Optimization of Conditions for Extractive Ethanol Fermentation in an Aqueous Two Phase System (수성이상계 에탄올 추출발효 조건의 최적화에 관한 연구)

  • This study was undertaken with objective of optimizing the conditions of fermentation in an aqueous two-phase system which is composed of polyethylene glycol (PEG) 20000 and crude dextran (Dx). The data were obtained and analyzed using the Box-Wilson's experimental design protocol and the response surface methodology. To reach this end a multilinear polynomial regres- sion model was developed, which can be utilized for the purpose of optimizing the extractive fermentation. Optimum conditions for batch fermentation with aqueous two phase system were found to be at 4.2~5.4% PEG/3.2~4.2% Dx range. The composition of the center was 4.8% PEG/ 3.6% Dx. Optimum operating conditions for initial sugar concentration and fermentation time were approximately 160 g/l, and 21~22 hr, respectively. Fermentation in the aqueous two phase system composed of 5% PEG/4% Dx showed increase of 23% in ethanol concentration, of 9.5% in ethanol yield, and of 19% in ethanol productivity as compared to the case of fermentation of neat Jerusalem artichoke juice.

Alcohol Production from Whey in Batch and Continuous Culture of Kluyveromyces fragilis.

  • In order to develop the whey beverage, we examined the optimum conditions for alcohol fermentation by Kluyveromyces tragilis ATCC 46537. The optimum conditions for alcohol production by K. fragilis ATCC 46537 were as follows; pH 4.5, $30^{\cir}C$, with a supplement of 50 g/l of lactose. To develop a continuous production of alcohol from whey, we compared batch fermentation with continuous iermentation in conjunction with UF system. Batch fermentation produced 11.0 g/l of alcohol, whereas pseudocontinuous and continuous fermentation with UF system produced 8.5 g/l of alcohol. To increase the alcohol production, we added 50 g/l of lactose to both fermentations. Batch fermentation with lactose supplement produced 15.7 g/l of alcohol and continuous fermentation with lactose supplement in conjunction with UF system produced 15.0 g/l of alcohol. These results clearly demonstrate that the UF system can be used to increase the alcohol production from whey, supplemented with exogenous lactose.

  • Recently, people are consuming more fermented food for its health benefits. When we think of fermented food, skate comes to mind. However, there are different preferences depending on the degree of fermentation. Thus, if there is a system that shows the degree of the fermentation on skate, people can eat according to their taste. This paper proposes a design of skate fermentation monitoring system for consumer taste using the smart RFID tag. The proposed system consists of the RFID tag, $NH_3$ sensor, reader, and server. In order to confirm the usefulness of the proposed system, we performed experiments on the skate fermentation. The proposed monitoring system can show the skate fermentation at 4 grades: None, Low, Medium, and Strong. With the smart RFID tag, we successfully estimated the skate fermentation.

Effects of Artificial Filaments Equipped in the Aeration Tank of Aerobic Fermentation System on the Removal Efficiency of Nitrogen of Swine Wastewater Containing High Nitrogen (담체설치가 고질소함유 양돈폐수의 호기발효에 미치는 영향)

  • This study was conducted to investigate the effects of artificial filaments equipped in the aeration tank of aerobic·fermentation system on the removal efficiency of swine wastes which were fermented an aerobically and thus containing high nitrogen. Two aerobic fermentation system each consist4s of 4 tanks ; storage tank, 1st and 2nd aeration tank and settling tank were run before and one or three weeks after equipment of artificial filament in the aeration tanks. Total solids concentration tended to increase by aerobic fermentation in all running periods. However, decreased(P<0.05) total nitrogen concentration was shown three weeks after the equipment of artificial filament. Ammonia nitrogen concentration also largely decreased(p<0.05) in both running periods of one and three weeks after equipment of artificial filaments. These results suggest that the artificial filaments may improve the removal efficiency of nitrogen in swine wastewater containing high nitrogen during aerobic fermentation.

