육상 사육 수조에서 쥐노래미 치어의 적정 사육 밀도 (Influence of Stocking Density on Growth, Feed Efficiency and Body Composition of Juvenile Fat Cod (Hexagrammos otakii Jordan et Starks) in Indoor Culture System)

  • 육상 사육 수조에서의 쥐노래미 적정 사육 밀도를 구명하기 위해 13 g 전후의 치어를 대상으로 250 l FRP 수조(수용적 : 200 l)에 각각 2반복으로 50 (330 g/100 l), 100 (660 g/100 l), 150 (990 g/100 l) 및 200 (1330 g/100 l)마리씩 수용하여 냉동 전갱이와 분말사료를 1 : 1의 비율로 혼합한 moist pellet으로 2개월간 사육 실험하였다. 사육실험한 후의 평균체중, 증체율, 사료효율, 일일사료섭취율 및 단백질 축적율이 수용 밀도가 증가됨에 따라 이 값들이 감소하여 실험 시작시 수용적 100 l당 330 g을 수용한 실험구가 $660\~990 g을 수용한 실험구와는 유의차가 없었지만, 1330 g을 수용한 실험구와는 차이를 보였다(P>0.01).

Investigation of variations in energy, macronutrients and sodium intake based on the places meals are provided - Using the Korea National Health and Nutrition Examination Survey (KNHANES, 1998-2009) -

  • This study was conducted to investigate nutrient consumption by Korean adults in various places. To accomplish this, we used the 1998-2009 Korea National Health and Nutrition Examination Survey (KNHANES). Subjects of this investigation were over 19 years and the study included 37,160 people. The meals were categorized as breakfast, lunch, dinner, and snacks consumed at home, or while eating-out. Investigation of the rate of consumption at serving places based on daily meals and years showed that eating-out generally increased with time. The consumption of meals prepared at home was higher than that of meals consumed anyplace else in 1998, 2001, 2005, and 2007-2009. However, the rate of consumption of home meals decreased from 1998 to 2007-2009, while the rate of eating-out increased during this period. Annual nutrient intake according to serving places with respect to meals, energy, fat, and sodium were significantly lower in home meals than those consumed elsewhere in 2007-2009 relative to 1998. The sodium intake and energy distribution ratio of fat in meals consumed while eating-out increased significantly from 1998 to 2007-2009. The energy, fat and sodium intake and energy contribution ratio of fat consumed in meals at institutions was significantly higher in 2007-2009 than in 1998. Based on these results, additional research is required to develop guidelines for dietary life improvement at each serving place and to address education and policies for balanced nutrition intake.

고지방식이 마우스 동물모델에서 갯방풍 부탄올 분획물의 심혈관 보호 효과 및 주요 성분 (Cardiovascular Protective Effects of the n-Butanol Fraction from Glehnia littoralis Fr. Schm. in a High-Fat-Diet Mouse Model and Its Composition)

  • In our search for natural products affecting blood circulation, the n-butanol fraction from whole plant of Glehnia littoralis Fr. Schm. (GLB) improved blood lipid parameters, and ameliorated obesity in high-fat-diet (HFD)-fed C57BL/6 mouse model. Hyperlipidemia was induced by high-fat-diet for 4 weeks, and then GLB was orally administrated with 400 mg/kg/day for 4 weeks. GLB-treated group showed that the gain in body weight was significantly attenuated, the levels of total cholesterol and triglyceride significantly lowered on blood chemical analysis, and significantly prolonged the mice bleeding time when compared with those of HFD control group. Concomitantly, phytochemical composition of GLB was investigated by HPLC-hyphenated spectroscopy, and two major phenolic compounds, rutin and chlorogenic acid were identified in the GLB. Taken together, these results indicate that GLB has cardiovascular protective effects and could be a natural medicine candidate for the prevention of cardiovascular disease.

High fat diet-induced brain damaging effects through autophagy-mediated senescence, inflammation and apoptosis mitigated by ginsenoside F1-enhanced mixture

  • Background: Herbal medicines are popular approaches to capably prevent and treat obesity and its related diseases. Excessive exposure to dietary lipids causes oxidative stress and inflammation, which possibly induces cellular senescence and contribute the damaging effects in brain. The potential roles of selective enhanced ginsenoside in regulating high fat diet (HFD)-induced brain damage remain unknown. Methods: The protection function of Ginsenoside F1-enhanced mixture (SGB121) was evaluated by in vivo and in vitro experiments. Human primary astrocytes and SH-SY5Y cells were treated with palmitic acid conjugated Bovine Serum Albumin, and the effects of SGB121 were determined by MTT and lipid uptake assays. For in vivo tests, C57BL/6J mice were fed with high fat diet for 3 months with or without SGB121 administration. Thereafter, immunohistochemistry, western blot, PCR and ELISA assays were conducted with brain tissues. Results and conclusion: SGB121 selectively suppressed HFD-induced oxidative stress and cellular senescence in brain, and reduced subsequent inflammation responses manifested by abrogated secretion of IL-6, IL-1β and TNFα via NF-κB signaling pathway. Interestingly, SGB121 protects against HFD-induced damage by improving mitophagy and endoplasmic reticulum-stress associated autophagy flux and inhibiting apoptosis. In addition, SGB121 regulates lipid uptake and accumulation by FATP4 and PPARα. SGB121 significantly abates excessively phosphorylated tau protein in the cortex and GFAP activation in corpus callosum. Together, our results suggest that SGB121 is able to favor the resistance of brain to HFD-induced damage, therefore provide explicit evidence of the potential to be a functional food.

