Effects of Dietary Lipid Sources and Levels on Lecithin : Cholesterol Acyltansferase Activity and Cholesterol Metabolism in Rats (식이지방의 종류와 수준이 흰쥐의 Lecithin : Cholesterol Acyltransferase 활성 및 콜레스테롤대사에 미치는 영향)

  • This study was carried out to investigate the effects of different sources and level of dietary lipid on lecithin : cholesterol acyltrasferase activity and cholesterol metabolism in male rats of Sprague-Dawley strain. The effects of different lipid sources was compared with sardine oil($\omega$3 EPA and DHA), beef tallow(SFA), perilla oil($\omega$3 linolenic acid) and corn oil($\omega$6 linoleic acid). Diets were formulated in such a way that 10%, 20% and 40% dietary energy were supplied with each of four experimental lipid sources. Control diet contained only non-lipid energy. A total number of 78 rats, equally divided into 13 groups, were fed the experimental diets for a period of 6 weeks. In vitro cultures were also carried out to study the cholesterol synthetic activity in the liver prepared from rats used in feeding trials. The concentration of plasma total cholesterol, HDL-cholesterol, LDL-cholesterol and HDL-C/T/C(total cholesterol) ratio were significantly (p<0.001) influenced by dietary lipid sources. Higher HDL-cholesterol and lower LDL-cholesterol concentration in plasma were obtained in rats fed $\omega$3 fatty acid supplemented diets(sardine oil and perilla oil group) compared to diets containing $\omega$6 and saturated fatty acid(corn oil and beef tallow group). In total cholesterol concentration of plasma, beef tallow group was significantly (p<0.001) higher than other lipid groups, and non-lipid group was significantly(p<0.05) higher than the lipid supplemented groups. The activity of lecithin : cholesterol acyltransferase(LCAT) in plasma was greatly(p<0.001) affected by dietary lipid sources and levels. In LCAT acivity of plasma, lipid supplemented groups were significantly(p<0.05) higher than non-lipid group, vegetable oil groups were significantly (p<0.001) higher than animal fat groups, and sardine oil group were significalylty (p<0.001) higher than beef tallow group. Also perilla oil group was significanlty (p<0.05) higher than corn oil group, and sardine oil group was significantly (p<0.05) higher than perilla oil group. Low lipid group, compared with medium or high lipid group, showed higher activity of LCAT in plasma. In cholesterol synthetic activity of liver tissues culture, sardine oil group($\omega$3 EPA and DHA) was significantly(p<0.001) higher than other lipid groups, non-lipid group was significantly(p<0.001) higher than the lipid supplemented groups, and amimal fat group were significantly(p<0.001) higher than vegetable oil groups, but the synthetic activity was not affected by dietary lipid levels.

Comparative Analysis of Bone Mineral Contents with Dual-Energy Quantitative Computed Tomography (이중에너지광자선의 전산화단층촬영술을 이용한 정량적 골무기물함량의 비교분석)

  • Purpose : The Dual-Energy Quantitative Computed Tomography(DEQCT) was compared with bone equivalent $K_2HPO_4$ standard solution and ash weight of animal cadaveric trabecular bone in the measurement of bone mineral contents(BMC). Method and Materials : The attenuation coefficient of tissues highly depends on the radiation energy density and effective atomic number of composition, The bone mineral content of DEQCT in this experiments was determined from empirical constants and mass attenuation coefficients of bone,fat and soft tissue equivalent solution in two photon spectra. In this experiments, the BMC of DEQCT with 80 and $120kV_p$ X rays was compared to ash weight of animal trabecular bone. Results : We obtained the mass attenuation coefficient of 0.2409 0.5608 and 0.2206 in $80kV_p$, and 0.2046, 0.3273 and $0.1971cm^2/g$ in $120kV_p$ X-ray spectra for water bone and fat equivalent materials, respectively. The BMC with DEQCT was acomplished with empirical constants $K_1=0.3232,\;K_2$=0.2450 and mass attenuation coefficients has very closed to ash weight of animal trabecular bone The BMC of empirical DEQCT and that of manufacturing DEQCT were correlated with ash weight as a correlation r= 0.998 and r= 0.996, respectively. Conclusion : The BMC of empirical DEQCT using the experimental mass attenuation coefficients and that of manufacture have showed very close to ash weight of animal trabecular bone.

