국제표준화기구 기준에 의한 의료기기용 하이드로겔의 세포독성 평가 (Cytotoxicity Evaluation on Hydrogels for Medical Devices based on the International Organization for Standardization)

  • Hydrogels for medical devices such as hydrophilic dressing, moisturizing healing band, hydrophilic intravenous catheter and soft contact lens were evaluated for their cytotoxicity according to the International Organization for Standardization (ISO) procedures. To test indirect cytotoxicity of hydrogel products, dissolution medium and dissolution condition were selected based on the guideline for medical devices. Cytotoxicity was low in all the case of hydrogel products. Soft contact lens showed no significant difference in dissolution between complete medium and saline. Currently, there is no specific guidelineto test hydrogel for medical devices in Korea with consideration of characteristics of hydrogel. Thus, proper method of cytotoxicity evaluation should be selected depending on the characteristics and usages of hydrogels for medical devices.

Micronutrients as Supportive Care for Gastrointestinal Cancer Patients; Benefits and Concerns

  • Cancer is the first leading cause of death in Korea and the second leading cause of death in the USA. There is extensive research into prevention of cancer and the support of oncology patients with diet or dietary supplements. In vitro and in vivo animal studies have indicated that antioxidants, including beta-carotene, alpha-tocopherol, and ascorbic acid, can yield anti-cancer effects in addition to providing protection against oxidative damage. Although many observational studies have shown that consuming fruits and vegetables can reduce the risk of some cancers, the results of several large-scale human intervention trials testing the benefits of a single or combined higher-dose of individual micronutrients have been inconsistent. Cancer can cause profound metabolic and physiological changes which may affect patients' nutrient requirements. Although the optimal route of nutrient delivery is through diet, cancer patients often suffer symptoms that disrupt their food intake, including anorexia, premature satiety, altered taste and smell, and changes in bowel mobility. In particular, micronutrient deficits can slow postoperative healing, contribute to depression symptoms, and decrease immune competence. Cancer patients are generally motivated to take dietary supplements to improve responses to treatment and quality of life. The Physician's Health Study II (PHS II) randomized controlled trial reported recently that daily multivitamin supplementation significantly, albeit modestly, reduced the risk of total cancer. Although evidence of multivitamin use benefits is limited in cancer patients, taking dietary supplements with constituents in the range of the recommended daily allowance according to the Dietary Reference Intake (DRI) recommendation is generally considered to be safe.

Physiochemical and Organoleptic Properties of Kefir Containing Different Concentrations of Hyaluronic Acid : A Preliminary Study

  • Recently, hyaluronic acid, which is the most popular among various biomaterials, has been used in osteoarthritis treatment, eye surgery, and wound healing. This study was carried out to determine the physicochemical and organoleptic properties of kefir containing various concentrations of hyaluronic acid. The pH value of hyaluronic-acid-containing kefir ranged from 3.58 to 3.46. Moreover, for hyaluronic-acid-containing kefir, the score of taste ranged from 4.75 to 4.0, the flavor ranged from 4.0 to 3.38, the color ranged from 4.38 to 4.0, the texture ranged from 4.38 to 3.75, and the overall acceptability ranged from 4.59 to 3.72. Overall, the organoleptic properties of hyaluronic-acid-containing kefir were better than those of the control. Because this is the first study involving hyaluronic acid and kefir, the results are valuable. The addition of hyaluronic acid did not significantly affect the physicochemical and organoleptic properties of kefir. Therefore, the commercial availability of various hyaluronic-acid-added dairy food products must be investigated.

