UV Spectrometric and DC Polarographic Studies on Apigenin and Luteolin

  • Remarks on polyphenolic compounds has been arisen since past few years. the flavonoids appears to be the important groups of compounds with their capability to inhibit DNA damage, lipid peroxidation, to quench free radicals and, at least, anticarcinogenic and antiproliferative effects. On the other hand, their mechanism of action is still unexplained. Apigenin and luteolin are the most wide-spread flavones and they exhibited to be useful in chemoprevention. UV spectrometric and DC polarographic studies on these two compounds have been carried out with regard to changing pH. The most significant changes were observed at basic pH. These results could aid to elucidation of their mechanism of action as pH is one of the important factors for bioprocesses passing in living organisms.

감귤 내 생리활성성분이 간암세포의 생존에 미치는 영향 (Effect of active ingredients in the Citrus fruits on the proliferation of hepatocellular carcinoma cells)

  • Previous studies have suggested that Citrus fruits might suppress the proliferation of various cancer cells. However, little is known about any specific ingredients in the extract of Citrus fruits to exert its anti-proliferative activity in cancer cells. The present study aimed to identify the active ingredients in Citrus fruits to suppress the proliferation of rat hepatocellular carcinoma cells. Among tested compounds, two polymethoxylated flavones (nobiletin and tangeritin) showed significant anti-proliferative activity whereas other compounds (synephrine, rutin, hesperidin) did not. Interestingly, nobiletin as well as tangeritin also decreased the protein amount of gluconeogenic enzymes, PEPCK and G6Pase. The possible involvement of gluconeogenic activity in the proliferation of hepatocellulacarcinoma cells are further to be investigated.

초석잠(Stachys sieboldii MIQ) 뿌리 추출물의 항산화 활성 고찰 (Antioxidant Activities of Stachys sieboldii MIQ Roots)

  • 약용 식물로서 그 효능이 알려져 있으나 다른 약용 작물에 비해 성분 연구가 알려져 있지 않은 초석잠에 대한 항산화 활성도에 시험하였다. 뿌리의 메탄올 추출물 100 mg을 메탄올 20 mL에 녹여 만든 샘플에서 total poplyphenols과 flavonoids가 각각 14.3 $\mug/mL$ (3.02%), 9.9$\mug/mL$ (1.97%)씩 함유되어 있음을 알 수 있으며 각 유기 용매층의 추출물의 항산화 활성도를 비교해 본 결과 DPPH법을 통해 기존의 항산화제인 $\alpha-tocopherol$ (21.8 $\mug$), BHT (100 $\mug$) 그리고 BHA (20.6 $\mug$)와 비교해서 에틸아세테이트(2.8 $\mu\textrm{g}$), 부탄올(10$\mug$_ 추출물이 보다 우수한 항산화능을 보였다. 과산화 지질 형성 억제능과 아질산염 소거능 조사에서는 에틸아세테이트의 추출물이 다른 용매 분획물과 비하여 가장 높은 활성도를 나타내었으며 기존의 항산화제들과 비슷한 효능을 나타내었다. 이를 실리카겔칼럼 크로마토그래피를 이용해 다시 분획하여 6개의 분획을 얻었으며 UV/VIS 흡광광도계를 통해 최대 홉광도의 파장은 280∼330 nm사이에 있음을 확인하였다. 그 중 최대 흡광도가 띠 I 이 325 nm, 띠 II가 289 nm 인 ES-R5에서 가장 높은 항산화 활성을 보였다. 이는 일반적으로 알려져 있는 phenolic compounds의 UV-VIS spectral data와 일치하며 특히 flavones의 UV- VIS spectral data와 유사한 경향을 보이고 있으므로 flavones 계통의 phenolic compounds가 초석잠(stachys sieboldii MIQ)속에 함유되어 있음을 알 수 있었다.향을 미치는 인자로서는 술전의 증상의 정도가 중요하다. 중증 근무력증으로 흠선절제술을 시행받은 환자들에 대한 중장기 추적 결과 술전에 비해 우수한 증상의 호전을 보였다.하는 중요한 역할을 담당하고 있다. 따라서 주부의 바람직한 식생활 행동과 건강상태는 자신뿐만 아니라 가족 구성원 개개인의 영양소 섭취상태에 직접, 간접적으로 큰 영향을 미칠 수 있으므로 이들에 대한 영양교육은 그 중요성이 크다고 볼 수 있다. 중요성이 크다고 볼 수 있다.였다. 이와 같은 영향이 계속된다면 동아시아 유황부족은 2005년에는 1.05백만 톤으로 증가될 것으로 예상된다. 그러나 중국에서 황화철에서부터 공급되는 유황함량이 많아 이 지역에서의 원소유황의 진정한 균형에 대해서는 정확하게 평가되지 못한다. 1995년 동아시아에서는 1.3백만 톤의 원소유황을 일본, 캐나다, 미국에서 수입했다. 이들 국가는 앞으로도 이 지역의 주요 공급자가 될 것이다. 황산의 많은 양은 일본에서 이 지역으로 수출되는데 그 양은 1995년 10만톤 이상에 이른다. 더욱이 경제?환경적 이점 때문에 중국이 황화물에서 회수하는 유황대신 원소 유황의 수입을 지속적으로 증가시키고 있어 지역내의 유황 부족이 증가 될 것이다. 이 같은 상황진전으로 앞으로 10년 이내에 2.5백만 톤의 추가시장이 있게 될 것이다. 이 기간내 한국으로서 현재 326,000톤의 부족에서 2005년에는 309,000톤의 과잉으로 유황균형이 변할 수 있는 주요계기가 될 것이다 이 같은 과잉은 회수 유황생산이 1995년 333,000톤에서 2005년 870,000톤으로 $161\%$가 증가될 것으로 예상된다. 동기간 내에 기타 유황생산은 280,000톤에서 320,000톤으로 $14\%$ 증가되는 것으로 추정된다. 그리하여 2005년 한국에서 유황 공급은 1.19백만 톤이고 수요는 881,000톤으로 추정 된다. 미래 한국에서 유황의 또 다른 잠재시장은 식물양분으로서