  • The underlying action of policosanol in lowering cholesterol level has not yet been clearly elucidated. Several recent studies have suggested that sterol regulatory element-binding proteins (SREBP)-1c play a role in the regulation of cholesterol synthesis via the fatty acid synthesis pathway. To date, no study has evaluated the effects of policosanol on SREBP-1c-mediated fatty acid synthesis. Therefore, this study aimed to investigate whether the SREBP-1c-mediated fatty acid biosynthetic pathway is associated with the cholesterol-lowering effects of policosanol. Seven-week-old C57BL/6 male mice were randomly divided into 7 groups (n=7 per group) and treated for 8 weeks as follows: 1) normal diet (normal control), 2) high-fat diet (HFD), 3) HFD+ethanol (Pol-0), 4) HFD+policosanol 1 mg/kg (Pol-1), 5) HFD+policosanol 2 mg/kg (Pol-2), 6) HFD+policosanol 4 mg/kg (Pol-4), and 7) HFD+simvastatin 50 ㎍/kg (positive control). Policosanol and simvastatin were administered at the same time every day while maintaining the HFD. Body weight and food intake were measured weekly for 8 weeks. After 8 weeks, serum cholesterol levels were measured, histological analysis was carried out, and the expressions of SREBP-1c and fatty acid synthase (FAS) in the liver tissues were examined. Policosanol reduced body weight and the amount of food intake in a dose-dependent manner. Serum cholesterol levels were significantly lowered in the Pol-1 and Pol-4 groups. The expression of SREBP-1c and FAS was also significantly decreased in the Pol-4 group. These results suggest that the cholesterol-lowering effects of policosanol can occur due to the inhibition of the expression of SREBP-1c and FAS.

Effects of Organic Selenium Mix on the Performance, Carcass Characteristics, Tissue Selenium Distribution, and Economic Value in Finishing Hanwoo Steers (유기셀레늄 혼합제 급여가 비육말기 거세한우의 성장, 도체성적, 체내 셀레늄 분포 및 경제성에 미치는 영향)

  • This study fulfilled to investigate the feed efficiency, tissue selenium distribution, carcass characteristic and economic value in finishing Hanwoo steers fed organic selenium mix (OSM) which included seleno-yeast, rumen culture and other microbial supplements. Forty five finishing Hanwoo steers were tested for 4 months dividing to three feeding groups: OSM add as 0.5 ppm Se of DM feeds (0.5 ppm OSM), OSM enriched add as 1.0 ppm Se of DM feeds (1.0 ppm OSM) and basal diet without OSM (control). The total weight gains, the average daily gains and the feed intakes were not differ in treatments (p > 0.05). No differences (p > 0.05) were noted for hot carcass weight, loin eye area, backfat thickness, meat yield index, meat color, fat color, tenderness and maturity. However, the 1.0 ppm OSM showed better performances for feed requirement, TDN per gain, meat yield grade and meat quality grade compared to other groups. Tissue selenium distribution was increased by organic selenium feeding: higher Se concentration in liver and rump of 0.5 ppm OSM (p < 0.05), and kidney, liver, sirloin and rump of 1.0 ppm OSM (p < 0.05) than the tissues of control group. Generally, tissue selenium was the highest value in 1.0 ppm OSM and showed higher concentrate in order; kidney, liver, sirloin and rump. The income over feed cost was 1.06-fold higher in 1.0 ppm OSM than control group. In conclusion, organic selenium mix supplementation and its amounts were not influenced to feed intake, body gain and carcass characteristic but significantly increased tissue selenium. Therefore, these results suggest that finishing Hanwoo steer fed an enriched organic selenium mix with proper probiotics is able to produce “high-Se” beef as high bioavailable form as well as create a beneficial opportunity on Hanwoo farm.

