A Useful Material Production from Whey : Effect of Carbon Sources on Zooglan Production by Zoogloea ramigera (유청으로부터 유용물질 생산 : Zoogloea remigera에 의한 Zooglan 생산에서 탄소원의 영향)

  • Effects of carbon sources on zooglan production by Zoogloea ramigera were investigated. The production of zooglan was varied according to the carbon sources used. The largest amount of zooglan was obtained when lactose was used as carbon source and fermentation broth with lactose showed the higher viscosity. The effects of carbon sources were in decreasing order of lactose, glucose, galactose and sucrose. The viscosities of purified zooglan solutions (5g/L) obtained from different carbon sources were measured. When lactose was used, the viscosities of zooglan solutions was quite high and other carbon sources such as glucose and galactose gave little lower viscosities than lactose but sucrose gave very low values. On the other hand, it could be postulated that most of lactose is hydrolyzed by intracellular ${\beta}$-galactosidase.

  • A mixed Lactobacillus acidophilus and Propionibacterium freudenreichii was cultured in the $10\%$ whey broth with $0.5\%$ yeast extract for 4 days. Viable cell numbers of Lactobacillus acidophilus and Propionibacterium freudenreichii were $2.4\ties10^9$ cfu/mL after 36hr and $9.42\times10^8$ cfu/mL after 96 hr. Consumption rate of lactose during fermentation was $87.5\%$. Propionic acid was produced 18.5g/L and acetic acid was produced 4.8g/L, as a result of that, comparison of propionic acid and acetic acid was 3.8:1 Volume of white pan bread was decreased as added amount of whey was increased. Hardess of white pan bread was decresed as added amount of whey was increased. Preservation period of white pan bread with $10\%$ whey fermented product elongated 2 day compared to control.

Control of Both Foam and Dissolved Oxygen in the Presence of a Surfactant for Production of $\beta$-Carotene in Blakeslea trispora

  • A production of $\beta-Carotene$was attempted in a fed-batch culture of Blakeslea trispora by controlling both foam and dissolved oxygen in the presence of surfactant, Span 20. Results obtained from the shake flask cultures indicated that a high concentration of dissolved oxygen was needed for both cell growth and $\beta-Carotene$ synthesis, and the optimal concentration of glucose was found to be in the range of 50-100 g/l. In order to maintain the dissolved oxygen concentration level at higher than 50% of air saturation, pure oxygen was automatically sparged into the medium with air. Foam was controlled by bypassing air from the submerged aeration to the headspace in response to the foam that was caused by Span 20. High agitation speed was found to be detrimental to the cell growth due to shear damage, even though it provided sufficient dissolved oxygen. On the other hand, a low aeration speed caused stagnant regions in the fermentor because of improper mixing. Thus, for the fed-batch operation, agitation speed was increased gradually from 300 to 700 rpm to prevent cell damage at the initial stage of fermentation and to give efficient mixing for a viscous culture broth as the culture proceeded. By controlling dissolved oxygen and foam, a high concentration of $\beta-Carotene$otene (1,190 mg/l) was obtained in 6 days of the fed-batch culture of B. trispora with 2.5% of the dry cell weight, which was approximately 5 times higher than that of the batch cultures.

  • The mycotoxins, $Aflatoxin\;B_1(B)$ and Ochratoxin A(A), were measured from the various mejus manufactured under the artificial or natural conditions by the indirect competitive ELISA; The various fungi isolated from the Korean traditional home (KTH) made meju collected were observed to produce each mycotoxin mentioned above in the toxin producing broth, but only few in the sterilized cereals of soybean under the artificial conditions. Thus, the isolated fungi were not found to produce both A and B toxins in the artificial conditions. Particularly, the any mycotoxin was not determined at the range of 0.01 to 100 ng per gm of the mejus made under the conditions of KTH widely collected in Korea. The mycotoxins produced by the meju-fermenting fungi were seemed or speculated to be degraded in KTH's mejus under the natural conditions. The species of Mucor involved in the initial stage of fermentation were discussed to be important in the fermentations of KTH mejus.

