출혈성 쇼크로 인한 급성 폐손상의 발병기전과 아스피린의 효과 (Effects of Aspirin on the Pathogenesis of Acute Lung Injury in Rats Subjected to Hemorrhage)

  • 연구배경 : 아스피린이 출혈성 쇼크시 나타나는 급성 폐손상에 미치는 효과와 혈장 및 폐세척액 내 ferritin 농도변화를 알아보고자 본 연구를 시행하였다. 방 법 : 실험동물은 체중 350g 내외의 Sprague-Dawley 종 흰쥐를 사용하였고, 혈압측정 및 출혈을 시키기 위하여 catheter를 양쪽 대퇴동맥에 삽입하였다. 수술 후 polygraph를 이용하여 평균동맥압을 기록하였으며, 출혈은 withdrawal pump를 이용하여 5분간 체중 kg당 20 ml의 혈액을 출혈시켰다. 실험군은 대조군, 출혈군과 아스피린 처치군으로 분류하였다. 대조군은 출혈군과 동일하게 수술하고 출혈은 시키지 않았으며 나머지 과정은 출혈군과 동일하게 처리하였다. 아스피린 처치군은 출혈 30분 전 대퇴정맥으로 아스피린(10mg/kg)을 주입하였고, 출혈군은 체중 당 동일한 양의 생리식염수를 주입하였다. 출혈 2시간 후의 폐손상 정도와 아스피린이 이에 미치는 효과를 알아보기 위하여 폐장내 myeloperoxidase 활성도와 폐세척액 내의 단백함량과 백혈구 수 및 혈장 ferritin 농도와 폐세척액 내 ferritin 농도를 측정하였다. 결 과 : 폐장내 myeloperoxidase 활성도와 폐세척액 내의 단백함량과 백혈구수는 출혈 후 대조군에 비해 유의하게 증가하였다. 이러한 반응은 아스피린 전처치에 의하여 효과적으로 차단되었다. 혈장 및 폐세척액 내 ferritin 농도는 출혈 후 크게 증가하였는데, 아스피린 전처치로 반응이 억제되었다. 결 론 : 심한 출혈 후에 생기는 급성 폐손상은 아스피린 전처치로 효과적으로 예방될 수 있으며, 출혈 후 증가하는 혈장 및 폐세척액 내 ferritin 농도는 급성폐손상이 나타날 수 있는 환자에서 조기진단을 위한 생체지표 로 활용될 수 있다고 생각된다.

대한민국 성인에서 대사증후군 및 대사증후군 구성요소의 증가와 혈청 Ferritin의 관련성 (The Association of Serum Ferritin and Metabolic Syndrome and Metabolic Syndrome Score in Korean Adults)

  • 본 연구는 국가자료인 2010~2012년도 국민건강영양조사 자료를 이용하여 제 2형 당뇨병을 가진 대한민국 20세 이상 성인 16,096명(남성, 6,840명; 폐경 전 여성, 4,916명; 폐경 후 여성, 4,340명)을 대상으로 대사증후군(MetS) 및 대사증후군 위험요소의 증가(MSS)와 혈청 Ferritin수준의 관련성을 평가하고자 실시하였다. 대사증후군의 발생률은 16,096명 중 3,978명으로 24.7%(남성, 24.6%; 폐경 전 여성, 11.1%; 폐경 후 여성, 40.3%)이었다. 본 연구의 주요결과는 첫째, 관련변수를 보정한 후의 결과에서, 혈청 ferritin 수준은 비 대사증후군(남성, $111.08{\pm}1.01ng/mL$; 폐경 전 여성, $32.26{\pm}0.50ng/mL$; 폐경 후 여성, $63.26{\pm}0.98ng/mL$)에 비하여 대사증후군(남성, $132.25{\pm}1.98ng/mL$; 폐경전 여성, $39.89{\pm}1.49ng/mL$; 폐경 후 여성, $73.45{\pm}1.14ng/mL$)에서 유의하게 증가하였다(p<0.001). 둘째, 남성과 폐경 전 및 폐경 후 여성 모두에서, 혈청 ferritin 수준은 대사증후군 위험요소의 증가함에 따라 증가하였다(p<0.001). 결론적으로, 대한민국 성인에서 대사증후군 및 대사증후군 위험요소의 증가는 혈청 ferritin 수준을 증가시킨다.

한국 성인에서 사구체여과율 및 요 중 미세알부빈/크레아티닌 비율과 페리틴의 관련성 (Relationship between the Estimated Glomerular Filtration Rate and the Urine Microalbumin/Creatinine Ratio and Ferritin in Korean Adults)

  • 본 연구는 대한민국 성인에서 eGFR 및 uACR과 Ferritin의 관련성에 대한 연구이다. 2012년 국민건강영양조사자료에서 20세 이상의 4,948명을 대상으로 관련변수를 보정한 후, 만성신장질환(CKD, eGFR<$60mL/min/1.73m^2$) 및 알부민뇨($uACR{\geq}30mg/g$)에 따른 페리틴 수준을 분석하였다. 만성신장질환군의 ferritin 수준($M{\pm}SE$) [$103.04{\pm}6.59mL/min/1.73m^2$; 95% confidence interval (CI), 90.12~115.96]은 정상군($84.87{\pm}1.16mL/min/1.73m^2$; 95% CI, 82.59~87.14)에 비하여 유의하게 높았다(P=0.007). 그러나 정상군($82.72{\pm}4.09mg/g$; 95% CI, 74.71~90.73)과 알부빈뇨군($82.72{\pm}4.09mg/g$; 95% CI, 74.71~90.73)의 ferritin 수준은 유의한 차이가 없었다(P=0.487). 결과적으로, 대한민국 성인에서 만성신장질환과 ferritin수준은 양의 상관관계가 있었지만, 알부빈뇨에서는 유의한 차이가 없었다.

