Genetic Variability of Pleurotus ostreatus Monospore Isolates by Random Amplified Polymorphic DNA Analysis (DNA 다형성 분석에 의한 느타리버섯 단포자 분리주의 유전적 변이성)

  • This study was carried out to obtain data concerning the genetic variability of Pleurotus ostreatus. Monospores of P. ostreatus were isolated and cultured to estimate differences in the rate of mycelial growth and genetic similarity among the isolates. Although the overall growth rates were slow compared to their parental dikaryon except 2-MI 4, significant differences in the rate of mycelial growth were observed among isolates. Random amplified polymorphic DNA (RAPD) analysis using twenty six random primers showed 345 polymorphic DNA bands from 35 monospore isolates and their parental dikaryon. DNA bands ranged from 0.1 to 4.0 Kb in size. Most various polymorphic DNA bands within monospore isolates were obtained when we used J (OPA-01) or W (OPB-04). The 36-MI 103 showed totally different band patterns from those of the others. RAPD analysis revealed that there is approximatly 75% genetic similarity between monospore isolates and their parental dikaryon except 36-MI 103, which showed only 49% genetic similarity. In addition, genetic similarity degrees were classified into four groups: I (parental dikaryon), II (fast growing type), III (moderate growing type), and IV (slow growing type). However, there is no correlation between mating type and mycelial growth rates.

  • The foreign superior players and national team players' turning phase was measured, compared and analyzed to help the representative players improve the skill of turn. The underwater video camera used to analyze and evaluate the representative players' skill of turn in detail and the result is as follows. 1. The record for the phase of turn was similar to the rank of the last record. The improvement of the skill of turn was required because Korean players' record was lower than the foreign players' one. In case of 200m events the 1st turn was the fastest and it took more time as the turn is repeated. 2. It shows that the preparation phase and turing motion cause the difference between the players and within one player. 3. The horizontal movement of center of gravity moves to turning point slowly in the preparation phase, does not move nearly in the turning phase and increase again in the propulsion phase. Good record has short time for turn phase. The result means that the shorten the turning phase is the most important factor. Therefore the preparation for this is required. The vertical movement is maintained or increase a little and then move to from the turning phase. 4. The characteristic of horizontal velocity in center of gravity is that there is any big changes at the preparation phase, the faster velocity is found from the better record and the accelerating time is fast at the propulsion phasen. The wrong motion is made by not using the swimming velocity for fuming and waiting and more time is required by this. 5. The angle of knee when the player touch the turning point is 106.22-135.56 and the maximum angle of knee during the driving after the touch of tuning point is full extension. The size of maximum angle of knee did not match with the required time of propulsion phase. It seems that the individual difference is big when the players touch the turning point the angle of knee and the research for the individual angle which can reveal the maximum power should be carried out. The national team player's skill for the him is behind the foreign players' one and a lot of problems were found. It shows that the players could not practice the skill for turn during the training. The 1st reason for it is the various facilities like underwater window or analyzing equipment like underwater camera with which the coaches can teach and correct the players' detailed skill. It is need to evaluate the players' detailed skill exactly and correct it by objective data to complete the good skill in the future. In this regard, the investment for the facility is necessary.

The Practical Suggestion for the Production of Educational Animation for Prevention of Safety Accidents (안전사고 예방을 위한 교육용 애니메이션 제작에 대한 실천적 제안)

  • The Korean society has become increasingly concerned about the safety consciousness because of the Sewol Ferry disaster in April 2014. This shows the absence of social system and safety consciousness of Korean society that has been rapidly growing through fast industrialization. The purpose of this paper is to suggest guidelines for the production of a safety animation by approaching the discourse of public value. For this research process, first, literature review will be conducted by focusing on the safety culture, causes of safety accidents, safety education and educational effect using animation. Second, I will analyze 11 safety animation films by focusing on (1) construction of accident environment(narrative, character and background design) (2) exploration of dangerous elements and accident happening(accident type and dangerous elements, method of direction for accident happening, damage degree) (3) the guideline for safety behaviour. Third, guidelines for the production of safety animation will be suggested by analyzing the results of literature reviews and case studies in a comprehensive way. It is expected that this paper will be used as a basic data for academic research and animation production related to the safety accident.

