Riparian Connectivity Assessment Using Species Distribution Model of Fish Assembly (어류군집의 종분포모형을 이용한 수변지역 연결성 평가)

  • River corridors facilitate dispersal and movement and prevent local extinction of species. As a result of stream restoration projects, which include installation of waterfront and flood control structures, the number of animals, which rely on river corridor, is decreasing. For the study, factors affecting fish assembly were extracted by a species distribution model with the fish data collected from the Seom River in Hoengseong County and City of Wonju, Ganwon Province, Korea between March to October 2013. The riparian connectivity was assessed using species richness and rarity. According to result of the field survey, there were 38 species and 7,061 individuals for fish. The analysis suggests the following. Firstly, factors affecting fish richness in species distribution model results are shown to be velocity, riffle, riparian width, and water width. The accuracy of the model proves to be suitable with the correlation coefficient of 0.83 and MAPE of 19.2%. Secondly, the low rarity area is shown to be straight streams in Jeon river near to Hongseong County and the high rarity area to be streams with large width, existing alluvial area at channel junction between Jeon river and Seom river. Thirdly, according to connectivity results, areas where weirs are installed or riparian buffer area is removed showed low connectivity. The areas where farmland near riparian and forest areas showed high connectivity. The results of this study can be utilized to improve current facilities and enhance connectivity as a restoration guide.

  • The present study aims to optimise the process parameters of fish glue and adhesion property through room temperature in terms of curing time and opened assembly time for attaining best adhesion strength to wood (Yellow birch, B. alleghaniensis). The uncured and cured fish glue have been characterised by TGA, DSC, FT-JR. It is observed that with increasing curing time up to 48 H, tensile lap shear strength of adhesive joint of fish glue to wood increases and decreases after 48 H. At 15 min. opened assembly time, lap shear strength increases up to 4833 kgf/cm2 and decreases after 15 min.

Analysis of Spiramycin in Fish Using High Performance Liquid Chromatography (HPLC를 이용한 어류 중의 Spiramycin 분석)

  • A high performance liquid chromatography assay method for spiramycin in fish muscle was developed. The developed method was evaluated and validated by monitoring spiramycin in olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli) and in live conger eel (Anguilla japonica) in fish farms and distribution centers. Using the developed method, the recovery rate was up to 82.4-88.8%, which was higher than that of conventional methods (77.6-87.1%). In particular, the proposed sample treatment protocol was suitable for use with fish samples to remove low molecular weight materials and pigments that could interfere an accurate analysis. The prepared stock solution was very stable, and it remained chemically stable for 5 weeks at $4^{\circ}C$. The performance limit of the developed method for spiramycin acid in fish muscle was 0.05 ppm. One hundred thirty-four fish samples including olive flounder, black rock fish and live conger eel were analyzed to evaluate the overall efficiency of the modified method and to monitor the actual condition of spiramycin usage in fish farms. Olive flounder and black rock fish were collected from fish farms in coastal areas of Korea, and live conger eels were purchased from a fish market in the Busan area from September 2001 to March 2002. According to the overall performance of the developed method, it was considered suitable for the monitoring of spiramycin in fish muscle. The suggested method of analysis for spiramycin in fish muscle is registered in the Korean Official Methods of Food Analysis and is currently tieing utilized for fishery products inspection by the Korea Food and Drug Administration and the National Fisheries Products Quality Inspection Service.

