Molecular Cloning and Characterization of a Vitellogenin of the Bumblebee Bombus ignitus

  • A vitellogenin cDNA was cloned from the bumblebee Bombus ignitus. The cDNA encoding B. ignitus vitellogenin (BiVg) is 5473 bases long with an open reading frame of 1773 amino acid residues. BiVg possesses two consensus (RXXR/S) cleavage sites and has the conserved DGXR and GL/ICG motif near its C-terminus. The deduced amino acid sequence of BiVg cDNA showed significant similarity with hymenopteran Vgs (51% identity to Apis mellifera Vg, and $33{\sim}36%$ to other insect Vgs). The BiVg cDNA was expressed as a 200-kDa polypeptide in baculovirus-infected insect Sf9 cells. Northern and Western blot analyses showed the expression of BiVg in fat bodies of pupae and adults. In queens, the expression of BiVg was first detected in late pupal stage fat bodies, and secreted BiVg was also observed in late pupal stage hemolymph.

  • This paper proposes a multimodal registration method for segmentation of the Visible Human color images, in which color characteristics of bones are very similar to those of its surrounding fat areas. Bones are initially segmented in CT images, and then registered into color images to lineate their boundaries in the color images. For the segmentation of bones in CT images, a thresholding method is developed. The registration method registers boundaries of bodies in CT and color images using a cross-correlation approach, in which the boundaries of bodies are extracted by thresholding segmentation methods. The proposed method has been applied to segmentation of bones in a head and legs whose boundary is ambiguous due to surrounding fat areas with similar color characteristics, and produced promising results.

  • We attempted in this study to understand the hypoglycemic action of the fat soluble fraction of red ginseng roots in streptozotocin injected diabetic rats, through its actions on several enzymes relating to carbohydrate metabolism of the 1eve1 to compare with those of ginsenosides in streptozotocin injected diabetic rats. It was realized that the increased level of glucose, ketone bodies, lactate, nonesterified fatty acids and triacylglycerol in blood was significantly decreased and the decreased liver glycogen content of streptozotocin injected rats were appreciably moderated by intraperitoneal injection of the fat soluble fraction of red ginseng roots as shown in the saponin injected diabetic rats. The deceased activities of liver enzymes relating to carbohydrate metabolism such as phosphofructokinase, glucokinase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of streptozotocin induced diabetic rats were also sufficiently modified by the intraperitoneal injection of the above fat soluble fraction as shown in the ginsenoside injected streptozotocin induced rats.

A Study of the Anthropometric Indices and Eating Habits of Female College Students

  • A study was conducted to investigate the anthropometric indices, fat distribution, eating habits and levels of daily physical activity based on the body mass index (BMI ; kg/㎡) of 180 female college students. This stady was based on survey 2001 using a questionnaire. The results are as follow ; The mean age, height, weight and BMI were 20.41 ${\pm}$ 1.82 years, 161.86 ${\pm}$ 4.51cm, 53.49 ${\pm}$ 7.02kg, and 20.43 ${\pm}$ 2.65kg/㎡, respectively. The group with BMI of under 20 (the lean group) comprised 48.9% : the group with BMI of 20 to 25 (the normal group) comprised 44.4% and the group with BMI of over 25 (the obese group) comprised 6.7% of the subjects. Even though the lean and the normal groups were determined according to BMI. 12.5% of the lean group and 31.3% of the normal group had more than 25% body fat. Of the students, 11.5% of the lean group, 25.0% of the normal group and 58.3% of the obese group had of more than 0.8 waist/Hip Ratio (WHR). The medical problems suffered by the subjects were constipation (24.4%), gastroenteritis (20.6%), anemia(15.0%) and edema(13.3%), which was a result of their eating habits. Out of all the subjects, 27.2% ingested their meals irregularly, 74.4% skipped breakfast, and 92.4% of the lean group, 89.2% of the normal group and 81.8% of the obese group ingested snacks on a daily basis. Their favorite types of snacks were cookies breads and cakes (33.7%), spicy sliced rice cakes pork sausage and boiled fish cakes (18.5%), fruits (15.2%), juices and sodas (13.5%) and hamburgers and pizzas (3.9%). Their consumption of high calorie and processed foods was high, Of the subjects, 38.9% ate out daily. 45.1% had tried weight loss. Those with large upper bodies comprised 11.5%, large intermediate bodies 18.5% and large lower bodies 69.0%. Most of the students were dissatisfied with their body snaps. Of the subjects, 95.1% exercised irregularly. Daily physical activity tended to increase significantly with an increase in BMI : the lean group's average energy consumption was 1509.40㎉ the normal group's was 1792.21㎉ and the obese group's was 2334.63㎉, which were below the recommended value. This study suggests that the maintenance of an ideal weight, the improvement of eating habits and adequate exercise are needed for female college students to improve their health and nutritional status. Effects of Low-carbohydrate and High-fat Diet Supplemented with Ketogenic Drink on Cognitive Function and Physical Performance in the Elderly at High Risk for Dementia (케톤음료를 보충한 저탄수화물·고지방식이 섭취가 치매고위험 노인의 인지기능 및 신체활동 능력 변화에 미치는 영향)

