Effect of culture pH and media composition on the ratio of tcicoplanin $A_1$ and $A_2$ biosynthesis

  • Teicoplanin is a glycopeptide antibiotic produced by Actinoplanes teichomyceticus novo sp. A TCC 31121. It is active against Gram-positive bacteria and it is under evaluation for use in man. Teicoplanin mixture in fermentation broth contains major amounts of teicoplanin $A_1$ and $A_2$ and a minor amount tcicoplanin of $A_3$. Commercial teicoplanin product is composed of five major components of very similar polarity, designated T-$A_2$-l, 2, 3, 4 and 5, and the more polor component, designated T -$A_3$. The culture conditions were studied in order that hydrophilic teicoplanin $A_2$ components are more produced but hydrophobic teicoplanin $A_1$ with lower bioactivity are less produced in submerged culture. Effects of culture pH and nutrients on the biosynthes ratio of teicoplanin $A_1$ and $A_2$ were confirmed in flask culture using MOPS buffer system through TLC, bioautography and bioassay. It was elucidated that optimal pH is 7.4 for teicoplanin $A_2$ biosynthesis but is 5.2 for teicoplanin $A_1$ biosynthesis, and that trace elements stimulate $A_2$ production but malt extract stimulate $A_1$production.

Evaluation of the role of Lactobacillus casei on alcohol metabolism and liver functions of rats

  • Alcohol consumption causes numerous consequences on the health of the human body. Heavy drinking on a daily base has caused liver diseases. Furthermore, some products such as acetaldehyde produced from alcohol metabolism are more toxic than alcohol itself. This study was carried out to evaluate the role of Lactobacillus casei on alcohol metabolism, especially, the removal of the toxic effect of alcohol. The maximum alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities from L. casei were observed at 4 hr of culture. L. casei was confirmed to produce the ADH and ALDH by the SDS-PAGE. From in vivo test using SD rats with 22% alcoholic drink, blood alcohol concentration (BAC), glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) of the rats feeding the medium containing L. casei were lower than those of the rats feeding the medium containing an alcoholic drink only This demonstrates that the ADH and ALDH produced by L. casei have virtual functions to detoxicate the alcohol in vivo and the fermentation broth of L. casei can be used as an alcohol detoxification drink.

Characterization of Kombucha Beverages Fermented with Various Teas and Tea Fungus

  • Kombucha beverages were made from sweetened tea by Oriental, European and Tibetan tea fungus starters. The hot water extracts o green tea, black tea, Gugija and Omija were mixed with white and/or brown sugar, and were fermented under a static culture at 3$0^{\circ}C$. Titrable acidity, pH, color and cellulose production in kombucha beverages were evaluated. All tea fungus starters showed a higher acid production in green/black tea extracts rather than Gugija and Omija extracts. In green/black tea extracts Oriental tea fungus produced a kombucha beverage with a higher titrable acidity and lower pH than those of European and Tibetan tea fungus starters. By the static fermentation of green/black tea extract for 18 days, Oriental, Tibetan and European tea fungus starters produced cellulose pellicles of 0.43g, 0.16g, and 0.19 g (dry weight) on the top in the culture, respectively. As a mother starter, the cellulose pellicle was more efficient in acid production compared with tea fungus broth. Oriental/Tibetan mixed tea fungus showed the best acid production in the green/black tea extract supplemented with brown sugar.

  • Cheese 제조시 부산물로 생성되는 whey를 배지로 사용하여 Aspergillus niger를 이용하여 citric acid를 생산하는데 영향을 미치는 여러 가지 요인 중 중요한 요인인 온도와 pH의 영향에 대하여 고찰하였다 15일간 27, 30, 33, $36^{\circ}C$와 pH 2, 3, 4, 5에서 각각 배양하면서 소비된 lactose의 양과 생산된 citric acid의 양을 측정하였다. 생산된 citric acid의 최대 농도는 33.9 g/l(구연산 생산에 쓰여진 유당을 기준으로 할 때 68.26%)이었으며, shaking speed는 citric acid 생산에 직접 영향을 주기보다는 pellet 형성시 그 형태에 영향을 미치는 것으로 나타났다. 배양 온도가 $33^{\circ}C$, pH는 3일때 가장 많은 양의 citric acid가 생산되었다.

미생물이 생산하는 새로운 대사길항물질에 관한 연구 (Studies on the New Antimetabolites Produced by Microorganisms)

  • 미생물이 하는 생산하는 대사길항물질을 screening 한 결과 최소검정배지상에서 Gram 양성균과 Gram 음성균에 다같이 강한 항균작용을 나타내고 L-aspar tic acid와 L-g1utamic acid에 의하여 길항당하는 물질 N-2292를 생산하는 방선균을 분리하였다. 본 생산균을 분류동정한 결과, Streptomyces albulus 로 보거나 혹은 그 유연균으로 동정하였다. N-2292물질의 배양조건을 검토하여 최적조건에서 대량 배양하고 주로 ion 교환수지, Dowex 50을 사용하여 활성물질을 분리정제하였으며 배양액 약 10ι에서 약 40mg의 백색분말을 얻었다. 본물질은 그 의 이화학적 성질로 보아 indole 화합물을 함유하는 peptide 양 물질임을 추정할 수가 있었다. 생물학적 성질은 최소검정배지상에서 Gram 양성 및 Gram 음성세균에 항균력을 나타냈다. 이러한 항균력은 L-Asp. 및 L-Glu.의 첨가에 의해서 조해당하였다.