  • 본 연구는 PCOS 환자를 대상으로 MetS 여부에 따른 플라보노이드 섭취량의 차이를 분석하고 MetS과 플라보노이드 섭취와의 관련성을 알아보기 위해 수행되었다. 이 연구를 통해 관찰된 결과는 다음과 같다. 첫째, MetS이 없는 군에 비해 MetS이 있는 군은 총 6종의 플라보노이드군 중 플라보놀 (flavonols)을 유의적으로 적게 섭취하는 것으로 나타났다. 둘째, 간이식생활진단 (MDA)을 통한 식습관을 조사한 결과, '과일이나 과일주스를 매일 먹는다'라는 질문에 대해 '아닌 편이다'라고 응답한 비율이 MetS이 없는 군에 비해 MetS이 있는 군이 높았다. 셋째, 플라보놀 (flavonols)의 섭취량과 MetS의 교차비 (OR)를 분석한 결과, 모든 혼란변수를 보정한 후, 최하위 섭취군 대비 최상위 섭취군에서 MetS에 대한 교차비가 유의적으로 낮았다. 이상 PCOS 환자를 대상으로 MetS과 플라보노이드 섭취와의 관련성을 조사한 결과, MetS이 있는 군이 MetS이 없는 군에 비해 플라보놀 (flavonols)의 섭취량이 유의적으로 낮았고 플라보놀 (flavonols)의 섭취량은 MetS의 위험도와 역의 관계를 나타냈다.

Effects of Naturally Occurring Flavonoids on Inflammatory Responses and Their Action Mechanisms

  • Flavonoids are natural polyphenolic compounds widely distributed in plant kingdom. Although many flavonoids were found to show anti-inflammatory activity in vitro and in vivo, the potency of anti-inflammatory activity was not enough for a clinical trial. Thus, a search for finding potential flavonoid molecules is continuing. In this review, in vivo anti-inflammatory activity of various flavonoid derivatives is summarized mainly based on the results obtained in authors' laboratories. Among them, several biflavonoids such as amentoflavone and ginkgetin were found to possess anti-inflammatory activity on animal models of acute/chronic inflammation comparable to nonsteroidal and steroidal anti-inflammatory drugs currently used. In respect of their action mechanisms, the effects on arachidonic acid metabolism and nitric oxide production were described. Some flavonoids directly inhibit cyclooxygenase and/or lipoxygenase. Biflavones such as ochnaflavone and ginkgetin are inhibitors of phospholipase $A_2$. In recent studies, certain flavonoids were also found to suppress cyclooxygenase-2 and inducible nitric oxide synthase expression induced by inflammatory stimuli. Therefore, it is suggested that anti-inflammatory activity of the certain flavonoids (mainly flavones, flavonols and biflavonoids) may be mediated by direct inhibition of arachidonic acid metabolizing enzymes as well as suppression of the enzyme expression involved in inflammatory responses.

