Chemical Composition and Size of Floating and Sunken Eggs of Olive Flounder Paralichthys olivaceus

  • Olive flounder (Paralichthys olivaceus) is one of the most commercially important species of farmed marine fish in Korea. Although techniques for rearing the larva of this species are improving, production costs are also increasing due to microbial influences and genetic degradation caused by successive culturing. Since the demand for healthy seed, which requires eggs of good quality of, is high, we examined the effects of nutrient composition on the size of P. olivaceus eggs. We analyzed floating (live) and sunken (dead) eggs of P. olivaceus from five different hatcheries for their size and amino and fatty acid composition. The sizes of eggs and oil globules from floating vs. sunken eggs were significantly different at p<0.05. No significant relationships were observed, however, among larval length, hatching percentage, and egg and oil globule size. The dry weight and amino acid levels of floating eggs were greater than those of sunken eggs (p<0.05), but no difference in fatty acid content was observed.

  • 본 연구는 2015-2018년 5월 기간 동안 납자루아과 어류의 서식 집단 중 강원도 홍천 내촌천(HN), 덕치천(HD), 정선 골지천(JG) 및 조양강(JJ)을 대상으로 기 개발된 제한절편 길이 다형성(restriction fragment length polymorphism; RFLP) 분자마커를 이용하여 숙주조개 속 난 및 치어에 대한 정확한 종 동정을 수행 후 납자루아과 어류의 숙주조개 크기에 대한 산란양상을 파악하고자 하였다. 연구대상 지역 내 납자루아과 어류는 내촌천과 골지천에서 1종(묵납자루; Acheilognathus signifer), 덕치천에서 3종(각시붕어; Rhodeus uyekii, 묵납자루; A. signifer, 줄납자루; Acheilognathus yamatsutae), 조양강에서 2종(묵납자루, 줄납자루)으로 확인되었고, 네 지역에서 모두 동서하고 있는 숙주조개인 작은 말조개(Unio douglasiae sinuolatus) 982개체를 채집하였다. RFLP 분자마커를 이용하여 납자루아과 어류의 난 및 치어가 확인된 작은말조개(N=163; 16.6%)에서 총 646개체의 납자루아과 어류의 난 및 치어(묵납자루 454, 줄납자루 43, 각시붕어 149개체)를 확인하였다. 각 지역 숙주조개 크기에 따른 산란선호도를 조사하기 위해 난 및 치어가 확인된 숙주조개(mussels with [presence] eggs/fry)와 확인되지 않은 숙주조개(mussels without [absence] eggs/fry)의 각장(shell length), 각고(shell height) 및 각폭(shell width)의 평균 크기를 비교하였다. 그 결과 3종의 납자루아과 어류가 동서하는 덕치천의 경우 난 및 치어가 확인된 숙주조개가 확인되지 않은 숙주조개보다 각장(1.98mm), 각고(0.85mm), 각폭(0.73mm)의 크기가 통계적으로 유의하게 크게 나타났으며(Mann-Whitney U tests, P=0.002, P=0.012, P=0.009), 다른 세 개의 지역에서도 난 및 치어가 확인된 조개의 각장, 각고, 각폭의 크기가 큰 결과를 보였으나, 통계적으로 유의하지는 않았다. 추가적으로 종 간 숙주조개 당 평균산란 난 및 치어의 수를 분석한 결과 각시붕어 $9.31{\pm}5.94$개, 묵납자루 $2.86{\pm}2.45$개, 줄납자루 $2.50{\pm}1.32$개로 각시붕어는 묵납자루와 줄납자루보다 숙주조개 당 평균 6.45~6.81개 더 많이 산란하였고, 통계적으로 유의한 결과를 나타내었다(Kruskal-Wallis test, P<0.001). 이 결과는 본 연구 대상 납자루아과 어류 3종에서 크기가 큰 작은말조개를 산란을 위한 숙주로서 선호함을 의미하고, 조개 크기에 대한 선호도 차이가 동서하는 납자루아과 종의 수가 많을수록 크게 나타났다. 또한, 묵납자루와 줄납자루의 경우 많은 숙주조개에 적은 양의 난을 추가적으로 고르게 산란하는 반면에 각시붕어는 비교적 적은 수의 숙주조개에 많은 양의 난을 산란하는 번식전략을 나타내었다. 2종 이상의 납자루아과 어류가 서식하는 덕치천(HD)과 조양강(JJ)에서 묵납자루와 줄납자루 2종이 동일한 조개에 산란하는 것이 관찰되었다(N=4). 이는 납자루아과 어류가 2종 이상 동서할 때, 동일한 자원인 작은말조개를 자신의 산란숙주로 이용하기 위한 종간경쟁(interspecific competition)이 일어나고 있는 것으로 판단된다. 본 연구에서는 기존 연구된 생태학적 연구에 유전학적인 방법을 추가하여 각 집단 간, 종 간 숙주조개 크기에 대한 산란양상을 보다 정확히 규명하여 숙주조개를 이용하는 납자루아과 어류의 생태적 적응양상을 명확히 파악하고 더 나아가 숙주조개와 납자루아과 어류의 공생(mutualism) 혹은 숙주-기생의 상호관계(host-parasite relationship)를 규명하는데 기여할 수 있을 것으로 기대한다.

