Near-infrared reflectance spectroscopic (NIRS) calibrations were developed for accurate and fast prediction of whole broiler chicken carcass composition. The Feed and Forage Foss systems Model 5000 Reflectance Transport Model 5000 with near-infrared reflectance spectroscopy (NIRS)-WinISI II windows software was used for this purpose. One equation was developed for the prediction of each carcass component. One hundred and fifty freeze dried broiler whole carcass samples were ground in a Cyclotech 1,093 sample mill and analyzed for dry matter, protein, fat, calcium and phosphate. Samples were divided into two sets: a calibration set from which equations were derived and a prediction set used to validate these equations. The chemical analysis values (mean${\pm}$SD) were calculated based on dry matter basis as follows: dry matter: 33.41${\pm}$2.78 (range: 26.41-43.47), protein: 54.04${\pm}$6.63 (range: 36.20-76.09), fat 35.44${\pm}$8.34 (range: 7.50-55.03), calcium 2.55${\pm}$0.65 (range: 0.99-4.41), phosphorus 1.38${\pm}$0.26 (range: 0.60-2.28). One hundred and three samples were used to calibrate the equations and prediction values. The software used was modified to obtain partial least square regression statistics, as it is the most suitable for natural products analysis. The coefficients of determination ($R^2$) and the standard errors of prediction were 0.82 and 1.83 for the dry matter, 0.96 and 1.98 for protein, 0.99 and 1.07 for fat, 0.90 and 0.30 for calcium and 0.91 and 0.11 for phosphorus, respectively. The present study indicated that NIRS can be calibrated to predict the whole broiler carcass chemical composition, including minerals in a rapid, accurate, and cost effective manner. It neither requires skilled operators nor generates hazardous waste. These findings may have practical importance to improve instrumental procedures for quick evaluation of broiler carcass composition.
This study investigated the effects of tannic acid (TA) in the diets for weanling pigs prepared with/without supplemental Fe on performance, hematology, fecal microflora and diarrhea incidence. Limestone and calcium phosphate used in Experiment 1 and 2 were of semi-synthetic and feed-grade quality, respectively; while the trace-mineral premix used in both the experiments was prepared without any added Fe source. In Experiment 1, 108 weaned pigs (Landrace ${\times}$ Yorkshire ${\times}$ Duroc, initially 6.46 ${\pm}$ 1.04 kg BW) were allotted to 3 treatments including control (diet added with $FeSO_4$ and antibiotic), T1 (diet devoid of $FeSO_4$ and antibiotic) and T2 (T1 diet added with 125 mg/kg TA). Each treatment had 4 replicates with 9 pigs in each pen. Feeding of T1 diet had a negative effect on the performance and plasma Fe status of pigs, while addition of TA to T1 diet resulted in performance of pigs comparable to pigs fed the control diet, reduced diarrhea incidence but had a negative influence on the hematological and plasma Fe indices. Additionally, pigs fed T2 diet had fewer (p<0.05) total anaerobic bacteria, Clostridium spp. and coliforms than pigs fed T1 diet, and greater number of Bifidobacterium spp. and Lactobacillus spp. in feces when compared with pigs fed control and T1 diets. In Experiment 2, 144 weaned pigs (Landrace ${\times}$ Yorkshire ${\times}$ Duroc, initially 6.00 ${\pm}$ 1.07 kg BW) were allotted to 4 dietary treatments including control (diet added with $FeSO_4$ and antibiotic) and diets devoid of supplemental Fe added with antibiotic (An), TA and both (AnTA). Each treatment had 4 replicates with 9 pigs in each pen. Addition of An, TA or both to diets devoid of supplemental Fe did not have any effect on performance, blood hematology and plasma Fe but resulted in reduced (p<0.05) diarrhea incidence and lower (p<0.05) fecal coliform population than pigs fed the control diet. These results suggest that TA has a negative influence on blood hematology and plasma Fe status when diets are inadequate in Fe; however, TA reduced diarrhea incidence and might have antimicrobial activity.
We have previously reported recombinant productions of bacteriocins using yeast expression plasmid pAUR123, which contains the alcohol dehydrogenase (ADH) promoter, in Saccharomyces cerevisiae cells and their antibacterial activities. In order to improve the antibacterial activities of bacteriocidal yeast cells, a strong glyceraldehyde phosphate dehydrogenase (GPD) promoter gene of S. cerevisiae was amplified and inserted upstream into bacteriocin genes such as the OR-7, Subpeptin JM4-A or JM4-B gene in the corresponding recombinant yeast plasmid. Yeast cells transformed by the recombinant plasmid containing the GPD promoter represented higher antibacterial activities against both Gram positive B. subtilis and Gram negative E. coli cells compared to those transformed by the corresponding recombinant plasmid containing the ADH promoter. Thus, yeast cells harboring the recombinant plasmid containing the GPD promoter constructed in this study could be applied in the food preservative or animal feed industries.

