단일 및 분할 방사선조사에 의한 백서절치의 급성반응에 관한 연구 (ACUTE RESPONSE OF THE RAT INCISOR BY SINGLE AND FRACTIONATED IRRADIATION)

  • Six to eight-month-old female albino rats were used as experimental animals. As an irradiation equipment, a Co-60 was used. The experimental animals were divided to; 6 of the control group, 12 of the 500cGy single irradiation group, 12 of the 1000cGy fractionated irradiation group, and 12 of the 1500cGy fractionated irradiation group. From the first week to the forth, 3 rats were picked from each group every week to be sacrificed and fixed with formalin. Those rats were observed by means of H-E stain after being taken radiograph and decalcified. The analysis of radiographic findings and light microscopic findings gives results as follows: 1. The delay of dental eruption rate was found in every group which underwent the irradiation experiment. Dentin niche, osteodentin, and dentin island were formed in the parts which were damaged by the irradiation. 2. The longer the observation period was, the more deposit of osteodentin and dentin island was formed. 3. In the single irradiation group, the damage effect was in proportion to the increase of radiation dose, whereas the damage was much less in the fractionated group receiving the same dose. 4. The 500cGy single irradiation group got temporary repairable damage, while the 1000cGy single irradiation group got considerable damage and showed much slower eruption rate than the 500cGy single irradiation group. The basal portion of the 1500cGy single irradiation group, whose growth was arrested, was destroyed. 5. The fractionated group were irradiated 500cGy everyweek. Repair was visible during the interval periods. The damage was accumulated as irradiation repeated, but degree of damage was lower than that of the 1000cGy and 1500cGy single irradiation group.

백서 유방암의 단일 및 분할 조사시 방사선치료 효과 (Single and Fractionated Irradiation of Mammary Tumor of Rat)

  • The therapeutic effect of mammary breast cancer of rat (Sprague Dawley) was estimated by single and 5 fractionated irradiation of $Co^{60}-\gamma-ray$. Response rates over 50a were 20, 43, 67, $80\%$ respectively by single dose irradiation of 800, 1,200, 1,600, 2,000 rad, and 20,38, 57, $88\%$ by 5 fractionated irradiation of 1,400, 2,100, 2,800, 3,500 rad. $50\%$ tumor control dose$(TCD_{50})$ were 1,282rad, 2,312rad respectively with single and fractionated irradiation.

단일 및 분할 방사선조사가 백서 악하선 미세혈관계에 미치는 영향 (EFFECTS OF THE SINGLE AND FRACTIONATED IRRADIATION ON THE MICROVASCULATURE OF THE RAT SUBMANDIBULAR GLAND)

  • The purpose of the study was to investigate the effects of the single and fractionated irradiation on the microvascular structure of the submandibular gland in rats. For this study, 90 Sprague-Dawley strain rats were irradiated to their neck region with equal split doses of 9Gy for a 4 hours interval and 15Gy single dose by 6MV X-irradiation and sacrificed on the 1st, 3rd, 7th, 14th and 27th day after irradiation. The author observed histological changes at Hematoxylin and Eosin staining and PAS staining under a light microscope, and also observed distribution and structural changes of the microvasculature in rat submandibular gland using a scanning electron microscope by forming vascular resin casting. The results were as follows: 1. In the light microscopic examination, the microvasculature was slightly dilated and decreased in number on the 1st day after irradiation, and increase in number of microvasculature was observed on the 3rd day after irradiation. And then distribution of microvasculature was markedly increased on the 7th day after iradiation, but decreased on th 14th day after irradiation again. Such changes were greater in the single irradiated group than in the fractionated irradiated group. 2. The reaction to PAS staining on glandular cell was decreased on the 1st and the 3rd day after irradiation, and recovered on the 7th day after irradiation. The reaction was decreased on the 14th day after irradiation again, and recovered on the 28th day after irradiation. Changes were more apparent in the single irradiated group. 3. In the scanning electron microscopic examination, early changes of microvasculature were decreased capillary density, dilation of conduits and meandering. Increased capillary dentsity or anastomosis due to vascular reproduction and smooth curved running were observed on the 7th and 14th day after irradiation. Decreased capillary and smooth running tendency were observed on the 28th day after irradiation again. Such changes were greater in the single irradiated group than in the fractionated irradiated group.