어류 알로부터 Protease Inhibitors의 단백질 용해도 차이에 의한 분획 특성 (Fractionation and Characterization of Protease Inhibitors from Fish Eggs Based on Protein Solubility)

  • A protease inhibitor was fractionated from fish eggs using methods based on protein solubility. Fractionation efficiency was evaluated with regard to percent recovery and total inhibitory activity (U). The fractionation of protease inhibitor (PI) from egg extracts of skipjack tuna (ST, Katsuwonus pelamis), yellowfin tuna (YT, Thunnus albacores), and Alaska pollock (AP, Theragra chalcogramma) was performed by precipitation with cold acetone or ammonium sulfate (AS). Fractions exhibiting the strongest inhibitory activity contained 20-40% (v/v) cold acetone or 40-60% saturated AS fractions. AS fractionation was more effective in isolating PI than was precipitation with acetone. The total inhibitory activity and percent recovery of fraction obtained with AS 40-60% toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 4,976 U and 24.2% for ST, 3,331 U and 38.1% for YT, and 4,750 U and 43.8% for AP, respectively. In comparisons against six commercial proteases, 40-80% AS fractions, made by combining the 40-60% and 60-80% AS fractions from fish egg extract, exhibited the strongest inhibition of trypsin when using a casein substrate. These results suggest that fish eggs act as serine protease inhibitors and may be useful for protease inhibition in foodstuffs.

명태(Gadus chalcogrammus) 친어 사료내 명태유 첨가가 수정란과 자어 품질에 미치는 효과 (Effects of the Dietary Inclusion of Pollock Oil on the Quality of Eggs and Larvae of Walleye Pollock Gadus chalcogrammus)

  • This study investigated the effects of inclusion of pollock oil (PO) in the diet of broodstock on the egg and larval quality of Walleye Pollock Gadus chalcogrammus. The experimental diet comprised of semi-moist pellets prepared either with or without PO (control; Con). The fish assigned to each diet were fed to visual satiation (two meals per day) for 12 weeks. Eggs spawned from both groups of fish were sampled during the spawning season (three months) and the egg and larval quality was determined. Total egg volume, fertilization, and hatchability of eggs from the PO group were higher than those of the Con group. Fatty acid composition of fertilized eggs was not affected by the experimental diet. Oleic acid, eicosapentaenoic acid, and docosahexaenoic acid content in larvae of the PO group was higher than that in larvae of the Con group. These results demonstrate the beneficial effects of including PO in the broodstock diet of Walleye Pollock, specifically on the quality of the eggs (total egg volume, fertilization, and hatchability) and larvae (fatty acid composition).