Apostichopus japonicas (Echinodermata; Holothuroidea)에서 온도 스트레스에 의한 Hsp90 및 Ferritin 유전자의 발현 (The Expression of Hsp90 and Ferritin Genes under Thermal Stress in the Sea Cucumber (Apostichopus japonicas))

  • The Apostichopus japonicus is an important species in some Asia countries including Korea, China and Japan. The purpose of the present study was to investigate the differential gene expression of heat shock protein90 (Hsp90) and ferritin as a biomarker for the thermal stress during water temperature rising in the sea cucumber, A. japonicus. The A. japonicus (1.4 g) was cultured in incubator of separate temperature ($15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$ and $30^{\circ}C$) for each 0, 3, 6, 12, 24, 48 hours. The mRNA expression levels of Hsp90 and ferritin were examined using RT-PCR assay. Results showed that, the expression of Hsp90 mRNA was not significantly changed at $15^{\circ}C$. The expression of Hsp90 mRNA was significantly increased at high temperature such as $20^{\circ}C$ and $25^{\circ}C$. Furthermore, Hsp90 mRNA was early increased at $25^{\circ}C$ than $20^{\circ}C$. The ferritin mRNA was similar expression pattern with Hsp90. But, Hsp90 mRNA was more sensitive than ferritin mRNA at high thermal stress. These results indicate that Hsp90 and ferritin mRNAs were involved in the temperature changes response and may be play an important role in mediating the thermal stress in A. japonicas.

저열량 생즙 식이가 성인의 혈청 페리틴 농도에 미치는 영향 (Effects of a Low-Calorie Raw Juice Diet on the Level of Serum Ferritin in Korean Adults)

  • This study examines the effects of a low-calorie raw juice diet on the level of serum ferritin in adults and analyzes nutrient intake from the diet. There were significant differences between juices; the highest calorie was provided by pear juice, highest crude protein, vitamin A, and vitamin B2 levels were from green Juice 1; and highest vitamin C and vitamin B1 levels were from fruit juices. The ratio of estimated energy requirements (EER) for the participants was 56.2% from the raw juice diet. The percentages of recommended intake (RI) from the raw juice diet of protein (57.9%), dietary fiber (19.1%), niacin (6.2%), calcium (0.1%), and magnesium (0.2%) were lower than 75%. However, those of RI of vitamin A, vitamin B1, vitamin B2, vitamin B6, and vitamin C were 1796.5%, 7481.7%, 1915.5%, 30858.7%, and 7500%, respectively, exceeding the tolerable upper intake level (UL) for vitamin A, vitamin B6, and vitamin C. There were significant decreases in weight, the body mass index (BMI), body fat mass, and skeletal muscle mass in males and females. After the diet program, serum iron and SOD (superoxide dismutase) showed significant decreases, whereas RBC, hemoglobin, hematocrit, and serum ferritin showed significant increases. There were negative correlations between serum ferritin and weight and between serum ferritin and skeletal muscle mass for all participants. There were negative correlations between serum ferritin and skeletal muscle mass for males and between serum ferritin and body fat mass for females. These results suggest that a raw juice diet can supplement a regular diet to prevent excess or deficient nutrient intake.

Ferritin Light Heavy Chain 유전자가 도입된 인삼형질전환체의 단일배발생을 통한 식물체의 기내증식 (In vitro Propagation of Transgenic Ginsengs Introduced with Ferritin Light Heavy Chain Gene through Single Embryo Culture)

  • Ferritin light heavy chain (FLHC) gene는 일부 중금속과 결합, 저장 및 운반하여 무독화 시킬 수 있는 것으로 알려져 있다. Fe 관련 유전자인 FLHC유전자를 식물 발현용 promoter인 35S promoter와 Tnos를 사용하여 식물 형질전환용 vector를 재조합하였다. 식물세포형질전환용 binary vector는 상기 cassette vector가 조립이 매우 양호하며 border sequence를 가지고 있는 pRD400 binary vector를 사용하여 최종적으로 가나마이신 내성 유전자 (NPT II gene)와 tadpole ferritin heavy chain gene 및 human ferritin light chain gene를 함유하고 있는 binary vector를 재조합하였다. Binary vector의 아그로박테리움에 도입은 triparental mating 방법에 의하여 수행하여 AB배지 및 가나마이신 함유 배지에서 disarmed Ti-vector를 가지고 있는 Agrobacterium tumefaciens MP90/FLHC을 선발하였다. FLHC 유전자 도입된 식물형질전환용 binary vector를 이용하여 형질전환방법을 변형하여 많은 embryo를 유도하였으며 유도된 embryo들은 GA 10mg/L가 첨가된 배지에 지상부를 유도하였다. 형질전환체식물체의 정상적인 생장을 유도하기 위해 최적의 배양조건을 조사하였던 바, 비교적 1/3 MS배지에서 뿌리의 생장과 지상부의 생장이 균일하게 생장하는 경향을 보였으며, 뿌리와 줄기가 잘 발달된 약 7cm의 유식물체를 대량으로 증식하여, 모래와 흙이 1:1로 혼합된 토양에 옮겼다.