Optimization of Culture Conditions for the Bioconversion of Vitamin $D_3\;to\;1{\alpha}$,25-Dihydroxyvitamin $D_3$ Using Pseudonocardia autotrophica ID9302

  • We assessed the ability of a Pseudonocardia sp. from soil samples to bioconvert vitamin $D_3$. The optimal culture conditions for the bioconversion of vitamin $D_3$ to active $1{\alpha}$,25-dihydroxyvitamin $D_3$ were investigated by varying the carbon and nitrogen sources, the metal salt concentrations, the initial pH, and the temperature. Microbial transformations were carried out with the addition of vitamin $D_3$ dissolved in ethanol. They were sampled by extraction with methanol-dichloromethane and the samples were examined by HPLC. Optimum culture conditions were found to be 0.4% yeast extract, 1% glucose, 3% starch, 1% fish meal, 0.2% NaCl, 0.01% $K_2HPO_4$, 0.2% $CaCO_3$, 0.01% NaF, and pH 7.0 at $28^{\circ}C$. The optimal timing of the addition of vitamin $D_3$ for the production of calcitriol by Pseudonocardia autotrophica ID9302 was concurrent with the inoculation of seed culture broth. Maximum calcitriol productivity and the yield of bioconversion reached a value of 10.4mg/L and 10.4% respectively on the 7th day in a 75L fementer jar under the above conditions.

Mass Cultivation and Secondary Metabolite Analysis of Rhodobacter capsulatus PS-2 (광합성세균 Rhodobater capsulatus PS-2의 대량배양 최적화 및 대사산물 분석)

  • Plant growth promoting (PGP) hormones, which are produced in a small quantity by bacteria, affect in plant growth and development. PGPs play an important role on the crop productivity in agricultural field. In this study, a photosynthetic bacterial strain producing the PGP was isolated from paddy soil. Bacterial isolate was gram negative, rod-shaped and motility positive. From the 16s rRNA gene sequence analysis, the isolate was identified as Rhodobacter capsulatus PS-2. The mass cultivation of R. capsulatus PS-2 was optimized by considering of the carbon, nitrogen and inorganic salt sources. Optimal medium composition was determined as Na-succinate 4.5 g, yeast extract 5 g, $K_2HPO_4$ 1 g, $MgSO_4$ 5 g, per liter. From the result of 500 L fermentation for 2 days using the optimal medium, the viable cells were $8.7{\times}10^9cfu/mL$. R. capsulatus PS-2 strain produced the carotenoid and indole-3-acetic acid (IAA). The carotenoid extraction and quantitative analysis were performed by HCl-assisting method. Total carotenoid contents from R. capsulatus PS-2 culture broth were measured as $7.02{\pm}0.04$ and $6.93{\pm}0.05mg/L$ under photoheterotrophic and chemoheterotrophic conditions, respectively. To measure the productivity of IAA, colorimetric method was employed using Salkowski reagent at optical density 535 nm. The results showed that the highest content of IAA was $197.44{\pm}5.92mg/L$ in the optimal medium supplemented with 0.3% tryptophan.

Statistical Selection of Amino Acids Fortifying a Minimal Defined Medium for a High-level Production of the Kringle Fragments of Human Apolipoprotein(a)

  • A synthetic defined medium, fortified with amino acids, was developed for the stable production of the kringle fragments of human apolipoprotein(a) (apo(a)), rhLK68. Using a complex rich medium containing yeast extract and a high-cell-density fed-batch culture, the expression level of rhLK68 reached 17% of the total cellular protein, which corresponded to $5\;g\;l^{-1}$ of the culture. To replace the complex media with chemically defined media, several amino acids that positively affect cell growth and gene expression were chosen by a statistical method. The various combinations of the selected amino acids were tested for its fortifying effect on a minimal defined medium. When glutamine only was added, the overall expression level of rhLK68 reached 93% of the complex rich medium increasing the specific expression level by 22.4% and decreasing the cell growth by 24%. Moreover, the addition of glutamine resulted in a 2-fold increase in the concentration of rhLK68 in the culture broth, compared with the minimal defined medium. The synthetic defined media developed in this study could be generally applied to high-cell-density cultures of the recombinant Escherichia coli BL21(DE3), especially for the production of therapeutic proteins that require a strict quality control of the culture media and fermentation processes.