3종의 연어과 어류와 수정난으로부터 분리한 물곰팡이병 원인 진균의 분류와 약물 효과 (Identification and chemotherapeutic effects of the fungi from three salmonid species and their eggs)

  • 2005년 11월과 12월에 걸쳐 연어 수정란 (ChS-E0511, 강원 양양), 무지개송어 수정란 (RaT-E0511, 경남 밀양), 무지개 송어 성어 (Rat-A0512, 강원 평창) 및 산천어 자어 (MaS-F0512, 강원 평창)에서 물곰팡이를 분리하였다. 분리 곰팡이의 분류 결과, 형태학적 및 유전학적 연구를 통하여 4종 모두 Saprolegnia parasitica로 확인되었으며, 연어 수정란으로부터 분리된 ChS-E0511 균주는 조란기 (oogonia)와 무성아 (gemmae)를 형성하므로 S. parasitica group 1으로 분류되었다. 분리균주인 ChS-E0511을 이용하여 다양한 약물의 성장 억제 효과를 측정한 결과, 사용금지 약물인 malchite green에 대한 약물 효과가 높게 나타났으며, 손바닥선인장 (Opuntia ficus-indica) 추출물 (MBT-01108), 2-bronopol (BNP) 및 sodium chloride 등에 대한 무지개송어 수정란의 상대발안율이 높게 나타났다. 본 연구의 결과, 국내 연어과 어류의 질병과 관련한 곰팡이는 S. parasitica로서 2가지 이상의 group으로 구분되며 다양한 약물들에 대한 평가가 지속적으로 이루어 져야 할 것으로 사료된다.

능성어(Epinephelus septemfasciatus) 양식장에서의 바이러스성신경괴사증(VNN) 예방대책 (Prevention Strategies for Viral Nervous Necrosis (VNN) in Sevenband Grouper Epinephelus septemfasciatus Aquaculture Farms)

  • Viral nervous necrosis (VNN) is a serious disease of sevenband grouper Epinephelus septemfasciatus in Korean aquaculture farms. However, we suggest the following preventative methods for hatcheries: 1) disinfecting rearing water, 2) selecting spawners via ELISA and PCR, 3) selecting eggs via PCR, 4) disinfecting fertilized eggs, and 5) proper facilities management. When these methods are implemented, nervous necrosis virus (NNV)-free fish are produced because vertical and horizontal transmission is prevented. However, horizontal transmission of NNV through rearing seawater sourced from the environment during grow-out stages in sea cages can still occur. Live NNV vaccines with a low rearing temperature or Poly(I:C) immunization are very effective at preventing horizontal transmission of NNV in rearing farms. Furthermore, even after VNN is contracted, fish mortality can be reduced by administering Poly(I:C).

조선시대 좌반류(佐飯類)의 종류에 대한 문헌적 고찰 (A Literature Review on the Type of Joseon Dynasty Jwabans)

  • This article examines the types of Jwabans as recorded in 21 old books of the Joseon dynasty (1392-1909). The ingredients used in Jwabans during the Joseon dynasty were root vegetables, sea algae, seeds nuts, bird, meat, and fish. In the early, middle, and late eras of the Joseon dynasty, 10, nine, and 181 kinds of Jwabans were prepared, and two, one, and seven kinds of Jwabans were prepared using root vegetables (根菜類). During the early and late eras of the Joseon dynasty, one and 14 kinds of Jwabans were prepared using sea algae (海藻類), respectively, and four kinds of Jwabans were prepared using seeds nuts during late eras of the Joseon dynasty (種實類). During the early, mid and late eras, one, two, and 17 kinds of Jwabans were prepared using bird (鳥類), three, one, and 47 kinds of Jwabans were prepared using meat (肉類), and one, five, and 81 kinds of Jwabans were prepared using fish (魚類). The frequency of the Jwabans ingredients in order were fish (30.5%), meat (23.5%), pheasant (7%), root vegetables (5%), abalone (全鰒) (5%), laver (海苔) (4%), shellfish (貝類) (3%), fish eggs (魚卵) (2.5%), fleshy prawn (大蝦) (2.5%), sea tangle (昆布) (2%), dried tangle (海草) (1.5%), sparrow meat (雀肉) (1.5%), and etc during the Joseon dynasty. It seems that the appearance and supplementation with different ingredients increased throughout the Joseon dynasty. This may be associated with the commercial industrial development that prevailed during the late Joseon dynasty. Further study will be conducted on recipes and ingredients recorded in these old books to develop a standardized recipe to globalize Jwabans.