Continuous immobilized-cell culture was carried out for the production of kasugamycin, a secondary metabolite by a filamentous bacteria, Streptomyces kasugaensis, with an intention of reducing waste generation. A sporulation medium was developed for production of bulk amounts of spores, and the spores were entrapped into celite biosupports for immobilization. It was possible to effectively keep the immobilized-cells inside the reactor during the continuous culture by an efficient immobilized cell separator of decantor type on the outlet of the fermentor. Using this continuous immobilized-cell fermentor system, we investigated the effects of feed substrate and phosphate concentrations on kasugamycin production and chemical oxygen demand(COD). Comparing with the conventional suspended-cell batch culture, the kasugamycin productivity was observed to increase by 2.5 times, whereas COD per unit kasugamycin production decreased by 2.3 times in the continuous immobilized-cell culture. Based on these results, the continuous immobilized-cell system was considered to be a cleaner bioprocess than the conventional batch suspended-cell system.

Effect of the egg yolks from laying hens intubated, p.o., astaxanthin (designated AEY) on the catabolic response overcome of mice was examined. Female ICR mice (6~7 weeks of age) were adapted in a temperature- and humidity-controlled house for one week and randomly divided into 5 groups (6 mice/cage/treatment). Mice were intubated p.o., AEY (5, 10 and 15 mg), control egg yolk (CEY, 10 mg), or fish oil (5 mg) dissolved in 0.2 mL phosphate buffered saline (PBS) every two days for 14 days. At day 15, the 0.1 mL of lipopolysaccharide solution (LPS, 30$\mu\textrm{g}$/0.1 mL 10 mM HEPES) was injected through tail vein, and then, the body weight of mouse and the amount of feed intake were measured over a period of 72 hours. Control group mice were received only PBS and LPS. AEY treatment suppressed the loss of mice body weight in a dose-response manner. Twenty four hours post LPS injection, the reduced body weight per mouse of AEY 5, AEY 10, and AEY 15 mg treatment groups was 3.70, 3.54, and 3.25 g, respectively. Body weight suppression effect of AEY treatment was greater than that of CEY, but less than fish oil. AEY treatment did not alter thymus weight, but increased the weight of spleen or liver. These results indicate that AEY suppressed the loss of body weight by LPS via any function of the spleen and/or liver.
The objective of the present study was to investigate the effect of dietary supplementation of cultured wild-ginseng powder or its fermented culture byproduct on growth performance, blood parameters, carcass and meat quality in finishing pigs. The animals used in the experiment were a total of 36 Landrace×Yorkshire and weighted 65.81±2.02kg. The experimental diets were basis diet, 2.5% wild-ginseng fermented culture byproduct of B. subtilis replaced lupin in basis diet and 0.2% cultured wild-ginseng powder replaced lupin in basis diet to CON, T1 and T2 for 60 days, respectively. The pigs were allotted at 4 pigs per pen with three replicate pens per treatment by completely randomized design. In growth performance, ADG was not significantly different between treatments. ADFI was significantly lower (P<0.05) in T1 and T2 than in CON. Feed/Gain was not different between treatments. In plasma's biochemical composition, total protein was significantly higher (P<0.05) in T1 than in CON. Blood urea nitrogen was not different between treatments. Glucose and albumin were significantly higher (P<0.05) in T1 than in other treatments. Calcium was significantly higher (P<0.05) in T1 than in CON. Inorganic phosphate was significantly higher in T1 than in other treatments. In plasma's lipid composition, triglyceride was significantly higher (P<0.05) in T1 than in other treatments. Total cholesterol was not different between treatments. HDL cholesterol was significantly higher (P<0.05) in T1 than in other treatments. In carcass and meat quality, carcass weight, dressing precent, meat precent and back-fat thickness were not significantly different between treatments. Moisture and crude fat were also not significantly different between treatments. The results indicate that growth performance, carcass and meat quality were not affected but plasma's biochemical and/or lipid composition were affected when replaced with wild-ginseng fermented culture byproduct of B. subtilis and cultured wild-ginseng. Our research indicates that wild-ginseng fermented culture byproduct of B. subtilis and cultured wild-ginseng powder were able to using with pig's diet in finishing period.
This study, including two in vitro experiments and an in vivo experiment were conducted to evaluate effects of Passtein$^{(R)}$ on crude protein degradability, ruminal fermentation characteristics and nutrient digestibility. In in vitro experiment protein degradability was examined using borate-phosphate buffer and neutral detergent, and using protease from Stroptomyces griseus at 39$^{\circ}C$ for 0, 2, 4, 8, 12, and 48 h. In addition, an in vivo experiment was conducted in a switch back design and ruminal fermentation and nutrient digestibility were determined. Four ruminal-fistulated Holstein cows weighing 300kg in mean body weight randomly allotted to 2 treatments (control and Passtein$^{(R)}$ supplementation). Although there was no significant difference on protein fraction between treatments, it appears that Passtein$^{(R)}$ supplementation decreased buffer soluble protein fraction compared to control. Protein degradability was not affected by Passtein$^{(R)}$ from 0 h to 4 h, but decreased at 12 h and 48 h compared to control. Degradation of immediately degradable fraction was higher in Passtein$^{(R)}$ treatment, but degradation of fermentable fraction was lower in Passtein$^{(R)}$ treatment compared to control. The pH and $NH_3$-N concentration tended to increase in Passtein$^{(R)}$ treatment, but VFA production, microbial counts and enzyme activity tended to decrease in Passtein$^{(R)}$ treatment compared to control. In addition, nutrient digestibility in the total tract tended to increase in Passtein$^{(R)}$ treatment compared to control.

