비상품 양파추출물의 Mouse 피부암 및 위장안 억제효과 (Reduction Effect of Carcinogen-induced Mouse Epidermal and Forestomach Carcinogenesis by the Extract of Onion Wastes)

  • 상품가치가 없는 비상품구로부터 추출한 OWE를 mouse의 피부암 및 위장암 저해 효과에 관한 실험을 한 결과 피부암의 경우 control구에서는 6주부터 종양이 발생하여 23주에는 mouse 당 2.9개의 종양이 발생하였으나 20mg OWE 처리구에서는 8주부터 종양이 발생하여 23주에는 mouse 당 1.3개의 종양이 발생하여 control에 비해 55.2%의 종양 억제효과가 있었다. 20mg quercetin 처리에서는 6주부터 종양이 발생하기 시작하여 23주에는 mouse 당 1.2개의 종양이 발생하였다. 처리별 암 발생율은 20mg OWE 처리에서 85.7%, 20mg quercetin 처리에서는 76.2%의 발생율을 보여 control의 95.2%에 비해 낮은 암발생율을 보였다. 위장암의 경우 control에서는 mouse 당 9.2개의 종양이 발생하였고, 50mg OWE 및 25mg OWE 처리에서는 각 6.1, 6.3개의 종양이 발생하여 control에 비해 각각 33.7%, 31.5%의 종양억제효과가 있었다. 그리고 25mg quercetin 처리에서는 mouse 당 5.3개의 종양이 발생하였다. 처리별 암발생율은 50mg OWE 및 25 mg OWE 처리에서 각 88.2%, 94.1%, 25mg quercetin 처리에서 83.3%의 발생율을 보여 control 100%에 비해 낮은 암발생율을 보였다.

  • Ground beef에서 처음으로 분리된 conjugated linoleic acid (CLA)는 mouse epidermal carcinogenesis, mouse forestomach tumor, rat mammry carchnogenesis 및 colon carcinogenesis를 억제하는 항암성 효과를 지니고 있다. CLA는 항암성뿐만 아니라 anti-atherosclerosis, immune function과 항산화성 등의 생리활성 효능이 있는 것으로 밝혀졌으며, CLA의 주요 source는 반추동물에서 유래된 meat, 우유 및 유제품이며 일반 식물성유에도 미량으로 존재한다. 최근 국내외에서 CLA를 이용한 기능성 meat 및 식품을 생산하기 위한 연구가 수행되고 있다. 본 발표에서는 CLA에 관한 간단한 review와 본 연구진의 연구결과를 소개하고자 한다.

내관혈(內關穴) 자침(刺鍼)이 급성 역류성 식도염 백서(白鼠)에 미치는 영향 (Inhibitory Effects of Naegwan-acupuncture($PC_6$) on Acute Reflux Esophagitis Rat)

  • Objectives : This study was to evaluate inhibitory effects of Naegwan-acupuncture($PC_6$) on acute RE(reflux esophigitis) rat induced by pylorus and forestomach ligation operation. Methods : Twenty seven SD rats were divided three groups (intact normal rat; RE control rat; RE control rat respectively stimulated by Naegwan point($PC_6$)). All rats was fasted for 18 h but free water, we induced RE by pylorus and forestomach ligation operation. Six hour after the operation, rats were sacrified, collected bloods in the abdominal vein, dissected a esophagus and stomach. The stomach was washed a 1 ml PBS to research gastric volume, pH, acidity and mucin release of gastric juice, esophagus was cut longitudinally and pictured a innter mucosa area to research damages in esophagus. The proinflammatory cytokine and chemokine including IFN-${\gamma}$, TNF-${\alpha}$, IL-$1{\beta}$, IL-6 and MCP-1 were analyzed by ELISA kit. Results : 1. Significantly, death rate of $PC_6$ acupuncture rat group was decreased compared to that of RE control group. 2. Gastric Volume, gastric injury and esophageal mucosa demage were decreased significantly, too. 3. Compared with RE, all of the proinflammatory cytokine and chemokine analyzed in serum of $PC_6$ were decreased remarkably. Especially, there were significant meanings TNF-${\alpha}$, IL-6 and MCP-1 in serum of $PC_6$ were decreased. Conclusion : The results suggest that antiinflammatory and protecting effects of PC6 could attenuate the severity of reflux esophagitis and prevent the esophageal mucosal damage, and validate its therapeutic use in esophageal reflux disease.