단회(單回) 및 분획(分劃) X선(線) 흉부(胸部) 조사(照射)가 가토(家兎) 폐포(肺胞) 표면활성(表面活性) 물질(物質)에 미치는 영향(影響) (Effect of Single or Fractionated X-Irradiation on the Pulmonary Surfactant in Rabbits)

  • In an attempt to clarity the effect of X-irradiation on the activity of surfactant in rabbits, and also to observe the possible difference when the irradiation was made in single or fractionated dose, X-ray in dose of 900 r was irradiated to rabbits either in single or fractionated dose of 300 r each day for three day, Tension-area diagram of lung extract was recorded automatically by a modified Langmuir-Wilhelmy balance with a synchronized recording system designed in this department. The surface tension of lung extract was measured at 1,3,5,24, and 48 hours post-irradiation, and the results were compared with the non-irradiated normal group. The result obtained are summerized as follows: 1) The maximal surface tension, minimal surface tension, width of the tension-area diagram at the surface area of 40% in lung extract, and stability index of the normal rabbit lung extracts were 40.73 dynes/cm, 8.96 dynes/cm. 20.71 dynes/cm, and 1.28 respectively. 2) Activity of surfactant was significantly depressed by X-irradiation, and the pattern of depression was more prominent in the single irradiation group than in the fractionated group. 3) It was found that the changes of the width of the tension-area diagram at the surface area of 40% in lung extract and stability indices corresponded well with that of the maximal or minimal surface tension.

탄소 빔 분할조사 시 Linear-Quadratic모델, Incomplete-Repair모델, Marchese 모델 결과 비교 (Comparison of Linear-Quadratic Model, Incomplete-Repair Model and Marchese Model in Fractionated Carbon Beam Irradiation)

  • 본 연구는 탄소 빔의 분할조사 후 세포생존율 (Surviving Fraction, SF) 값에 따른 Linear-Quadratic model, Incomplete Repair model, Marchese model의 결과값을 비교하기 위해 진행하였다. 탄소 빔을 4fraction까지 조사한 후 얻은 세포생존율 값을 바탕으로 mathematica 프로그램 (ver 9.0)을 이용하여 각각의 모델로 결과값을 얻어 비교해 보았다. 그 결과 즉시 NB1RGB를 시딩한 값은 repair가 감안되지 않은 LQ 모델이 적합하였지만 fraction 시행한 후의 결과값은 오차를 보였다. 따라서 Potentially Lethal Damage Repair (PLDR)과 Sublethal Damage Repair (SLDR)의 발생을 각각 감안한 repair 모델을 이용하여 적합한지 판단하였다. 이를 바탕으로 탄소 빔의 분할 조사 시 LQ 모델에 각각의 repair의 양을 감안한 새로운 회복 관련 모델의 적용 가능성을 보고자 하였다.

  • In view of the importance of sulfhydryl groups in producing a chemical protection against ionizing radiation, an attempt was made to evaluate the changes of intrinsic non-protein sulfhydryl (NP-SH) and non-Protein disulfide(NP-SS) of the lung and liver tissues of rabbits following the whole body X-irradiation with 900 r either in single or fractionated $(300\;r{\times}3)$ dose. NP-SH was measured by Ellman's method, and NP-SS was measured by the electrolytic reduction method described by Dohan. Experiment was performed at 1,3,5,24 and 48 hours post·irradiation, and the results were compared with the control. The results thus obtained are summarized as follows; 1) Intrinsic levels of NP-SH and NP-SS of normal rabbits were $0.77{\pm}0.10$ and $0.61{\pm}0.07\;{\mu}mol/gm$ wet weight in liver, and $0.28{\pm}0.03$ and $0.54{\pm}0.03$ in lung tissues respectively. 2) NP-SH of liver after single X-irradiation showed no significant change in general, but at 48 hours post·irradiation, it was elevated comparing with the normal value. 3) Levels of NP-SS in liver was decreased than the normal value in the irradiated groups, and the lowest level was observed at 3 hours after single X-irradiation and at 5 hours after fractionated irradiation. 4) In lung tissues, levels of NP-SS showed no significant change from the control at earlier experimental hours, but a great decrease was observed at later Part of the experiment.