고지방식이 공급에 따른 성장기 마우스의 골의 형태학적 미세구조와 염증지표 변화 (Relationship between Bone Morphological Microstructure and Inflammatory Markers in Growing Mice Fed a High Fat Diet)

  • 본 연구에서는 성장기 마우스 모델을 이용하여 고지방식 이를 제공한 후 이에 따른 염증지표와 골의 형태학적 미세구조의 변화를 살펴보았다. C57BL/6J 4주령 수컷 마우스를 난괴법에 의해 대조군 (n = 6)과 실험군 (n = 6)로 분류하여 대조군에는 10% Kcal fat 식이와 고지방식이군에는 45% Kcal fat 식이를 12주 동안 자유급식 방법으로 제공하였다. 혈액검사와 염증지표를 분석하였으며 micro-CT를 이용하여 대퇴부 뼈의 형태학적 미세구조를 측정하였다. 대조군과 고지방식이군의 체중 증가는 각각 $5.85{\pm}1.84g$, $16.06{\pm}5.64g$로 유의한 차이를 보였으며 (p < 0.01), 혈당은 각각 $115.00{\pm}16.88mg/dL$, $188.33{\pm}13.29mg/dL$ (p < 0.01), 중성지방은 각각 $65.00{\pm}6.19mg/dL$, $103.33{\pm}8.02mg/dL$ (p < 0.05)로 나타났다. 렙틴과 IL-6는 고지방식이군에서 유의적으로 높게 나타났다 (p < 0.01). 골대사의 생화학적지표 분석 결과 오스테오칼신은 고지방식이군에서 낮게 나타났으나 유의적이지 않았으며, CTx은 고지방식이군에서 유의적으로 높게 나타났다 (p < 0.01). 골밀도는 고지방식이군에서 낮게 나타났으나 유의적인 차이는 보이지 않았다. 그러나 골의 형태학적 미세구조 분석결과 골소주의 두께는 고지방식이 군이 대조군보다 유의적으로 좁게 나타났으며 (p < 0.05), 골소주의 간격은 고지방식이군이 유의적으로 넓게 나타났다 (p < 0.05). 골의 형태학적 미세구조인 골소주의 간격과 IL-6가 양의 상관성이 나타났다 (p < 0.05). 본 연구결과 최대골밀도가 형성되는 단계의 성장기 마우스에서 고지방식이 공급을 통한 비만 유도 현상은 골소주의 수와 골소주가 차지하는 비율의 변화를 유발하여 골 미세구조에 영향을 미치는 것으로 나타났으며, 염증지표와 상관성이 나타났다. 이에 성장기에 염증지표 증가를 억제하고 정상적인 골형성을 위하여 과잉의 지방섭취 제한이 필요할 것으로 사료된다.

Reduction of Mouse Body Fats by Water Extract of Pleurous Ostreatus

  • Body fat-reducing ability of oyster mushroom (Pleurotus ostreatus) water extract (OMWE) was investigated of mice by supplying it drinking water. OMWE(2.95% solid content ) was prepared by extracting a low grade of the mushroom at 12$0^{\circ}C$ for 10min. The solid material of OMWE was composed of 65.2% reducing sugar, 0.23% crude fat, 0.5%total protein, 1.2% ash and 32.9% others. OMWE was appropriately diluted with drinking water. Seventy two male ICR mice(25$\pm$1 g, 7~8 weeks of age, 6 mice/cage, 18 mice/treatment) housed in polycarbonate cages containing $\beta$-chips were adopted in a temperature-and humidity-controlled facility with free access to water and diet. One week later, the mice were subjected to one of the treatments for 36days : 0 (control), 10, 50 and 100% OMWE. Drinking wter with or without OMWE was supplied twice (40ml each, 80ml in total ) daily per cage. Body weight and fed intake were recorded every three days. At the end of the experiment, mice were sacrificed to determine the chemical composition (fat, protein, ash and water). Body weight of mice treated with OMWE (10, 50 and 100%) at day 36 was 35.9, 35.9and 35.5g per mouse , respectively, and not significantly reduced as compared to that (36.5g/mouse) of control mice. Average body fat of 0,10,50 and 10% OMWE -treated mice was 14.3, 13.1, 10.7 and 12.0% , respectively. Body fat reduction by 50% OMWE treatment was 25.2% (p<0.05) relative to control. OMWE did not affect feed intake. The contents of body protein and ash were increased with respect to body fat decrease, while water content was not changed much. These results suggest that OMWE could reduce body fat of the mice without body weight change, giving the best effect by 50% OMWE.