Morphological Development of Eggs, Larvae and Juveniles of the Far Eastern Catfish, Silurus asotus in Korea (Pisces: Siluridae)

  • This study was conducted to investigate egg development and larvae morphological development of catfish and to provide basic data to clarify the genetic relationship with Siluriformes fish. The mother fish that was used in this study was caught in the stream of Nakdong River in Uiseong-gun, Gyeongbuk. The temperature range of the breeding was $23.0-25.0^{\circ}C$ (mean $24.0{\pm}1.0^{\circ}C$) and egg size was 1.62-1.70 mm (mean $1.66{\pm}0.05$, n=30). Eggs of catfish began hatching at 54 hours and 40 minutes after fertilization. Immediately after hatching, the total length of larvae was 3.60-3.65 mm (mean $3.62{\pm}0.03$, n=5) and had an egg yolk without swimming ability. On the third day after hatching, the larvae at the medium stage was 8.00-8.65 mm (mean $8.32{\pm}0.45$) in total length, and two pairs of whiskers formed around the mouth were elongated. On the 12th day after hatching, the larvae at the juvenile stage was 16.5-17.0 mm (mean $16.7{\pm}0.35$) in total length, and the stem of each fin was in the range, and the juvenile at this period was morphologically similar to the mother fish.

서해연안 해역에 분포하는 부유성 난 및 자치어의 분포특성 (Distribution of ichthyoplankton in the West coastal waters of Korea)

  • The ichthyoplanktons were sampled by Bongo net to study the distribution of the eggs and larvae in the West coastal waters of Korea during the study period (February, May, August and October of 2017). Collected fish eggs were divided into Engraulis japonicus and unidentified species. The most dominant species, Engraulis japonicus accounted for 79.52% of the total fish egg, which was followed by unidentified species (20.48%). The total of 36 taxa in fish larvae were identified based on morphological and molecular characteristics. Fish larvae were dominant by Gobiidae species. 48.30% of the total catch in abundance, which was followed by Engraulis japonicus (31.86%). These two taxa composed 80.16% of the total collected fish larvae. The seasonal diversity index of species from the collected fish larvae in the West coastal waters in 2017 was 0.625-1.883 and the evenness index was 0.380-0.865. In addition, dominance index was 0.618-0.920 and richness was 0.573-3.189.

Spawning Characteristics and Artificial Hatching of Female Mottled Skate, Beringraja pulchra in the West Coast of Korea

  • The gonadsomatic index (GSI) of mottled skate was the highest in April, GSI and HSI showed a reverse phase for its reproductive cycle. The fish had one pair of egg capsules, having 1 to 7 fertilized eggs, and spawned all the year round. When surveying the reproductive characteristics of females over 63 cm in disc width, we found the spawning peak was between April to June, and the appearance ratio of egg capsules was the highest in May (32.1%). The eggs were hatched at $8^{\circ}C$, $13^{\circ}C$, $18^{\circ}C$, water temperature (12.8 to $24.2^{\circ}C$), and the best hatching temperature was $18^{\circ}C$. The number of fish hatched was 4 to 5 fish/egg capsules, and the hatching rate was 100%. The sex ratios of hatching larvae were 45.5% female and 54.5% male. Therefore this study will provide fundamental data and information for artificial reproduction of the mottled skate.
