Effects of Calcium Ion Concentration on Starch Hydrolysis of Barley ${\alpha}$-Amylase Isozymes

  • Barley ${\alpha}$-amylase genes, amy1 and amy2, were separately cloned into the expression vector of $pPICZ{\alpha}A$ and recombinant Pichia strains were established by homologous recombination. Both AMYs from Pichia shared almost identical hydrolysis patterns on short maltooligosaccharides to result in glucose, maltose, or maltotriose. Against insoluble blue starch, AMY1 showed the highest activity at 0.1-5 mM calcium concentration, whereas 15-20 mM was optimal for AMY2. On the hydrolysis of soluble starch, unexpectedly, there was no significant difference between AMYs with increase of calcium. However, the relative activity on various starch substrates was significantly different between AMYs, which supports that the isozymes are clearly distinguished from each other on the basis of their unique preferences for substrates.

  • In this study, the DongPieHong Ditch were taken as the research object, five sampling points were set to measure the COD, NH3-N,TNandTPindexes. The correlation and principal component analysis were used to judge the main pollution sources and calculate pollution contribution rate. According to the population in the basin, the load of point source pollution into the river was estimated. As a result, the load of COD, NH3-N and TP into the river was 323.04t/a, 43.8t/a and 3.9t/a, respectively. According to the statistics of the rainfall in the basin, the concentrations of COD, TP and NH3-N in the initial rainwater were measured and calculated for non-point source pollution, and the results shown that the inflow loads of COD, NH3-N and TP into the river were 34.59t/a, 0.12t/a and 0.71t/a, respectively. It was found that the main cause of the pollution in the east flash flood gully was point source pollution, and the proportions of COD, NH3-N and TP into the river were 90.33%, 99.72% and 84.61%, respectively.

A Study on the Job Satisfactions of School Food Service Employees (학교 급식 종사자의 직무만족도에 관한 연구)

  • The objective of this study is to identify factors affecting the job satisfaction of cook(n= 173) employed in school food service and to propose a plan to improve school foodservice quality. The subjects were cook employed by school in Chungchon province, Gwangju area. The average of cook's job satisfaction was 3.06, the rank factor for nature of work(4.25). the co-workers(3.28), the organic environment(3.17), promotion welfare payment(2.31), the physical environment(2.29) shown respectively. The highest satisfaction with nature of work and organic environment was indicated in the areas of the group of higher age, and cooks who received college education showed significantly lower job satisfaction in physical environment. The highest satisfaction for promotion welfare payment, and physical environment shown in the group of higher length of service who receive higher wages. The groups of regular job shown higher result in the nature of work(3.46>2.94), organic environment(3.46>2.94), promotion welfare payment(2.88>1.94) comparing to irregular job. It is necessary to elevate cook's low satisfaction for improving their work efficiency and the quality of elementary school foodservice.

Determination of 4-Methylimidazole and 2-Acetyl-4(5)-tetrahydroxybutylimidazole in Caramel Color and Processed Foods by LC-MS/MS

  • In this study, the quick HPLC-MS/MS method for the simultaneous separation of 2-acetyl-4(5)-tetrahydroxybuthylimidazole (THI) and 4-(5)-methylimidazole (4-MI) in alkaline medium was used for caramel color and processed foods in Korea. After a simple sample pretreatment, 51 4-MI-labeled samples were positive for 4-MI and 2 also contained THI. The concentration of 4-MI was 260.5~24, $499.3{\mu}g/kg$ in caramel color, less than $LOD{\sim}1,712.5{\mu}g/kg$ in sauce, 1,242.3, $5,972.2{\mu}g/kg$ in balsamic vinegar, $2,118.3{\sim}5,802.4{\mu}g/kg$ in complex seasoning, $82.7{\sim}5,110.6{\mu}g/kg$ in curry, and $29.9{\sim}464.4{\mu}g/kg$ in soft drinks. The recovery rate of 4-MI was 97.1~111.0% in sauce and 81.9~110.0% in powder and that of THI was 83.6{\sim}106.4% in sauce and 61.2{\sim}99.4% in powder. Our results concluded a safe amount of 4-MI and THI compared to the limit of Korea additive code but the processed foods do not have a limit of caramel color and 4-MI in Korea. Therefore, research and monitoring of 4-MI and THI is needed for processed foods in Korea.

  • Organizations should constantly modify and refine the mechanism by which they achieve their purposes- no only by scanning the environmental change and uncertainty, but also by choosing the right strategies. As scanning largely perceived environment, the management need to maintain a systemized perspective on their environment in order to make profitable decisions(strategic choice). Accordingly, this article presents a theoretical framework that top managers or managers of food service organizations can use to analyze their environment as an integrated and systemized whole - a model that take into account the interrelationships among domains of environment and strategic choice. Also, the results of this study will be a guideline for further expanded environmental scanning.

