Development of Bovine Embryos Reconstructed by Microinjection of Cultured Fetal Fibroblast Cells into In-Vitro Matured Oocytes

  • Bovine cumulus-oocyte complexes were recovered from ovaries at a slaughter and then divided into five groups: control group(unvitrified oocytes), 0 hr. group(composed of oocytes vitrified before the onset of maturation) and 10, 14, and 20 hrs groups(vitrified respectively at 10, 14 and 20 hrs after the onset of maturation). The oocytes remained vitrified for 24 hrs, and then were thawed in 30℃ water bath. Survival and cleavage rates were defined as development rate on in vitro culture and stained with aceto-orcein or FDA test.

The Comparison of Commercial Serum-Free Media for Hanwoo Satellite Cell Proliferation and the Role of Fibroblast Growth Factor 2

  • Fetal bovine serum (FBS), which contains various nutrients, comprises 20% of the growth medium for cell-cultivated meat. However, ethical, cost, and scientific issues, necesitates identification of alternatives. In this study, we investigated commercially manufactured serum-free media capable of culturing Hanwoo satellite cells (HWSCs) to identify constituent proliferation enhancing factors. Six different serum-free media were selected, and the HWSC proliferation rates in these serum-free media were compared with that of control medium supplemented with 20% FBS. Among the six media, cell proliferation rates were higher only in StemFlexTM Medium (SF) and Mesenchymal Stem Cell Growth Medium DXF (MS) than in the control medium. SF and MS contain high fibroblast growth factor 2 (FGF2) concentrations, and we found upregulated FGF2 protein expression in cells cultured in SF or MS. Activation of the fibroblast growth factor receptor 1 (FGFR1)-mediated signaling pathway and stimulation of muscle satellite cell proliferation-related factors were confirmed by the presence of related biomarkers (FGFR1, FRS2, Raf1, ERK, p38, Pax7, and MyoD) as indicated by quantitative polymerase chain reaction, western blotting, and immunocytochemistry. Moreover, PD173074, an FGFR1 inhibitor suppressed cell proliferation in SF and MS and downregulated related biomarkers (FGFR1, FRS2, Raf1, and ERK). The promotion of cell proliferation in SF and MS was therefore attributed to FGF2, which indicates that FGFR1 activation in muscle satellite cells may be a target for improving the efficiency of cell-cultivated meat production.

Gene Transfer into Pig and Goat Fetal Fibroblasts by Co-transfection of tPA Transgene and $Neo^r$ Gene

  • The transfection efficiency of a transgene into pig and goat fetal fibroblast cells (PFF and GFF, respectively) was tested using co-transfection of a human tissue-type plasminogen activator (tPA) transgene and neomycin-resistant ($Neo^r$) gene, followed by G418 selection. To initially test G418 resistance, GFF and PFF were incubated in culture medium containing different concentration of G418 for 2 weeks, and cell survival was monitored over time. Based on the obtained results, the concentrations chosen for G418 selection were 800 ug/ml and 200 ug/ml for GFF and PFF, respectively. For co-transfection experiments, the pBC1/tPA and $Neo^r$ vectors were co-transfected into GFF and PFF ($1{\times}10^6$ cells in each case) using the FuGENE6 transfection reagent, and resistant colonies were obtained following 14 days of G418 selection. We obtained 96 and 93 drug-resistant colonies of GFF and PFF, respectively, only 54 and 39 of which, respectively, continued proliferating after drug selection. PCR-based screening revealed that 23 out of 54 analyzed GFF colonies and 5 out of 39 analyzed PFF colonies contained insertion of the tPA gene. Thus, the experimentally determined transfection efficiencies for tPA gene co-transfection with the $Neo^r$ gene were 42.6% for GFF and 12.8% for PFF. These findings suggest that co-transfection of a transgene with the $Neo^r$ gene can aid in the successful integration of the transgene into fetal fibroblast cells.

태아 섬유아세포로 재구성된 핵치환 소 수정란의 체외발달 (In Vitro Development of Bovine Nuclear Transfer Embryos Reconstructed with Fetal Fibroblasts)

