Effects of Medium Supplements on Seed-Derived Callus Culture and Regeneration of Orchardgrass (오차드그래스의 종자유래 캘러스배양 및 재분화에 미치는 배지첨가물질의 영향)

  • In order to optimize tissue culture conditions for genetic transformation of orchardgrass (Dactylis glomerata L.), the effects of culture medium supplements on tissue culture responses were investigated with mature seeds of a cultivar, 'Roughrider', as explant tissues. The optimal concentration of 2,4-D for the induction of embryogenic callus from mature seeds was 3 mg/L. Plant regeneration frequency was 36.3% when embryogenic calli were cultured on the regeneration medium supplemented with 1mg/L 2,4-D and 3mg/L BA. Addition of 1 g/L casein hydrolysate and 300 mg/L L-proline improved frequencies of embryogenic callus induction and plant regeneration up to 57.3 and 60.7%, respectively. Supplementation of the media with 10 mga $\textrm{AgNO}_3$ and 40 mg/L cysteine enhanced frequencies of callus induction and plant regeneration. Efficient regeneration system established in this study will be useful for molecular breeding of orchardgrass through genetic transformation.

Vetch Effects for the Low-input No-till Direct-Seeding Rice-Vetch Cropping System

  • A field experiment was carried out to observe the weed control effects of vetch and to evaluate vetch characteristics on clay loam soil in no-till direct-sown rice-vetch cropping systems. The effects of weed control, forage productivity, and N content of vetch plants were investigated. With the progress of plant growth, density of Chinese milk vetch (Astragalus sinicus L.) gradually decreased, but densities of foxtail and other weeds decreased steeply due to the depression by the over-shaded vetch canopy in a no-till direct-sown rice-vetch cropping paddy field. The vetch density in tillage systems was lower than in notillage cropping systems. Lower vetch density occurred with an increase in foxtail density and other weeds. Weed control effect increased by the progress of vetch growth, which indicated that the vetch canopy over-shaded the weeds. Vetch straw was degraded rapidly submergence after with water at the time of wet sowing of rice. Early harvesting of vetch seed resulted in lower seed germination. To acquire enough seedlings without re-sowing, the harvesting of seed should be delayed at least 28 days after the flowering stage in order to ensure the vetch population is sustainable in a no-till direct-sown rice-vetch cropping system. In order to improve the survival of vetch plants, vetch seeds should germinate from the heading .stage to before the full-ripening stage of rice plants. To enhance the percent of over-wintering survival, vetch seeds should germinate no later than the end of October in southern Korea. The dry weight of vetch plants increased with the progress of vetch growth until the flowering stage but N content decreased for 30 days from before the flowering stage (2.9%) until the ripening stage (1.8%). We concluded that Chinese milk vetch could have an effect on weed control before the flowering stage, sustainability without re-sowing of seed annually, and effective green manure for rice pre-crop in no-till direct-sown rice-vetch relay cropping systems.

Effects of Dried Whole Crop Barley Treated with Cellulolytic Microorganisms on In Vitro Fermentation Characteristics in Swine (섬유소 분해균을 이용한 건조 청보리 발효사료가 돼지의 In vitro 발효 특성에 미치는 영향)

  • The experiment was conducted to observe the effects of dried whole crop barley treated with cellulolytic microorganisms (Aspergillus niger KCCM 60357 and Bacillus licheniformis KCCM 40934) on the chemical composition, in vitro colonic fermentation and whole tract digestibility in swine. Whole crop barley were fermented with no microorganism addition (control), A. niger, B. licheniformis and co-culture of A. niger and B. licheniformis (Mixture) for 3 days at $30^{\circ}C$. In the feed chemical composition, CP contents of whole crop barley treated with A. niger (7.52%) and B. licheniformis (7.77%) were significantly higher than control (6.81%) (p<0.05). The in vitro colonic fermentation of dried whole crop barley fermented with control showed significantly higher $CH_4$ contents than A. niger, B. licheniformis and Mixture at 18h incubation (p<0.05). Dry matter (DM) digestibilities of A. niger (55%) and Mixture (57.42%) treatments were significantly higher than control (43.74%) (p<0.05). Ammonia-N was significantly increased in A. niger, B. licheniformis and Mixture relative to control at 24 hour incubation (p<0.05). Xylanase activities in A. niger, B. licheniformis and Mixture treatments were significantly higher than control at 24 hour incubation (p<0.05). Concentrations of total VFA were significantly increased in B. licheniformis (12.61 mM) at 24hour incubation (p<0.05). In vitro whole tract digestibility was significantly increased in B. licheniformis (49.61%) compared with the control (45.65%) (p<0.05). In conclusion, whole crop barley treated with cellulolytic microorganisms improved whole tract digestibility and colonic fermentation for swine.

