Analysis of Prevalence and Survival pattern of Staphylococcus aureus from Dried Seasoned Fishes (조미건어포의 Staphylococcus aureus 오염도 및 생존패턴 분석)

  • In this study, the contamination levels of total aerobic bacteria, E. coli, total coliform and S. aureus of seasoned dried fishes (SDF) in Korea were investigated. A total of 81 SDF samples were purchased randomly from 28 stores. Contamination range of total aerobic bacteria, total coliform and S. aureus were 150~1,700,000, 10~31,000 and 10~220 CFU/g, respectively. E. coli was detected in only one samples in the qualitative test. We have analyzed quantitatively Staphylococcal enterotoxins (SE-type A, C and D) produced by S. aureus contaminated in SDF using a TECRA kit and standard curve. The curve equation was Y = 0.1499 * X + 0.1183 and maximum amount of SEs in SDF was 0.71 ng/ml. Reduction speed of S. aureus in SDF stored at $37^{\circ}C$ was the highest among the samples stored for 8 days at different temperature of 7, 18 and $37^{\circ}C$. On the basis of the results, SDF in Korea can be contaminated by a variety of pathogenic bacteria. Therefore, precautionary measures are necessary for consumer protection, including the improvement of sanitary conditions in the processing plants in Korea.

Comparison of Liquefying Efficiency of Mixed Organic Fertilizer as Affected by Aeration Time and the Ratio of Organic Fertilizer to Water (폭기시간과 유기질비료 농도에 따른 혼합유기질비료의 액비화 특성비교)

  • This study was conducted to evaluate the liquefying efficiency of mixed organic fertilizer in different conditions. The organic fertilizer was composed of sesame oil cake, rice bran, fish meal, ground bone meal etc, and made by fermenting process. It included $23g\;kg^{-1}$, $17.0g\;kg^{-1}$, $23.9g\;kg^{-1}$, $290g\;kg^{-1}$ of N, $P_2O_5$, $K_2O$, organic matter, respectively. In one test, the mixed organic fertilizer was added in the proportion of 10% to water 90% and aerated continuously, for 2, 8 hours per day, and not aerated as control. In the other test, ratios of organic fertilizer to water were 5%, 10%, 20% and aerated for 2 hours a day. With the increase of liquefying time, pH, EC and $NH_4-N$ increased without relation to aeration time. After 10 days, liquid organic fertilizer aerated for 2 hours a day contained $634mg\;N\;kg^{-1}$, $68.1mg\;P_2O_5\;kg^{-1}$, $453mg\;K_2O\;kg^{-1}$, which was not significantly different from 8 hours a day or continuous aeration. Then extraction ratios of inorganic contents were 27.6%, 4.0% and 18.9%, respectively. Continuous aeration resulted in increasing the viable number of aerobic bacteria, spore forming bacteria and fungi in liquefied solution. Higher ratio of organic fertilizer to water increased EC, $NH_4-N$ and other inorganic matter contents, but decreased extraction ratio of nutrients in liquid fertilizer. The liquid organic fertilizer of 20% contained $1,140mg\;N\;kg^{-1}$, $35.4mg\;P_2O_5\;kg^{-1}$, $544mg\;K_2O\;kg^{-1}$ after 10 days. Then extraction ratios were 24.8%, 2.4% and 13.6%, respectively. The ratio of organic fertilizer to water was positively correlated with only spore forming bacteria, Pseudomonas spp. among microorganisms.

