Precise and fast identification of crop cultivars is essential for efficient breeding and plant breeders' rights. Traditional methods for identification of jujube cultivars are based on the evaluation of morphological characteristics. However, due to time constraints and environmental influences, it is difficult to distinguish cultivars using only morphological traits. In this study, we cloned fragments from improved inter simple sequence repeats (ISSR) analysis, and developed stably diagnostic sequence-characterized amplified region (SCAR) markers. The specific ISSR bands of jujube cultivars from Dalizao and Boeundaechu were purified, cloned, and sequenced. As a result, four clones labeled 827Dalizao550, 827Boeun750, 846Boeun700, and 847Dalizao850 were identified. In order to investigate whether they were specific for the jujube cultivar, four pairs of SCAR primers were then designed and polymerase chain reaction (PCR) amplifications were conducted to analyze 32 samples, including jujube and sour jujube. In the PCR amplification of the 827Dalizao550 SCAR marker, the specific bands with 550 bp were amplified in six samples (Dalizao, Sandonglizao, Dongzao, Yuanlin No. 2, Suanzao 2, Suanzao 4), but unexpected bands (490 bp) were amplified in the others. Moreover, in the PCR amplification of the 847Dalizao850 SCAR marker, the specific bands with 850 bp were found in three samples (Dalizao, Sandonglizao, and Dongzao) and 900 bp unexpected bands were amplified in five samples (Pozao, Suanzao 1, Suanzao 2, Suanzao 3, Suanzao 4). These results showed that newly developed markers could be useful as a fast and reliable tool to identify jujube cultivars. However, further identification of polymorphic information and the development of SCAR markers are required for the identification of more diverse cultivars.
The jujube is an important fruit tree species in Korea. Traditionally, classifications of jujube cultivars have been based on morphological characters; however, morphological identification can be problematic because morphological traits are affected by environmental conditions. Therefore, DNA markers are now being used for the rapid and accurate identification of plant species. Inter-simple sequence repeat (I-SSR) is one of the best DNA-based molecular marker techniques, which is useful for studying genetic relations and for the identification of closely related cultivars. In this study, 5 Korean jujube trees and 1 jujube tree imported from China were analyzed for 16 I-SSR primers. Amplification of the genomic DNA of jujube cultivars by using I-SSR analysis generated 100 bands, with an average of 6.25 bands per primer, of which 45 bands (45%) were polymorphic. The number of amplified fragments with I-SSR primers ranged from 2 to 13. The percentage of polymorphism ranged from 10% to 100%. I-SSR finger printing profiles showed that 'Boeun jujube' and 'Daeri jujube' had characteristic DNA patterns, indicating unequivocal cultivar identification at molecular level. According to the results of clustering analysis, the genetic similarity coefficient ranged from 0.68 to 0.92. 'Boeun jujube' and 'Daeri jujube' were divided into independent groups, and 'Bokjo jujube', 'Geumseong jujube', 'Wolchul jujube', and 'Mudeung jujube' were placed in the same group. Therefore, I-SSR markers are suitable for the discrimination of 'Boeun jujube' and 'Daeri jujube' cultivars.
This study was carried out to prove the factor properties of the soil affected by a forest fire through the physical and chemical analysis and the data from the conclusion of those analysis are applied to the development of technics that reduces a forest fire-induced 2nd damage. The forest fire was in December 2000 at Gangreung city and Donghae city, Gangwon provinces, Korea. Soil samples were collected at upper layers (0-5 cm) and bottom layers (5-40 cm) in November 2001 from the burned and control sites. Values of pH in burned soils of the upper layers affected by forest fire are higher than those in control soils. Both the fragments of fire-burned plant and differences of geological properties are resulted in a class of soil. Contents of organic matters in burned soils are higher than those in control soils, exceptionally the contents of organic matters in burned soils that contain coaly shale are lower than those in control soils. Weathering indices in burned soils are higher than those in control soils and it concerned with loss of soil. Iron ions Fe(Fe$^{2+}$ or Fe$^{3+}$) are easily extracted from the burned soils by rainfall, but Mn ions are straightly exist in the burned soils by physiochemical adsorption of colloid. Through the sequential extraction in the burned soils and control soils, we are certificate the extraction of Fe ions and the disturbance of Mn ions from the burned soils. As a consequence of factor analysis in burned soil and control soil, we are certificate that the influence of forest fire results in a disturbance of positive correlation factors.
