Effects of Meju Powder Supplementation on Lipid Metabolism in Rats Fed Hypercholesterolemic Diet (고콜레스테롤 식이를 섭취한 흰쥐에 메주 분말 급여가 지질대사에 미치는 영향)

  • This study was performed to examine the effects of Meju supplementation to high cholesterol diet on lipid metabolism in rats. Male Sprague-Dawley rats were divided into two groups(Steamed soybean, Meju) and were fed with 0.5% cholesterol diet. Serum and liver lipid profiles and fecal bile acids excretion were examined after four weeks of experimental diet. Food intake, weight gain and liver weight were not significantly different between the two groups. Total cholesterol concentrations in serum was significantly lower in Meju group compared to steamed soybean group(p<0.05). HDL-cholesterol, triglyceride, phospholipids and nonestrified fatty acid concentrations in serum were not significantly different between the two groups. Cholesterol, triglyceride, and phospholipids concentrations in liver were not significantly different between the two groups. Fecal bile acid excretion were markedly higher in Meju groups than the group fed steamed soybean(p<0.05). These results suggest that the cholesterol lowering effect by Meju supplementation might be resulted from the modulation of fecal bile acid excretion in rats.

Rapid Gas Chromatographic Screening of Alcoholic Beverages for Volatile and Nonvolatile Organic Acids (기체크로마토그래피법에 의한 알코올 음료내 휘발성 및 비휘발성 유기산의 신속한 스크리닝)

  • A rapid gas chromatographic (GC) profiling method for the simultaneous analysis of volatile and nonvolatile organic acids was applied to alcoholic beverages (white wine, red wine, brandy, and beer). It involves the solid-phase extraction of organic acids using Chromosorb P as the sorbent and diethyl ether as the eluent with subsequent triethylamine treatment. The resulting triethylammonium salts of acids were directly converted to volatile tert.-butyldimethylsilyl derivatives, which were analyzed by dual-capillary column GC and GC-mass spectrometry. From the alcoholic beverages studied, more than 29 organic acids were detected. When the simplified retention infer (RI) spectra of organic acids, and the direct comparisor method between alcoholic beverages and a test sample were attempted to identify a test sample, it was quickly recognized to be a red wine with the 998 ppt match quality value.

Effect of Orostachys malacophyllus by Fermented Lactic Acid Bacteria on Plasma Levels of Lipid and Lipid Peroxidation in Alcohol Feeding Rats (유산균 발효 와송이 알코올성 유발 지방간 흰쥐의 조직 과산화지질 및 혈중 지질농도에 미치는 영향)

  • The purpose of this study was to identify the effect on plasma levels of lipid and lipid peroxidation by administration of lactic acid bacteria (Lactobacillus bulgaricus, Streptococcus thermophilus, Lactobacillus acidophilus) fermented Orostachys malacophyllus (FOM) in alcohol fed rats. Male Sprague-dawley rats were randomly divided into seven groups: normal diet group (N), alcohol treated group (C), 2.5% (w/w) or 5% (w/w) OM treated group (2.5OM, 5OM), 2.5% (w/w) or 5% (w/w) fermented OM treated group (2.5FOM, 5FOM) and silymarin treated group (SM) as a positive control. As a result of measuring serum total lipid, C group were increased total lipid, free fatty acid content and were decreased HDL-cholesterol content, but 5FOM group were significantly decreased lipid content and were increased HDL-cholesterol content and accordingly reduced the incidence of atherosclerosis. Serum total protein content was similarly measured in all groups and serum albumin content was decreased in alcohol feeding groups compared to the N group. The 5FOM group had significantly decreased liver and serum triglycerides compared to the C group. The TBARS content in the liver, serum, testis, kidney, spleen, and heart were slightly decreased in the 5FOM group compared to the C group and the 5FOM group had an increased glutathione concentration. The 5FOM treatment was showed analogous results to those of the SM treatment, suggesting that FOM is can improve the lipid profiles of alcohol-fed rats.

Comparison of the dietary intake and clinical characteristics of obese and normal weight adults