매복 하악 제3대구치 발치와에 Atelo-collagen Sponge 삽입이 제2대구치 예후에 미치는 영향 (THE EFFECTS OF ATELO-COLLAGEN SPONGE INSERTION ON THE PERIODONTAL HEALING OF SECOND MOLARS AFTER IMPACTED MANDIBULAR THIRD MOLAR EXTRACTION)

  • Extracellular matrix(ECM) is known to function as a reservoir of endogenous growth factors, can be an effective delivery system of growth factor that easily lost bioactivity in solution. Fibrillar collagens like type I collagen, are the major constituent of the ECM and structural protein of bone. Also, it can be a scaffold for osteoblast migration. The purpose of this study was to compare the effects of absorbable Atelo-collagen Sponge($Teruplug^{(R)}$) insertion in tooth extraction sites on periodontal healing of the mandibular second molar after the extraction of the impacted third molar. The study population comprised 31 cases who had been scheduled for surgical removal of impacted mandibular third molars. All patients were in good general health and were not using any medication that would influence wound healing after surgery. In 15 cases control group, none was inserted into the tooth extraction site. In 16 cases experimental groups, $Teruplug^{(R)}$ was inserted into the tooth extraction site. We evaluated tooth mobility, pocket depth, gingival margin level preoperatively and 1 week, 2 weeks, 4 weeks, and 3 months postoperatively. The change was compared with two groups using Mann-Whitney test. The results were as follows. 1. There was no significant change of tooth mobility on both groups. 2. There was tendency of decreasing of previous pocket depth causing tooth extraction on both groups. 3. On gingival margin level, there was various change according to initial swelling and loss of attachment on both groups. 4. There was tendency of decreasing of gingival margin level on both groups because of removal of inflammation and decreasing of previous pocket depth. 5. There was large change of pocket depth on buccal middle, distal, lingual distal area because of tooth extraction and bone reduction. Compared with the control group and experimental group, we observed significant difference during some periods. The results of this study suggest that absorbable atelo-collagen sponge($Teruplug^{(R)}$) is relatively favorable bone void filler with prevention of tissue collapse, food packing and enhance periodontal healing.

The purpose of this study is to estimate the effects of absorbable atelo-collagen sponge (Teruplug$^{(R)}$, Terumo biomaterials Co., Tokyo, Japan) insertion in tooth extraction sites on periodontal healing of the second molar, healing of the fractured mandibular bone and new bone formation of third molar socket after the extraction of the impacted third molar with mandibular angle fracture. Methods: In our study of six cases of mandibular angle fractures, all of them underwent the extraction of the third molar tooth & absorbable atelo-collagen sponge insertion in tooth extraction site. Three of them had a intraoral infection & oral opening to fracture site, two of the six had dental caries, and only one had reduction problem due to third molar position. Six consecutive patients with noncomminuted fractures of the mandibular angle were treated by open reduction and internal fixation using one noncompression miniplates and screws placed through a transoral incision. Results: All of the patients have showed good postoperative functions and have not experienced complications requiring second surgical intervention. There was well healing of the mandibular bone and the most new bone formation of third molar socket after the extraction of the impacted third molar with mandibular angle fracture. Conclusion: The results of this study suggest that absorbable atelo-collagen sponge is relatively favorable bone void filler with prevention of tissue collapse, food packing, and enhance periodontal healing. Thus, the use of atelo-collagen sponge and one noncompression miniplate seems to be relatively easy, safe, and effective for the treatment of fractures of the mandibular angle and third molar extraction.

인간 섬유육종세포에서 비쑥 추출물과 유기용매 분획물의 암전이 억제 효과 (Anti-invasive Effect of Artemisia scoparia Halophyte Extract and its Solvent-partitioned Fractions in Human Fibrosarcoma Cells)