Induction of Ornithine Decarboxylase and Tumor Promotion by N-Methyl-N′-Nitro-N-Nitrosoguanidine, Sodium Chloride, and Dimethyl Itaconate

  • The possible tumor-promoting activities of sodium chloride (NaCl) and dimethyl itaconate (DMI), one of the quinone reductase inducers, were examined on stomach of male Wistar rats treated with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). Administrations of NaCl and DMI after the initiation by MNNG resulted in various sized masses in the rat forestomach. Histopathologic studies showed that the combination of NaCl and DMI made an enhancing effect on the MNNG-induced carcinogenesis, resulting in papilloma in 5 weeks and squamous cell carcinoma in 20 weeks in submucosal area of forestomach. We also used an in vivo shortterm method for evaluating possible tumor-promoting activity with ornithine decarboxylase (ODC) as a marker. The markable inductions of the ODC activities by MNNG, NaCl, and DMI were found in the pyloric mucosa of rat stomach in time-dependent manners. A single administration of MNNG induced ODC activity up to 288 pmol $CO_2$/hr/mg protein at 24 hr after the administration. NaCl caused induction of ODC with a maximum of 179 pmol $CO_2$/hr/mg protein at 8 hr after the administration. ODC was induced up to 539 pmol $CO_2$/hr/mg protein at 16 hr after the administration of DMI. Additional treatment of NaCl and NaCl plus DMl caused 2 fold and 7 fold increases, respectively, in the ODC activity of the MNNG-alone group at 24 hr after the administration. These results suggest that NaCl and DMI have promoting activities in the rat gastric carcinogenesis initiated by MNNG.

역류성(逆流性) 식도염(食道炎)에 대한 이진탕가미방(二陳湯加味方) 구성(構成) 개별(個別) 한약재의 효과(效果) (Effects of Individual Herbal Components of Yijintang-gamibang in the Rat Reflux Esophagitis)

  • Objectives : The object of this study was to observe the protective effects of 8 types of individual herbal components of Yijintang-gamibang, on the reflux esophagitis (RE) in rats as compared with omeprazole. Methods : Each 100 mg/kg individual herbal component was orally pretreated, at 1 hr before and 6 hrs after pylorus and forestomach ligation, the changes on the esophageal lesion areas, gastric volumes, acid and pepsin outputs, antioxidant effects, esophageal total hexose and sialic acid contents were observed with changes on the esophageal histopathology. The results were compared with an omeprazole 10 mg/kg treated group. Results : Pylorus and forestomach ligation-induced RE were inhibited by treatment of all 8 herbal components and omeprazole except for Pinella Rhizoma and Massa Meicata Fermentata, in order of Atractylodis Rhizoma, Citri Pericarpium, omeprazole, Glycyrrhizae Radix, Hordei Fructus Germiniatus, Holelen and Citri Pericarpium, as compared with RE control. Pinella Rhizoma and Massa Meicata Fermentata did not show any favorable protective effects against RE nor antioxidant effects in the present study. Conclusions : The main active herbal components showing favorable protective effects on RE of Yijintang-gamibang mediated by antioxidant effects, were Atractylodis Rhizoma, Citri Pericarpium, Glycyrrhizae Radix, Hordei Fructus Germiniatus, Holelen and Citri Pericarpium. Furthermore Atractylodis Rhizoma and Citri Pericarpium showed more favorable protective effects as compared with omeprazole, therefore, it is expected that Atractylodis Rhizoma and Citri Pericarpium have potential as new alternatives or safe therapeutics against RE.

이진탕가미방(二陳湯加味方)의 항산화 효과를 통한 역류성 식도염 억제효과 (Protective Effects of Yijin-tang-gamibang Aqueous Extracts on Reflux Esophagitis Mediated by Antioxidant Defense Systems)