  • Obesity contributes to an increased risk for chronic diseases, including diabetes, cardiovascular diseases, and certain types of cancer. The prevalence of obesity has increased in Korea. We compared the clinical and dietary characteristics of obese adults (n = 30, 17 men and 13 women, mean age 29.9) to those with a normal weight (n = 15, 8 men and 7 women, mean age 26.5). We determined lipid profiles, fasting blood sugar (FBS), blood pressure, and serum free fatty acid (FFA). Dietary intake was estimated using a food frequency questionnaire (FFQ) and a 3-day dietary record. Exercise patterns and average alcohol intake were determined. The average body mass index was 28.3 kg/$m^2$ in the obese and 21.2 kg/$m^2$ in the normal weight groups. The obese group had significantly higher levels of total cholesterol, LDL cholesterol, and triglycerides, lower levels of HDL cholesterol, and higher blood pressures compared to the normal weight group. FBS was not significantly different between the two groups. The obese group had significantly more subjects with metabolic syndrome (26.7%) compared to the normal weight group (0%). Serum FFA levels tended to be higher in the obese (P = 0.087). No significant difference in caloric intake was observed between the two groups. No differences in carbohydrate, protein, or fat intake between two groups were observed from the FFQ. However, results from the 3-day dietary record showed that the percentage of energy from fat was significantly higher in the obese group. The frequency and amount of exercise did not differ between the two groups. Alcohol consumed per drinking episode was significantly higher in the obese group. These results confirm that excessive weight is associated with disturbances in lipid metabolism in these fairly young and otherwise healthy adults. Dietary factors, including higher fat intake and alcohol consumption, seem to be contributing to the obesity of these subjects.

Diversity Analysis of Diazotrophic Bacteria Associated with the Roots of Tea (Camellia sinensis (L.) O. Kuntze)

  • The diversity elucidation by amplified ribosomal DNA restriction analysis and 16S rDNA sequencing of 96 associative diazotrophs, isolated from the feeder roots of tea on enriched nitrogen-free semisolid media, revealed the predominance of Gram-positive over Gram-negative bacteria within the Kangra valley in Himachal Pradesh, India. The Gram-positive bacteria observed belong to two taxonomic groupings; Firmicutes, including the genera Bacillus and Paenibacillus; and Actinobacteria, represented by the genus Microbacterium. The Gram-negative bacteria included ${\alpha}$-Proteobacteria genera Brevundimonas, Rhizobium, and Mesorhizobium; ${\gamma}$-Proteobacteria genera Pseudomonas and Stenotrophomonas; and ${\beta}$-Proteobacteria genera Azospira, Burkholderia, Delftia, Herbaspirillum and Ralstonia. The low level of similarity of two isolates, with the type strains Paenibacillus xinjiangensis and Mesorhizobium albiziae, suggests the possibility of raising species novum. The bacterial strains of different phylogenetic groups exhibited distinct carbon-source utilization patterns and fatty acid methyl ester profiles. The strains differed in their nitrogenase activities with relatively high activity seen in the Gramnegative strains exhibiting the highest similarity to Azospira oryzae, Delftia lacustris and Herbaspirillum huttiense.

Seasonal Difference of Nutrient Intake, Serum Lipid and Antioxidative Index in Female College Students (여대생의 영양소 섭취, 혈청 지질 및 항산화 지표의 계절별 차이)

  • To evaluate the seasonal differences of nutrient intake and serum biochemical indices (total cholesterol, HDL-, LDL- cholesterol, TG, MDA : malondialdehyde, TAS : total antioxidant status, folate) in Korean college women with a mean age of 20.15y, this study was conducted twice: once in winter and a second time in summer. Anthropometric assessment was also measured in the two seasons. As a result body fat mass (p<0.05), body fat% (p<0.05), and abdominal fat% (p < 0.01) in the summer were significantly lower than those in winter. Intake of fat (p < 0.05), polyunsaturated fatty acid (p < 0.01), zinc (p < 0.05), and vitamin E (p < 0.05) were at significantly higher levels in the summer period compared with the winter period. In addition, nutrients with an intake level less than 75% RI of KDRIs were folate in winter and calcium, and folate in the summer. Total cholesterol (p < 0.05) and HDL-cholesterol in summer, according to lipid profiles, were significantly lower than those in winter. Although MDA, one of the products of lipid peroxidation, remained unchanged, TAS was significantly higher in summer than in winter. The level of folate in summer showed significantly lower than that in winter. The present study suggests that nutrient intake of college women women differs according to the seasons and anthropometric indices and serum biochemical indexes were associated with seasons. Thus nutritional education programs on the basis of season are needed for college women.

Effects of Rumen pH on Degradation Kinetics and Fermentation Indices of Corn Silage Ensiled with Antifungal and Carboxylesterase Producing Inoculants

  • The present study investigated effects of antifungal and carboxylesterase inoculant on rumen fermentation with different rumen pH. Corn silage was treated without inoculant (CON) and with a mixed Lactobacillus brevis 5M2 and L. buchneri 6M1 (MIX). Rumen fluid was collected from two cannulated Hanwoo heifers before morning feeding (high rumen pH at 6.70) and 3 h after feeding (low rumen pH at 6.20). Dried corn silage was incubated in the rumen buffer (rumen fluid + anaerobic culture medium at 1:2 ratio) for 48 h at 39℃. Eight replications for each treatment were used along with two blanks. Both in a high and a low rumen pH, MIX silages presented higher (p<0.05) the immediately degradable fraction, the potentially degradable fraction, total degradable fraction, and total volatile fatty acid (VFA) than those of CON silages. Incubated corn silages in a low rumen pH presented lower (p<0.05) total degradable fraction, ammonia-N, total VFA (p=0.061), and other VFA profiles except acetate and propionate, than those in a high rumen pH. The present study concluded that application of antifungal and carboxylesterase inoculant on corn silage could improve degradation kinetics and fermentation indices in the rumen with high and low pH conditions.

