Due to the fact that an inactivated anthrax vaccine may lark its immunogenicity and stability of immunogen a number of spore vaccines were exclusively used worldwide. In these studies a number of important factors were emphasized to achieve the following: selection of non or less allergic strain of anthrax bacillus, capsulation of bacteria. obtaining of non sporulating but vegetative organisms, adequate inactivation of B. anthraccis by means of formalin, adsorption of immunogen to aluminum hydroxide gel. Non or less allergic strains of anthrax bacillus which is inactivated with formalin was selected by a hyperimmunization and shock test in rabbits. Obtaining capsular material and vegetative immunogen, a virulent anthrax organisms were cultivated on sodium bicarbonate medium with of without adding of l-alanine in which B, anthracis grew luxuriantly without forming spores. Inactivation was carried out at $37^{\circ}C$ water bath for 3 days after the bacterial culture was mixed with formalin, in a final concentration of two per cent of formalin. Aluminum hydroxide gel was added to the mixture of anthrax and blackleg bacterin. Vaccines were injected guinea pig via subcutaneous or intramusoular route and challenged after three weeks and the possibilities of protection was tested. Throughout the studies. the above mentioned vaccines possibly protected the vaccinated guinea pigs more than 80 per cent compared to that of the controls. This experimental results strongly suggest that the vaccine may possibly applicable to bovine.

To clarify the distinction between three difference acupoints, the effects of acupuncture on a behavioral performance were evaluated following formalin test. Male Sprague-Dawley rats were used. Each rat received a manual acupuncture at ST36 (zusanli), SP9 (yinlingquan) or BL60 (kunlun) acupoint before formalin injection. The flinching and licking responses were counted by two blinder investigators. The pretreatment of BL60 acupoint was showed significantly inhibition in flinch behavior as compared with control group. These results suggest that acupuncture at BL60 acupuncture may be effective in relieving inflammatory pain.
A comparative experiment on the duration of immunity and antigenicity of the Black-leg vaccine was conducted and following results were obtained. Animals vaccinated with formalin and modified living vaccine were equally survived when they were challenged 12 months after the vaccination. It was further found that the rate of survival was 50 % on both groups when the animals were challenged 16 months after the vaccination, and no distinctive difference was observed between the two groups by means of protection power.
When glutamate was infected intrathecally, the result is similar to those produced by TPA injected. The involvement of protein kinase C (PKC) in the nociceptive responses in rat dorsal horn neurons of lumbar spinal cord was studied. In test with formalin, a PKC inhibitor (chelerythrine) inhibited dose-dependently the formalin-induced behavior response. Neomycin also inhibited it significantly. But, a PKC activator (12-O-tetradecanoylphorbol-13-ester, TPA) showed reverse effect. When gluatamate was injected intrathecally, we observed the result is smilar to those produced by TPA injection. On the other hand, intrathecal injection of glutamate induced thermal and mechanical hyperalgesia. In Tail-flick test, we examined the involvement of PKC on the glutamate-indeced thermal hyperalgesia. Chelerythrine showed an inhibitory effect and TPA enhanced thermal response. Glutamate decreased the mechanical threshold significantly. A pretreatment of chelerythrine and neomycin inhibited glutamate-induced mechanical hyperalgesia, but the effect of neomycin was not significant. TPA had little effect on the mechanical nociceptive response. These results suggest that the PKC activation through metabotropic receptor at postsynaptic region of spinal cord dorsal horn neurons may influence on the persistent nociception produced by chemical stimulation with formalin, thermal and mechanical hyperalgesia induced by glutamate.
Background: The authors showed in a previous study that some novel triazine derivatives had an anti-inflammatory effect. The present study was designed to evaluate the antinociceptive effect of five out of nine compounds including two vanillintriazine (5c and 5d) and three phenylpyrazole-triazine (10a, 10b, 10e) derivatives which showed the best anti-inflammatory effect. Methods: Male Swiss mice (25-30 g) were used. To assess the antinociceptive effect, acetic acid-writhing, formalin, and hot plate tests were used after intraperitoneal injection of each compound. Results: All compounds significantly (P < 0.001) reduced acetic acid-induced writhing at tested doses (50, 100, and 200 mg/kg). Also, the percent inhibition of writhing in the acetic acid test showed that at the maximum tested dose of these compounds (200 mg/kg), the order of potencies is as follows: 10b > 10a > 10e > 5d > 5c. In the formalin test, compounds 5d, 10a, and 10e showed an antinociceptive effect in the acute phase and all compounds were effective in the chronic phase. In the hot plate test, compounds 5c, 5d, and 10a demonstrated an antinociceptive effect. Conclusions: The results clearly showed that both vanillin-triazine and phenylpyrazole-triazine derivatives had an antinociceptive effect. Also, some compounds which showed activity in the early phase of formalin test as well as in the hot plate test could control acute pain in addition to chronic or inflammatory pain.
Objectives : The objective of this study is to investigate Anti-nociceptive and anti-inflammatory effects of Clematidis Radix (CR) herbal-acupuncture on the test rats with induced acute pain. Methods : The effects of Clematidis Radix (CR)-distillates were investigated in three types of models with three different pain. Highly purified distillate of CR called CR herbal-acupuncture was injected to Zusanli ($ST_{36}$) acupoint. In the tail flick test, the CR herbal-acupuncture treatment did not show a significant effect of relieving acute pain. To investigate the anti-inflammatory effect of CR herbal-acupuncture, the second testing model'pain was induced by injecting formalin to its planter. For the last model, carrageenan was injected into tarsal joint. the medicinal effect of CR herbal-acupuncture was evaluated through the behavioral analyses such as licking time, weight distribution ratio and ankle circumference. Results : In the formalin test, the analgesic effect of CR herbal-acupuncture was more pronounced in the late phase (for 20 min after the early phase) than in the early phase (for the first 10 min post formalin injection). It was proven by weight distribution ratio testing and ankle edema testing that herbal-acupuncture of CR inhibited arthritis caused by the carrageenan. Conclusions : These results revealed that CR herbal-acupuncture was effective to alleviate the inflammatory pain and could be used as an analgesic treatment with an anti-inflammatory effect.