Evaluation of Fermentation Ability of Microbes for Whole Crop Rice Silage Inoculant (총체 벼 사일리지용 미생물의 발효능력 평가)

  • This experiment was conducted to study on the evaluation of fermentation ability of microbes for whole crop rice silage Inoculant at National Institute of Animal Science, RDA from 2004 to 2005. We collected 28 strains of microbes from whole crop rice silage. According to acidity and growth ability, 5 strains of microbes was isolated (R4-1, R7-1, R7-2, R10-1, R12-1). The cultures of 4 strains were identified to be Lactobacillus plantarum (R4-1, R7-1, R7-2 and R10-1) and one was identified to be Lactobacillus pentosus (R12-1). Whole crop rice was harvested at the yellow ripen stage. It was ensiled in experimental silos (20ℓ capacity) with or without microbial additives (R4-1, R7-1, R7-2, R10-1, R12-1 and three commercial inoculant) and stored at room temperature for 60d. The pH value and acetic acid content of additivetreated silages were lower and lactic acid content was higher than those of the control (p<0.05). There was a trend for acetic acid content to be lowest and lactic acid to be highest in R7-1 treated silage. Crude protein (CP) contents of R7-2 treated silage was higher and acid detergent fiber (ADF) and neutral detergent fiber (NDF) contents of R7-1 treated silage was lower (p<0.05). Although some strains of inoculant could improve silage quality, L. plantarum R7-1 was more effective as an inoculant for whole crop rice silage. This microbe was named NLRI 401 and registered in the Korea Agricultural Culture Collection.

Influences of Feeding Seleniferous Whole Crop Barley Silage on Growth Performance and Blood Characteristics in Growing Hanwoo Steers (셀레늄함유 청보리 사일리지급여가 육성기 거세한우의 생산성 및 혈액성상에 미치는 영향)

  • This study was conducted to investigate effects of feeding seleniferous whole crop barley silage (WCBS) on growth performance and blood characteristics in growing Hanwoo steers. A total of 20 growing Hanwoo steers, initially weighing on average 208.8 kg of body weight, were used and treatments were consisted of 1) controls : 0.1 mg/kg Se, 2) T1 : 0.3 mg/kg Se, 3) T2 : 0.9 mg/kg Se by combining seleniferous and/or non-seleniferous WCBS, and 4) T3 : 0.9 mg/kg Se of inorganic Se treatment. Five steers were allocated to each treatment, and the trial was lasted for 90 days. All experimental diets were included to 30% of combination of seleniferous and/or non-seleniferous WCBS, and in T3 diet, sodium selenite that corresponds to 0.9 mg/kg Se was added to control diets. Also, the diets were isonitrogenous and isocaloric among treatments. Dietary level and type of selenium did not affect feed intakes and daily gain, and blood glucose concentration was significantly (p<0.05) lower for controls than T1 and T2 treatments. Blood total lipid concentration was significantly (p<0.05) decreased with increasing levels of dietary selenium, and also that of T2 and T3 was significantly (p<0.05) lower than controls. LDL-cholesterol concentration was significantly (p<0.05) lower for treatments including dietary selenium than controls, and also blood triglyceride concentration was significantly (p<0.05) lower for T2 than controls. Overall, it was tended that feeding seleniferous WCBS or inorganic Se increased blood IgG concentration, and in the same dietary selenium levels, T2 treatment was higher for IgG than T3 group (p>0.05). Blood selenium concentration was significantly (p<0.05) increased by feeding increasing levels of seleniferous WCBS, but there was no significant difference between controls and T3 group. These results showed that feeding seleniferous WCBS to growing Hanwoo steers was responsible for saccharide and lipid metabolism, and in particular, it reduced their total lipid and blood LDL-cholesterol concentrations. Furthermore, selenium present in seleniferous WCBS rather than inorganic selenium was better available in intestinal absorption, and it might help to enhance immunity in growing and younger stages of Hanwoo steers.

Effect of Nitrogen Top Dressing Levels on Productivity, Feed Value, and Anthocyanin Content of Colored Barley (질소 추비량이 유색 보리의 생산성, 사료가치 및 안토시아닌 함량에 미치는 영향)

  • his experiment was carried out to evaluate the effects of nitrogen top dressing levels on the growth, feed value, and anthocyanin content for developing functional feed of colored barley. A colored barley cultivar, Boanchalbori, was tested in this experiment. Nitrogen top dressing levels was six (0, 20, 40, 60, 80, 100%) and top dressing time was a regeneration time. In case of productivity, heading data was get behind and dry matter rate was significant decreased with higher nitrogen top dressing levels (p<0.05). Fresh yield, dry matter yield and TDN yield was increased with higher nitrogen top dressing levels, but not significant. In case of feed value, crude protein content was significant increased with higher nitrogen top dressing levels (p<0.05), and higher in the order of spike, whole and leaf culme of the plant. Percent NDF and ADF was decreased with higher nitrogen top dressing levels in leaf culme, but no difference in spike and whole (p<0.05), and higher in the order of leaf culme, whole and spike of the plant. TDN was increased with higher nitrogen top dressing levels in leaf culme, but no difference in spike and whole (p<0.05), and higher in the order of spike, whole and leaf culme of the plant. Total anthocyanin content was significant decreased with higher nitrogen top dressing levels in leaf culme and whole (p<0.05), and higher in the order of leaf culme, whole and spike of the plant. Specially, cyanidin-3-glucoside (C3G), delphinidin (Del), malvidin-3-glucoside (M3G) and malvidin (Mal) show a significant decrease. So there are an accumulation of anthocyanin in the culm, and standard nitrogen top dressing levels dressing on the regeneration time for produces high anthocyanin content of the colored barley.