  • 염생식물인 비쑥은 식용이 가능한 약용식물로서 살충, 항염, 항콜레스테롤, 해열, 항균 활성 등이 알려져 있다. 본 연구에서는 phorbol-12-myristate-13-acetate (PMA)로 자극된 인간 섬유육종 HT-1080 세포에서 5가지 활성검색방법 즉 : gelatin zymography, MMPs ELISA, wound healing assay, reverse transcription-polymerase chain reaction (RT-PCR), Western blot을 이용하여 비쑥의 조추출물과 그 용매 분획물의 MMP-2와 MMP-9 효소 활성에 대한 저해 효과를 평가하였다. 비쑥 시료들을 메틸렌 클로라이드(MC)와 메탄올(MeOH)로 각각 2번 추출하여 합한 것을 조추출물로 사용하였으며 이 조추출물은 MMP-2와 MMP-9 효소활성에 대해 유의한 억제 효과를 나타내었다. 이 조추출물은 용매극성에 따라 다시 n-hexane, 85% aq.MeOH, n-butanol 및 물 분획층으로 분획되었으며 이렇게 얻어진 4개의 용매 분획물들중에 n-hexane과 85% aq.MeOH 분획이 gelatin zymography와 MMP ELISA assay에서 MMP-2와 MMP-9의 활성을 효과적으로 억제하였다. 또한 wound healing assay, RT-PCR 및 Western blot assay에서 H2O 분획을 제외한 모든 용매 분획물들이 세포 이동, 그리고 MMP-2 및 MMP-9의 mRNA와 단백질 발현을 유의하게 억제하였다.

산림치유 프로그램 개발을 위한 프로그램 선호도 분석 (The Analysis of Program Preferences for the Development of Forest Therapy Program)

  • 본 연구의 목적은 산림치유 프로그램 개발을 위한 기초자료를 제공하는데 있다. 본 연구는 탐색적 연구로 산림치유 잠재수요자의 산림치유 프로그램 선호도가 무엇인지 파악하게 함으로써, 산림치유 프로그램을 개발하고 운영하는데 중요한 기초자료가 될 것이라고 생각된다. 일반 성인 남 여 620명을 대상으로 산림치유 프로그램의 선호도에 관한 설문조사를 실시하였다. 자료분석은 SPSS 21.0 프로그램을 사용하여 빈도분석(Frequency analysis), T검정(T-test), 일원분산분석(One-way ANOVA)을 통해 성별, 연령, 직업에 따른 산림치유 프로그램 선호도의 차이를 살펴보았다. 산림치유 프로그램 선호도가 성별, 연령, 직업 등에 따라 다양하다는 것을 알 수 있었다. 첫째, 성별에 따라, 남성은 여성에 비해 숲속 운동회, 캠핑을 선호하고, 여성은 남성에 비해 전반적인 프로그램에서 선호도가 높은 것으로 나타났다. 둘째, 연령에 따라서 다른 연령과 비교해서 20대는 스트레스 평가와 진단, 음식 만들기, 숲속 사진치료 등, 30대는 자유롭게 숲속 걷기, 상담, 강의 듣기(스트레스, 대인관계 관련), 비전 세우기 등, 40대는 명상, 경관보기, 삼림욕, 풍욕, 일광욕, 맨발로 숲속 걷기 등, 50대는 호흡법 호흡체조, 요가, 등산, 묵언 산행, 자연음식 먹기 등의 프로그램을 각각 특히 선호하는 것으로 나타났다. 셋째, 직업에 따른 산림치유 프로그램 선호도를 살펴보면 다음과 같다. 다른 직업과 비교해서 학생은 스트레스 평가와 진단, 음식 만들기 등을, 교사는 자유롭게 숲속걷기, 등산, 숲속 독서, 경관보기, 삼림욕 풍욕 일광욕, 캠핑 등을, 주부는 요가, 물 흐르는 소리 듣기, 허브차 마시기, 자연음식 먹기 등을, 전문 연구직은 호흡법 호흡체조, 묵언산행, 명상, 숲속 잠자기 등을 각각 선호하는 것으로 나타났다. 본 연구의 분석 결과가 성인을 대상으로 하는 산림치유 프로그램 개발을 위한 기초자료로 활용되기를 기대한다.