The Dynamic Split Policy of the KDB-Tree in Moving Objects Databases (이동 객체 데이타베이스에서 KDB-tree의 동적 분할 정책)

  • Moving object databases manage a large amount of past location data which are accumulated as the time goes. To retrieve fast the past location of moving objects, we need index structures which consider features of moving objects. The KDB-tree has a good performance in processing range queries. Although we use the KDB-tree as an index structure for moving object databases, there has an over-split problem in the spatial domain since the feature of moving object databases is to increase the time domain. Because the over-split problem reduces spatial regions in the MBR of nodes inverse proportion to the number of splits, there has a problem that the cost for processing spatial-temporal range queries is increased. In this paper, we propose the dynamic split strategy of the KDB-tree to process efficiently the spatial-temporal range queries. The dynamic split strategy uses the space priority splitting method for choosing the split domain, the recent time splitting policy for splitting a point page to maximize the space utilization, and the last division policy for splitting a region page. We compare the performance of proposed dynamic split strategy with the 3DR-tree, the MV3R-tree, and the KDB-tree. In our performance study for range queries, the number of node access in the MKDB-tree is average 30% less than compared index structures.

  • Layered-earth Green's functions in electormagnetic (EM) surveys play a key role in modeling the response of exploration targets. They are computed through the Hankel transforms of analytic kernels. Computational precision depends upon the choice of algebraically equivalent forms by which these kemels are expressed. Since three-dimensional (3D) modeling can require a huge number of Green's function evaluations, total computational time can be influenced by computational time for the Hankel transform evaluations. Linear digital filters have proven to be a fast and accurate method of computing these Hankel transforms. In EM modeling for 3D inversion, electric fields are generally evaluated by the secondary field formulation to avoid the singularity problem. In this study, three components of electric fields for five different sources on the surface of homogeneous half-space were derived as primary field solutions. Moreover, reflection coefficients in TE and TM modes were produced to calculate EM responses accurately for a two-layered model having a sea layer. Accurate primary fields should substantially improve accuracy and decrease computation times for Green's function-based problems like MT problems and marine EM surveys.

Glycosyl glycerides from hydroponic Panax ginseng inhibited NO production in lipopolysaccharide-stimulated RAW264.7 cells

  • Background: Although the aerial parts of hydroponic Panax ginseng are reported to contain higher contents of total ginsenosides than those of roots, the isolation and identification of active metabolites from the aerial parts of hydroponic P. ginseng have not been carried out so far. Methods: The aerial parts of hydroponic P. ginseng were applied on repeated silica gel and octadecylsilane columns to yield four glycosyl glycerides (Compounds 1-4), which were identified based on nuclear magnetic resonance, infrared, fast atom bombardment mass spectrometry, and gas chromatography/mass spectrometry data. Compounds 1-4 were evaluated for inhibition activity on NO production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Results and conclusion: The glycosyl glycerides were identified to be (2S)-1-O-7(Z),10(Z),13(Z)-hexadecatrienoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (1), (2S)-1-O-linolenoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (2), (2S)-1-O-linolenoyl-2-O-linolenoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (3), and 2(S)-1-O-linoleoyl-2-O-linoleoyl-3-O-${\beta}$-$\small{D}$-galactopyranosyl-sn-glycerol (4). Compounds 1 and 2 showed moderate inhibition activity on NO production in LPS-stimulated RAW264.7 cells [half maximal inhibitory concentration ($IC_{50}$): $63.8{\pm}6.4{\mu}M$ and $59.4{\pm}6.8{\mu}M$, respectively] without cytotoxicity at concentrations < $100{\mu}M$, whereas Compounds 3 and 4 showed good inhibition effect ($IC_{50}$: $7.7{\pm}0.6{\mu}M$ and $8.0{\pm}0.9{\mu}M$, respectively) without cytotoxicity at concentrations < $20{\mu}M$. All isolated compounds showed reduced messenger RNA (mRNA) expression of interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, and tumor necrosis factor-${\alpha}$ in LPS-induced macrophage cells with strong inhibition of mRNA activity observed for Compounds 3 and 4.

Land Use Changes and Climate Patterns in Southeast Korea (우리나라 동남부 지역의 토지 이용과 기후 패턴 변화 분석)

  • Landscape structure changes over the past three decades were determined with land use and land cover(LULC) maps, and their relationships with mean air temperature time series were the analyzed for the Busan metropolitan area and South Kyeongsang Province, Korea. The geometric structures of the LULC data were quantitatively represented based on FRAGSTATS, a spatial pattern analysis program for quantifying landscape structure. FRAGSTATS-derived landscape metrics confirmed that there were major changes in LULC and landscape fragmentation in the region. Meteorological observation records showed that mean air temperature had increased from $14.1^{\circ}C$ in the 1990's to $14.8^{\circ}C$ in the 2000's in Busan. For South Kyeongsang Province, they increased from $13.2^{\circ}C$ to $13.9^{\circ}C$ during the same time period. These long-term temperature changes are correlated with typical spatial pattern changes of LULC in the southeastern region of the country. Spatial metrics analysis showed that urban area expanded from 9.7% to 26.8% of Busan while forest and agricultural land decreased by 9.6% and 14.9%, respectively over the past thirty years. The significant urbanization are tightly associated with deforestation, removal of agricultural land, and fast temperature increases since the 1990's. The urban area of South Kyeongsang Province rapidly increased, and it became 12 times as large as it was. The degree of temperature increases differed among three different sub-regions. The temperature increasing rate was lowest in the coastal region while the colder mountainous region had the highest figure.