  • The object of this study was to observe the protective effects of Yijin-tang-gamibang (YJGMB), Yijin-tang has been traditionally used in the Korean Medicine for treating various digestive diseases. The study showed that it is effective on reflux esophagitis induced by pylorus and forestomach ligation in rats. Three different dosages of YJGMB extracts, 200, 100 and 50 mg/kg, were orally pretreated, once a day for 28 days before pylorus and forestomach ligation. Seven groups, each of 8 rats per group were used in the present study. The results were compared with omeprazole, antioxidant and proton pump inhibitor, 30 and 10 mg/kg treated group. YJGMB 200 mg/kg were showed similar protective effects as compared with 30 mg/kg of omeprazole but more favorable effects were observed in 200, 100 and 50 mg/kg of all YJGMB treated rats as compared with omeprazole 10 mg/kg in the present study. In addition, YJGMB 200 mg/kg were showed more favorable antioxidant effects as compared with 30 mg/kg of omeprazole in the present study. Detail mechanism studies should be conduced in future with the screening of the biological active chemical compounds in herbs.

역류성 식도염 랫트에 미치는 금은화(金銀花) 물 추출물의 치료 효과 (Effects on Rats with Reflux Esophagitis Treated with Lonicerae Flos Extract)

  • Because Lonicerae Flos has effects of antiinflammatory and antioxidant, we studied an effect of Lonicerae Flos on reflux esophagitis (RE) through those effects. Rats were treated with three different dosages of LF (500, 250 and 125 mg/kg) orally for 14 days before pylorus and forestomach ligation. Six hrs after pylorus and forestomach ligation, we dissected a stomach and examined a stomach volume, gastric acid output, pepsin release in the stomach, total hexose, sialic acid in stomach tissue and histamine contents of sera. The results were compared with an ${\alpha}$-tocopherol (once orally, 1hr before operation, 30 mg/kg) treated group in which the effects on RE were already confirmed. Lonicerae Flos extract (LE) reduced gastric volumes compared to RE control. This indicate that LE protect a stomach mucosa by depressing of gastric acid release and corresponse with a reducing histamine content of serum. And LE decreasd a volume of pepsin in stomach compraed to RE control, LE increased contents of total hexose and sialic acid based on esophageal and gastric mucus. This indicated that an increased mucus by LE protected inflammation of esophagus mucosa and gastric mucosa induced by gastric acid. So, LE suppressed a gasric acid by decreasing a pepsin release in stomach, suppressed an injury of esophagus inducted by gastric acid with increasing esophageal mucus and a minimum dose of LE to RE was 250 mg/kg. The results suggest that antioxidant effects of LF could attenuate the severity of reflux esophagitis and prevent the esophageal mucosal damage, and validate its therapeutic use in esophageal reflux disease.

Three different dosages of Yijin-tang-gamibang extracts, 300, 150 and 75 mg/kg, were once orally administered 1 hour before pylorus ligation. 6 groups, each of 8 rats per group were used in the present study. The results were compared with Omeprazole, antioxidant and proton pump inhibitor, 30 mg/kg treated group. Yijin-tang-gamibang extracts 300 mg/kg were showed similar or slightly lower effects as compared with 30 mg/kg of omeprazole in the present study. Detail mechanism studies should be conduced in future with the efficacy tests of individual herbal composition of Yijin-tang-gamibang and the screening of the biological active compounds in herbs.

Protective Effects of Chlorogenic Acid against Experimental Reflux Esophagitis in Rats

  • Esophageal reflux of gastric contents causes esophageal mucosal damage and inflammation. Recent studies show that oxygen-derived free radicals mediate mucosal damage in reflux esophagitis (RE). Chlorogenic acid (CGA), an ester of caffeic acid and quinic acid, is one of the most abundant polyphenols in the human diet and possesses anti-inflammatory, antibacterial and anti-oxidant activities. In this context, we investigated the effects of CGA against experimental RE in rats. RE was produced by ligating the transitional region between the forestomach and the glandular portion and covering the duodenum near the pylorus ring with a small piece of catheter. CGA (10, 30 and 100 mg/kg) and omeprazole (positive control, 10 mg/kg) were administered orally 48 h after the RE operation for 12 days. CGA reduced the severity of esophageal lesions, and this beneficial effect was confirmed by histopathological observations. CGA reduced esophageal lipid peroxidation and increased the reduced glutathione/oxidized glutathione ratio. CGA attenuated increases in the serum level of tumor necrosis factor-${\alpha}$, and expressions of inducible nitric oxide synthase and cyclooxygenase-2 protein. CGA alleviates RE-induced mucosal injury, and this protection is associated with reduced oxidative stress and the anti-inflammatory properties of CGA.
