Phenolic Compounds from Fruits of Amorpha fruticosa L.

  • This study was carried out to examine the constituents of Amorpha fruticosa (Leguminosae), a shrub originated from North Africa. Dried and ground fruit of A. fruticosa were extracted with methanol and then concentrated to give the crude extracts. The crude extracts was successively fractioned with organic sovents, such as n-hexane, CH$_2$Cl$_2$ and EtOAc. Seven compounds were isolated from the fruits of A. fruticosa. On the basis of spectrosopic data, the structures of these compounds were determined as: kaempferol 7-O-${\alpha}$-L-rhamnopyranoside (I), methyl 3,4,5- trihydroxybenzoate (methyl gallate, II), tephrosin (III), dalbinol (IV), gallic acid (V), 2",4",5",7-tetramethoxyisofavone (Ⅵ) and Dalbinol 2"-O-${\beta}$-D-Glucopyranoside (Ⅶ) respectively. Lignans from the Fruits of Schizandra chinensis and Their Inhibitory Effects on Dopamine Content in PC12 Cells

  • Five lignans including gomisin N (1), wuweizisu C (2), gomisin L1 (3), (+)-deoxyschizandrin (4), and gomisin J (5) have been isolated from the fruits of Schizandra chinensis. The structures of the isolated compounds were elucidated by analyzing MS and NMR spectra. Effects of the compounds isolated in this study on the dopamine content in PC12 cells were investigated to evaluate their inhibitory effectiveness. The gomisin N, wuweizisu C, and gomisin J showed 25.4%, 39.8%, and 35.1 %, respectively, inhibition effect on dopamine content in PC12 cells at the concentration of $50\;{\mu}g/ml$.

Effects of Liriodenine on Dopamine Biosynthesis in PC12 Cells (Liriodenine이 PC12 세포중의 Dopamine 생합성에 미치는 영향)

  • The effects of liriodenine, an aporphine isoquinoline alkaloid, on dopamine content in PCl2 cells were investigated. Treatment of PC12 cells with liriodenine decreased dopamine content in a dose-dependent manner (33.6% inhibition at $10\;{\mu}M$ for 12 h). The $IC_{50}$ in value of liriodenine was $8.4\;{\mu}M$. Dopamine content decreased at 3 h and reached a minimal level at 12 h after the exposure to liriodenine. Under these conditions, the activities of tyrosine hydroxylase and aromatic L-amino acid decarboxylase were also inhibited at $10\;{\mu}M$ of liriodenine by 10.1% and 20.2% relative to control, respectively. In addition, liriodenine inhibited the increase in dopamine content induced by L-DOPA Treatments $(50-100\;{\mu}M)$ in PC12 cells. These results suggest that liriodenine inhibited dopamine biosynthesis and L-DOPA-induced increase in dopamine content by reducing the activities of tyrosine hydroxylase and aromatic L- amino acid decarboxylase in PC12 cells.

Detection, isolation, and characterization of the cucumber mosaic virus in Pseudostellaria heterophylla from Korea

  • Weeds play an important role in the survival of viruses and are potential inoculum sources of viral diseases for crop plants. In this study, specimens of Pseudostellaria heterophylla exhibiting symptoms of the cucumber mosaic virus (CMV) were collected in Bonghwa, Korea. The characteristics of the disease were described and leaf RNA was extracted and sequenced to identify the virus. Three CMV contigs were obtained and PCR was performed using specific primer pairs. RNA from positive samples exhibiting CMV leaf symptoms was amplified to determine the coat protein. A sequence comparison of the coat protein gene from the CMV BH isolate shared the highest nucleotide identity (99.2%) with the CMV ZM isolate. Phylogenetic analysis showed that CMV-BH belonged to subgroup IA and that the most closely-related isolate was CMV-ZM. All test plants used for the biological assay were successfully infected with CMV and exhibited CMV disease symptoms such as blistering, mosaic, and vein yellowing. To our knowledge, this is the first report of CMV infection in P. heterophylla from Korea.

Differences in Habitat Structure and Herpetofauna Populations Caused by Thinning (간벌에 의한 서식지 구조와 양서·파충류 개체군의 특성 차이)

  • This study was conducted to clarify the differences in habitat structure and amphibian and reptile (herpetofauna) populations between before and after thinning in the coniferous and deciduous forests from June to September 2014 and 2015 at the Mt. Maewha of Hongcheon, Gangwon province, Korea. Forest structure was dramatically changed after thinning. Coverage of understory and mid-story were decreased while volume and number of downed trees were increased after thinning. We used line transect sampling method on a total of 4 transect lines, 2 lines in each forest type. 119 individuals of 5 amphibian species and 5 individuals of 3 reptile species were recorded. Mean number of amphibian species were decreased after thiining in the coniferous forest. There was no observed reptiles after thinning in the coniferous and deciduous forests. Detailed monitoring with different area, frequency and intensity might be needed to clarify more precise differences characterisitics of herpetofauna by thinning.

