Rapid discrimination of commercial strawberry cultivars using Fourier transform infrared spectroscopy data combined by multivariate analysis

  • To determine whether pattern recognition based on metabolite fingerprinting for whole cell extracts can be used to discriminate cultivars metabolically, leaves and fruits of five commercial strawberry cultivars were subjected to Fourier transform infrared (FT-IR) spectroscopy. FT-IR spectral data from leaves were analyzed by principal component analysis (PCA) and Fisher's linear discriminant function analysis. The dendrogram based on hierarchical clustering analysis of these spectral data separated the five commercial cultivars into two major groups with originality. The first group consisted of Korean cultivars including 'Maehyang', 'Seolhyang', and 'Gumhyang', whereas in the second group, 'Ryukbo' clustered with 'Janghee', both Japanese cultivars. The results from analysis of fruits were the same as of leaves. We therefore conclude that the hierarchical dendrogram based on PCA of FT-IR data from leaves represents the most probable chemotaxonomical relationship between cultivars, enabling discrimination of cultivars in a rapid and simple manner.

Prediction and discrimination of taxonomic relationship within Orostachys species using FT-IR spectroscopy combined by multivariate analysis (FT-IR 스펙트럼 데이터의 다변량 통계분석 기법을 이용한 바위솔속 식물의 분류학적 유연관계 예측 및 판별)

  • To determine whether pattern recognition based on metabolite fingerprinting for whole cell extracts can be used to discriminate cultivars metabolically, leaves of nine commercial Orostachys plants were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data from leaves were analyzed by principal component analysis (PCA) and Partial least square discriminant analysis (PLS-DA). The dendrogram based on hierarchical clustering analysis of these PLS-DA data separated the nine Orostachys species into five major groups. The first group consisted of O. iwarenge 'Yimge', 'Jeju', 'Jeongsun' and O. margaritifolius 'Jinju' whereas in the second group, 'Sacheon' was clustered with 'Busan,' both of which belong to O. malacophylla species. However, 'Samchuk', belong to O. malacophylla was not clustered with the other O. malacophylla species. In addition, O. minuta and O. japonica were separated to the other Orostachys plants. Thus we suggested that the hierarchical dendrogram based on PLS-DA of FT-IR spectral data from leaves represented the most probable chemotaxonomical relationship between commercial Orostachys plants. Furthermore these metabolic discrimination systems could be applied for reestablishment of precise taxonomic classification of commercial Orostachys plants.

Rapid discrimination system of Chinese cabbage (Brassica rapa) at metabolic level using Fourier transform infrared spectroscopy (FT-IR) based on multivariate analysis (배추 대사체 추출물의 FT-IR 스펙트럼 및 다변량 통계분석을 통한 계통 신속 식별 체계)

  • To determine whether FT-IR spectral analysis based on multivariate analysis could be used to discriminate Chinese cabbage breeding line at metabolic level, whole cell extracts of nine different breeding lines (three paternal, three maternal and three $F_1$ lines) were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data of Chinese cabbage plants were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA), and hierarchical clustering analysis (HCA). The hierarchical dendrograms based on PLS-DA from two of three cross combinations showed that paternal, maternal, and their progeny $F_1$ lines samples were perfectly separated into three branches in breeding line dependent manner. However, a cross combination failed to fully discriminate them into three branches. Thus, hierarchical dendrograms based on PLS-DA of FT-IR spectral data of Chinese cabbage breeding lines could be used to represent the most probable chemotaxonomical relationship among maternal, paternal, and $F_1$ plants. Furthermore, these metabolic discrimination systems could be applied for rapid selection and classification of useful Chinese cabbage cultivars.

