The Distribution of Barbiturates in Model Membranes of Total Lipids and Total Phospholipids Extracted from Brain Membranes

  • The distribution of barbiturates in the model membranes of total lipids (SPMVTL) and total phospholipids (SPMVPL) extracted from synaptosomal plasma membrane vesicles was determined by employing a fluorescent probe technique. The two fluorescent probes 2-(9-anthroyl)stearic acid and 12-(9-anthroyl)stearic acid were utilized as probes for the surface and the hydrocarbon interior of the outer monolayer of the SPMVTL and SPMVPL, respectively. The Stern-Volmer equation of fluorescent quenching was modified to calculate the relative distribution. The analysis of preferential quenching of these probes by barbiturates indicates that pentobarbital, hexobarbital, amobarbital and phenobarbital are predominantly distributed on the surface area, while thiopental sodium has an accessibility to the hydrocarbon interior of the outer monolayer of the SPMVTL and SPMVPL. From these results, it is strongly suggested that the more effective penetration into the hydrocarbon interior of the outer monolayer of the membrane lipid bilayer could result in a higher general anesthetic activity.

  • The Fluorescent 7-aminodipyrido[3,2-a:2',3'-c]phenazine (7-amino-dppz, 1) is functionalized as an europium ion ($Eu^{3+}$) sensor, which showed the effective emission quenching when europium cation is chelated to the bpy site of 1 compound. The complexation ratio indicated that the 1 compound forms a 1 : 1 complex with $Eu^{3+}$.

CdTe Quantum Dots as Fluorescent Probes for Josamycin Determination

  • A new method for the determination of josamycin has been developed based on quenching of the fluorescence of 3-mercaptopropionic acid-capped CdTe quantum dots (MPA-CdTe QDs) by josamycin in ethanol. Reaction time, interfering substances on the fluorescence quenching, and mechanism of the interaction of CdTe QDs with josamycin were investigated. Under optimum conditions, the relative fluorescence intensity was linearly proportional to the concentration of josamycin between 12.0 and 120.0 ${\mu}g\;mL^{-1}$ with a correlation coefficient of 0.9956 and a detection limit of 2.5 ${\mu}g\;mL^{-1}$. The proposed method was successfully applied to commercial tablets, and the results were satisfactory, i.e. consistent with those of high-performance liquid chromatography (HPLC).

Fluorescence Intensity Changes for Anthrylazacrown Ethers by Paramagnetic Metal Cations

  • Three anthrylazacrown ethers in which the anthracene fluorophore π system is separated from the electron donor atoms by one methylene group were synthesized, and their photophysical study was accomplished. These fluorescent compounds showed a maximum fluorescence intensity at pH=5 in aqueous solutions and a decrease in fluorescence intensity upon binding of paramagnetic metal cations (Mn 2+ (d 5 ), Co 2+ (d 7 ), Cu 2+ (d 9 )). The decrease in fluorescence intensity may be attributed to the paramagnetic effect of metal cations to deactivate the excited state by the nonradiative quenching process. The benzylic nitrogen was found to play an important role in changing fluorescence intensity. From the observed linear Stern-Volmer plot and the fluorescence lifetime independence of the presence of metal ions, it was inferred that the chelation enhanced fluorescence quenching (CHEQ) mechanism in the system is a ground state static quenching process. Enhanced fluorescence was also observed when an excess Na + ion was added to the quenched aqueous solution, and it was attributed to cation displacement of a complexed fluorescence quencher.

  • By using the general theoretical framework proposed recently for treating the fluorescence quenching kinetics, we investigate the effect of light pulse intensity on the decay of fluorescence which follows excitation of fluorophors by the light pulse of very short but finite duration. It is seen that conventional theory breaks down when the exciting light pulse has a pulse width comparable to the fluorescent lifetime and its intensity is very high. We also find that even when the light intensity is not too high, conventional theory may fail in either of the following cases: (i) when the quencher concentration is high, (ii) when there is an attractive potential of mean force between the fluorophor and quencher, or (iii) when the energy transfer from the fluorophor to the quencher may also occur at a distance, e.g., via dipole-dipole interaction. The validity of the predictions of the present theory may thus be tested by fluorescence quenching experiments performed under such situations.