In the present study, the anti-inflammatory effect of Jungcheonhwadamgangki-tang plus antler water extract was tested in formalin-injected chronic inflammation model in mouse hind paw. The test articles were dosed once a day for 10 days, and changes on the body weight, paw weights were observed with histopathology of induced paw dorsum pedis. In addition, histomorphometry was also monitored at sacrifice. 15 mg/kg/10 mL of dexamethasone (DEXA) and diclofenac (DICLO) intraperitoneally dosed groups were used as reference groups. A significantly decrease of both absolute and relative paw weights were observed in all dosing groups including DEXA and DICLO groups compared to that of control, and a significantly decrease of the differences between intact and induced paw weights were also observed Jungcheonhwadamgangki-tang plus antler dosing groups compared to that of control. These histological signs-hypertrophy of paw dorsum pedis tissues were detected results from edematous changes on the cutaneous and subcutaneous tissues with severe infiltration of chronic inflammatory cells-were dramatically decreased in all dosing groups including DEXA and DICLO dosing groups compared to that of control. Especially, dose dependently decreases were detected in Jungcheonhwadamgangki-tang plus antler dosing groups compared to that of control. Base on these aforementioned results, it is concluded that Jungcheonhwadam- gangki-tang plus antler have clear anti-inflammatory effect on the chronic inflammation induced by formalin injection.
Acute toxicity of formalin (37% formaldehyde) was conducted to determine the median lethal concentration (LC50) on eel (avarage weight 96 ± 3.6 g, average total length 43 cm), Anguilla japonica at concentrations ranging from 0 to 500 ppm. In particular, this study was designed to estimate the safety concentrations of formalin in testing eels to eradicate Pseudodactylogyrus. All fish died after 10 hours and 24 hours at 500 ppm and 400 ppm, respectively. After 24 hours, cumulative mortality was 96.6% and 13.3% at 300 ppm and 200 ppm formalin, respectively. However, all experimental fish were alive after 24 hours at 100 ppm. The lethal concentration values were computed by using non-linear least square method. At the start of the test, water temperature, pH and dissolved oxygen level were 27~28℃, 7.4 and 5.55 ppm, respectively. The 24 hr-LC50 were 269 ppm.

Voltage dependent calcium channel (VDCC), one of the most important regulator of $Ca^{2+}$ concentration in neuron, play an essential role in the central processing of nociceptive information. The present study investigated the antinociceptive effects of L, T or N type VDCC blockers on the formalin-induced orofacial inflammatory pain. Experiments were carried out on adult male Sprague-Dawley rats weighing 220-280 g. Anesthetized rats were individually fixed on a stereotaxic frame and a polyethylene (PE) tube was implanted for intracisternal injection. After 72 hours, 5% formalin ($50 \;{\mu}L$) was applied subcutaneously to the vibrissa pad and nociceptive scratching behavior was recorded for nine successive 5 min intervals. VDCC blockers were administered intracisternally 20 minutes prior to subcutaneous injection of formalin into the orofacial area. The intracisternal administration of 350 or $700{\mu}g$ of verapamil, a blocker of L type VDCC, significantly decreased the number of scratches and duration in the behavioral responses produced by formalin injection. Intracisternal administration of 75 or $150 \;{\mu}g$ of mibefradil, a T type VDCC blocker, or 11 or $22\; {\mu}g$ of cilnidipine, a N type VDCC blocker, also produced significant suppression of the number of scratches and duration of scratching in the first and second phase. Neither intracisternal administration of all VDCC blockers nor vehicle did not affect in motor dysfunction. The present results suggest that central VDCCs play an important role in orofacial nociceptive transmission and a targeted inhibition of the VDCCs is a potentially important treatment approach for inflammatory pain originating in the orofacial area.