털부처꽃 에탄올 추출물이 고지방식이를 급여한 흰쥐의 지질대사 및 항비만에 미치는 영향 (Effect of Lythrum salicaria L. Ethanol Extract on Lipid Metabolism and Anti-Obesity in Rat Fed High Fat Diet)

  • This study was carried out to investigate the effect of Lythrum salicaria L. ethanol extract on anti-obesity effects in rat fed a high fat diet for 8 weeks to induce obese rat model. Male SD rats were divided into normal group, control (high fat diet) group, positive control (Garcinia Cambogia extracts) group, high fat group supplemented with ethanol extracts of Lythrum salicaria L. (EELS). The body weight gain and control (high fat diet) were increased by a high fat diet, but decreased in the EELS. At the end of the experiment, the body weight in high fat diet groups was higher than that of normal diet group, while the body weights of EELS and positive control group were significantly reduced by 16.62%, as compared with that of high fat diet group (p < 0.05). The levels of serum triglyceride, total cholesterol in EELS group were significantly decreased as compared with high fat diet group (p < 0.05). The liver and mesenteric adipose tissue weights of control (high fat diet) increase than that for normal group, whereas EELS and positive control group were significantly decreased (p < 0.05). Levels of triglyceride in liver were significantly lower in EELS group than those in high fat diet group (p < 0.05). These results indicate that Lythrum salicaria L. extract may improve lipid metabolism and reduce fat accumulation and body weight.

Utilization of Transglutaminase for the Development of Low-fat, Low-salt Sausages and Restructured Meat Products Manufactured with Pork Hams and Loins

  • This study was performed to determine whether transglutaminase (Aciva-TG, TGase) can be used to reduce the salt level in low-fat sausages and to replace emulsified meats (10%) for the manufacture of restructured meat products (RMP). Pork hams and loins were collected from a local retail market in Gwangju, Korea and used for the manufacture of sausages and RMPs, respectively. TGase at the level of 0.1% can permit the reduction of the salt level from 1.5% to 1.0% in low-fat comminuted sausages without any quality defects, however a crumbly texture was found if the salt level was reduced below 1.0% even though it combined with certain amounts of TGase. No differences in chemical composition and physical properties were observed (p>0.05) among treatments. Approximately 0.3% of TGase can replace 10% emulsified meats, which are normally used for improvement of binding capacity to manufacture RMPs, without quality defects. This study suggests that TGase could be used for the manufacture of low-fat, low-salt functional meat products for the improvement of textural characteristics and binding capacity without adverse effects.

Theracurmin (Highly Bioavailable Curcumin) Prevents High Fat Diet-Induced Hepatic Steatosis Development in Mice

  • Curcumin, a hydrophobic polyphenol isolated from the Curcuma longa L. plant, has many pharmacological properties, including antioxidant, anti-inflammatory, and chemo-preventive activities. Curcumin has been shown to have potential in preventing nonalcoholic fatty liver disease (NAFLD). However, the low bioavailability of curcumin has proven to be a major limiting factor in its clinical adoption. Theracurmin, a highly bioavailable curcumin that utilizes micronized technology showed improved biological absorbability in vivo. The aim of this study was to investigate the role of theracurmin in modulating hepatic lipid metabolism in vivo. A fatty liver mouse model was produced by feeding mice a high fat diet (HFD; 60% fat) for 12 weeks. We found that treatment for 12 weeks with theracurmin significantly lowered plasma triacylglycerol (TG) levels and reduced HFD-induced liver fat accumulation. Theracurmin treatment lowered hepatic TG and total cholesterol (T-CHO) levels in HFD-fed mice compared to controls. In addition, theracurmin administration significantly reduced lipid peroxidation and cellular damage caused by reactive oxygen species in HFD-fed mice. Overall, these results suggest that theracurmin has the ability to control lipid metabolism and can potentially serve as an effective therapeutic remedy for the prevention of fatty liver.

삼채의 급여가 제 2형 당뇨마우스의 지질대사에 미치는 영향 (Effects of Allium hookeri on Lipid Metabolism in Type II Diabetic Mice)

  • Allium hookeri is a plant species native to India, Sri Lanka, Myanmar, and China. The plant is widely cultivated in Korea lately as a medicinal food item. This study was conducted to evaluate the effects of Allium hookeri (A. hookeri) on lipid metabolism in Type II diabetic mice (n=8/group, 5 groups). High fat diets with dextrin as a positive control (Dex), leaf (AL), root (AR), and fermented root (FAR) at 3% of diet were fed to all experimental mouse, respectively for 8 weeks. Body weight gain, liver and epididymal fat weights, and excreted fecal lipid levels were measured. Serum and hepatic lipid profiles were analyzed, and fat accumulation in liver was evaluated. In this study, body weight gain and epididymal fat weight were lower in the FAR group, while serum HDL-cholesterol level and excreted fecal total lipid and triglyceride levels were higher in AL or FAR groups. These results suggest that A. hookeri, specially fermented root can be a useful food item to control lipid metabolism in diabetic mice.