  • D. bryoniae를 원인균으로 하는 참외 만고병에 대해서 항만고병 활성물질을 생산하는 미생물을 방선균으로부터 선별 분리한 후 배양액으로부터 항균활성물질의 규명을 시도하였다. 항진균 물질과 같은 이차대사산물의 생산에 증가시키는 $K_2$$HPO_4$와 칼슘이온이 포함된 GSS배지에서 방선균 SKM338 균주를 180 rpm, $30^{\circ}C$, 5일 동안 배양하여 얻어진 배양 상등액으로부터 물리화학적인 방법으로 항진균 활성이 있는 물질을 분리 정제한 결과 참외의 만고병에 대한 생물농약으로 개발 가능한 방선균 유래의 항진균성 물질은 NMR, IR, UV 및 Mass spectral data 분석 등을 통해 polyene macrolide계에 속하는 항생물질인 Flavofungin, Fungichromin, Filipins로 밝혀졌으며 이들의 응용을 기대해 본다.

  • 김치가 젖산발효식품으로서의 특성을 유지하면서 보존성을 증진시키는 방안의 일환으로, 김치의 젖산 생성량이 낮은 초기 숙성균인 Leuconostoc mesenteroides의 생육에는 무관하면서 김치의 산패에 관여하는 Lactobacillus plantarum을 선택적으로 저해할 수 있는 항균제를 탐색하기 위하여 10여종의 향신료로부터 얻은 에탄올추출물의 항균효과를 조사하였다. Paper disc법에 의한 향신료 추출물의 항균력을 측정한 결과 10여종의 향신료 추출물중 cardamon, thyme 및 cumin의 에탄올추출물은 Lac. plantarum을 선택적으로 저해시키는 항균효과를 나타내었다. 또한 농도별에 따른 cardamon 및 thyme 추출물은 4%에서, cumin 추출물은 1% 이상의 농도에서 항균효과를 나타내었다. SDS-PAGE 및 투과전자현미경의 결과의 분석을 통하여 향신료 에탄을 추출물의 주요 항균작용은 균체 세포벽 및 세포막을 약화시키며 이로 인하여 균체세포의 내용물을 세포외로 유출시킴으로서 성장을 저해 또는 사멸시키는 것으로 나타났다.

나노여과를 이용한 Sodium Lactate의 회수 (Recovery of Sodium Lactate Using Nanofiltration)

  • The effects of operating pressure, lactate concentration, impurities, and pH on solution flux and lactate rejection in nanofiltration were investigated with model sodium lactate solutions (lactate 10~200g/L) as a model system. In the tested range of pressure(80~140 psig), the solution flux was observed to be proportional to the operating pressure and the rejection of lactate increased only slightly with the pressure. Both of the flux and the rejection decreased with lactate concentration, while the recovery rate of lactate increased. The effects of glucose and yeast extract as impurities on lactate rejection were negligible, but the flux decreased significantly with the addition of yeast extract. At low lactate concentrations, the rejection of lactate increased with pH due to the increased repulsion (Donnan exclusion effect) between lactate ions and membrane surface. But, at high lactate concentrations, the donnan effect was observed to be overwhelmed by the effect of sodium ions added to adjust the pH, and the rejection of lactate decreased with pH. When fermentation broth containing about 89g/L of lactate was nanofiltered, the flux and the rejection of lactate were 2.8L/$m^2$h and 5%, respectively at 120psig. Both of them were slightly lower than those with model solutions. The recovery rate was 2.6mol/$m^2$h.

회분식 발효에서의 전기투석에 의한 젖산의 동시분리 (In situ Separation of Lactic Acid by Electrodialysis in Batch Culture)

  • Effects of lactate concentration, temperature, counter ions, pH as well as voltage (current) in batch electrodialysis (ED) experiments with a 3-compartment unit were investigated. The applied voltage was found to be the most critical factor as expected. The electrodialysis rate increased with the lactate concentration of the source solution. The amount of lactate transferred was limited by the lactate concentration difference between cathode and permeate compartments. The electrodialysis rate did not heavily depend on temperature change. The electrodialysis rate of NH$_{4}$-lactate was faster than that of Na-lactate and both lactates showed the highest electrodialysis rate at a pH of 4.0. A little amount of non-ionic glucose diffused through the anionic membrane to the permeate compartment. To test the effectiveness of the in situ recovery of lactic acid from fermentation broth by ED, three cases of batch culture were carried out; pH control only, ED only, and pH control and ED. The case with both pH control and ED was more efficient than that with pH control only in the aspects of productivity and product yield.

Identification of Phenylethyl Alcohol and Other Volatile Flavor Compounds from Yeasts, Pichia farinosa SKM-l, Pichia anomala SKM-T, and Galactomyces geotrichum SJM-59

  • Three strains of yeasts, Pichia farinosa SKM-1, Pichia anomala SKM-T, and Galactomyces geotrichum SJM-59, produced volatile flavor compounds during fermentation. To investigate these volatile flavor compounds, the liquid culture broth of the three yeast strains were extracted with methylene chloride, and then GC and GC-MS analyses were conducted. Flavor analyses revealed that 5, 12, and 15 kinds of volatile compounds were isolated, and 4, 8, and 11 volatile flavor compounds were identified, respectively. Phenylethyl alcohol was identified with the common volatile flavor compound of Pichia farinosa SKM-1, Pichia anomala SKM-T, and Galactomyces geotrichum SJM-59. 1H-indole-3-ethanol, a precursor of plant growth hormone, was identified from Pichia anomala SKM-T.