분할방사선조사와 SR 4233 병용에 의한 저산소분압 종양의 반응증강 (Hypoxic Tumor Can be More Responsive to Fractionated Irradiation Combined with SR 4233 (Tirapazamine))

  • 마우스 종양인 SCCVll과 RIF-1에서 선택적 저산소세포 치사제인 SR 4233 (tirapazamine)과 분할 방사선조사를 병용하면 저산소분획이 큰 종양에서 방사선 감수성이 더욱 증강될 것인가를 확인하기 위하여 본 연구를 실시하였다. 종양세포 이식이전에 방사선조사를 받은 정상조직에서 성장한 종양의 저산소세포 분획이 크다는 사실을 이용하여 저산소세포분획이 큰 종양 모델을 수립 사용하였다. 일정 용적의(100$mm^{3}$) 종양에 대하여 1회 2 Gy를 1일 2회씩 6회 또는 1회 2.5 Gy를 1일 2회씩 8회 조사하는 등의 분할조사법을 사용하여 방사선조사를 시행하였다. SR 4233를 병용한 경우에는 매회 방사선조사 30분 전에 0.08 mmol/kg를 복강내 주사하였다. 정상조직에서 성장한 종양에 비하여 방사선 조사를 받은 조직에서는 성장한 종양은 분할 방사선조사에 대하여 약 5배의 내성을 보였으나 SR 4233 분할 치료에 대하여는 약 5배의 감수성을 나타내었다. 정상 종양 및 저산소세포 분획이 큰 종양의 방사선 감수성은 공통적으로 SR 4233의 병용후 증강되었으며 저산소세포 분획이 큰 종양에서의 증강율이 정상종양에서의 증강율과 동일하거나 더욱 높았다. 따라서 선택적 저산소세포 치사제인 SR 4233과 분할 방사선조사를 병용할 경우 저산소분획이 큰 종양에서 치사효과가 더욱 증강된 사실을 확인하였다. 이러한 결과는 높은 저산소세포 분획에 의한 방사선내성을 보이는 종양중 일부는 저산소세포치사제와 방사선치료를 병용함으로써 국소적 종양관해 실패의 요인인 저산소 세포를 극복하는 임상 적용 가능성을 시사한다.

방사선조사가 백서 협점막에 미치는 영향에 관한 전자현미경적 연구 (AN ELECTRON MICROSCOPIC STUDY ON THE EFFECTS OF IRRADIATION ON THE BUCCAL MUCOSA OF RAT)

  • The author studied the acute reaction of cobalt-60 irradiation to buccal mucosa in rats and difference of the effects of single versus fractionated exposure. 195 Sprague Dowley strain rats, weighing about 120gm, were used in this experiment. 3 rats served as controls and the remaining 192 rats were divided into six groups of 32 rats each. Experimental group Ⅰ, Ⅱ, Ⅲ were received a single dose of 15Gy, 16.5Gy, 18Gy and group Ⅳ, Ⅴ, Ⅵ were received two equal sized fractionated dose of 9Gy, 9.75Gy, 10.5Gy at 4 hour intervals, respectively. The experimental groups were irradiated with cobalt-60 teletherapy unit, Picker model 4M 60 (Field size, 12x5 cm, SSD, 50㎝, Dose rate, 222cGy/min, Depth, 1㎝). The animals were sacrificed at 1, 2, 3, 6, 12 hours, 1, 3, 7 days after irradiation and the changes of the irradiated buccal mucosa were observed by electron and light microscopy. The results were as follows: 1. A single exposure was more damaging than fractionated exposure, and as the radiation dose increased, the changes of cell organelles became faster, but the healing of radiation-induced damage in fractionated exposure was faster than in single exposure. 2. The radiation-induced changes of the basal cells were the most prominent in 18Gy-single exposure group, and the least in 18Gy-fractionated exposure group. 3. Electron-microscopically, there appeared nuclear changes, swelling of mitochondria and rough endoplasmic reticulum, decrease of free ribosome, presence of vesicles, widening of intercellular space, and loss of basal lamina. The early remarkable changes were partly loss of nuclear membrane and swelling of mitochondria. 4. Light-microscopically, derangement and pyknosis of basal cells, hydropic changes of spinous cells, enlargement of granular cells, indistinctness of basement membrane, and proliferation of epithelium were observed.