  • In this study, we developed a "Korean-style button mushroom cultivation Compost post-fermentation system." The purpose is to increase farm income by reducing the labor force of button mushrooms farmers and shortening the production cycle. The "Korean-style button mushroom cultivation Compost post-fermentation system" was designed to reflect the reality of domestic button mushroom farmers. By reducing the temperature difference of the fermentation Compost in the fermentation system, the company produces a button mushroom Compost that ensures uniform quality. As a result of the performance experiment, the working time of the Compost post-fermentation system was shortened by 40 hours. The number of aerobic bacteria and actinomyces that help the button mushrooms Compost increased. Filamentous bacteria that deteriorate the quality of mushrooms have been sterilized.

Indirect Estimation of CH4 from Livestock Feeds through TOCs Evaluation

  • Thirty-five available feeds were fermented in vitro in order to investigate their soluble total organic carbon (TOCs) and methane ($CH_4$) production rate. A fermentation reactor was designed to capture the $CH_4$ gas emitted and to collect liquor from the reactor during in vitro fermentation. The results showed that $CH_4$ production rate greatly varied among feeds with different ingredients. The lowest $CH_4$-producing feeds were corn gluten feed, brewer's grain, and orchard grass among the energy, protein, and forage feed groups, respectively. Significant differences (p<0.05) were found in digestibility, soluble total organic carbon (TOCs), and $CH_4$ emissions among feeds, during 48 h of in vitro fermentation. Digestibility and TOCs was not found to be related due to different fermentation pattern of each but TOCs production was directly proportional to $CH_4$ production (y = 0.0076x, $r^2$ = 0.83). From this in vitro study, TOCs production could be used as an indirect index for estimation of $CH_4$ emission from feed ingredients.

Preliminary survey of swine internal parasites at the sawdust fermentation floor system (발효톱밥돈사에 대한 돼지 내부기생충 조사)

  • Preliminary survey on the prevalence of swine internal parasites was carried out at 91 pens of 4 piggeries being operated by the sawdust fermentation floor system in Kyunggi province. To investigate the effects of viability of eggs., larva and cysts, temperatures at the underlayer of the sawdust floor and rooms of pens were taken three times a day for 8 days. In a pig farm which has been operated by the sawdust fermentation floor system for G years, eggs and larva of Ascaris suum, Trichuris suis, Oesophagostomum sp., Hyostrongylus sp. and Strongyloides ransomi were detected from 42(70.0%) among 60 pens. Cysts and oocysts of coccidia, Balantidium coli, Entamoeba sp. were also detected from 50(83.3%) among 60 pens. In three pig farms which have been operated by the sawdust fermentation floor system for one year, eggs and larva of nematoda were not detected at all, and the contamination rates of protozoan cysts and oocysts was relatively low situation. The change of the temperature in the pig room was variable according to outside condition, whereas that in underfloor was consistently stable between $28^{\circ}C$ to $30^{\circ}C$ which was strictly suitable for the development of eggs, larva and cysts.

  • The acetone-butanol fermentation by C. acetobutylicum has gained increasing attention for the following reasons. First, the finite supply of petrochemical resources, combined with increasing concern over global environmental effects and the unstable nature of the price of petroleum has renewed interest in the development of fermentation technology that allows utilzation of biomass wastes for the production of alcohol. Second, it serves as excellent model system for understading the regulation and molecular biology of tightly regulated complex primary metabolism, and for applications of metabolic engineering. In this review various aspects of acetone-butanol fermentation by C. acetobutylicm including strain and fermentation characteristics, enzyme regulation, and solvent formation mechanism, and product recovery and summarized.

  • Bioethanol has recently attracted much attention as a sustainable and environmentally friendly alternative energy source. This study aimed to develop a potential process for bioethanol production by fed-batch fermentation using instant dry yeast. To obtain the highest cell growth, we studied the influence of the initial sugar concentrations and pH of sugarcane molasses in batch fermentation. The batch system employed three levels of sugar concentrations, viz. 10%, 15%, 20% (w/v), and two levels of pH, 5.0 and 5.5. The highest cell growth was achieved at 20% (w/v) and pH 5.5 of molasses. The fed-batch system was then performed using the best batch fermentation conditions, with a molasses concentration of 13% (w/v) which resulted in high ethanol concentration and fermentation efficiency of 15.96% and 89%, respectively.
