Saururus chinenesis Extracts Scavenge Reactive Oxygen Species and Modulate Nitric Oxide Production in Raw 264.7 Macrophages

  • Saururus chinensis Baill has been used in Korean folk medicine for the treatment of various diseases such as edema, Jaundice, and furuncle. The components of this plant were extracted into four fraction. Among the four fraction, hexane and ethyl acetate fraction were highly toxic to 3T3 mouse embryo fibroblast and Raw 264.7 mouse macrophage, but n-butanol and residue fraction did not show any toxic effect to those cell lines. n-Butanol and residue fraction exhibited antioxidant effects on hydro-gen peroxide, hydroxyl radical, and superoxide anion directly in vitro and in the 3T3 fibroblasts. All the four fractions inhibited lipid peroxidation measured by thiobarbituric acid reactive substances (TBARS) formation. In addition, n-butanol and residue fraction showed inhibitory effects on lipopolysaccharide (LPS)-induced nitric oxide production, and also down-regulated inducible nitric oxide synthase (iNOS) mRNA transcription 6 h after LPS stimulation in Raw 204.7 cells. Only n-butanol fraction, which mainly consists of flavonoids, inhibited NF-kB activation by decreasing IkBa degradation 90 min after LPS stimulation. horn the results, it is suggested that this plant could be a good candidate material for drug development based on its antioxidant and/or anti-inflammatory constituents.

Sweet Basil (Ocimum basilicum): much more than a condiment

  • Ocimum basilicum belongs to the family Lamiaceae and can be found in tropical Asia, Africa, Central America and South America. It is widely cultivated commercially as a condiment or for the production of essential oil. In this review, this plant is accredited with important effects on health, in addition to being a condiment. In folk medicine it is used to treat vomiting, intestinal colic and diarrhea, menstrual pains and to improve kidney function. Some studies have demonstrated that it can be used for the treatment of hyperlipidemia and also that it has antioxidant, neuroprotective, anti-inflammatory and vasodilator properties as well as hepatoprotective effects. In view of its broad range of effects, further studies should be conducted to ascertain the possible use of this plant in clinical trials, not only for prevention but also as an adjuvant in the treatment of different diseases.

무궁화나무의 성분 및 생물활성에 관한 연구(I) (Studies on the Constituents of Hibiscus syriacus (I))

  • Hibiscus syriacus L. (Malvaceae) is widely distributed over Korean, China, India and Siberia. The dried flower of Hibiscus syriacus is used as a folk medicine for curing of hematochezia, dysentery, obstruction due to wind-phlegm, regurgitation, and vomiting of food, and the dried root bark is used antipyretic, anthelmintic and antifungal agents. From a chloroform extract of root bark of this plant, compound I, II, and III were isolated and the structures were elucidated by various spectroscopic analyses. These compounds were identified as syringaresinol. E-N-feruloyltyramine, and Z-N-feruloyltyramine, respectively and were isolated from this plant for the first time. Compound II and III exhibited lipid peroxidation inhibitory activities with $IC_{50}$ of 15.5 and 28.6 ${\mu}g/ml$, respectively.

$C_{18}$ and $C_{19}$ Quassinoids from Eurycoma Longifolia

  • Two new 1,2-seco-1-nor-6((5 $\mapsto$ 10)abeo-picrasan-2,5-olideskeleton quassinoids, eurylactones A [1] and B [2], and two new $C_{19}$ skeleton quassinoids 3 and 4 were isolated from Eurycoma longifolia together with two known $C_{18}$ quassinoids 5 and 6. Their sttuctures were established by the spectral evidence.Many quassinoids have already been isolated (1-3) from the roots, wood, and leaves of Eurycoma longiolia Jack (Simaroubaceae) and have been used as a folk medicine insoutheast Asia. As part of a series of studies, we have further undertaken the chemicalinvestigation of the n-BuOH-soluble fraction of the MeOH extract and isolated two new1,2-seco-1-nor-6(5 $\mapsto$ 10)abeo-picrasane skeleton quassinoids 1 and 2, which have beenknown only in shinjulactone B (4,5) and yadanfiolide (6), two new $C_{19}$-type quasslnolds3 and 4, and two known C18-type quassinoids 5 and 6. In this paper, the structureelucidation and biosenetic relation of these compounds are described.