Analysis of Oxolinic Acid in Fish Products Using HPLC (HPLC를 이용한 어류 중의 Oxolinic Acid 분석)

  • A high-performance liquid chromatography assay method for oxolinic acid in fish products was developed, evaluated and validated through the monitoring of oxolinic acid based on farming and distribution. The recovery rate of the developed method was $102.3-106.7\%$ as compared to conventional methods. The stock solution was stable for 3 weeks under refrigerated condition at $4^{\circ}C$ The performance limit was evaluated as 0.01ppm of oxolinic acid in fish muscle. 478 fish samples such as olive flounder, genuine porgy, common sea bass and black rock fish collected from fish farms in the coastal area from September 2001 to October 2004 were analyzed to evaluate overall efficiency of the modified method and to monitor the actual condition of oxolinic acid usage in fish farm. According to the monitoring results, the modified method was suitable for analysis of oxolinic acid in fish muscle and oxolinic acid might be used in a small portion of fish farms. The suggested analysis method of oxolinic acid was registered in the Korean Official Methods of Food Analysis and is being utilized for fishery products by the Korea Food and Drug Adminstration and the National Fisheries Products Quality Inspection Service.

State of the art on the physical mapping of the Y-chromosome in the Bovidae and comparison with other species - A review

  • The next generation sequencing has significantly contributed to clarify the genome structure of many species of zootechnical interest. However, to date, some portions of the genome, especially those linked to a heterogametic nature such as the Y chromosome, are difficult to assemble and many gaps are still present. It is well known that the fluorescence in situ hybridization (FISH) is an excellent tool for identifying genes unequivocably mapped on chromosomes. Therefore, FISH can contribute to the localization of unplaced genome sequences, as well as to correct assembly errors generated by comparative bioinformatics. To this end, it is necessary to have starting points; therefore, in this study, we reviewed the physically mapped genes on the Y chromosome of cattle, buffalo, sheep, goats, pigs, horses and alpacas. A total of 208 loci were currently mapped by FISH. 89 were located in the male-specific region of the Y chromosome (MSY) and 119 were identified in the pseudoautosomal region (PAR). The loci reported in MSY and PAR were respectively: 18 and 25 in Bos taurus, 5 and 7 in Bubalus bubalis, 5 and 24 in Ovis aries, 5 and 19 in Capra hircus, 10 and 16 in Sus scrofa, 46 and 18 in Equus caballus. While in Vicugna pacos only 10 loci are reported in the PAR region. The correct knowledge and assembly of all genome sequences, including those of genes mapped on the Y chromosome, will help to elucidate their biological processes, as well as to discover and exploit potentially epistasis effects useful for selection breeding programs.

Persistent Infection of Marine Birnavirus and its Status of Infection in Cells (잠복감염시의 해양버나바이러스의 세포내에서의 동태)

  • The objective of the study was to clarify the mechanism of persistent infection of marine birnavirus (MABV) in various nonpermissive cell lines. It was observed in CHSE-214, RTG-2 and RSBK-2 that the virus produced at high yield with typical cytopathic effect (CPE). On the contrary, the CPE was not produced in EPC, FHM and BF-2 cells. However amount of virus protein in both permissive and nonpermissive cell lines detected by ELISA was almost the same. Electron microscopy showed virions in permissive cells but not in nonpermissive cells. From the results, it is clear that virus protein and RNA were produced in nonpermissive cells as observed in permissive cells; however, assembly of the virus particles did not occur in nonpermissive cells.

Analysis of Tetracyclines Using Righ-Perforklance Liquid Chromatography for Fishery Products (HPLC를 이용한 어패류 중의 테트라사이클린계 항생제 분석방법 개발)

  • An analysis method of tetracyclines far fish and shellfish products containing large amount of low molecular materials and pigments was established. The recovery of the established analysis method for four tetracycline samples was $72-100\%$ and higher than other methods reported. Especially, proposed sample treatment protocol was shown to be effective for the removal of low molecular materials and pigments that tend to interfere with accurate analysis. The detection limit of oxytetracycline (OTC) and tetracycline (TC) from the sample was 0.02 ppm, and the detection limit of chlortetracycline (CTC) and doxycycline (DC) from the sample was 0.1ppm, To examine the efficiency of the established method and identify tetracycline usage in fish farms, tetracycline group antibiotics in the flounder being cultured was monitored. The improved method can be used for fish and shellfish products effectively and all surveyed fish farms have used tetracycline all the year round. The proposed method was adopted as official method for fishery products by Korean Food and Drug Administration in 2003 and it is being used by regulatory authority as National Fishery Products Quality Inspection Service.