The purpose of the precent investigation was to investigate diversity patterns in a fragmented Young-do forested landscape. It was conducted on sloping 18 plots (twelve 50$\times$50 m plots and six 10$\times$100 m plots). Total 1562 individuals were identified and measured in the 50$\times$50 m plots. These were a total of 68 species of 23 families present in the three sites. Least significant differences (LSD) by post hoc analysis revealed that fragments site A had significantly greater densities than site B. Shannon-Wiener functions differed significantly among forests (F=4.12, p<0.05), with site A forest having significantly higher value (3.401) than the others (2.590 for site B and 2.648 for site C). Pinus thunbergii and Quercus aliena were dominant on most sites. site. The Pinus densiflora forest of sites A and C is distributed better in the NE exposures compared with the opposite ones. Ficus erecta and Pinus densiflora largely occupied the southern exposures. The spatial distribution in southern Young-do's littoral forest was very heterogeneous, reflected by most species having low densities and patchy distributions.
An extracellular xylanase from the brown-rot fungus Fomitopsis palustris grown on rice straw culture was purified to a single protein band. On SDS-PAGE, the molecular mass of purified enzyme was estimated to be about 43 kDa. The amino acid sequence of the proteolytic fragments showed high homology with fungal glycoside hydrolase family 10 xylanases. The $K_m$, $K_{cat}$ and $V_{max}$ for birch xylan were $31mg/m{\ell}$, $2.3{\times}10^4/min$ and 252.3 U/mg, respectively. The optimal activity of the purified xylanase from F palustris was observed at pH 4.0~5.0 and $70^{\circ}C$.

This study established the genetic characterisation of 10 microsporidian isolates infecting non-mulberry silkworms (Antheraea assamensis and Samia cynthia ricini) collected from biogeographical forest locations in the State of Assam, India, using PCR-based markers assays: inter simple sequence repeat (ISSR) and random amplified polymorphic DNA (RAPD). A Nosema type species (NIK-1s_mys) was used as control for comparison. The shape of mature microsporidian spores were observed oval to elongated, measuring 3.80 to $4.90{\mu}m$ in length and 2.60 to $3.05{\mu}m$ in width. Fourteen ISSR primers generated reproducible profiles and yielded 178 fragments, of which 175 were polymorphic (98%), while 16 RAPD primers generated reproducible profiles with 198 amplified fragments displaying 95% of polymorphism. Estimation of genetic distance coefficients based on dice coefficients method and clustering with un-weighted pair group method using arithmetic average (UPGMA) analysis was done to unravel the genetic diversity of microsporidians infecting Indian muga and eri silkworm. The similarity coefficients varied from 0.385 to 0.941 in ISSR and 0.083 to 0.938 in RAPD data. UPGMA analysis generated dendrograms with two microsporidian groups, which appear to be different from each other. Based on Euclidean distance matrix method, 2-dimensional distribution also revealed considerable variability among different identified microsporidians. Clustering of these microsporidian isolates was in accordance with their host and biogeographic origin. Both techniques represent a useful and efficient tool for taxonomical grouping as well as for phylogenetic classification of different microsporidians in general and genotyping of these pathogens in particular.
Soils in the sloping uplands in Korea are subject to intensive land use with high input of agrochemicals and are vulnerable to soil erosion. Development of the environmentally sound land management strategy is essential for a sustainable production system in the sloping upland. This report addresses the status of upland agriculture and the best management practices for the uplands toward the sustainable agriculture. More than 60% of Korean lands are forest and only 21% are cultivating paddy and upland. Uplands are about 7% of the total lands and about 62% of the uplands are in the slopes higher than 7%. Due to the site-specificity of the upland, many managerial and environmental problems are occurring, such as severe erosion, shallow surface soils with rocky fragments, and loadings of non-point source (NPS) contaminants into the watershed. Based on the field trials, most of the sloping uplands were classified as Suitability Class III-V and the major limiting factor was slope and rock fragments. Due to this, soils were over-applied with N fertilizer, even though N rate was the recommendation. This resulted in decreases in yield, degradation of soil quality and increases in N loading to the leachate. Various case studies drew management practices toward sustainable production systems. The suggested BMP on the managerial, vegetative, and structural options were to practice buffer strips along the edges of fields and streams, winter cover crop, contour and mulching farming, detention weir, diversion drains, grassed waterway, and slope arrangement. With these options, conservation effects such as reductions in raindrop impact, flow velocity, runoff and sediment loss, and rill and gully erosion were observed. The proper management practice is a key element of the conservation of the soil and water in the sloping upland.