Effect of High Pressure Processing on Freshness of Meat Products (육류가공품의 고압처리가 신선도에 미치는 영향 평가)

  • The high pressure processing (HPP) is a technology which can preserve the quality of foods, such as the fresh taste, incense, texture, vitamin content, and so on, by minimizing the heating process. It does so by applying an instantaneous and uniform pressure that is the same as the water pressure that is 60 km deep in the sea. HPP is a technology that can inhibit food poisoning and spoilage caused by microorganisms and is currently an actively studied area. In this study, we investigated the effects of a high pressure treatment (0, 4, 6 min) on sliced ham, which is a typical meat product, at 600 MP a were tested for their effect on freshness. Moisture contents varied from 48 to 69%, salinity varied from 1.07 to 1.11%, and the pH decreased from 6.4~6.5 to 6.1~5.15. However, there was no difference between the control and treatment groups. General bacteria stored at $20^{\circ}C$ after hyper-pressure treatment were found to have no significant microorganisms in all groups until 4 weeks. but exceeded $10^5$ in control group and HPP 6 min treatment group from 5 weeks, At week 7, it was found to exceed $10^6$. The results indicate it was not possible to ingest food in the 4-and 6 minute treatment groups. Coliform was not observed in all groups despite observing for a total of 7 weeks at $20^{\circ}C$ weight test. VBN, a method used to determine the protein freshness of meat, showed a VBN value of less than 1 mg% until the fourth week and a value of 1 to 2 mg% after 5 weeks. The TBA was used as an index of the degree of fat acidosis in the meat tissues. The results showed it was below 0.18 mgMA / kg until the end of 7 weeks; this value was within the range for fresh meat, and there was no difference in treatment group. In this experiment, deformation of the packaging material did not occur and no swelling occurred due to the generation of gas. It is believed that the basic preservation effect was achieved only by blocking with the air due to the close contact of the packaging material.

Analysis of Global Gene Expression Profile of Human Adipose Tissue Derived Mesenchymal Stem Cell Cultured with Cancer Cells (암세포주와 공동 배양된 인간 지방 조직 유래 중간엽 줄기 세포의 유전자 발현 분석)

  • Mesenchymal stem cells (MSC) are multipotent and can be isolated from diverse human tissues including bone marrow, fat, placenta, dental pulp, synovium, tonsil, and the thymus. They function as regulators of tissue homeostasis. Because of their various advantages such as plasticity, easy isolation and manipulation, chemotaxis to cancer, and immune regulatory function, MSCs have been considered to be a potent cell source for regenerative medicine, cancer treatment and other cell based therapy such as GVHD. However, relating to its supportive feature for surrounding cell and tissue, it has been frequently reported that MSCs accelerate tumor growth by modulating cancer microenvironment through promoting angiogenesis, secreting growth factors, and suppressing anti-tumorigenic immune reaction. Thus, clinical application of MSCs has been limited. To understand the underlying mechanism which modulates MSCs to function as tumor supportive cells, we co-cultured human adipose tissue derived mesenchymal stem cells (ASC) with cancer cell lines H460 and U87MG. Then, expression data of ASCs co-cultured with cancer cells and cultured alone were obtained via microarray. Comparative expression analysis was carried out using DAVID (Database for Annotation, Visualization and Integrated Discovery) and PANTHER (Protein ANalysis THrough Evolutionary Relationships) in divers aspects including biological process, molecular function, cellular component, protein class, disease, tissue expression, and signal pathway. We found that cancer cells alter the expression profile of MSCs to cancer associated fibroblast like cells by modulating its energy metabolism, stemness, cell structure components, and paracrine effect in a variety of levels. These findings will improve the clinical efficacy and safety of MSCs based cell therapy.

