Fish Passage Evaluations in the Fishway Constructed on Seungchon Weir (승촌보에 설치된 어도에서 어류의 이동성 평가)

  • The objective of this study was to evaluate fish passage efficiency, based on fish-trap monitoring methodology, in the fishway of Seungchon Weir, which was constructed on the lower region of Yeongsan River in 2011. Seasonal patterns and diel variations of fish movements, fish fauna, and compositions in the fishway were analyzed in relation to the current velocity and location of the trap-setting in the fishway. For the analysis, we conducted seven times surveys in 2012 on the fishway and also conducted intensive monitoring of the fishway as 3 hrs interval for the diel variations of fish fauna and compositions in the fishway. According to the fish-trap monitoring methodology, the total number of fish species was 13 species, which was a 43.3% of the total. Most dominant fish used the fishway was Squalidus chankaensis tsuchigae and the relative abundance of the species used the fishway was 33.5% of the total. The season and time zone (in diel variation) observed most frequently in the fishway were July and 18:00-21:00 PM, respectively. The fish movements and use-rates of fishway varied depending on the locations of trap-setting; Fish biomass and the number of species were statistically (p < 0.05) greater in the most right or left-sided traps than in the mid-traps. Also, fish movements and use-rates of fishway were influenced by current velocity on the fishway; fish in the fishway preferred the low current velocity (mean 0.71 m/sec) than the high current velocity (mean 1.13 m/sec). Further long-term studies should be monitored for the efficiency evaluations of the fishway.

Attractants and Trap Development for Ussur Brown Katydid, Paratlanticus ussuriensis (Orthoptera : Tettigoniidae) (갈색여치의 유인물질 탐색과 트랩개발)

  • Recently, Ussur brown katydid outbreaks caused a serious pest problem in areas of Yeongdong, Chungbuk. This study was performed to control the pest with environment-friendly method. Trap was made of PET plastic bottles that easily found near farmhouse. Attractant materials such as oak (Quercus acutissima) leaf, fruits (peach, apple, grapefruit and pear) sarcocarp or its juices, rice wine (makgeolli) and fish meal were directly applied into the manufactured trap and investigated for the attraction efficacy compared with the funnel trap. During one day, manufactured trap (fish trap) attracted the Ussur brown katydid more than funnel trap. The efficacy of attractant materials were as follows: peach juice (32.7 adults) > rice wine+fish meal (31.3 adults) > rice wine (27.0 adults) > pear juice (19.0 adults) > apple juice (17.2 adults) > fish meal (16.7 adults) > grapefruit juice (14.4 adults) > oak leaf (2.3 adults). The attractive efficacy of fruit juices to ussur brown katydid was more than fruit carcocarps, and the trap hangover 1m in height more than that on ground. The composition of rice wine and fish meal prolonged its efficacy when treated with disinfectant.

  • The process of producing a fish trap frame is very complicated and manual, so it is expensive, the quality of the product is unstable, the quality is not guaranteed, the durability is weak, and it cannot be used for a long time. Therefore, we made a fish trap frame-making machine to reduce the manpower and costs and to make robust products. This machine cuts the wire of mild steel to a certain size and then makes the connecting parts into a trapezoid shape by spot welding. In this study, the weld point shear strength between manual and automatic operation was compared and analyzed.

Trap identification of the constitutive promoter-like sequences from the bacterial fish pathogen, as exemplified by Edwardsiella tarda

  • A trap identification system for isolating functional sequences to allow the constitutive expression of foreign protein from Edwardsiella tarda was developed. Using the green fluorescent protein (GFP) reporter-based trap system, various functional sequences to drive heterologous expression of the GFP were selectable in Escherichia coli host. However from the bioinformatic sequence analysis, all the segments predicted as regulatory regions were not native promoters actually existing upstream of endogenous E. tarda genes. Instead, a number of non-authentic sequences, possibly resulted from the random shuffling and/or intermolecular ligation were also proven to be able to display a potent GFP expression in the recombinant E. coli. Further analysis with selected clones showed that both authentic and non-authentic sequences could function in as a constitutive promoter, leading quite a consistent and stable GFP expression after repetitive subcultures. Microscopic examination also confirmed the uniform pattern of GFP expression in every host bacterium. Semi-quantitative assay of GFP showed that there was no clear relationship between expression levels and organizational features of the promoters trapped. Functional promoter-like elements achieved in the present study could be a good starting material for multivalent genetic engineering of E. tarda in order to produce recombinant vaccines in a cost-effective fashion.

Biomass estimation of fish in Samcheok marine ranching area (MRA) of Korea using the scientific echosounder (과학어군탐지기를 이용한 삼척 바다목장 해역의 어류 자원량 추정)

  • In this study, we aimed to determine the seasonal distribution and biomass of fish in Samcheok marine ranching area (MRA) of Republic of Korea using the scientific echosounder. Fish trap and gillnets were used to identify fish species in the survey area, and dB-difference method was used to estimation the spatio-temporal distribution and density of fish. The results showed that the dominant species in Samcheok marine ranching area were Chelidonichthys spinosus, Sebastes inermis, Hexagrammos otakii and Tribolodon hakonensis. The spatio-temporal distribution of fish showed that fish had a relatively higher distribution at night than during the day. In addition, the density of fish by season was highest at night in July at 34.22 g/m2 and lowest in April at 0.42 g/m2.