일엽초 추출물의 항산화 및 항암 효과 (Antioxidant and Anticancer Properties of the Extracts from Lepisorus thunbergianus (Kaulf.) Ching)

  • Lepisorus thunbergianus (Kaulf.) Ching has been used in folk medicine in Korea. In this study, a L. thunbergianus methanol extract and its fractions were investigated for their antioxidant properties. The results showed that the ethyl acetate and butanol fractions of L. thunbergianus possess potent DPPH radical scavenging activities. Both fractions also possessed reducing power and inhibited reactive oxygen species formation. In addition, the cytotoxic activity of the L. thunbergianus n-hexane fraction (HF) was investigated. The results suggested that the HF remarkably suppressed proliferation of human breast, liver and colon cancer cells. These results demonstrate, for the first time, that L. thunbergianus extract induces apoptosis in SW620 cells, suggesting that L. thunbergianus may have potential as a therapeutic agent for colon cancer.

  • Menthol (M) is a cyclic monoterpenode and is a major component of essential oils. Menthol, along with menthol, isomenton, etc., gives taste and odor of the mint plant, and when it comes to menthol in general, L- or (-) -menthol is usually used. Included in pharmaceuticals, cosmetics and pesticides. It has antimicrobial, antibacterial, antioxidant properties. It is also known that the licorice plant (Glycyrrhiza Glabra L.) differs from other types of plants by its medicinal properties. For many years it has been used in folk medicine. Extraction of licorice root revealed up to 25% glycyrrhizinic acid (GA). Its aglycone - glycyrrhizic acid is notable for its structural similarity to the adrenal cortex hormones. Currently, GA and glycyrrhizic acid are widely used in medicine as a remedy for colds, allergies, viral diseases, tumors. The biological activity of menthol and GA-based supramolecular compounds has been poorly studied, and their effect on the functional parameters of rat liver mitochondria has been studied little. For this purpose, in our experiments, the effect of menthol (M), glycyrrhizinic acid (GA) and their supramolecular complexes obtained in different proportions on in vitro and in vivo studies on rat liver mitochondria was studied.

Antioxidant Activity of Medicinal Plant Extracts Used as Folk Remedies by Diabetic Patients

  • The aim of this study was to investigate the antioxidant effect of medicinal plants used by diabetic patients. Fourteen medicinal plants were selected and antioxidant activity such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, inhibition effect of linoleic acid autoxidation and low density lipoprotein (LDL) oxidation, thiobarbituric acid (TBA) value were measured. The Cornus officinalis had DPPH radical scavenging activity of 84.79%, which was higher than the 78.79% for $\alpha$-tocopherol. Rosa rugosa Thunberg, Pueraria thundergiana Bentham, Artemisia princeps var. orientalis and Sasamorpha purpurascens Nakai also had high values. Extracts with higher DPPH radical scavenging activities had higher total phenol concentrations, and positive correlations between these parameters were found. Inhibitory activities of linoleic acid autoxidation, LDL oxidation and TBA value, used as indices of oxidative stress, were observed in most of the selected medicinal plants. The highest inhibitory activity for TBA value was observed in the extract of Pueraria thundergiana Bentham. Rosa rugosa Thunberg (75.50 %), Sasamorpha purpurascens Nakai (74.00 %), and Cornus officinalis (73.00 %) all had high antioxidant activity against linoleic acid autoxidation, similar to that of $\alpha$-tocopherol. The same 3 plants also had high in inhibitory activity against LDL oxidation. Therefore, we demonstrated that medicinal plants used as folk remedy by diabetic patients had antioxidant activity. Especially, Rosa rugosa Thunberg, Cornus officinalis, Pueraria thundergiana Bentham, Arlemisia princeps var. orientalis, and Sasamorpha purpurascens Nakai had high phenol concentrations which resulted in high values for DPPH radical scavenging. These same plants exhibited high values for inhibitory activities for TBA value, linoleic acid autoxidation and LDL oxidation, indices associated with lipid peroxidation.