  • Antibiotics have greatly contributed to the treatment and prevention of bacterial diseases in humans, animals, and fish. However, antibiotic misuse has led to the emergence and spread of multidrug-resistant bacteria. In addition to antibiotic discovery research, efforts are being made to combat such multidrug-resistant bacteria using antimicrobial agents, antioxidants, host immune enhancement, probiotics, and bacteriophages, as well as various symptomatic therapies. To discover novel bioactive compounds, it is crucial to adopt approaches that incorporate fresh ideas, new targets, innovative techniques, and untapped resources. Halophytes are plants that grow in high-salt soils and are known to adapt to salt-induced stress through unique metabolic processes that produce secondary metabolites. This study aimed to investigate the effects of extracts of halophytes native to Korea on oxidative stress and to determine whether they exert inhibitory activity against biofilms, which are major pathogenic factors of infectious bacteria. The Acinetobacter baumannii strain ATCC 17978, a representative drug-resistant bacterium, was used to measure anti-biofilm activity. The results showed that Aster spathulifolius, Carex kobomugi, Rosa rugosa, and Asparagus cochinchiensis exerted strong antioxidant and anti-biofilm effects without affecting bacterial growth itself. The halophytes used in this study are promising candidates for the development of pharmaceutical agents with antioxidant and antimicrobial properties.

Effects of Protein and Carbohydrate Supplementations on Fibre Digestion and Microbial Population of Sheep

  • The effects of two types of protein, soybean meal (SBM) and fish meal (FM); and two types of energy supplements, corn flour (CF) and paper pulp (PP), on intake of guinea grass (Panicum maximum), fibre digestion and microbial activities in four Merino rams with an average weight of $54.4{\pm}4.5kg$ were studied. Each animal was fitted with a ruminal cannula and a duodenal cannula at the proximal position. The animals were fed twice daily with chopped guinea grass (5 cm) ad libitum and one of the four dietary supplements: 170 g FM+268 g PP; 170 g FM+268 g CF; 200 g SBM+200 g PP or 200 g SBM+200 g CF. All the supplements were mixed with 100 g molasses. In sacco and in vivo digestibilities, digesta flow rates, fermentation and microbial population were studied in a $4{\times}4$ Latin square design with a $2{\times}2$ factorial arrangement of dietary treatments. The effects of energy or protein sources were not significant on grass intake of sheep. The potential degradabilities of NDF and ADF were not significantly affected by any of the supplements. However, the energy and protein sources had significant efects on disappearance rate of NDF and ADF. The disappearance rate of both NDF and ADF were significantly (p < 0.05) higher in animals fed PP when compared to animals fed CF. Animals fed FM also showed significantly (p < 0.03) higher disappearance rate of ADF than those fed SBM. Animals fed PP showed better digestion in the rumen and total tract. Total flow of NDF and ADF through the duodenum was not significantly affected by the various supplements. The mean rumen pH values (5.8-6.1) were not significantly different among the four different diets. The concentration of rumen ammonia was significantly (p < 0.0001) higher in animals fed SBM (235-266.4 mg N/L) supplement than in animals fed FM (174.9-179.7 mg N/L), while total VFA concentration was not significantly affected by both energy and protein supplements. Mean values of total VFA ranged from 72.5-82.3 mM. Molar proportions of acetate, propionate and butyrate were typical of a roughage type fermentation. Molar proportion of acetate was significantly (p < 0.0001) higher in sheep fed PP when compared to sheep fed CF. Animals fed FM had higher total viable bacterial counts, while animals fed CF showed higher protozoal numbers. Proportions of cellulolytic bacteria were only slightly higher in animals fed SBM or PP.

Inhibitory Effect of Garlic Extract on Histamine Accumulation in Mackerel Meat (마늘 추출물의 고등어육에서의 히스타민 생성 억제)

  • This study was carried out to investigate the effects of garlic extract on histamine reduction in mackerel meat stored at $4^{\circ}C$ and $15^{\circ}C$, respectively. The number of total bacteria in mackerel meat treated with 7.5% garlic extract was ten times lower than that of the control stored for 6 days of $4^{\circ}C$. However, there was no difference among the samples after 9 days of storage. Reduction of eicosapentaenoic acid (C20:5) and docosahexaenoic acid (C22:6) contents was observed in the normal control group at both temperatures. However, contents of these two polyunsaturated fatty acids slightly increased in garlic extract-treated fish. Amounts of volatile basic nitrogen increased over time in the control group, but decreased in the garlic extract-treated meat, indicating that garlic extract might suppress decomposition. No histamine was detected initially in all samples. However, a small amount of histamine (42.87 mg/kg) was detected in the control at 3 days after storage at $4^{\circ}C$. Histamine content increased continuously with storage period but was lower in mackerel meat containing garlic extract, in which histamine suppression was proportional to the concentration of garlic extract. It can be concluded that garlic extract could be utilized to extend the storage period of mackerel.