  • 본 연구는 태아 섬유아세포를 이용한 소 핵치환 난자의 융합에 있어서 여러 조건의 전기자극 처리 후 그에 따른 발달율을 조사하였다. 임선 45 일령 태아로부터 섬유아세포를 분리하여 3~4 차례 계대배양후, 이들 단일 섬유아 세포들을 미세조작기를 이용하여 제핵된 난자의 위란강 내에 주입하였다. 본 실험에서는 첫째로, 다른 전기자극 조건의 적용에 따른 융합율 및 발달율을 비교 조사하였다. 180 V/mm 의 조건하에서 다른 조건의 전기자극 시간 (15, 30, 45 $\mu$see)을 처리하였을 때, 15 (45.5%, 120/264)나 30 $\mu$see (43.9%, 106/241) 처리구가 45$\mu$see (23.2%, 58/250) 처리구 보다 높은 융합율을 나타내었다 (P>0.05). 그러나, 융합된 난자의 발달율에 있어서는 처리구 간에 차이가 나타나지 않았다. 다음 실험으로, 핵치환 난자의 체외발달에 있어서 15 $\mu$see 의 전기자극 시간의 조건하에서 다른 전장 (1.5, 1.8, 2.1 kV/cm)에 따른 효과를 조사하였다. 핵치환 난자의 융합이나 발달율에 전장에 따른 차이는 나타나지 않았다 (P>0.05). 마지막으로, 체세포 핵치환과 체외수정에 의한 수정란의 체외 발달율을 비교 조사하였다. 태아 섬유아세포로 재구성된 수정란의 배반포로의 발달율은 27.4% (75/274) 로 나타났으며, 이는 체외수정에 의해 발달한 배반포의 비율(24.5%, 58/237)과 유사하다는 것을 알 수 있었다. 그러나, 핵치환에 의해 유도된 배반포의 평균 세포 수는 체외수정에 의해 유도된 배반포 보다 낮게 나타났다. 결론적으로, 본 실험에서는 체세포로 핵치환된 소 난자의 전기적 융합에 대한 조건 (1.8 kV/cm, 15 $\mu$see)을 확립하였다. 또한, 본 실험에서 나타난 결과들은 체세포로 재구성된 소 난자들이 비록 체외수정에 의해 생산된 배반포보다는 적은 세포수를 보여주었지만, 체외에서 정상적으로 배반포 단계까지 발달할 수 있다는 것을 제시하고 있다.

Characterization and Genetic Profiling of the Primary Cells and Tissues from Mandible of Mouse Fetus and Neonate

  • The stem cell research is emerging as a cutting edge topic for a new treatment for many chronic diseases. Recently, dental stem cell would be possible for regeneration of tooth itself as well as periodontal tissue. However, the study of the cell characterization is scarce. Therefore, we performed the genetic profiling and the characterization of mouse fetus/neonate derived dental tissue and cell to find the identification during dental development. We separated dental arch from mandibles of 14.5 d fetal mice and neonate 0 d under the stereoscope, and isolated dental cells primarily from the tissues. Then, we examined morphology and the gene expression profiles of the primary cells and dental tissues from fetus/neonate and adult with RT-PCR. Primary dental cells showed heterogeneous but the majority was shown as fibroblast-like morphology. The change of population doubling time levels (PDLs) showed that the primary dental cells have growth potential and could be expanded under our culture conditions without reduction of growth rate. Immunocytochemical and flow cytometric analyses were performed to characterize the primary dental cell populations from both of fetus (E14.5) and neonate. Alpha smooth muscle actin (${\alpha}-SMA$), vimentin, and von Willebrand factor showed strong expression, but desmin positive cells were not detected in the primary dental cells. Most of the markers were not uniformly expressed, but found in subsets of cells, indicating that the primary dental cell population is heterogeneous, and characteristics of the populations were changed during culture period. And mesenchymal stem cell markers were highly expressed. Gene expression profile showed Wnt family and its related signaling molecules, growth factors, transcription factors and tooth specific molecules were expressed both fetal and neonatal tissue. The tooth specific genes (enamelin, amelogenin, and DSPP) only expressed in neonate and adult stage. These expression patterns appeared same as primary fetal and neonatal cells. In this study we isolated primary cells from whole mandible of fetal and neonatal mice. And we investigated the characteristics of the primary cells and the profile of gene expressions, which are involved in epithelial-mesenchymal interactions during tooth development. Taken together, the primary dental cells in early passages or fetal and neonatal mandibles could be useful stem cell resources.

소구경 폴리우레탄 인공혈관의 개발을 위한 세포외기질위의 혈관내피세포 배양 (Endothelial Cell Seeding Onto the Extracellular Matrix of Fibroblasts for the Developement of Small Diameter Polyurethane Vessel)

  • A variety of experiments of endothelial cell seeding onto artificial vessels have been performed. To improve endothelialization, one or two components of the extracellular matrix (ECM) have been used as an underlying matrix. In this study, the whole ECM excreted from fibroblasts was used as an underlying matrix. Fetal human fibroblasts were cultured on a polyurethane (PU) sheet. After a conflu; ence was attained, the cytoskeleton and the nuclei of the fibroblast were destroyed using Triton-X. Mitomycin, or irradiation. Omental microvascular endothelial cells from adult human were seeded onto various substrates. After 12 days in culture, the cells were counted. It was observed that the ECM treated by irradiation had the highest cell number. In addition, the cells on this substrate exhibited the most typical endothelial cell morphology. For preliminary animal experiments the PU vessels (inner diameter, 1.5mm) coated with ECM were implanted in the infrarena] abdominal aorta of rat. After the vessels had been implanted for 5 weeks, it was found that the surface of the PU vessels was completely covered with endothelia] cells. In conclusion, we can state that the fibroblast-derived whole ECM makes a better underlying substrate for the endothelialization of small diameter artificial vessels.

