Inhibitory Effects of Soyasaponins on Antigen-induced Degranulation in RBL-2H3 Cells (Soyasaponin의 RBL-2H3 세포에서 탈과립화 억제 효과)

  • Soyasaponins are glycosylated, which gives rise to a wide diversity of structures and functions. We evaluated for inhibitory effects of 4 soyasaponins on antigen-induced degranulation in RBL-2H3 Cells. 4 soyasaponins had shown dose-dependently inhibited histamine and ${\beta}$-hexosaminidase.

A report of 42 unrecorded bacterial species belonging to the Alphaproteobacteria in Korea

  • As a subset study to discover indigenous prokaryotic species in Korea, a total of 42 bacterial strains assigned to the class Alphaproteobacteria were isolated from diverse environmental habitats including plant roots, ginseng soil, forest soil, marsh, mud flat, freshwater, and seawater. From the high 16S rRNA gene sequence similarity (>99.1%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 42 species have been described in Korea; therefore 4 species of 1 genera in the order Caulobacterales, 18 species of 10 genera in the order Rhizobiales, 7 species of 5 genera in the order Sphingomonadales and 13 species of 11 genera in the order Rhodobacterales within the Alphaproteobacteria are reported for alphaproteobacterial species found in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are also described in the species description section.

Composition and Diversity of Gut Bacteria Associated with the Eri Silk Moth, Samia ricini, (Lepidoptera: Saturniidae) as Revealed by Culture-Dependent and Metagenomics Analysis

  • The polyphagous eri silk moth, Samia ricini, is associated with various symbiotic gut bacteria believed to provide several benefits to the host. The larvae of S. ricini were subjected to isolation of gut bacteria using culture-dependent 16S rRNA generic characterization, metagenomics analysis and qualitative enzymatic assays. Sixty culturable aerobic gut bacterial isolates comprising Firmicutes (54%) and Proteobacteria (46%); and twelve culturable facultative anaerobic bacteria comprising Proteobacteria (92%) and Firmicutes (8%) were identified inhabiting the gut of S. ricini. The results of metagenomics analysis revealed the presence of a diverse community of both culturable and un-culturable gut bacteria belonging to Proteobacteria (60%) and Firmicutes (20%) associated with seven orders. An analysis of the results of culturable isolation indicates that these bacterial isolates inhabited all the three compartments of the gut. Investigation on persistence of bacteria coupled with metagenomics analysis of the fifth instar suggested that bacteria persist in the gut across the different instar stages. In addition, enzymatic assays indicated that 48 and 75% of culturable aerobic, and 75% of anaerobic gut bacterial isolates had cellulolytic, lipolytic and nitrate reductase activities, thus suggesting that they may be involved in food digestion and nutritional provision to the host. These bacterial isolates may be good sources for profiling novel genes and biomolecules for biotechnological application.

A report of 17 unrecorded bacterial species of Korea belonging to the phylum Bacteroidetes

  • While screening indigenous prokaryotic species in Republic of Korea in 2017, a total of 17 bacterial strains assigned to the phylum Bacteroidetes were isolated from a variety of environmental habitats including water of fountain, tidal flat, plant root, soil, the gut of Russian grayling butterfly, ginseng field, seawater, lagoon and seashore sand. From the 16S rRNA gene sequence similarity of more than 98.7% and the formation of a robust phylogenetic clade with the closest species, it was found that the 17 strains belong to independent and recognized bacterial species. There has been no official report that the identified 17 species have been previously isolated in the Republic of Korea. Thus, 15 species in 10 genera of one family in the order Flavobacteriales, one species in one genus of one family in the order Cytophagales, and one species in one genus of one family in the order Sphingobacteriales are proposed as unrecorded species of the phylum Bacteroidetes found in the Republic of Korea. Gram reaction, colony and cell morphology, basic phenotypic characteristics, isolation source, taxonomic status, strain ID and other information are described in the species descriptions.