Inhibitory Effects of (-) Epigallocatechin Gallate and Quercetin on High Glucose-induced Endothelial Cytotoxicity

  • Functional damage to microvascular endothelial cells by hyperglycemia is thought to be one of the critical risk factor.; in the impaired wound healing seen with diabetes mellitus. It is also thought that oxidative stress plays a significant role in this endothelial cell dysfunction. The present study examined the differential effects of flavonoids on endothelial cell dysfunction under high glucose conditions. Human endothelial cells exposed to 30 mmol/L glucose for 7 d were pre-treated with various flavonoids and pulse-treated with 0.2 mmol/L $H_2O_2$ for 30 min. High glucose markedly decreased cell viability with elevated oxidant generation and nuclear condensation. $H_2O_2$ insult exacerbated endothelial cytotoxicity due to chronic exposure to high glucose. (-)Epigallocatechin gallate and quercetin improved glucose-induced cell damage with the disappearnnce of apoptotic bodies, whereas apigenin intensified the glucose cytotoxicity. In addition, cell viability data revealed that these flavonoids of (-)epigallocatechin gallate and quercetin substantially attenuated both high glucose- and $H_2O_2$- induced dual endothelial damage. These results suggest that (-)epigallocatechin gallate and quercetin may be beneficial agents for improving endothelial cell dysfunction induced by high glucose and may prevent or reduce the development of diabetic vascular complications.

LC-MS/MS를 이용한 식품 중 천속단과 한속단 지표성분 동시분석 (Simultaneous determination of Phlomis umbrosa and Dipsacus asperoides in foods using LC-MS/MS methods)

  • 천속단과 한속단은 명칭이 유사하여 '속단'으로 통칭하여 판매되고 있으나, 종과 속이 서로 다른 약재일 뿐만 아니라 함유 성분이 다르므로 주의가 필요하다. 천속단은 골격강화, 골절치료 작용이 있는 한약재로 식품원료로는 사용할 수 없으나, 한속단의 뿌리와 잎은 식품원료로 사용이 가능하다. 본 연구에서는 식품 중 천속단 혼입여부 판별을 위하여 LC-MS/MS를 이용한 동시분석법을 개발하였으며, 선택성, 직선성, 검출한계, 정량한계, 정확성, 정밀성을 검토하였다. 국내 유통중인 식품 중 어린이 키성장을 표방하는 제품 17건을 구매하여 천속단 혼입여부에 대한 실태조사를 하였다. 분석결과 17건의 시료 중 5건의 시료에서 천속단이 검출되었으며, 천속단 지표성분의 함량은 loganin $0.19-14.45{\mu}g/mL$, sweroside $0.13-4.61{\mu}g/mL$, akebia saponin D $0.59-19.29{\mu}g/mL$이었다. 본 연구에서 개발된 동시분석법은 식품 중 불법 혼입된 천속단을 간단하면서도 신속하게 판별하는데 도움이 될 것으로 사료된다.

두부가 인체에 미치는 영양학적 고찰 (The Nutritional Aspect of Tofu)

  • 일반적으로 두부는 만드는 방법이나 두부의 성질에 따라 다양한 형태의 두부가 있다. 두부의 효능으로는 혈중 지질, 지단백, 콜레스테롤 등의 농도를 감소시켜 동맥경화와 뇌혈관을 깨끗하게 하여 심뇌혈관계 질환을 예방하는 효과가 있다. 또한 장내 유용균의 증식을 촉진하는 역할을 하며 콜레스테롤 배설을 촉진시키고, 장 기능에 대한 생리적 효과와 식사 후 혈당 상승 억제와 인슐린 분비를 억제하는 효과가 있다. 그리고 두부에는 칼슘이 풍부해 뼈 손상을 막아주고, 뼈 조직의 생성에 도움을 주는 효과를 가지고 있어 골다공증 예방 및 치료에 효과가 있다. 반면, 빈혈이 있는 사람의 경우 두부 섭취 시 주의해야 한다. 콩에 들어있는 phytic acid라는 물질은 아연 및 철분과 같은 무기물의 흡수를 저해하기 때문에 철 결핍성 빈혈 환자는 더 심한 철분 부족이라는 결과를 초래할 수 있으므로 섭취 시 주의해야 한다. 따라서 건강식품으로 선호되고 있는 두부의 부족한 부분을 보완하기 위해 다양한 천연 첨가제를 활용한 추가적인 연구가 필요할 것으로 생각된다.