Chromosome numbers of eight taxa of Aconitum L. in Korea and their systematic significance (Ranunculaceae)

  • Various aneuploidy and polyploidy have been reported in the genus Aconitum L. (ca. 300 species worldwide, Ranunculaceae), and there is a demonstrated association between major lineage diversification and polyploidy. This study reports chromosome counts of eight Aconitum from Korea, including the first counts for A. japonicum Thunb. subsp. napiforme ($H. L{\acute{e}}v.$ & Vaniot) Kadota (2n = 32) and A. longecassidatum Nakai (2n = 16). The study also includes chromosome numbers for two taxa on the Critically Endangered species list in Korea. Among Korean native species, chromosome numbers in Aconitum subgenus Aconitum range from 2n = 16 to 2n = 64 with diverse levels of polyploidy (2x, 4x, and 8x), whereas Aconitum subg. Lycoctonum exhibits only diploids (2n = 16). Greater chromosome number diversity in subg. Aconitum than subg. Lycoctonum might explain higher species diversity within the former subgenus (more than 250 species worldwide). Investigating chromosome number diversity of Aconitum in a phylogenetic framework will be a critical step to understand species richness of the genus.

Ecological resilience of soil oribatid mite communities after the fire disturbance

  • This study investigated the impact of the fire disturbance and the pattern of recovery of soil dwelling oribatid mite communities with respect to the resilience from the fire disturbance. Oribatid mites are important decomposer animals of plant debris in soil with the feeding habits of saprophagy and mycophagy. Massive wild fire reduced soil oribatid mite abundance and diversity. The impact varied relative to the intensity of the disturbance. The proportion of the species common to the non-disturbed natural site increased as the time after the disturbance elapsed, which implying some degree of naturalness occurring in reorganization phase of the oribatid mite community. From the sites with different degree of fire impact, we found higher diversity in intermediately disturbed sites than in severely disturbed or non-disturbed site, supporting the intermediate disturbance hypothesis. Also this study showed that with differential degree of disturbance plots, resilience pattern after the disturbance can be explored even with shorter period research relative to the ecological succession of community.

The Development of Ecological Planting Model for the Make Up of Coastal Windbreak Forest on Suncheon Bay in Suncheon-si, Korea (순천만 해안방풍림 조성을 위한 생태학적 식재모델 개발)

  • This study was carried out to the development of ecological planting model to make up of coastal windbreak forest on the Suncheon-bay in Sucheon-si, Korea. Make up of coastal windbreak forest in this site was needed for appropriate bioresource, biodiversity and ecological structure, and for conservation of the eco-tour resource and protection of human life and property by the unforeseen disaster from the coast. Based on the plant-social principle, the planting model of windbreak forest was developed to facilitate growth of trees, considering planting locations. The ecological planting model for the coastal windbreak was composed of warm temperate evergreen and windbreak forest which is spreading around the inland area in Korea. The horizontal forest style was composed of forest edge community and inner forest community, and the vertical forest style was composed of upper, middle, low and ground planting class. The target of the present model was quasi-natural forest, and the species of tree were selected based on the adaptability to surroundings depending on a goal to create a forest and forest style. To achieve both functions of wind break forest and visual effect in short period of time, small trees and seedlings were planted with high-density of 40,000/ha in an expectation of easy natural maintenance in the future. The significance of the present study is a suggestion for a guideline to create ecological coastal windbreak forest in the Suncheon-bay in which the harmony of human life and the ecological conservation is of great importance. Also, the ecological coastal windbreak forest model should be developed further through the long term monitoring after construction of forest.

Floristic study of Mt. Cheongtae area (Pyeongchang-gun, Gangwon-do) (청태산 지역(강원도 평창군)의 식물상)

  • The vascular flora in the Mt. Cheongtae area, located Pyeongchang-gun in Gangwon-do, were surveyed 14 times from April of 2008 to October of 2011. The result of this survey revealed 663 taxa in total, comprising 89 families, 342 genera, 577 species, 6 subspecies, 73 varieties and 7 forms. In the flora of this area, 19 taxa were Korean endemic plants and 24 taxa were rare and endangered plants of Korea. The floristic regional indicator plants found in this area were 136 taxa comprising 4 taxa of grade V, 16 taxa of grade IV, 31 taxa of grade III, 41 taxa of grade II and 44 taxa of grade I. Naturalized plants in Korea numbered37 taxa. In addition, 485 taxa were categorized by usage into 8 groups, as follows: 240 edible, 8 fiber, 177 medical, 60 ornamental, 245 pasturing, 2 industrial, 12 dyeing and 16 timer plants.

An Analysis on the Structural Changes of Rural Land Use According to Urbanization (도시화에 따른 농촌토지이용구조변화 분석)

  • This study aims to show how the urbanization of Korea has progressed for the last three decades, what its characteristics are, and how rural land use has changed by the national and district(cities and counties) level. The land use changes accompanying to the urbanization is analyzed through 3 indicators such as urbanization rate, the rate of cultivated and forest land and the rate of urbanized area. The statistical data are 30 years from 1976 to 2005 for time series analysis by the national level, and are for the two years of 1995 and 2005 by the district level. The relationship between urbanization and land use changes in the national level is analyzed using statistical analysis(Correlation Analysis). In order to analyze the dynamic and spatial urbanization and land use changes effectively in the district level, Z-score, Paired T-test, Correlation Analysis, Analysis of Variance and Chi-squire Test are used. The results show negative correlation between urbanization rate and the rate of cultivated and forest land, and positive correlation between urbanization rate and the rate of urbanized area respectively. In the aspect of the change of urbanization rate, four categories are examined. In addition, four types are characterized on the basis of the rate of cultivated and forest land and the rate of urbanized area between 1995 from 2005.