Variation of Caffeic acid, Rosmarinic acid, Luteolin and Apigenin Contents in Perilla Germplasm

  • Perilla frutescens, which belonging to the Labiatae family, is widely cultivated oil crop and have been used traditional herbal medicine in East Asia such as Korea, China, and Japan. Especially, the leaves and the seeds of this species are important in Korean traditional cooking, as one of the popular garnish and food colorants. Numerous studies have revealed that the beneficial health effects of perilla are due to its several phytochemicals contents, such as rosmarinic acid, caffeic acid, luteolin, and apigenin. For this reason, increasing the content of phytochemicals in perilla hasbecome a major breeding objective. The genetic diversity of the rosmarinic acid, caffeic acid, luteolin, and apigenin content in perilla seed is poorly documented. We analyzed the rosmarinic acid, caffeic acid, luteolin, and apigenin content of 203 accessions of perilla germplasm by high performance liquid chromatography (HPLC). The rosmarinic acid and luteolin contents ofgermplasms were ranged from $15.7{\mu}g/g$ to $2717.1{\mu}g/g$ and from $1.6{\mu}g/g$ to $582.4{\mu}g/g$ respectively.

Analysis of Gene Expression in Larval Fat Body of Plutella Xylostella Under High Temperature (고온에서 배추좀나방 유충 지방체의 유전자 발현 변화 분석)

  • BACKGROUND: Insects are ectothermic organisms in terrestrial ecosystems and play various roles such as controlling plant biomass and maintaining species diversity. Because insects are ectothermic, their physiological responses are very sensitive to environmental temperature which determines survival and distribution of insect population and that affects climate change. This study aimed to identification of genes contributing to fitness under high temperature. METHODS AND RESULTS: To identify genes contributing to fitness under high temperature, the transcriptomes of fat body in Plutella xyostella larva have been analyzed via next generation sequencing. From the fat body transcriptomes, structure-related proteins, heat shock proteins, antioxidant enzymes and detoxification proteins were identified. Genes encoding proteins such as structural proteins (cuticular proteins, chitin synthase and actin), stress-related protein (cytochrome P450), heat shock protein and antioxidant enzyme (catalase) were up-regulated at high temperature. In contrast expression of glutathione S transferase was down-regulated. CONCLUSION: Identifications of temperature-specific up- or down-regulated genes can be useful for detecting temperature adaptation and understanding physiological responses in insect pests.

Effect of Agricultural Land Use on Abundance, Community Structure and Biodiversity of Epigeic Arthropods (농경지의 이용형태가 토양성 절지동물 군집 및 다양성에 미치는 영향)

  • BACKGROUND: Epigeic arthropods participate in ecological functions as predators, decomposers and herbivores. The purpose of this study was to investigate the responses of some dominant arthropods in rice fields to different forms of agricultural land management. METHODS AND RESULTS: The abundance of microarthropods was compared between rice fields and uplands in the non-growing season. Collembola, Oribatida and Mesostigmata were more abundant in the upland fields than in the paddy fields. The community composition and diversity of epigeic arthropods were compared between fallow and rice fields. The total abundance and species richness of spiders and ground beetles were not significantly different in the two types of agricultural fields. The abundance of Arctosa kwangreungensis was greater in fallow fields than in cultivated fields. The community structure of arthropods was compared between paddy fields with and without barley. The cropping system altered the community composition of spiders but not their biodiversity. Barley cultivation increased the abundance of ground beetles but decreased that of spiders. We suggest that this contrast was partly due to the availability of plants that provided shelter and food for ground beetles. CONCLUSION: These results show that soil use intensity and cropping system alter the community composition of epigeic spiders and ground beetles. This could result in ecosystem-level alterations with respect to the control of pests and weeds. Our results also suggest that biodiversity of ground-dwelling arthropods may not increase during short fallow periods.