Characterization and Mapping of the Bovine FBP1 Gene

  • Fructose-1,6-bisphosphatase (FBP1) is a key regulatory enzyme of gluconeogenesis that catalyzes the hydrolysis of fructose-1,6-bisphosphate to generate fructose-6-phosphate and inorganic phosphate. Deficiency of fructose-1, 6-bisphosphatase is associated with fasting hypoglycemia and metabolic acidosis. The enzyme has been shown to occur in bacteria, fungi, plants and animals. The bovine FBP1 gene was cloned and characterized in this study. The full length (1,241 bp) FBP1 mRNA contained an open reading frame (ORF) encoding a protein of 338 amino acids, a 63 bp 5' untranslated region (UTR) and a 131 bp 3' UTR. The bovine FBP1 gene was 89%, 85%, 82%, 82% and 74% identical to the orthologs of pig, human, mouse, rat and zebra fish at mRNA level, and 97%, 96%, 94%, 93% and 91% identical at the protein level, respectively. This gene was broadly expressed in cattle with the highest level in testis, and the lowest level in heart. An intronic single nucleotide polymorphism (SNP) (A/G) was identified in the $5^{th}$ intron of the bovine FBP1 gene. Genotyping of 133 animals from four beef breeds revealed that the average frequency for allele A (A-base) was 0.7897 (0.7069-0.9107), while 0.2103 (0.0893-0.2931) for allele B (G-base). Our preliminary association study indicated that this SNP is significantly associated with traits of Average Daily Feed Intake (ADFI) and Carcass Length (CL) (p<0.01). In addition, the FBP1 gene was assigned on BTA8 by a hybrid radiation (RH) mapping method.

Domestic Test Species for Aquatic Toxicity Assessment in Korea (수생태계 독성평가에 적용 가능한 국내 시험종 선정)

  • The use of aquatic species in ecotoxicity research is well established in developed countries. However, there are limitations of using the species that are not native to Korea, and the toxicity data produced by domestic test species are significantly needed to reflect the domestic situation. The purpose of this study was to investigate the domestic species that can be applicable for the aquatic toxicity assessment in Korea. Aquatic toxicity data were collected in the framework of the project 'Development of integrated methodology for evaluation of water environment' to obtain a range of test species used for aquatic toxicity assessment internationally. The test species collected were evaluated in terms of domestic distribution based on the reliable references and the advices of experts. We figured out the 71 test species native to Korea. They included 7 fish, 26 invertebrates (2 annelids, 2 bryozoa, 13 crustaceans, 3 insects, 4 mollusc, 1 platyhelminth, and 1 protozoan), 26 plants (9 diatoms, 14 green algae, 3 macrophytes), and 12 others (2 amphibians, 3 bacteria, 6 blue-green algae, and 1 fungus). The result of this study should be a very useful information for ecotoxicity assessment in aquatic ecosystem, especially in choosing the test species applicable for the ecotoxicity in Korea hereafter.

Effects of Natural Compounds from Various Plant Eradicate the Persister Cell of Edwardsiella tarda Treated with Antibiotics of Florfenicol and Amoxicillin (천연 식물 추출물 첨가에 의한 어류 에드워드증(Edwardsiellosis) 발생균인 Edwardsiella tarda에 항생제 투여로 생성되는 persister cell 저감 효과)