  • Purpose: Our previous study demonstrated that persimmon (Diospyros kaki Thumb.) at different stages of ripening provided different protective effects against high-fat/cholesterol diet (HFD)-induced dyslipidemia in rats. In this study, we compared the metabolites profile and gene expressions related to triglyceride (TG)/cholesterol metabolism in vitro and in vivo after treating with persimmon water extracts (PWE) or tannin-enriched persimmon concentrate (TEP). Methods: Primary and secondary metabolites in test materials were determined by GC-TOF/MS, UHPLC-LTQ-ESI-IT-MS/MS, and UPLC-Q-TOF-MS. The expression of genes related to TG and cholesterol metabolism were determined by RT-PCR both in HepG2 cells stimulated by oleic acid/palmitic acid and in liver tissues obtained from Wistar rats fed with HFD and PWE at 0, 150, 300, and 600 mg/d (experiment I) or TEP at 0, 7, 14, and 28 mg/d (experiment II) by oral gavage for 9 weeks. Results: PLS-DA analysis and heatmap analysis demonstrated significantly differential profiling of metabolites of PWE and TEP according to processing of persimmon powder. In vitro, TEP showed similar hypolipidemic effects as PWE, but significantly enhanced hypocholesterolemic effects compared to PWE in sterol regulatory element-binding protein 2 (SREBP2), HMG-CoA reductase (HMGCR), proprotein convertase subtilisin/kexin type 9 (PCSK9), cholesterol $7{\alpha}-hydroxylase$ (CYP7A1), and low density lipoprotein receptor (LDLR) gene expression. Consistently, TEP and PWE showed similar hypolipidemic capacity in vivo, but significantly enhanced hypocholesterolemic capacity in terms of SREBP2, HMGCR, and bile salt export pump (BSEP) gene expression. Conclusion: These results suggest that column extraction after hot water extraction may be a good strategy to enhance tannins and long-chain fatty acid amides, which might cause stimulation of hypocholesterolemic actions through downregulation of cholesterol biosynthesis gene expression and upregulation of LDL receptor gene expression.

  • The sensory test and analysis of the proximate composition and hydroxyproline content of 4 kinds of Korean commercial sausage products (Beef, Hamburger, Frank, Hotdog) were carried out and the results were compared one another. The qualities of these sausage products were compared one another. The qualities of these sausage products were compared on the basis of the nutritive value of foreign sausages and according to the Korea Food Law. The results obtained were as follows. 1. The general conclusion drawn from the results of the proximate analysis was that the average composition of commercial sausages was adequate in view of the limits set by the Food Law. 2. The evaluation factors of sausages based on moisture-to-protein and protein-to-fat ratios were proposed tocompare the qualities of Korean commercial sausages with those of foreign commercial sausages. 3. Content of collagen as connective tissues in total protein of Korean commercial sausages were $2.1{\sim}15%$. The results of the present study appeared to suggest that meat contents of Korean commercial sausages used in this study were less than those of foreign sausages. 4. Among the 4 different kinds of sausages studied the qualities of Frank sausages were the best whereas those of Hotdog sausages were of the lowest quality. Beef sausages appeared better than Hamburger sausages from the view point of nutritive components.

The Separate and Combined Effects of Insulin, Dexamethasone and Growth Hormone on the OB Gene Expression and Leptin Secretion from Cultured Human Visceral Adipose Tissue (인체의 복강 내 지방조직 배양을 통한 OB 유전자 발현과 Leptin 분비에 미치는 인슐린, Dexamethasone과 성장호르몬의 단독 또는 복합적 영향에 관한 연구)

