast PCLC) was performed for the purification of Fraction-I (Fr-I) collected from the fraction 8 and 9 of flash chromatography. Fsat PCLC was performed on a Luna $C_{18}\;10{\mu}m,\;100{\AA}$, semipreparative reverse phase column ($250cm{\times}50mm$ i.d.) for the purification of isolated unknown compound (Fr-I-2). Chemical structure of acetyl soyasaponin $A_1\;(MW:1436.6,\;C_{67}H_{104}O_{33})$ was identified and determined by a combination of extensive NMR ($^1H-NMR$, 400 MHz; $^{13}C-NMR$, 100 MHz; DEPT), IR, UV, and ESI-MS analysis.