Walker 256 Carcinosarcoma의 원자력병원 싸이클로트론 속중성자선 분할조사에 대한 생물학적 효과비에 관한 연구 (The RBE of Fractionated Fast Neutron on Walker 256 Carcinosarcoma with KCCH-Cyclotron)

  • 원자력병원 싸이클로트론으로 발생되는 고에너지 속중성자선의 생물학적효과에 대한 연구의 일환으로 RBE를 측정하였다. 대상은 Walker 256 carcinosacoma 및 정상조직으로 마우스 위장관 및 골수를 이용하였고 단일 및 분할조사에 대한 반응을 관찰하였다. 마우스 공장소낭선세포의 단일전신조사에 의한 RBE는 2.8이었다. 단일조사시 이식암 Walker 250의 TCD 50에 대한 RBE, 마우스 골수 및 위장관의 RBE는 각각 1.9, 1.9, 1.5이었다. 분할조사시 Walker 256의 RBE는 분할횟수의 증가에 따라 감소하였고 1회 분할조사량의 증가에 따라 증가하였다.

마우스에서 전신 저선량 분할 방사선 조사에 의한 면역학적 변화 평가 (Effects of Low-Dose Fractionated Total Body Irradiation on Murine Immune System)

  • 방사선요법은 항암 치료에서 널리 이용되는 요법으로, 항암치료에 저선량 방사선을 이용하는 것에 대한 관심이 증가되고 있고, 저선량 방사선의 다양한 생물학적 효과가 있음이 보고되고 있다. 그러므로 본 연구에서는 마우스 모델에서 저선량 방사선이 면역반응에 어떠한 영향을 미치는지, 또한 이를 감지할 수 있는지를 조사하였다. C57BL/6 마우스에 $^{137}Cs$ 선원을 이용하여 연속 3일간 총 90 mGy의 저선량 방사선을 전신 조사한 후 마지막 방사선 조사 2, 14, 28일 후에 마우스를 희생시켜 말초 혈액 세포수, 비장 세포수, 비장 내 면역세포의 비율과 활성화 정도를 분석하였다. 말초 혈액 검사를 통해, 저선량 방사선 조사군에서 적혈구와 혈소판 수의 유의적 변화는 관찰할 수 없었으며, 백혈구 수는 마지막 방사선 조사 후 2일째에 선량-의존적인 감소를 보였으나, 점차 회복되는 경향을 나타냈다. 비장세포 수도 2일째 감소를 보였지만 서서히 그 수가 증가됨을 확인하였다. 2일과 14일째에 비장세포의 Foxp3 mRNA가 감소된 반면, CD4 T 세포와 CD69 양성세포가 증가되었다. 마우스에서 분할 저선량 방사선을 전신조사한 결과, 방사선조사군에서 특이적인 임상 증상이나 유의적인 체중감소를 보이지 않았다. 본 연구에서는 마우스를 대상으로 저선량을 분할 조사하였을 경우에도 면역학적 변화를 확인할 수 있으며 이를 통해 향후 저선량 방사선의 생물학적 효과를 뒷받침하는 자료로 활용할 수 있으리라 기대한다.