1-Ethyl-3-methylimidazolium acetate known as efficient biomass pretreatment reagent was used for the extraction of lignin from rigida pine wood (pitch pine), which was called to ionic liquid lignin (ILL), and chemical structural features of ILL were compared with the corresponding milled wood lignin (MWL). The amounts of phenolic hydroxyl groups (Phe-OH) was determined to 10.0% for ILL and 7.2% for MWL, respectively, where those of methoxyl groups (OMe) were 4.9% for ILL and 11.0% for MWL, respectively. The weight average molecular weight (Mw) of ILL (3,995) were determined to ca. 1/2 of that of MWL (8,438) and polydispersity index (PDI: Mw/Mn) suggested that the lignin fragments were more uniform in the ILL (PDI 1.36) than in the MWL (PDI 2.64). The temperature (Tm) corresponding to maximum decomposition rate (Vm) of ILL ($306.6^{\circ}C$) was ca. $35^{\circ}C$ lower than that of MWL ($341.9^{\circ}C$), suggesting that ILL was thermally unstable than MWL, as evidence from the lower Tm for ILL. Moreover, the structural characteristics of ILL and MWL were confirmed by spectroscopic analyses (FT-IR and $^{13}C$-NMR), and these results indicated ionic liquid (1-ethyl-3-methylimidazolium acetate) was chemically or physically bound to ILL.
An isolate of Peanut stunt virus (PSV) isolated from black locust tree (Robinia pseudo-acacia L.) showing severe mosaic and malformation symptoms, was designated as PSV-Rp. PSV-Rp was characterized by the tests of host range, physical properties, RNA and coat protein composition and RT-PCR analysis. Nucleotide sequences of the cucumoviruses CP genes were also used for identification and differentiation of PSV-Rp. Six plant species were used in the host range test of PSV-Rp. PSV-Rp could be differentiated from each Cucumovirus strain used as a control by symptoms of the plants. The physical properties of PSV-Rp virus were TIP $65^{\circ}C$, DEP $10^{-3}$, and LIP $2{\sim}3$ days. In dsRNA analysis, PSV-Rp consisted of four dsRNAs, but satellite RNA was not detected. Analysis of the coat proteins by SDS-PAGE showed one major protein band of about 31 kDa. RT-PCR using a part of Cucumovirus RNA3 specific primer amplified ${\sim}950bp$ DNA fragments from the crude sap of virus-infected black locust leaves. RFLP analysis of the RT-PCR product could differential PSV-RP from CMV The nucleotide sequence identity between the PSV-Rp CP and the TAV-P CP genes and the PS-V-RP CP and CMV-Y CP genes were 61.6% and 40.5%, respectively. On the other hand, the nucleotide sequence identity of the PSV-Rp CP gene was $70.9%{\sim}73.4%$ in comparison with those of PSV subgroup I (PSV-ER and PSV-J) and 67.3% with that of PSV subgroup II(PSV-W). Especially, the nucleotide sequence identity of PSV-Rp CP gene and that of PSV-Mi that was proposed recently as the type member of a novel PSV subgroup III was 92.4%.

Genus Sorbus is a long lived woody species. Plants of this genus are primarily distributed patchy throughout Asia and Europe. Sorbus commixta is primarily distributed throughout Europe. Eastern Asian Sorbus species are regarded as very important herbal medicines in Korea and China. Random amplified polymorphic DNA (RAPD) was used to investigate the genetic variation and phylogenetic analysis of four species of this genus in Korea. Although some Korean populations of these species were isolated and patchily distributed, they exhibited a high level of genetic diversity. Twenty-six primers revealed 205 loci, of which 128 were polymorphic (62.4%). S. commixta showed the highest diversity (0.165), whereas S. aucuparia showed the lowest diversity (0.109). The estimated gene flow (Nm) was low high among intra-species (mean Nm=0.755). A similarity matrix based on the proportion of shared fragments (GS) was used to evaluate relatedness among species. The estimate of GS ranged from 0.786 to 0.963. The molecular data allowed us to resolve well-supported clades in Korean taxa and European species. An addition, especially, species-specific markers for genus Sorbus by RAPD analysis may be useful in germ-plasm classification and agricultural process of several taxa of this genus.