  • Objectives: Reduced glucose utilization in the main parts of the brain involved in memory is a major cause of Alzheimer's disease, in which ketone bodies are used as the only and effective alternative energy source of glucose. This study examined the effects of a low-carbohydrate and high-fat (LCHF) diet supplemented with a ketogenic nutrition drink on cognitive function and physical activity in the elderly at high risk for dementia. Methods: The participants of this study were 28 healthy elderly aged 60-91 years showing a high risk factor of dementia or whose Korean Mini-Mental State Examination (K-MMSE) score was less than 24 points. Over 3 weeks, the case group was given an LCHF diet with nutrition drinks consisting of a ketone/non-ketone ratio of 1.73:1, whereas the control group consumed well-balanced nutrition drinks while maintaining a normal diet. After 3 weeks, K-MMSE, body composition, urine ketone bodies, and physical ability were all evaluated. Results: Urine ketone bodies of all case group subjects were positive, and K-MMSE score was significantly elevated in the case group only (p=0.021). Weight and BMI were elevated in the control group only (p<0.05). Grip strength was elevated in all subjects (p<0.01), and measurements of gait speed and one leg balance were improved only in the case group (p<0.05). Conclusions: We suggest that adherence to the LCHF diet supplemented with a ketogenic drink could possibly influence cognitive and physical function in the elderly with a high risk factor for dementia. Further, we confirmed the applicability of this dietary intervention in the elderly based on its lack of any side effects or changes in nutritional status.

A Study on the Method of Culture for Paecilomyces japonica Using an Egg (계란을 이용한 눈꽃동충하초 재배방법에 관한 연구)

  • This study was conducted to established method of culture for Paecilomyces japonica using an egg. Mycelia grew favorably at the temperature of 22~26$^{\circ}C$ on eggs. 5.1g of dry matter basis(average 7.2cm of longer and 199.6 of numbers) of artificial fruiting bodies were harvested at 60 days after inoculation from one of egg. Commercial fruiting bodies of Paecilomyces japonica from silkworms was used for comparative nutriental contents. Cordycepin contents of fruit bodies of Paecilomyces japonica cultivated on eggs and silkworms were not significantly different. Crude fat contents of fruiting bodies of Paeilomyces japonica cultivated from eggs was significantly higher than from silkworms(P<0.05). Mn and Cu contents of fruiting bodies of Paecilomyces japonica cultivated from silkworms were significantly higher than from eggs(P<0.05), but Na, Mg, Fe and Zn contents were significantly higher from eggs(P<0.05). Glycine, Arginine and Proline contents in the fruiting bodies of Paecilomyces japonica cultivated from silkworms were tend to higher than from eggs, but Serine, cystein. methionine, isoleucine and phenylalanin were tend to higher from eggs. These results were made possible that possible mass production of artificial fruiting bodies of Paecilomyces Japonica cultivated on eggs.