  • High concentration of antibiotics has been used to treat the outbreak of edwardsiellosis caused by Edwardsiella tarda in aquaculture. However, not all of the bacteria have been killed with high concentrations of antibiotics treatment by the formation of persister cells with a dormant state. The main objective of this study was to kill persister cell using antibiotics with the addition of natural plant compounds. Antibiotics used in this study consist of 100 mg/ml florfenicol and 100 mg/ml amoxicillin. Ten natural plant compounds with persister cell inhibitor activity to E. coli were obtained from Protein Engineering and Systems Biology Lab. of Sungkyunkwan University. The persister cell inhibition activities of those natural plant compounds were evaluated in test tube. Concentrations of the antibiotics were in the ranges of 25~200 ${\mu}g/ml$. The persister cell formation was observed after 16 hours of culture. Persister cells were killed by antibiotics with natural plant compounds. Among ten natural plant compounds, Gynostemma pentaphyllum, Mallotus japonicus, and Orixa japonica showed persister cell formation inhibition activities. The optimal concentrations of G. pentaphyllum, M. japonicus, and O. japonica for the inhibitor of persister cell formation were 100 ${\mu}g/ml$, 100 ${\mu}g/ml$, and 200 ${\mu}g/ml$, respectively. In vivo study was carried out to evaluate the effect of the antibiotics with natural plant compounds using aquacultural fish, olive flounder, as test animals. G. pentaphyllum, M. japonicus, and O. japonica of 30 ${\mu}g/ml$, 10 ${\mu}g/ml$, and 10 ${\mu}g/ml$ with antibiotics reduced cumulative mortalities, showing the effectiveness of persister cell inhibition.

Effects of Gamma Irradiation on the Decontamination of Animal Feeds : Sterilization of Protein Sources (배합사료 원료에 대한 방사선 살균 효과 -단백질원의 살균-)

  • The effect of gamma irradiation of microbiological and physicochemical properties of raw ingredients (thirteen kinds of protein sources) for the mixed feed were investigated. The total aerobic bacteria counts in the samples was ranged from $10^2$ to $10^7$cells/g. After 5 to 7 kGy irradiation, the total count was reduced by 3 to 4 log cycles. Coliforms were detected in seven samples and the range was from $10^4$ to $10^6$cells/g. Enteric pathogens were found only in fish meal and poultry byproduct as $10^2$ to $10^6$cells/g. They were sensitive to radiation and completely sterilized by 3 to 5 kGy irradiation. Total fungi count was ranged from $10^1$ to $10^4$cells/g in all samples. They were osmophiles such as Aspergillus, Penicillium, Cladosporium, Aureobasidium and Rhizopus and were destroyed by 3 to 7 kGy irradiation. Five species of potential mycotoxin producers including Aspergillus flavus were also identified. Physicochemical properties, such as total amino acid content, TBA value, POV and color difference showed that irradiation with optimum does was less detrimental than autoclaving.

The Distribution of Microflora in the Viscera of Sardine, Sardinops melanosticta by the storage Temperature (저장온도에 따른 정어리 내장 미생물상의 변화에 대하여)

  • As the putrefaction of fish is greatly relied on the microorganisms inhabited in the viscera of them, we investigated the microfloral changes in the viscera of sardine, Sardinops melanosticta, which has been caught a lot in adjacent sea of Korea but showed rapid spoilage, after storages with various temperature. The following results were obtained. Viable cell counts at $25^{\circ}C$ of the viscera of sardine were $1.6\times10^5/g$ at the fresh sample, $1.5\times10^5/g$ at the frozen sample, $2.9\times10^8/g$ at the spoiled samples. The most predominant microbial genera from the fresh sardine were Moraxella spp.($31.4\%$) and Pseudomonas spp.($28.6\%$), but Enterobacteriaceae($83.1\%$) was in spoiled sample. While Moraxella spp.($46.2\%$) and Flavobacterium-Cytophaga($21.0\%$) were predominant in the frozen sample and Enterobacteriacear($69.6\%$) was in the thawed-spoiled sample. The rates of proteolytic enzyme producing bacteria were $20\%$ in the fresh sample, $22\%$ in the frozen sample but the rates were increased to $52\%,\;29\%$ in the spoiled sample and the thawed-spoiled sample respectively.