The objective of the present study was to investigate the effect of dietary supplementation of the ginseng by-product on growth performance and meat quality in finishing pigs. The animals used in the experiment were a total of 24 Landrace×Yorkshire and weighted 65.81±2.02kg. The experimental diets were basal diet(CON) and 2.5% ginseng by-product(GBP), which replaced lupin in basal diet. The pigs were allotted at 4 pigs per pen with three replicate pens per treatment by completely randomized design. In growth performance, ADFI was significantly lower(P<0.0001) in GBP than in CON. In plasma biochemical composition, total protein(P<0.01), blood urea nitrogen(P<0.03), glucose(P<0.01), albumin(P<0.02), calcium(P<0.01) and inorganic phosphate(P<0.01) were significantly higher in GBP than in CON. Carcass and meat quality were not significantly different between treatments. Total ginsenoside content on meat was significantly higher(P<0.0001) in GBP than in CON. TBARs was significantly lower in GBP than in CON for 6 days(P<0.03) and 12 days (P<0.06), respectively. Our research indicates that plasma biochemical composition, total ginsenoside content and TBARs were affected when replaced with ginseng by-product. Ginseng by-product in the pig diet increased pig muscle ginsenoside, indicating that ginseng by-product can be used as a feed additive.
To meet the need of protein feed and fine more efficient ways of returning waste to resources, we have carried out the study of the production of yeast for foods and feeds from the corn starch cake. The present study includes the method for acid-hydrolysis, the selection of yeast capable of utilizing hydrolyzate of the corn starch cake, and culture condition of Candida tropicalis under the liquid culture and the semisolid culture. Obtained results were as follows. 1. Hydrochloric acid was more excellent on the hydrolysis of the corn starch cake than sulfuric acid, and the yield of sugar was maximum, 57.2%, when the corn starch cake was hydrolyzed with 1.0% of hydrochloric acid at 2.0kg/cm for 30 minutes. 2. As the acid solution content was increased, more sugar was liberatedfrom the mixture, until the acid solution-substrate ratio reached 10:1. Beyond this point, no further increase was observed. To prepare the cultural medium of semisolid fermentation, a acid solution to substrate ratio of 3:1 appeared to be optimum. 3. Out of 6 yeast strains, Candida tropicalis had excellent growth on the hydrolyzate of the corn starch cake, and optimum temperature and initial pH were $30^{\circ}C$ and 6.0 respectively. 4. Optimum liquid medium of Candida tropicalis is ures 0.3%, potassium phosphate monobasic 0.15g and magnesium sulfate 0.04g in 100ml of the hydrolyzate of the corn starch cake, while optimum semisolid medium is ammonium chloride 0.4g, potassium phosphate monobasic 0.1%, magnesium sulfate 0.04%. 5. Candida tropicalis could assimilate the sugar in the hydrolyzate up to more than 88.75%, and a yield of dry yeast reached 19.13% to the corn starch cake under the liquid culture. 6. Compared to the that of the untreated corn starch cake, the cellulose content of the semisolid fermented cake decreased by 3.76% to 14.7%, whereas dry yeast contents increased by 13.89%.
This study was conducted to investigate liquid pig manure (LPM) application rates on the growth characteristics, yield, and feed value of whole crop barley in Yeongsangang and Saemangeum reclaimed tidal land. Electronic conductivity (EC), organic matter (OM), and available phosphate (Av. $P_2O_5$) increased in chemical properties of Yeongsangang and Saemangeum soil as raising LPM application level. As increasing LPM application level, exchangeable $Na^+$ significantly increased in Yeongsangang, while exchangeable $K^+$ significantly increased in Saemangeum. Plant height was not significantly different from LPM 100% to LPM 200% in Yeongsangang and in Saemangeum. Dry matter yield of whole crop barley increased steadily, but crop yield of LPM 200% in Yeongsangang ($10.5ton\;ha^{-1}$) was as much as that of LPM 150% ($10.0ton\;ha^{-1}$). Yield of LPM 200% ($11.2ton\;ha^{-1}$) in Saemangeum was similar to that of LPM 150% ($10.5ton\;ha^{-1}$). Crude protein (CP) increased depending on LPM application level, but total digestible nutrients (TDN) increased regardless of LPM application level. LPM 200% was the highest in TDN yield (Yeongsangang: $7.4ton\;ha^{-1}$, Saemangeum: $6.9ton\;ha^{-1}$), but there was no statistical difference between LPM 150% (Yeongsangang: $6.9ton\;ha^{-1}$, Saemangeum: $6.6ton\;ha^{-1}$) and LPM 200%. From the results described above, optimum rate of LPM for cultivating whole crop barley is considered 100% in Yeongsangang reclaimed tidal land and 150% in Saemangeum reclaimed tidal land, showing that the effect of LPM application is better in Segmentation than that in Yeongsangang for yield of whole crop barley.