The Effect of Feeding Whole-crop Barley Silage and Grass Hay on Productivity and Economic Efficiency in Elk (Cervus canadensis) (청보리 사일리지 및 목건초 급여가 엘크 사슴의 생산성 및 경제성에 미치는 영향)

  • This study was conducted to determine the effect of dietary whole-crop barley silage on the daily feed intake, daily weight gain, velvet antler yield and economic efficacy in elk during growth. A total of 21 elk (average BW 303 kg, 5 years old) were allotted randomly to three treatments. The three treatments were T1 (hay diet), T2 (whole-crop barley silage), and T3 (50% hay + 50% whole-crop barley silage). The average daily weight gain of T3 was higher than the others throughout the entire experimental period. The average daily feed intake of elk in decreasing order was as follows; T1>T3>T2. The velvet antler yield was higher in T3 group (8,585 g) compared with T1 (8,037 g) and T2 (7,713 g). However, there were no significant differences in the average daily feed intake, average daily weight gain, or velvet antler yield (p>0.05). In economic efficacy, T3 was gained about 29~43% more value than T1 or T2. In conclusion, the mixed feeding of 50% hay and 50% whole-crop barley silage was more effective than feeding hay of barley alone, in terms of average daily weight gain, velvet antler yield, and economic efficacy in elk.

Effect of Lactic Acid Bacteria and Formic Acid on the Silage Quality of Whole Crop Rice at Different Maturity (유산균 및 개미산 첨가가 수확시기별 벼 사일리지의 발효 품질 및 사료성분에 미치는 영향)

  • Silage additives are needed to increase the quality of whole crop rice silage which seldom produce without the additives due to both high pH and butyric acid concentrations. Little information, however, is available about the silage fermentation of whole crop rice added with silage additives in Korea. This study was conducted to determine the optimum levels of silage additives by evaluating the effects of latic acid bacteria (LAB) and formic acid concentrations on the silage quality of whole crop rice harvested at different mature stages. Field study was established early in May until October 7th on a rice field at Yupori, Sinbuk-yeup, Chunchon, Kangwon-Do. "Ilpum" mutant rice was harvested at six different mature stages; booting stage (17 Aug.), milk-ripe stage (27 Aug.), dough stage (7 Sep.), yellow ripe stage (17 Sep.), dead ripe stage (27 Sep.) and full ripe stage (7 Oct.). Each sample was ensiled in three different ways; with 1) LAB (0.05, 0.1 and 0.2% of sample wt), 2) formic acids (0.2, 0.3 and 0.4% of sample wt.) and 3) no additive. The additive levels did not affect dry matter content, crude protein, fiber and total digestable neutriant concentrations at all stages. Addition of additives significantly decreased the silage pH and butyric acid concentrations which tended to be more decreased with higher levels of additives. Latic acid concentrations were higher with the use of additives, especially with LAB. The lower concentrations of ammonia-N were observed in additive treatments at all stages, but the concentrations of ammonia-N did not differ according to the additve levels after yellow ripe stage (0.69, 0.60 and 0.71% of DM in 0.05, 01 and 0.2% of LAB, respectively; 0.64 0.59 and 0.75% of DM in 0.2, 0.3 and 0.4% of formic acid, respectively). These results indicate that the optimum addition levels of LAB and formic acid are 0.5∼0.1% and 0.2∼0.3%, respectively, on which the high quality of rice whole crop silage was produced. produced.

Antioxidant Activities and Quality Characteristics of Germinated Rough Rice Tea according to Roasting Temperature, Time and Leaching Condition (볶음 조건 및 침출 조건에 따른 발아 벼차의 항산화 활성 및 품질 특성)

  • To verify the possibility of manufacturing a germinated rough rice tea, germinated rough rice was roasted at 200, 220, and $250^{\circ}C$ for 10, 20, and 30 min. The treated rice powder was then put into tea bags and leached for 1, 3, and 5 min, after which their antioxidant, physicochemical, and sensory characteristics were compared. The total polyphenol content and 2,2'-Azino-bis-(3-ethyl benzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities of the germinated rough rice tea increased as the roasting temperature increased as well as in response to increased roasting and leaching times. Furthermore, the greater the roasting temperature, roasting time, and leaching time, the greater the increase in total soluble solid contents. Moreover, the turbidity and browning index of the germinated rice tea rose as the roasting temperature, roasting time, and leaching time increased. Additionally, the pH tended to decrease as the roasting temperature, roasting time, and leaching time increased. Evaluation of the sensory characteristics of the germinated rice tea revealed that the formation of a Maillard reaction product in the course of heating the rice added a unique flavor, which led to increase preference for the color, flavor, and taste, and therefore, the overall preference.