  • Cheap, low quality food products that are unhealthy are prevailing among children. These low quality products are stirring up the urge to purchase among children and so it is pointed out as a problem. In order to arrive at a solution for the indication of negligence on the side of manufacturing, processing, circulation, and marketing management, a survey asking about interests and preferences on insanitary food products was held during May 2, 2005 to June 30. The subjects were 540 students and 510 parents form 5 elementary schools located at Gwangjin Gu, Seoul including Ja-yang elementary school, etc. This study aims at arranging measures against insanitary food products and contribution to maintenance of children's health.

Analysis on Consumption Behaviors Regarding Environment-Friendly Agricultural Products According to the Food-Related Lifestyles of Housewives in Yeungnam Region (영남지역 주부의 식생활 라이프스타일에 따른 친환경농산물 소비행동 분석)

  • The purpose of this study was to examine the consumption behaviors regarding environment-friendly agricultural products by food-related lifestyles of housewives. The data were collected from 298 housewives living in Yeungnam region through a self-administered questionnaire during April, 2010. Frequencies, Cronbach's $\alpha$, factor analysis, cluster analysis, one-way analysis of variance, Duncan's multiple range test and chi-square test were conducted by SPSS Windows. The results obtained from this study were as follows. Lifestyles were categorized into five factors by factor analysis: healthy-seeking type, popularity-seeking type, convenience-seeking type, safety-seeking type and taste-seeking type. The respondents belonged to one of three groups by cluster analysis: popularity-seeking group, convenience-seeking group, and wellbeing-seeking group. The main reason given for purchasing environment-friendly agricultural products was safety. The respondents were satisfied with the safety of environment-friendly agricultural products the most. Many respondents purchased environment-friendly agricultural products at large discount markets, and received information about them from mass media. There were significant differences among three groups in the place of purchase environment-friendly agricultural products, information source for environment-friendly agricultural products, most important factor when purchasing environment-friendly agricultural products, the level of satisfaction with environment-friendly agricultural products and the purchase rate of environment-friendly agricultural products among the monthly food expenses.

Antimicrobial Activity against Food-hazardous Microorganisms, Dermatophytes, and Pytopathogens and Antioxidative Activity of Sancho Oil (식품위해성균, 피부사상균 및 식물성 병원균에 대한 산초유의 항균 활성 및 항산화 활성)

  • Background: Although Sancho (Zanthoxylum schinifolium Siebold & Zucc) oil has traditionally been used for its antibiotics properties, there is currently a lack of scientific evidence regarding its biological activities. In this study, we investigated the antimicrobial and antioxidant activities of Sancho oil against food-hazardous microorganisms, phytopathogens, and dermatophytes. Methods and Results: We investiated the antimicrobial activity of Sancho oil against 11 food-hazardous microorganisms, nine phytopathogens, and six dermatophytes. The Sancho oil was found to show the strongest antibacterial activity against Shigella flexneri and Listeria spp. Sancho oil also showed high antifungal activity against plant pathogens, particularly Fusarium oxysporum, and showed antimicrobial activity against dermatophytes such as Trichophyton rubrum, Microsporum canis and Candida albicans. The antioxidant activity of Sancho oil was measured using the DPPH method, and was found to be stronger than that of unrefined oil. Moreover, this activity increased with increasing oil concentration. Conclusions: We found that Sancho oil showed differing antimicrobial activities against food-hazardous microorganisms, dermatophytes, and plant pathogens. The antimicrobial activity spectrum of Sancho oil was not broad and varied among microbial strains. On the basis of our findings, we consider that Sancho oil could be used an antibacterial material for food-borne S. flexneri and Listeria spp., a biopesticide for Fusarium spp., and a treatment for dermatophytes such as T. rubrum.

Investigation of microbial contamination and use of food additives for pet foods in Gwangju, Korea (광주지역 내 유통 중인 반려동물 사료·간식의 미생물 오염도 및 식품첨가물 사용실태 조사)

  • This study was conducted for safety evaluation on 130 pet food products, which are distributed in Gwangju, South Korea. The microbial contamination part and the usage of food additives part were mainly investigated. The five microorganisms that we tested were total viable cell counts (TVC), Coliforms, Salmonella spp., Campylobacter spp., pathogenic Escherichia coli and there were 15 products that exceed the microbial criteria or detected food poisoning bacteria. Specifically, Coliforms (13 products, 10%), TVC (9 products, 6.9%), Salmonella spp. (2 products, 1.5%), and E. coli (2 products, 1.5%) were followed. On the other hand, food additives such as preservatives, antioxidants and sodium nitrite were detected in 61 products. Among the preservatives, sorbic acid and benzoic acid were detected in 58 (44.6%) products. In antioxidants, Butylated hydroxytoluene (BHT) was detected in 3 (2.3%) products. In addition, preservatives and antioxidants were detected in 8 of 20 products labeled as 'additive-free'. Microbial contamination tended to occur mainly in small-scale individual homemade feed stores, while food additives were all detected in pet shops and supermarkets. Currently, the criteria for microorganisms and food additives for pet foods are insufficient in Korea. So, it is necessary to establish detailed feed standards and specifications for companion animals.