The R-Plasmid Transfer and Elimination of Shigella Cultures (Shigella균속의 항균제내성, 전달성 R-plasmid 및 제거에 관한 연구)

  • On hundred and forty stains of shigella cultures isolated from the twelve hygiene laboratories of cities and provincial general hospital laboratories in 1983 were tested for their resistance to thirteen antimicrobial drugs and their R-plasmid transfer. Antimicrobial drugs were used amikacin, ampicillin, cephalothin, chloramphenicol, gentamicin, kanamycin, nalidixic acid, rifampicin, streptamycin, tetracycline, tobramycin, cefoperazone and piperacillin. All strains were resistant to one or more of thirteen antimicrobial drugs but 94.3% were susceptible to amikacin, gentamicin and tobramycin of total isolated. The most strains commonly found resistance was to chloramphenicol (94%) followed by streptamycin (93%), tetracyline (92%) piperacillin (90%) ampicillin (83%), cefoperazone (42%), nalidixic acid (14%), cephalothin (17%), rifampicin (22%) and kanamycin (6%), sixty percent of strains among 140 were resistance to ampicillin, chloramphenicol, streptomycin, tetracycline at the same time. The transfer of drug resistance by conjugation was tested and ninety four strains (94.3%) were resistant to one or more drugs were found to transfer their drug resistance of E. coli. percentage of transfer frequency by conjugation was one strains (54%), the transfer frequency of drug resistance varied by donor strains and recipients, but not by selecting drugs. Resistance to nalidixic acid was not transferred by conjugation to recipients. Percentage of plasmid curing after the treatment of acriflavine, acridine orange was about 8%. Among strains cured two strains were tested compare original strains with them in biochemical properties in arginine dihydrolase and arabinose fermentation reaction. It was found to growth curves of No.2 shigella flexneri, serotype 1b, and its derivatives cured with acriflavine in $M{\ddot{u}}ller$ Hinton broth medium (pH 7.4, $38^{\circ}C$) by temperature Gradient Biophoto Recorder TN-1120 (Tokyo, Japan).

  • Bacillus licheniformis 9945a releases a natural ${\gamma}-poly(glutamic\;acid)({\gamma}-PGA)$ into fermentation broth and shows a mucoid phenotype on the solid agar medium. Transformation of mucoid cells of Bacillus species has not been simple and straightforward. The transpositional activity of Tn10 in B. licheniformis also has not been own either. Thus, a spontaneous non-mucoid derivative of the B. licheniformis was obtained first. Shuttle vector pHV1248 containing mini-Tn10 was introduced into the non-mucoid derivative by the method of protoplast transformation. The resulting transformant was reverted to the wild mucoid phenotype, and then mutated randomly with the mini-transposon by heat induction. Auxotrophs requiring arginine, lysine, or tryptophan were isolated by replica plating method. Southern blotting and DNA-DNA hybridzation analysis showed that these auxotrophs were generated by mini-Tn10 insertion into the chromosomal DNA. This method of transformation and mutation using pHV1248 would be useful for the generation of diverse mutants of B. licheniformis 9945a.(Received January 24,1997; accepted March 10, 1997)

Chnracterization and Inhibitory Activity on Staphylococcus aureus of a Bacteriocin Produced by Lactobacillus plantarum KU107 (Lactobacillus plantarum KU107이 생산하는 박테리토신의 특성 및 Staphylococcus aureus 억제 작용)

  • A bacteriocin producing lactic acid bacteria was isolated from ground beef and the strain was identified as Lactobacillus plantarum ssp. by use of API carbohydrate fermentation pattern and physiological tests. The bacteriocin produced by L. plantarum KU107 exhibited a good spectrum of activity against foodborne pathogens including Bacillus cereus, Escherichia coli, Listeria ivanovii, Listeria monocytogenes, Pseudomonas aeruginosa, Pseudomonas chlororaphis, Staphylococcus aureus, Staphylococcus intermedius, Salmonella typhimurium and Yersinia enterocolitica. The bacteriocin was active over a wide pH range and stable of heat treatment, and inactivated by treatment with proteases. A bacteriocin from L. plantarum KU107 was effetive in reducing S. aureus in tryptic soy broth. On the ground beef containing S. aureus was added with the crude bacteriocin, S. aureus was inhibited during storage period at 4$\^{C}$.