재조합 E. coli로부터 발현되는 철단백질의 분리 및 정제

  • Iron is an essential nutrient for most organisms, which supplied to them in a protein-iron complex known as ferritin. Ferritins are multimeric proteins found in prokaryotes, plants and animals. They are consisted of spherical shell of 24 subunits defining a cavity of about 8nm in diameter, where an iron core is laid down. Expression of ferritin in recombinant E. coli at $37^{\circ}C$ led to the accumulation of recombinant ferritin. Insoluble form of ferritin was separated from disrupted cells, followed by various primary separation steps with two kinds of buffers. Collected samples from the primary steps were purified by DEAE-cellulose gels packed in a column. The fractions from the DEAE column were assayed to gain the amount and the purity of ferritin by using HPLC and SDS-PAGE.

  • Ferritin, an iron-storage protein, is found in bacteria and some animal tissues such as liver, spleen, and bone marrow. It is more effective and causes less side reactions than the traditional ferrous sulfate, and thus has been used primarily to treat iron deficiency in pregnancy anaemia. Currently, the ferritin extracted from the bovine and equine spleens are sold as a commercial product. Its market is estimated as several hundreds of million US dollars. However, because of the recent warnings against the viral diseases of animal origins such as mad cow disease, a safer ferritin is sought after. Our research team has successfully developed a production process for recombinant human ferritin. Its expression titer from yeast is high enough to be economically viable, and its particle formation characteristics are as effective as well.

Influence of Site-Directed Mutagenesis on Protein Assembly and Solubility of Tadpole H-chain Ferritin

  • In order to understand the influence of ferroxidase center on the protein assembly and solubility of tadpole ferrin, three mutant plasmids, pTH58K, pTH61G, and pTHKG were constructed with the aid of site-directed mutagenesis and mutant proteins were produced in Eshcerichia coli. Mutant ferritin H-subunits produced by the cells carrying plasmids pTH58K and pTHKG were active soluble proteins, whereas the mutant obtained from the plasmid pTH61G was soluble only under osmotic stress in the presence obtained from the plasmid pTH61G was soluble only under osmotic stress in the presence of sorbitol and betaine. Especially, the cells carrying pTH61G together with the plasmid pGroESL harboring the molecular chaperone genes produced soluble ferritin. The mutant ferritin H-subunits were all assembled into ferritin-like holoproteins. These mutant ferritns were capable of forming stable iron cores, which means the mutants are able to accumulate iron with such modified ferroxidase sites. Further functional analysis was also made on the individual amino acid residues of ferroxidase center.

농촌 지역 주민의 혈청 Ferritin 수준과 대사증후군과의 관련성 (Serum Ferritin and Risk of the Metabolic Syndrome in Some Korean Rural Residents)

  • Objectives : The purpose of this study was to examine the relationship between serum ferritin and the metabolic syndrome (MS). Methods : We conducted a cross-sectional study of 1,444 adults over age 40 and under age 70 that lived in a rural area and participated in a survey conducted as part of the Korean Rural Genomic Cohort Study (KRGCS). The MS was defined as the presence of at least three of the followings : elevated blood pressure, low high density lipoprotein cholesterol, elevated serum triglycerides, elevated plasma glucose, or abdominal obesity. After adjustment for age, alcohol intake, menopausal status, body mass index (BMI), high sensitivity C-reactive protein (hs-CRP), and alanine aminotransferase (ALT), odds ratios (ORs) for the prevalence of the MS by sex were calculated for quartiles of serum ferritin using logistic regression analysis. Results : The MS was more common in those persons , with the highest levels of serum ferritin, compared to persons with the lowest levels, in men (37.1% vs. 22.4%, p=0.006) and women (58.8% vs. 34.8, p<0.001). In both sexes, the greater the number of MS components presents, the greater the serum ferritin levels. After adjustment for age, alcohol intake, and menopausal status, the OR for metabolic syndrome, comparing the fourth quartile of ferritin with the first quartile, was 2.21 (95% confidence interval; CI=1.26-3.87; p-trend=0.024) in men and 2.10 (95% CI=1.40-3.17; p-trend=0.001) in women. However, after further adjustment for BMI, hs-CRP, and ALT, the ORs were statistically attenuated in both sexes. Conclusions : Moderately elevated serum ferritin levels were not independently associated with the prevalence of the MS after adjusting for other risk factors. Further studies are needed to obtain evidence concerning the association between serum ferritin levels and the MS.