돼지 태아섬유아세포 Clonal Lines의 배양 (Culture of Clonal Lines in Porcine Fetal Fibroblast Cells)

  • This study was performed to establish the effective culture condition for the establishment of clonal lines from porcine fetal fibroblast cells. Fibroblasts derived from a pig fetus (Day 50) were cultured and passaged two times before use. A single cell was seeded in 96-well plates, cultured in medium supplemented with different concentrations of FBS, catalase or $\beta$-mercaptoethanol ($\beta$ME), and classified by cell size and morphology. Cells were passaged two times into 4-well dish before freezing. The establishment efficiencies were not different among different concentrations of FBS (0.3 to 5.1%). However, population doubling time (PDT) was significantly decreased by increasing the FBS concentration (P<0.05). The establishment efficiency of $\beta$ME-added group (10.4%) was significantly higher than those of catalase-added and control groups (3.5%, and 3.5%, respectively, p<0.05), and PDT was significantly decreased (23.6 vs 28.1, and 25.5 h, respectively, p<0.05). However, catalase did not show a positive effect on the establishment efficiency. Cell size and morphology did not affect the establishment efficiency and PDT of clonal lines. The result of present study shows that the establishment efficiency of clonal cell lines can be enhanced by the culture in media supplemented with 30% FBS and $\beta$ME.

Inhibition of Human $CD8^+$ Cytotoxic T Lymphocyte (CTL) -mediated Cytotoxicity in Porcine Fetal Fibroblast Cells by Overexpression of Human Cytomegalovirus Glycoprotein Unique Short (US) 2 Gene

  • Xenotransplantation of pig organs into humans is a potential solution for the shortage of donor organs for transplantation. However, multiple immune barriers preclude its clinical application. In particular, the initial type of rejection in xenotransplantation is an acute cellular rejection by host $CD8^+$ cytotoxic T lymphocyte (CTL) cells that react to donor major histocompatibility complex (MHC) class I. The human cytomegalovirus (HCMV) glycoprotein Unique Short (US) 2 specifically targets MHC class I heavy chains to relocate them from the endoplasmic reticulum (ER) membrane to the cytosol, where they are degraded by the proteasome. In this study we transfected the US2 gene into minipig fetal fibroblasts and established four US2 clonal cell lines. The integration of US2 into transgenic fetal cells was confirmed using PCR and Southern blot assay. The reduction of Swine Leukocyte Antigen (SLA)-I by US2 was also detected using Flow cytometry assay (FACS). The FACS analysis of the US2 clonal cell lines demonstrated a substantial reduction in SLA-I surface expression. The level (44% to 76%) of SLA-I expression in US2 clonal cell lines was decreased relative to the control. In cytotoxicity assay the rate of $CD8^+$ T cell-mediated cytotoxicity was significantly reduced to 23.8${\pm}$15.1% compared to the control (59.8${\pm}$8.4%, p<0.05). In conclusion, US2 can directly protect against $CD8^+$-mediated cell lysis. These results indicate that the expression of US2 in pig cells may provide a new approach to overcome the CTL-mediated immune rejection in xenotransplantation.

배양액내 지방함량의 변화가 신생흰쥐 피부섬유아세포의 노화와 지질과산화물 생성에 미치는 영향 (The Effect of Lipid Concentration in Culture Medium on Senescence and Lipid Peroxides Production of Fibroblast from Neonate Rats)

  • This study was performed to investigate the effects of lipid on cellular senescence, lipid peroxide production, and morphological changes. For this study we used primary skin fibroblasts from neonate rats grown in media various lipid contents. Fibroblasts were cultured until they lost their proliferation potential either in control medium (Dulbecco's modified Eagle's medium supplement with 10% fetal bovine serum) or in media supplemented with various concentrations of lipid-cholesterol rice component from bovine serum. Cumulative population doublings(CPD, as an index of cellular life span), and cellular thiobarbituric acid reactive substances (TBARS, as an index of lipid peroxide) concentrations were measured and morphological changes were observed. CPD were shortened with increasing lipid concentration in media ; 28.12 for cells grown in control medium and 13.42, 11.42, and 6.19 for those grown in 0.1%, 1% and 5% lipid rich components containing media, respectively. Cellular proliferation ratios were those grown in 5% lipid rich components containing media were delayed and they were degenerated soon. TBARS concentrations were increased with increasing concentration of lipid in media. Morphological changes were observed in cells grown in control medium by cellular senescence. Especially lipid droplets were observed in cells grown in 5% lipid rich components containing media. Therefore it seems that lipid contents in media had an effect on cellular proliferation and cellular life span, possibly via lipid peroxide production.