  • Purpose : We investigated the hormonal control of OB gene expression and leptin secretion in cultured human visceral adipose tissue. Methods : Visceral adipose tissues were cultured for up to 48 hrs in modified Eagle's medium with varying concentration of hormones : Control(no hormone), bovine insulin(100 nM), Dexamethasone(DEX, 100 nM), growth hormone(GH, 40 ng/mL), insulin+DEX(100 nM each), insulin+DEX+GH(100 nM insulin and DEX, 40 ng/mL GH). Quantitative analysis of leptin mRNA was performed by competitive reverse transcription polymerase chain reaction, and leptin secretion in culture medium was measured by IRMA using a commercial kit. Results : The addition of dexamethasone to the medium significantly increased OB gene expression and leptin secretion(P<0.05). Unlike dexamethasone, insulin did not affect OB gene expression and leptin secretion. Both insulin and dexamethasone, at high concentration, significantly stimulated leptin secretion compared with basal values(P<0.05). Leptin gene expression was not significantly increased by GH treatment alone, however GH, in combination with high concentrations of insulin and dexamethasone, attenuated the stimulatory effects of high concentrations of insulin and dexamethasone. Conclusion : Insulin cannot increase leptin secretion without the presence of dexamethasone. The mechanism suggested is that insulin may increase leptin secretion in cytoplasm only after dexamethasone increases the expression of OB gene. Further studies are necessary to elucidate the mechanism of the action of insulin on leptin secretion after increasing OB gene expression by dexamethasone.

Effects of Dietary Fatty Acids on fatty Acid Pattern in Development Rat Brain Phospholipids - Effects on P/M/S and $\omega$3/$\omega$6 Fatty Acid Ratios -

  • Docosahexaenoic acid(DHA), a $\omega$3 series fatty acid and arachidonic acid(AA). a $\omega$6 series fatty acid were found in relatively high concentrations in the phospholipids(PLs) of cell membranes of nerve tissues, and they can be affected by various factors. The present study examined the effects of dietary $\omega$6 and $\omega$3 fatty acid composition on P/M/S and on $\omega$3/$\omega$6 fatty acid ratios in brain PLs of 2nd generation rats. The expeimental diets consisted of 10% fat(by wt), which were computer- searched mixed oil('M') with P/M/S ratio, 1 : 1.4 : 1 and $\omega$6/$\omega$3 ratio, 6 : 1 and safflower oil('S') poor in $\omega$3 fatty acids. The experimental diets were started 3-4 wks prior to conception. During the lactation period, the feeding mothers were switched 1 wk after birth and provided the pups for 2 wks with milk which had compositions different from that of their natural mother. The same diet as their mothers was provided from weaning to 9 wks of age. The 'M'and 'S' rats were again subdivided into MM, MS, SS, SM rats according to diet which their lactating mothers were fed from the begining of the experiment. The relative percentage of P/M/S fatty acids in brain PLs in all experimental groups converged to a very similar value at 9 wks of age, indicating the existence of a control mechanism for the degree of fatty acids, unsaturation. The $\omega$3/$\omega$6 fatty acid ratios of brain PLs converged to about 1.0 in MM & SM groups and to 0.7 in SS & MS groups, suggesting also the existence of some balance between $\omega$3 and $\omega$6 fatty acids in developing rat brain. The concentrations of $\omega$3 fatty acids, especially DHA, in the SM group were increased and became similar to those in MM group at 9 wks of age. The increase in DHA of brain PLs was counterbalanced b)r a decrease in 22 5$\omega$6. Therefore, the ratios of 22 : 6$\omega$6/22 : 5$\omega$6 were higher in both MM & SM groups than those of SS & MS groups at 9 wak of age. Although dietary $\omega$3 and $\omega$6 fatty acids affected 22 : 6$\omega$S and 22 : 5$\omega$6 contained in rat brain PLs reciprocally, the relative percentage of AA did not appear to be significantly influenced by the diet in all groups at 9 wks of age, suggesting that a mechanism for the maintenance of a certain level of AA in brain PLs exists. In conclusion, the $\omega$3/$\omega$6 fatty acid and 22 : 6$\omega$3/22 : 5$\omega$6 ratios, but not P/M/S ratio, of rat brain PLs were affected by the postnatal dietary changes. Futher studies are required to clarify the mechanism(S) of ensuring a certain level of DHA and of maintaining a similar level of AA in rat brain PLs after. weaning(9 wk) regardless of prenatal and postnatal dietary changes.