Discrimination of Cultivars and Cultivation Origins from the Sepals of Dry Persimmon Using FT-IR Spectroscopy Combined with Multivariate Analysis (FT-IR 스펙트럼 데이터의 다변량 통계분석을 이용한 곶감의 원산지 및 품종 식별)

  • This study aimed to establish a rapid system for discriminating the cultivation origins and cultivars of dry persimmons, using metabolite fingerprinting by Fourier transform infrared (FT-IR) spectroscopy combined with multivariate analysis. Whole-cell extracts from the sepals of four Korean cultivars and two different Chinese dry persimmons were subjected to FT-IR spectroscopy. Principle component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) of the FT-IR spectral data successfully discriminated six dry persimmons into two groups depending on their cultivation origins. Principal component loading values showed that the 1750-1420 and $1190-950cm^{-1}$ regions of the FT-IR spectra were significantly important for the discrimination of cultivation origins. The accuracy of prediction of the cultivation origins and cultivars by PLS regression was 100% (p<0.01) and 85.9% (p<0.05), respectively. These results clearly show that metabolic fingerprinting of FT-IR spectra can be applied for rapid discrimination of the cultivation origins and cultivars of commercial dry persimmons.

Establishment of discrimination system using multivariate analysis of FT-IR spectroscopy data from different species of artichoke (Cynara cardunculus var. scolymus L.) (FT-IR 스펙트럼 데이터 기반 다변량통계분석기법을 이용한 아티초크의 대사체 수준 품종 분류)

  • To determine whether FT-IR spectral analysis based on multivariate analysis for whole cell extracts can be used to discriminate between artichoke (Cynara cardunculus var. scolymus L.) plants at the metabolic level, leaves of ten artichoke plants were subjected to Fourier transform infrared(FT-IR) spectroscopy. FT-IR spectral data from leaves were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). FT-IR spectra confirmed typical spectral differences between the frequency regions of 1,700-1,500, 1,500-1,300 and $1,100-950cm^{-1}$, respectively. These spectral regions reflect the quantitative and qualitative variations of amide I, II from amino acids and proteins ($1,700-1,500cm^{-1}$), phosphodiester groups from nucleic acid and phospholipid ($1,500-1,300cm^{-1}$) and carbohydrate compounds ($1,100-950cm^{-1}$). PCA revealed separate clusters that corresponded to their species relationship. Thus, PCA could be used to distinguish between artichoke species with different metabolite contents. PLS-DA showed similar species classification of artichoke. Furthermore these metabolic discrimination systems could be used for the rapid selection and classification of useful artichoke cultivars.

Establishment of rapid discrimination system of leguminous plants at metabolic level using FT-IR spectroscopy with multivariate analysis (FT-IR 스펙트럼 기반 다변량통계분석기법에 의한 두과작물의 대사체 수준 식별체계 확립)

  • To determine whether FT-IR spectroscopy combined with multivariate analysis for whole cell extracts can be used to discriminate major leguminous plant at metabolic level, seed extracts of six leguminous plants were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data from seed extracts were analyzed by principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). The PCA could not fully discriminate six leguminous plants, however PLS-DA could successfully discriminate six leguminous plants. The hierarchical dendrogram based on PLS-DA separated the six leguminous plants into four branches. The first branch was consisted of all three Vigna species including Vigna radiata var. radiate, Vigna angularis var. angularis and Vigna unguiculata subsp. Unguiculata. Whereas Pisum sativum var. sativum, Glycine max L and Phaseolus vulgaris var. vulgaris were clustered into a separate branch respectively. The overall results showed that metabolic discrimination system were in accordance with known phylogenic taxonomy. Thus we suggested that the hierarchical dendrogram based on PLS-DA of FT-IR spectral data from seed extracts represented the most probable chemotaxonomical relationship between six leguminous plants.

  • Silicon direct bonding technology is very attractive for both silicon-on-insulator devices and sensor fabrication because of its thermal stress free structure and stability. The process of SDB includes hydration of silicon wafer and heat treatment in a wet oxidation furnace. After hydration process, hydroxyl groups of silicon wafer were analyzed by using Fourier transformation-infrared spectroscopy. In case of hydrophilic treatment using a ($H_{2}O_{2}\;:\;H_{2}SO_{4}$) solution, hydroxyl groups are observed in a broad band around the 3474 $cm^{-1}$ region. However, hydroxyl groups do not appear in case of diluted HF solution. The bonded wafer was etched by using tetramethylammonium hydroxide etchant. The surface of the self etch-stopped silicon dioxide is completely flat, so that it can be used as sensor applications such as pressure, flow and acceleration, etc..