De novo Assembly and Analysis of Amur Sturgeon (Acipenser schrenckii) Transcriptome in Response to Mycobacterium Marinum Infection to Identify Putative Genes Involved in Immunity

  • Fish mycobacteriosis is a common bacterial disease in many species of freshwater and marine fish and has caused severe loss of fish production. Mycobacterium marinum has been the most prevalent pathogen observed in several outbreaks of mycobacteriosis of farmed sturgeons in China. However, the immune responses and pathology of sturgeons in mycobacterial infection are rarely studied. Therefore, we used the Illumina RNA-seq method to analyze the transcriptome profile of Acipenser schrenckii challenged with Mycobacterium marinum. To begin, 168,220 non-redundant contigs were acquired from the infection and control groups, and among these, 33,225 contigs have acquired annotations. A total of 4,043 differently expressed (DE) contigs between the two groups were identified, and among these, 2479 were up-regulated and 1564 were down-regulated in the infected fish. A total of 1,340 DE contigs with acquired annotations in KEGG were enriched for 124 pathways including the TNF signaling pathway, and the Toll-like receptor signaling pathway. The roles of DE genes involved in significant pathways and other processes were discussed. The 2,209 DE contigs that have yet to acquire proper annotation may represent candidate genes associated with infection in sturgeons and are expected to serve as immunogenetic resources for further study. To our best knowledge, this is the first transcriptome study on sturgeons under bacterial infection.

Production of virus-like particles of nervous necrosis virus displaying partial VHSV's glycoprotein at surface and encapsulating DNA vaccine plasmids

  • In order to use nervous necrosis virus (NNV) virus-like particles (VLPs) as a delivery tool for heterologous antigens or plasmids, we attempted to produce red-spotted grouper nervous necrosis virus (RGNNV) VLPs displaying a partial region of viral hemorrhagic septicemia virus (VHSV) glycoprotein at the surface and VLPs that are harboring DNA vaccine plasmids within the VLP. A peptide encoding 105 amino acids of VHSV glycoprotein was genetically inserted in the loop region of NNV capsid gene, and VLPs expressing the partial part of VHSV glycoprotein were successfully produced. However, in the transmission electron microscope analysis, the shape and size of the partial VHSV glycoprotein-expressing NNV VLPs were irregular and variable, respectively, indicating that the normal assembly of capsid proteins was inhibited by the relatively long foreign peptide (105 aa) on the loop region. To encapsulate by simultaneous transformation with both NNV capsid gene expressing plasmids and DNA vaccine plasmids (having an eGFP expressing cassette under the CMV promoter), NNV VLPs containing plasmids were produced. The encapsulation of plasmids in the NNV VLPs was demonstrated by PCR and cells exposed to the VLPs encapsulating DNA vaccine plasmids showed fluorescence. These results suggest that the encapsulation of plasmids in NNV VLPs can be done with a simple one-step process, excluding the process of disassembly-reassembly of VLPs, and NNV VLPs can be used as a delivery tool for DNA vaccine vectors.

Strain Analysis of a Six Axis Force-Torque Sensor Using Cross-Shaped Elastic Structure with Circular Holes (원구멍이 있는 십자형 탄성체를 가진 6축 힘, 토크 센서의 변형률 해석)

  • The necessity of six axis force-torque sensors is well recognized in the fields of automatic fine assembly, deburring polishing, and automatic fish processing using robotic manipulators. The paper proposes a simple and compact elastic structure of the force-torque sensor which senses externally applied three force and three torque components. Rough surface strain distribution of the elastic structure is examined analytically, and then more accurate surface strain are obtained from finite element analysis. The compliance matrix which is a linear relationship between force components and strain measurements is obtained for the proposed sensor. Some basic principles of measuring 3 force and torque components are also presented.