선학초(仙鶴草) 추출액이 흰쥐의 생리활성(生理活性)에 미치는 효과 (Effect of Extract Agrimonsa Pilosa L. on biological Activity in Rats)

  • 1998년 8월부터 11월까지 선학초(仙鶴草) 전초(全草) 추출액(抽出液)을 처리수준 0%, 0.02%, 0.04%, 0.06% 및 0.08%로 하여 흰쥐(Rat)에 경구투여(經口投與)하여 성장(成長) 및 생리활성상태(生理活性狀態)를 조사한 결과는 다음과 같다. 1. 선학초(仙鶴草) 전초(全草) 추출액(抽出液)을 흰쥐에 농도별(濃度別)로 투여한 결과 선학초(仙鶴草) 추출액(抽出液) 급여시(給與時) 증체양(增體量)은 무급(無給) 여구(與區) 대비(對比) 0.02% 급여구(給與區)가 353.2g으로 5% 성장하였으나 0.04% 급여구(給與區)에서는 5% 감소하는 경향을 보였다. 2. 사두효솔면(飼枓效率面)에서는 무급여구(無給與區)와 0.02% 급여구(給與區)에서는 0.14로 비슷한 경향을 보였으나 다른 수준에서는 7%정도 낮아지는 경향을 보였다. 3. 백혈구(白血球) 수(數)는 무급여구(無給與區)에 비해 0.02% 급여구(給與區)를 제외한 전(全) 급여구(給與區)에서 급여수준(給與水準)이 증가할 수록 백혈구(白血球) 수(數)가 증가하는 경향을 보였다. 4. 혈소판(血小板) 수(數)는 0.06% 급여구(給與區)에서 무급여구(無給與區)에 비해 20% 증가하는 경향을 나타냈다.

  • 우리 나라 자생 짚신나물(Agrimonia pilosa Ledeb.) 수집종의 분포, 자생지의 환경 및 자생지에서의 형태적 특성조사를 실시하고, 자생종 짚신나물와 도입 짚신나물 간의 형태적 특성 및 유연관계를 연구한 결과는 다음과 같다. 1. 수집된 종 중에서 하동지역의 화개면 쌍계사와 청암면 청학동에서 수집된 2종은 다른 수집종에 비하여 유전적 거리지수가 가장 가까운 수치에서 나타나 근연관계로 나타났으며, 충북 영동읍 비탄재와 경기도 강화읍 수집종간에는 유전적 거리지수는 최대값이 같은 것으로 보아 유연관계가 있는 것으로 판단된다. 2. 경남거제에서 수집(No. 7)된 것과 충북 보은에서 수집(No. 21)된 2종이 유전적으로 가장 가깝게 나타나 지역적 차이에 의한 진화에도 다양함을 보이고 있어 수집지역의 확대와 이에 따른 체계적인 선발을 통해서 우량종의 선발도 가능할 것으로 판단된다 3.증폭된 DNA band pattern에 있어서 절편크기는 300∼21000p로 PCR로 증폭된 종 상호간의 1-F값은 평균 0.624였고, 최소값 0.365, 최대값 0.827이었으며, 2개 군으로 분리되어 유전적으로 상당한 차이가 있는 것으로 나타났다.

The effect of Saururus chinensis Baill against oxidative damage and inflammation

  • Objectives : ROS are involved in a wide spectrum of diseases including chronic inflammation and cancer. S.chinensis Baill, a perennial herb commonly called Chinese lizard's tail or Sam-baek-cho in Korea, is used for the treatment of edema and inflammatory diseases in the Oriental folk medicine. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the two extracts, water(WE) and ethyl acetate(EAE) from S.chinensis Baill. Methods : Anti-oxidant activity was evaluated using Fe2+ chelating and hydroxyl radical scavenging assay. DNA cleavage assay, and western blot and immunostaining for phospho-p65 were performed to evaluate anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell. Results : In Fe2+ chelating activity and hydroxyl radical scavenging activity, WE showed more strong scavenging activity for hydroxyl radical than EAE. WE scavenged hydroxyl radical by 12% at 3.2 ${\mu}g/ml$, 21% at 16 ${\mu}g/ml$, 32% at 80 ${\mu}g/ml$, 66% at 400 ${\mu}g/ml$ and 82% at 2000 ${\mu}g/ml$, respectively. In addition, WE showed more strong chelating activity than EAE. WE chelated Fe2+ ion by 1.1% at 3.2 ${\mu}g/ml$, 8.2% at 16 ${\mu}g/ml$, 26.3% at 80 ${\mu}g/ml$, 72% at 400 ${\mu}g/ml$ and 89% at 2000 ${\mu}g/ml$, respectively. Also, WE inhibited oxidative damage via its anti-oxidant activity. In anti-inflammatory effect, EAE inhibited NO production and iNOS expression. In addition EAE suppressed the NF-${\kappa}B$ and MAPK signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, these data indicate that S. chinensis Baill, shows anti-oxidant activity and anti-inflammatory effect.