A Method for Absolute Determination of the Surface Areal Density of Functional Groups in Organic Thin Films

  • To develop a methodology for absolute determination of the surface areal density of functional groups on organic and bio thin films, medium energy ion scattering (MEIS) spectroscopy was utilized to provide references for calibration of X-ray photoelectron spectroscopy (XPS) or Fourier transformation-infrared (FT-IR) intensities. By using the MEIS, XPS, and FT-IR techniques, we were able to analyze the organic thin film of a Ru dye compound ($C_{58}H_{86}O_8N_8S_2Ru$), which consists of one Ru atom and various stoichiometric functional groups. From the MEIS analysis, the absolute surface areal density of Ru atoms (or Ru dye molecules) was determined. The surface areal densities of stoichiometric functional groups in the Ru dye compound were used as references for the calibration of XPS and FT-IR intensities for each functional group. The complementary use of MEIS, XPS, and FT-IR to determine the absolute surface areal density of functional groups on organic and bio thin films will be useful for more reliable development of applications based on organic thin films in areas such as flexible displays, solar cells, organic sensors, biomaterials, and biochips.

Grafting of Glycidyl Methacrylate upon Coralline Hydroxyapatite in Conjugation with Demineralized Bone Matrix Using Redox Initiating System

  • Grafting of glycidyl methacrylate (GMA) upon coralline hydroxyapatite in conjugation with demineralized bone matrix (CHA-DBM) using equal molar ratio of potassium persulfate/sodium metabisulfite redox initiating system was investigated in aqueous medium. The optimum reaction condition was standardized by varying the concentrations of backbone, monomer, initiator, temperature and time. The results obtained imply that the percent grafting was found to increase initially and then decrease in most of the cases. The optimum temperature and time were found to be 50 $^{\circ}C$ and 180 min, respectively, to obtain higher grafting yield. Fourier transform infrared (FT-IR) spectroscopy and X-ray powder diffraction (XRD) method were employed for the proof of grafting. The FT-IR spectrum of grafted CHA-DBM showed epoxy groups at 905 and 853 $cm^{-1}$ / and ester carbonyl group at 1731 $cm^{-1}$ / of poly(glycidyl methacrylate) (PGMA) in addition to the characteristic absorptions of CHA-DBM, which provides evidence of the grafting. The XRD results clearly indicated that the crystallographic structure of the grafted CHA-DBM has not changed due to the grafting reaction. Further, no phase transformation was detected by the XRD analysis, which suggests that the PGMA is grafted only on the surface of CHA-DBM backbone. The grafted CHA-DBM will have better functionality because of their surface modification and hence they may be more useful in coupling of therapeutic agents through epoxy groups apart from being used as osteogenic material.

Synthesis and Morphological Transformation of NaA Zeolite Crystals at High Temperature

  • Well-shaped NaA zeolite cubic crystals of a large size of $5-7{\mu}m$ were synthesized by a hydrothermal method in a mother solution having a $3.55Na_2O:Al_2O_3:1.6SiO_2:593\~2000H_2O$ composition. Thermal treatment of NaA zeolite crystals resulted in the formation of a crystalline phase of $NaAlSiO_4-Camegeite$ between 800 and $900^{\circ}C$. Even at $1000^{\circ}C,\;NaAlSiO_4$ phase was found as a major product. Environmental Scanning Electron Microscopy (ESEM), High Resolution Transmission Electron Microscopy (HRTEM), X-Ray powder Diffraction (XRD), Fourier Transform Infrared (FT-IR) spectroscopy, and DTA/TGA and BET analyses were used to characterize the initial materials and the obtained products after various heat treatments.