Ovarian and Fat Body Yolk Protein Synthesis in Culex piplens pallens (홍모기(Culex pipiens pallens) 지방체와 난소에서의 난황단백질합성에 관한 연구)

  • Ovarian Yolk protein (YP2) synthesis has been investigated in mosquito, Culex pipiens pallens. Yolk protein amount which was syntheized in fat body, accumulated into ovary were analyzed by Rocket immunoelectrophoresis and in vitro organ culture. The result was that yolk protein synthesis began to occur at 6hrs after blood meal, reached at maximum level by 24hrs, and was completed within 48hrs. Yolk protein accmulation into the ovary began to start at 6hrs and coutinued for up to 60hrs after blood meal. Extract from 0, 24, 48, 72hrs ovaries after blood meal were analyzed by electrophoresis and Western blotting. The result was that 24hrs ovary contain one yolk protein(YP1), and 48, 72hrs ovaries contain two kinds of yolk proteins(YPl and YP2). When 48hr ovaries and fat bodies were incubated in $^3$H-leucine contained medium, protein synthesis was not occurred in fat body, but ovary synthesized much protein contained yolk protein (YP2). The result of crossed immunoelectrophoresis represented the same immunity between YPl and YP2. The present data suggest that ovary synthesize yolk protein(YP2) in mosquito, Culex pipiens pallens.

Effects of Starvation on Lipid Metabolism and Gluconeogenesis in Yak

  • This research was conducted to investigate the physiological consequences of undernourished yak. Twelve Maiwa yak ($110.3{\pm}5.85kg$) were randomly divided into two groups (baseline and starvation group). The yak of baseline group were slaughtered at day 0, while the other group of yak were kept in shed without feed but allowed free access to water, salt and free movement for 9 days. Blood samples of the starvation group were collected on day 0, 1, 2, 3, 5, 7, 9 and the starved yak were slaughtered after the final blood sample collection. The liver and muscle glycogen of the starvation group decreased (p<0.01), and the lipid content also decreased while the content of moisture and ash increased (p<0.05) both in Longissimus dorsi and liver compared with the baseline group. The plasma insulin and glucose of the starved yak decreased at first and then kept stable but at a relatively lower level during the following days (p<0.01). On the contrary, the non-esterified fatty acids was increased (p<0.01). Beyond our expectation, the ketone bodies of ${\beta}$-hydroxybutyric acid and acetoacetic acid decreased with prolonged starvation (p<0.01). Furthermore, the mRNA expression of lipogenetic enzyme fatty acid synthase and lipoprotein lipase in subcutaneous adipose tissue of starved yak were down-regulated (p<0.01), whereas the mRNA expression of lipolytic enzyme carnitine palmitoyltransferase-1 and hormone sensitive lipase were up-regulated (p<0.01) after 9 days of starvation. The phosphoenolpyruvate carboxykinase and pyruvate carboxylase, responsible for hepatic gluconeogenesis were up-regulated (p<0.01). It was concluded that yak derive energy by gluconeogenesis promotion and fat storage mobilization during starvation but without ketone body accumulation in the plasma.

Genetic parameter estimation for milk β-hydroxybutyrate and acetone in early lactation and its association with fat to protein ratio and energy balance in Korean Holstein cattle

  • Objective: The objective of this study was to estimate the genetic parameters for milk ${\beta}$-hydroxybutyrate (BHBA), acetone (Ac), fat protein ratio (FPR), and energy balance (EB) using milk test day records and investigate the effect of early lactation FPR and EB on milk ketone body concentrations. Methods: Total 262,940 test-day records collected from Korea Animal Improvement Association during the period of 2012 to 2016 were used in this study. BHBA and Ac concentrations in milk were measured by Fourier transform infrared spectroscopy (FTIR). FPR values were obtained using test day records of fat and protein percentage. EB was calculated using previously developed equation based on parity, lactation week, and milk composition data. Genetic parameters were estimated by restricted maximum likelihood procedure based on repeatability model using Wombat program. Results: Elevated milk BHBA and Ac concentrations were observed during the early lactation under the negative energy balance. Milk FPR tends to decrease with the decreasing ketone body concentrations. Heritability estimates for milk BHBA, Ac, EB, and FPR ranged from 0.09 to 0.14, 0.23 to 0.31, 0.19 to 0.52, and 0.16 to 0.42 respectively at parity 1, 2, 3, and 4. The overall heritability for BHBA, Ac, EB and FPR were 0.29, 0.32, 0.58, and 0.38 respectively. A common pattern was observed in heritability of EB and FPR along with parities. Conclusion: FPR and EB can be suggested as potential predictors for risk of hyperketonemia. The heritability estimates of milk BHBA, Ac, EB, and FPR indicate that the selective breeding may contribute to maintaining the milk ketone bodies at optimum level during early lactation.