Analysis of Land Use Change within Four Major River Areas Using High-Resolution Air-Photographs: The Case of the Nakdong River Basin (고해상도 항공사진을 이용한 4대강 하천구역 내 토지이용변화 분석 - 낙동강 유역을 사례로)

  • Landuse changes and cadastral information error categories in the four major river areas were analyzed for the use of policy data as cadastral re-arrangement of national and public lands would be required, using high-resolution air-photographs and cadastral maps before and after the river development. The study sites were the river areas of 40km around four dams of the Nakdong river where their landuses were changed most. As the results, national and public lands reached 79.9% of land parcels and 93.3% of land areas of the study sites similar with those of the four river areas, 84.3% of land parcels and 85.5% of land areas. The landuse classification of the study sites before the four river development was consisted most of 'river'(71.6%) and 'rice field'(12.3%), but after the development the 'river' was reduced to 42.7% and 'park area'(19.6%) including sport fields and 'mixed lots'(20.8%) were increased. Also, 86.7% of land parcels before the development could be reduced after the development if administrative districts and land ownerships were not considered. Cadastral information error categories can be found as cadastral polygon missing, polygon overlap, location and boundary non-coincidence, small polygon generation, and non-coincidence between cadastral boundary and river boundary. Landuse change monitoring method using air-photographs will be useful to analyze landuse state through fast information aquisition and to manage properties of national and public lands such as river areas.

A Load Emulator for Low-power Embedded Systems and Its Application (저전력 내장형 시스템을 위한 부하의 전력 소모 에뮬레이션 시스템과 응용)

  • The efficiency of power supply circuits such as DC-DC converters and batteries varies on the trend of the power consumption because their efficiencies are not fixed. To analyze the efficiency of power supply circuits, we need the temporal behavior of the power consumption of the loads, which is dependent on the activity factors of the devices during the operation. Since it is not easy to model every detail of those factors, one of the most accurate power consumption analyses of power supply circuits is measurement of a real system, which is expensive and time consuming. In this paper, we introduce an active load emulator for embedded systems which is capable of power measurement, logging, replaying and synthesis. We adopt a pattern recognition technique for data compression in that long-term behaviors of power consumption consist of numbers of repetitions of short-term behaviors, and the number of short-term behaviors is generally limited to a small number. We also devise a heterogeneous structure of active load elements so that low-speed, high-current active load elements and high-speed, low-current active load elements may emulate large amount and fast changing power consumption of digital systems. For the performance evaluation of our load emulator, we demonstrate power measurement and emulation of a hard drive. As an application of our load emulator, it is used for the analysis of a DC-DC converter efficiency and for the verification of a low-power frequency scaling policy for a real-time task.

An Algorithm for Segmenting the License Plate Region of a Vehicle Using a Color Model (차량번호판 색상모델에 의한 번호판 영역분할 알고리즘)

  • The license plate recognition (LPR) unit consists of the following core components: plate region segmentation, individual character extraction, and character recognition. Out of the above three components, accuracy in the performance of plate region segmentation determines the overall recognition rate of the LPR unit. This paper proposes an algorithm for segmenting the license plate region on the front or rear of a vehicle in a fast and accurate manner. In the case of the proposed algorithm images are captured on the spot where unmanned monitoring of illegal parking and stowage is performed with a variety of roadway environments taken into account. As a means of enhancing the segmentation performance of the on-the-spot-captured images of license plate regions, the proposed algorithm uses a mathematical model for license plate colors to convert color images into digital data. In addition, this algorithm uses Gaussian smoothing and double threshold to eliminate image noises, one-pass boundary tracing to do region labeling, and MBR to determine license plate region candidates and extract individual characters from the determined license plate region candidates, thereby segmenting the license plate region on the front or rear of a vehicle through a verification process. This study contributed to addressing the inability of conventional techniques to segment the license plate region on the front or rear of a vehicle where the frame of the license plate is damaged, through processing images in a real-time manner, thereby allowing for the practical application of the proposed algorithm.