PMFs Analysis of Krachaidum Products by HPLC and GC

  • Polymethoxyflavones (PMFs) are a group of polymethoxylated bioactive flavones with diverse biological activities, including anticancer, anti-inflammatory, antibacterial, and antiviral activities. PMFs are found from various plants such as orange, tangerine, and krachaidum. To establish the simple quantitative analytical methods for PMFs, chromatographic analysis was applied to the selected krachaidum foods because krachaidum contains diverse PMFs compared to other PMF-containing foods. Krachaidum is the rhizome of Kaempferia parviflora, and many commercial krachaidum products, such as tea, juice and wine, are commercially available and consumed as health functional foods in Asian countries. Apart from the claimed health promoting benefits, reliable quality assurance and legal guideline for the registration of these products are not available yet. Twelve PMFs were analyzed from the commercial krachaidum foods by GC-FID and HPLC-DAD. No single chromatographic method could not analyze 12 PMFs simultaneously. HPLC-DAD method was found more sensitive to detect PMFs. Based on our analysis data, we proposed 5,7-dimethoxyflaone and 5,7,4'-trimethoxyflavone as index components for the food products.

Inhibition of Developmental Processes by Flavone in Caenorhabditis elegans and Its Application to the Pinewood Nematode, Bursaphelenchus xylophilus

  • Flavone (2-phenyl chromone) is a well-known plant flavonoid, but its bioactivity has been little explored. Treatment of Caenorhabditis elegans or C. brissage with flavones induced embryonic and larval lethality that was pronounced in early larval stages. This anti-nematodal effect was also observed in the pinewood nematode, B. xylophilus. $LD_{50}$ values were approximately $100{\mu}M$ for both B. xylophilus and C. elegans. Our results indicate that flavone is an active nematicidal compound that should be further investigated with the aim of developing a potent drug against B. xylophilus.

Docking Study of Biflavonoids, Allosteric Inhibitors of Protein Tyrosine Phosphatase 1B

  • Protein tyrosine phosphatase (PTP) 1B is the superfamily of PTPs and a negative regulator of multiple receptor tyrosine kinases (RTKs). Inhibition of protein tyrosine phosphatase 1B (PTP1B) has been proposed as a strategy for the treatment of type 2 diabetes and obesity. Recently, it has been reported that amentoflavone, a biflavonoid extracted from Selaginella tamariscina, inhibited PTP1B. In the present study, docking model between amentoflavone and PTP1B was determined using automated docking study. Based on this docking model and the interactions between the known inhibitors and PTP1B, we determined multiple pharmacophore maps which consisted of five features, two hydrogen bonding acceptors, two hydrogen bonding donors, and one lipophilic. Using receptor-oriented pharmacophore-based in silico screening, we searched the biflavonoid database including 40 naturally occurring biflavonoids. From these results, it can be proposed that two biflavonoids, sumaflavone and tetrahydroamentoflavone can be potent allosteric inhibitors, and the linkage at 5',8''-position of two flavones and a hydroxyl group at 4'-position are the critical factors for their allosteric inhibition. This study will be helpful to understand the mechanism of allosteric inhibition of PTP1B by biflavonoids and give insights to develop potent inhibitors of PTP1B.

Antimicrobial Effects of Flavone Analogues and Their Structure-Activity Relationships

  • It has been well known that the use of Saccharomyces cerevisiae can cause fungemia in critically ill patients and flavone shows an antimicrobial effect on S. cerevisiae. Therefore, we have investigated the activities of thirteen flavone analogues on S. cerevisiae in our studies. Because flavonoids including flavones have antioxidative effects, we try to carry out the activity studies of flavone analogues in vitro and in vivo. In addition, the relationships between the structures of flavone analogues and their biological activities, such as antimicrobial and antioxidative effects, were elucidated using Comparative Molecular Field Analysis calculations. Of the flavone analogues tested here, 3,2'-dihydroxyflavone showed both good antimicrobial and antioxidative activities.

한국인 상용식품의 플라보노이드 데이터베이스 구축 (Development of flavonoid database for commonly consumed foods by Koreans)

  • Flavonoids have been hypothesized to reduce the risk of chronic diseases, but the lack of a flavonoid database hampered epidemiological studies addressing this issue in Korea. In this study, we developed a flavonoid database, based on a systematic review. A total of 1549 food items containing flavonoids were selected using the Korean Nutrient Database. Among them, flavonoid contents for only 649 food items were evaluated with analytical values and the remaining 900 items were replaced with adaptations or calculations from similar items. The developed flavonoid database covered 93.2% of fruits and fruit juices, 76.1% of vegetables, 98.4% of legumes and legume products, and 85.0% of all plant foods overall (1,549 items) as reported by the 24-hr dietary recall method regarding the 2008 Korean National Health and Nutrition Examination Survey. We found that this flavonoid database, overall, included 95.6% of all mainly consumed plant foods by Koreans. This flavonoid database is expected to be useful in regards to the correlation study of flavonoid intake and chronic diseases.