  • There are more and more citizens suffering from severe fatigue, and they wish to escape from it and spend their leisure time for healing. As a result, buildings and complexes are being constructed nationwide with healing as their theme. Particularly, they tend to build facilities with concepts like a spa, beauty, healing, meditation, nature, or forest healing. The purpose of this study is to examine the concept of healing environment and the nationwide tendencies of building facilities with healing as their theme and also investigate the planning characteristics of complexes and architecture with three representative complexes as examples. Complexes intended for healing have immersion into nature being freed from one's routine as their concept. When planning the flow of human traffic within the complexes, they try to obtain the autonomy of choice as well as the diversity of space and experiential factors in order to provide opportunities for experiencing nature. In the complexes selected for a case study here, they have planned the factors of physical environment that are associated with one another based on architectural education programs using red clay, programs specializing poetry, and healing programs using food. Typically, this is centered around outdoor experiential space, indoor meditation and education space, or fitness space. Also, it is characterized by the planning of physical environment and the complex operation of programs. Particularly, public space is divided into communal space, resting space, and health and treatment space, and health/resting space is mainly intended for health and exercise, for example, fitness, spas, or jjimjilbang (Korean dry saunas). Also, it is characterized by the planning of pitched roofs harmonized with nature and also facade planning that can positively adopt the factors of natural environment.

Critical analysis on discourse of Single-Person Households in Korean newspapers: Implications to family studies (일인가구를 둘러싼 신문기사분석 : 가족학 연구에 관한 시사점을 중심으로)

  • Objectives: The purpose of this study is to critically review the recent discourse on single-person households (SPH) that appeared in Korean newspapers. Specifically, we analyzed articles in five major daily newspapers, which are representative of the media in Korean society, and examined the types of titles, subjects, and contents of articles in which SPH were mentioned. Method: For this purpose, 538 articles on SPH or "living alone" that appeared in the five major Korean newspapers during 2011-2017 were analyzed using content analysis methods. Results: Our analysis revealed eight major topics(living arrangement and household characteristics; financial instruments; food, clothing, and housing culture; policy; welfare; relationships; safety; values and attitudes). The results show that there is a surge of articles portraying neutral images of SPH in Korean newspapers, thereby emphasizing the recent growth in the number of SPH and the information relating to this population group. Although negative images of SPH continue to exist in the discourse, in recent times we found these attitudes to have become less prevalent compared to neutral images or even positive discourse. We also found that the discourse on these topics and arguments can be divided into four themes-the diversity of SPHs, the solo economy for single households, safety issues, and social relationships. Conclusions: Theoretical and policy implications on family studies relating to SPH are discussed.

Analysis of Plant Species in Elementary School Textbooks in South Korea

  • Background and objective: This study was conducted to find out the status of plant utilization in the current textbooks by analyzing the plants by grade and subject in the national textbooks for all elementary school grades in the 2015 revised curriculum in Korea. Methods: The data collected was analyzed using Microsoft Office Excel to obtain the frequency and ratio of collected plant data and SPSS for Windows 26.0 to determine learning content areas by grade and the R program was used to visualize the learning content areas. Results: A total of 232 species of plants were presented 1,047 times in the national textbooks. Based on an analysis of the plants presented by grade, the species that continued to increase in the lower grades tended to decrease in the fifth and sixth grades, the upper grades of elementary school. As for the number and frequency of plant species by subject, Korean Language had the highest number and frequency of plant species. The types of presentation of plants in textbooks were mainly text, followed by illustrations and photos of plants, which were largely used in first grade textbooks. In addition, as for the area of learning contents in which plants are used, in the lower grades, plants were used in the linguistic domain, and in the upper grades, in the botanical and environmental domains of the natural sciences. Herbaceous plants were presented more than woody plants, and according to an analysis of the plants based on the classification of crops, horticultural crops were presented the most, followed by food crops. Out of horticultural crops, flowering plants were found the most diversity with 63 species, but the plants that appeared most frequently were fruit trees that are commonly encountered in real life. Conclusion: As a result of this study, various plant species were included in elementary school textbooks, but most of them were horticultural crops encountered in real life depending on their use. Nevertheless, plant species with high frequency have continued a similar trend of frequency from the previous curriculums. Therefore, in the next curriculum, plant learning materials should be reflected according to social changes and students' preference for plants.