  • 세포막에서의 지방산 이동은 매우 빠르게 일어나므로 방사성 원소를 사용해서는 여러가지 단점이 있고, 정확한 이동속도 측정에도 어려움이 많았다 최근에 개발된 FRET assay는 형광성 물질과 형광성 물질을 상쇄시키는 quencher를 사용한 실험방법 이다. 이는 공명 에너지 이동의 원리를 이용한 것으로 형광광도계, stopped-flow장치를 사용하여 소수성 물질 이동을 직접 컴퓨터 모니터로 측정하는 방법으로 기존방법의 단점을 보완하였다. Donor막에는 형광성 표지를 붙인 지방산이 들어 있고 acceptor막에는 형광을 흡수하는 물질이 들어 있어서 형광성 지방산이 donor에서 acceptor로 이동하면 형광도가 감소하며, 시간에 따른 형광도 감소를 측정하여 지방산 이동속도를 측정하는 방법이다. 형광성 표지를 이용하여 소수성 물질 이동에 사용되는 또 다른 방법은 self-Quenching assay이다. 형광 물질의 농도가 높아지면 서로 형광을 흡수하는 성질을 이용한 방법으로 주로 micelle에서의 물질 이동에 많이 쓰인다. Donor micelle에는 높은 농도의 형광성 지방산이 들어 있고 acceptor micelle에는 형광성 지방산이 들어 있지 않을 때 형광성 지방산이 donor에서 acceptor로 이동하면 형광도가 증가하게 되고 시간에 따른 형광도 증가를 측정하는 방법이다.

New 7-Hydroxycoumarin-Based Fluorescent Chemosensors for Zn(II) and Cd(II)

  • Five new 4- or 8-substituted-7-hydroxycoumarin derivatives (1-5) were synthesized as fluorescent sensors for metal ions. Fluorescent changes and selectivity for metal ions were compared based on the introduction of different ligands and/or testing with different substitution positions of 7-hydroxycoumarin in aqueous solution. Especially, probes 2, 3 and 5 displayed large fluorescence enhancements with $Zn^{2+}$ and $Cd^{2+}$. Probes 2 and 3 showed moderate selectivity for $Zn^{2+}$ over $Cd^{2+}$. On the other hand, probe 4 showed large fluorescence quenching effects upon the addition of $Ag^+$ and $Hg^{2+}$.

A Thermodynamic Study on the Interaction of Quinolone Antibiotics and DNA

  • Fluorescence of quinolones including norfloxacin, ciprofloxacin and S- and R-ofloxacin is quenched upon association with single and double-stranded DNA (ss- and ds-DNA). The ratios of fluorescence intensity in the presence of DNA to its absent were plotted with respect to the DNA concentration to construct the Stern-Volmer plot. The slope of the Stern-Volmer plot become larger as the temperature is lowered, ensuring that the fluorescence quenching is static process, i.e., the fluorescence is quenched by formation of the non-fluorescent complex between quinolone and DNA. In the static quenching mechanism, the quenching constant which is equivalent to the slope of the Stern-Volmer plot, is considered as the equilibrium constant for the association of quinolones and DNA. From the temperature-dependent equilibrium constant, ${\Delta}H^0\;and\;{\Delta}S^0$ was obtained using the van’t Hoff relation. In general, association of the quinolone with ds- as well as ss-DNA is energetically favorable (an exothermic) process while the entropy change was unfavorable. Due to the steric effect of the substituents, the effect of the quinolone ring is smaller on the ss-DNA compared to ds-DNA.

Synthesis of Dendrimer Containing Dialkylated-fluorene Unit as a Core Chromophore via Click Chemistry

  • The convergent synthetic strategy for the emissive dendrimers having the chromophore at core via the coppercatalyzed 1,3-dipolar cycloaddition reaction between alkyne and azide was described. 2,7-Diazido-9,9-dioctyl-9H-fluorene, designed to serve as the core in dendrimer, was stitched with the alkyne-functionalized Frechettype and PAMAM dendrons by the click chemistry leading to the formation of the corresponding fluorescent dendrimers in high yields. The preliminary photoluminescence studies indicated that 2,7-diazido-9,9-dioctyl-9H-fluorene showed no fluorescence due to the quenching effect from the electron-rich ${\alpha}$-nitrogen of the azido group but the dendrimers fluoresced due to the elimination of the quenching through the formation of the triazole ring.

Detection of Nitroaromatic Compounds Based on Fluorescent Silafluorene Chemosensors

  • A simple and rapid method is described for detecting nitroaromatic explosives in air or seawater with the use of photoluminescent organosilicon compounds. The synthesis, spectroscopic characterization, and fluorescence quenching efficiency of silafluorenes are reported. Silafluorenes were synthesized from the reduction of dilithiobiphenyl with dichlorosilanes. Two silafluorenes were used for the detection of nitroaromatic compounds. Detection of nitroaromatic molecules, such as 2,4-dinitrotoluene (DNT), 2,4,6-trinitrotoluene (TNT), and picric acid (PA), has been explored. A linear Stern-Volmer relationship was observed for the first three analytes. Fluorescence spectra of silafluorenes obtained in either toluene solutions or thin films displayed no shift in the maximum of the emission wavelength. The photoluminescence quenching occurs by a static mechanism.