Characterization of Interphase Microbial Community in Luzhou-Flavored Liquor Manufacturing Pits of Various Ages by Polyphasic Detection Methods

  • It is vital to understand the changing characteristics of interphase microbial communities and interspecies synergism during the fermentation of Chinese liquors. In this study, microbial communities in the three indispensable phases (pit mud, zaopei, and huangshui) of Luzhou-flavored liquor manufacturing pits and their shifts during cellars use were first investigated by polyphasic culture-independent approaches. The archaeal and eubacterial communities in the three phases were quantitatively assessed by combined phospholipid ether lipids/phospholipid fatty acid analysis and fluorescence in situ hybridization. In addition, qualitative information regarding the microbial community was analyzed by PCR-denaturing gradient gel electrophoresis. Results suggested that the interphase microbial community profiles were quite different, and the proportions of specific microbial groups evolved gradually. Anaerobic bacteria and gram-positive bacteria were dominant and their numbers were higher in pit mud ($10^9$ cells/g) than in huangshui ($10^7$ cells/ml) and zaopei ($10^7$ cells/g). Hydrogenotrophic methanogenic archaea were the dominant archaea, and their proportions were virtually unchanged in pit mud (around 65%), whereas they first increased and then decreased in zaopei (59%-82%-47%) and increased with pit age in huangshui (82%-92%). Interactions between microbial communities, especially between eubacteria and methanogens, played a key role in the formation of favorable niches for liquor fermentation. Furthermore, daqu (an essential saccharifying and fermentative agent) and metabolic regulation parameters greatly affected the microbial community.

Screening and Characterization of Oleaginous Microalgal Species from Northern Xinjiang

  • A total of 646 strains, including green algae and diatoms, were isolated from 220 samples to screen microalgae with high lipid productivity (LP). The samples were obtained from nine habitats in Northern Xinjiang, China in June 2013. This study initially identified eight lipidrich strains, namely, Desmodesmus intermedius XJ-498, D. intermedius XJ-145, D. intermedius XJ-99, Monoraphidium pusillum XJ-489, M. dybowskii XJ-435, M. dybowskii XJ-151, Mychonastes homosphaera XJ-488, and Podohedriella falcata XJ-176, based on 18S rDNA sequencing. The strains were cultured in a photobioreactor for the same period. Results showed that the specific growth rate (day-1) of M. pusillum XJ-489 was the highest (1.14 ± 0.06), and the biomass concentration (g/l) of D. intermedius XJ-99 was the highest (2.84 ± 0.3). Futhermore, the lipid content (%) of M. dybowskii XJ-151 was the highest (33.5 ± 4.38), and the lipid productivity (mg l-1 day-1) of My. homosphaera XJ-488 was the highest (86.41 ± 9.04). C16 to C18 accounted for 86% to 98% of the total lipid, and the biodiesel qualities of the selected algae corresponded to international standards. This study suggests that My. homosphaera XJ-488, D. intermedius XJ-99, and M. dybowskii XJ-151 are the most potential strains for biodiesel production among all the isolated strains.

Influence of Lipids on the in Vitro Protein Digestibility of Dried Fish Meat (건어육 저장중 지질이 단백질 소화율 저하에 미치는 영향)

  • The interaction of myofibrillar protein with lipid or oxidized lipid was considered to be mostly contributing to the drop of digestibility of fish meat products. The digestibility of myofibrillar protein was $92.11\%$ for flounder and $88.04\%$ for hairtail fish, repectively, and as a rule it decreased as both the amount of lipid and reaction time increased. It also decreased with increase in the amount of added linoleate and oxidized linoleate. However, when the reaction continued for 6 hours or more the digestibility rather increased, which was provably due to the unfolding of protein structure. The hot air dried hairtail fish showed the lowest C-PER values among all dried fish products. The protein quality of flounder, hairtail fish and their dried ones except hot air dried ones measured by C-PER procedure were superior to that of ANRC casein. DC-PER values of all samples were greater than those of C-PER values and the greater discrepancies were noted in hairtail fish (fatty fish) products which possessed the lower in vitro protein digestibilities. Predicted diegstibilities, which were calculated using amino acid profiles, of all samples except raw ones were overestimated in comparison with in vitro protein digestibilities. From the observations so far, formation of complex of lipids and protein was thought to be the most important factor in lowering protein digestibility of the dried fish meat products.