  • Headspace volatile compounds of oxidized fish oil were analyzed by the combination of hexane solvent or solid phase microextraction, gas chromatography and mass selective detector. The optimum condition of headspace analysis by hexane trapping was 23 min absorption time, 96$^{\circ}C$ sample temperature and 20 mL/min air flow rate. The numbers of volatile compounds identified by solvent trapping and SPME were 35 and 14, respectively. Groups having the largest amount and many kinds were hydrocarbons and aldehydes, respectively. The numbers of aldehydes were 15 and 6 for solvent trap and SPME, respectively. These basic data could be used as indicators for the quality changes of fish oil.

Management of Golden Apple Snail (Pomacea canaliculata; Ampullariidae) Using Duck and Fish Trap (왕우렁이(Pomacea canaliculata: Ampullariidae) 방제를 위한 오리 및 통발 이용)

  • This study was investigated the potential of ducks and fish trap for the control of golden apple snail, Pomacea canaliculata, in paddy field and water canals. The smaller size of duck (700g) preyed snails than the bigger one (1,300g). The consumption of two ducks was bigger under plastic house. They preyed over 90% on the second days of release. The release time of ducks was more effective at seven days after inoculation of snails than simultaneous. Duck could suppress the density of snails at any time from month after inoculation. Four ducks per $30m^2$ could control snails in an irrigation canals. The number of attracted golden apple snail per week was 216 in big fish trap $({\Phi}15cm)$ with menthol paste which for a carp.

Preference evaluation of stink bugs to leguminous seeds by video tracking system (VTS를 이용한 두류종실에 대한 노린재류의 선호성 평가)

  • Preference of stink bugs to various leguminous seeds was evaluated by using VTS (Video Tracking System) in laboratory. Major soybean stink bugs such as bean bug, Riptortus pedestris (Fabricius), one-banded stink bug, Piezodorus hybneri (Gmelin), eastern green stink bug, Nezara antennata (Scott), and sole bug, Dolycoris baccarum (L.) were significantly most attracted to Cheongjakong, a soybean variety, baited fish-net trap, followed by soybean varieties Ilpumgeomjeongkong and Taekwangkong, a peanut variety Daekwangdangkong, a kidney bean variety Gangnangkong, and a adzuki bean variety Jungwonpat, respectively in a soybean field. VTS observation in laboratory showed that R. pedestris and D. baccarum had significantly higher frequency of visit on Cheongjakong, followed by Ilpumgeomjeongkong. However, P. hybneri, N. antennata and Halyomorpha halys (Stal) had significantly higher number of visits on Cheongjakong, Seonnogkong and Jinpumkong, followed by Ilpumgeomjeongkong. The sojourned time of stink bugs, however, was significantly longer on Cheongjakong regardless of species of the bugs. Accordingly, Cheongjakong was evaluated as the most preferred soybean seed by fish-net trap and VTS. Thus, VTS is found to be an effective means to evaluate the food preference of stink bugs.

Insecticidal Effect of Aggregation Pheromone Fish Net Trap using Residual Effect of Insecticides Against Bean Bug, Riptortus pedestris (Hemiptera: Alydidae) (살충제의 잔효성을 이용한 집합페로몬통발트랩에 유인된 톱다리개미허리노린재의 살충효과)

  • This study was examined for activity of aggregation pheromone fish net trap with residual effect of insecticides against Riptortus pedestris. Time of escape was from aggregation pheromone fish net trap researched 49.74 min ($ET_{99}$) and 65.01 min ($ET_{99}$) with 1st and 2nd instar nymph respectively. Six insecticides for the stink bugs were treated to 3 different materials (wood, metal and plastic). Bifenthrin and fenitrothion were showed 100% insecticidal activity at 48 h to all developmental stage of R. pedestris. Residual effect of bifenthrin and fenitrothion on plastic material were showed 100% insecticidal activity over 15 day after treatment with 1st, 3rd instar nymphs and adults except 5th instar nymph. In field test using plastic material trap with two pesticides, 1st instar nymphs and adults were showed 100% mortality until 10 day after treatment. These results indicate that R. pedestris might be managed using aggregation pheromone trap with insecticides.

Fish Passage Assessments in the Fishway of Juksan Weir Constructed in the Downstream Area of Youngsan-River Watershed (영산강수계의 죽산보에 설치된 어도에서 어류의 이동성 평가)

  • Fish passage asssessments were conducted in the fishway at Juksan Weir, which was constructed as a four-major rivers project in the downstream area of Youngsan-River Watershed. For the research, fish-movements/migrations were analyzed for seven times from April ~ October, 2013 using an approach of fish trap-setting. Fish fauna and compositions were analyzed in the fishway, and seasonal- and diel-movement patterns were analyzed in relation to current velocity in the fishway. Also, abundances of exotic fishes such as bluegill sunfish (Lepomis macrochirus), large-mouth bass (Micropterus salmoides), and white curcian carp (Carassius cuvieri) were monitored in the fishway. Current velocity(n = 18) in the fishway showed large variations ($0.82{\pm}0.63m/s$) depending on the location of the fish trap-setting and this physical factor influenced the fish movements. Fish movements, based on the CPUE of individuals, in the fishway was greater in slower velocity (mean: 0.36 m/s, range: 0.10~1.54 m/s) than faster velocity (mean: 1.51 m/s, range: 0.90~1.90 m/s). Seasonal analysis of fish movements showed that most frequent uses (8 speices and 591 individuals, 66.2% of the total) of the fishway occurred in spring period(i.e., June). Diel movement analysis, in the mean time, showed highest in the time period of 00:00 ~ 3:00 am (7 species and 281 individuals, 20.9% of the total). The efficient managements in the fishway at Juksan Weir are required in relation to the hydrological regime.
