Roles of Forkhead-box Transcription Factors in Controlling Development, Pathogenicity, and Stress Response in Magnaporthe oryzae

  • Although multiple transcription factors (TFs) have been characterized via mutagenesis to understand their roles in controlling pathogenicity and infection-related development in Magnaporthe oryzae, the causal agent of rice blast, if and how forkhead-box (FOX) TFs contribute to these processes remain to be characterized. Four putative FOX TF genes were identified in the genome of M. oryzae, and phylogenetic analysis suggested that two of them (MoFKH1 and MoHCM1) correspond to Ascomycota-specific members of the FOX TF family while the others (MoFOX1 and MoFOX2) are Pezizomycotina-specific members. Deletion of MoFKH1 (${\Delta}Mofkh1$) resulted in reduced mycelial growth and conidial germination, abnormal septation and stress response, and reduced virulence. Similarly, ${\Delta}Mohcm1$ exhibited reduced mycelial growth and conidial germination. Conidia of ${\Delta}Mofkh1$ and ${\Delta}Mohcm1$ were more sensitive to one or both of the cell cycle inhibitors hydroxyurea and benomyl, suggesting their role in cell cycle control. On the other hand, loss of MoFOX1 (${\Delta}Mofox1$) did not show any noticeable changes in development, pathogenicity, and stress response. Deletion of MoFOX2 was not successful even after repeated attempts. Taken together, these results suggested that MoFKH1 and MoHCM1 are important in fungal development and that MoFKH1 is further implicated in pathogenicity and stress response in M. oryzae.

Factors Related to the Commute Time of Dual-earner Couples: From a Family Developmental Perspective (맞벌이 부부의 통근시간 관련 요인: 첫 자녀의 연령에 따른 집단별 비교)

  • This study observed and investigated the commute time of dual-earner couples from a family developmental perspective. It tested whether the household responsibility hypothesis was effective in explaining the gender gap in the commute time for these couples. I extracted 2,103 time diaries written by 1,266 matched couples from the 2014 Korean Time Use Survey data for this study. The sample was categorized by the age of the eldest child into four age groups of 0-6, 7-12, 13-18, and 19-29. Analytic results from ANOVA, Scheffé test, and OLS regression are as follows. First, husbands traveled longer hours to work than wives in all age groups, while the commute time of couples tended to increase along with the child' age. However, couples in the child group aged 7 to 12 had the shortest commute time. Second, domestic labor time of wives were negatively associated with the commute time, which appeared to support the household responsibility hypothesis. Third, in the child group aged 7 to 12, wives spent more time for work commute as their income increased; however, wives with traditional gender role attitudes had a shorter commute time in the child group aged 0 to 6. Forth, neither the wives' nor husbands' work characteristics were related to the wives' commute time; however, both wives' and husbands' work characteristics were related to the husbands' commute time. The findings suggest the possibility of spatial entrapment by working wives throughout the family life cycle due to household responsibilities, which provides implications for policy intervention in consideration of the gender gap in commute time for dual-earner couples.

A Study on the Relationship of Gender Equity within Family and Second Birth (둘째 출산 계획의 결정요인과 가족내 성 형평성)

  • This research explores the relationship between gender equity within family and second birth in Korea. For a factor analysis on the intention of having additional children for women with one child, I have used the data from the 2005 National Survey on Marriage and Fertility Trend" of the Korea Institute for Health and Social Affairs. Among 516 samples(year of 20-44), women planning to have second birth are 54.7%, and others have answered that they have no intention to have second birth. As a result, main causes affecting progressing to second birth seemed to be family planning on family cycle, such as controlling age gaps between first and second child, composing children genders as they wish etc. The variable of "gender equity between couples" in progressing to second birth are of significant only with married working women group. In case of married working women, more the husband's houseworking hours increase, the more prone to progress to second birth. However, in case of housewives, gender equity level, such as husband's housework hours or share of housework are not of significant influence in progressing to second birth. Not only working mothers but also housewives need the realization of gender equity within the family and the expansion of social support system for work-family life balance.

  • The purpose of this study is to analyze the trend of changes in household labor among middle-aged generations who are facing dynamic changes in the middle of their life cycle. The research method is a literature study that examines and analyzes related data. Specifically, it aims to analyze the basic characteristics of middle-aged generations and trends in household labor, which are revealed through Time Use Survey, market trend reports, newspaper articles, and books. From the perspective of family resource management, housework is an unpaid activity performed by and for family members, and is an activity that can be replaced by purchasing goods or services from the market. This study focuses on the rapid growth of the home appliance market that replaces housework with the development of technology after industrialization, and how the change of technology affected the efficiency of housework. In addition, the composition and characteristics of middle-aged generations greatly changed according to changes in family structure, form, and family value. Examining the current state of housework and changing trends of middle-aged generations, which have different characteristics from previous generations, is significant in that it can predict future market trends and suggest implications for family policies that support the improvement of the quality of life of middle-aged generations.

Mediation Effect of Family Resilience between Family Stress and Social Problem Solving Ability of Women's Social Worker in Middle Age (중년기 여성 사회복지사의 가족스트레스와 사회적 문제해결능력 간 가족탄력성의 매개효과)

  • This study examined the mediation effect of the family resilience between the family stress and social problem solving ability of the women social worker and welfare specialist, considering that women's middle age needs the resilience including the adaptation, reestablishment in the family life cycle. The sampling target was the 328 women social workers in the middle age, and it performed the Sobel Test for the mediation effect verification after enforcing the multiple regression analysis, next, it is same. Firstly, the economic problem was most highly severe among the family stress subfactor. Secondly, there was altogether the part mediation effect of the family resilience' subfactors, ie, communication processes, belief systems and organizational patterns between the family stress and social problem solving ability. In conclusion, if the family stress which the middle aged women social worker confronted was accurately analyzed and the family is intervened so that the family resilience functions properly, the social problem solving ability would be able to be improved.

Activity Space of Yangban Landlords during the Colonial Era : Based on the Kieo written by Ryu Hyeong-eop - (일기를 통해 본 한말${\sim}$일제강점기 양반소지주의 활동공간 - 류형업의 "기어"를 자료로 -)

  • This paper uses the diary entitled Kieo, which was kept by Ryu Hyeong-eop for 38 years, to analyze the active space of small-scale Yangban landlords. Ryu lived during the period spanning from the end of the Taehan Empire through the colonial era. The researcher classified the contents of Kieo into different sections which were divided according to different stages of the writer's life, his objectives, destinations, and periods, and based on the writer's life-cycle, strived to recreate his spatial activities. As a result, researcher was able to ascertain that Ryu's spatial activities gradually increased, and that his active space also continuously expanded. Ryu's active space during his youth was narrow and centered around certain specific places. However, this active space gradually increased as time went by. It was during the middle stages of his life that the volume of his spatial activities was the heaviest. In addition, the objectives of his outings also became more diversified over time. This change was originated in the larger changes in his roles within the family and community, as well as by the changes in the periodic situation. Moreover, the regional sphere of Ryu's outings, including his day and over nights trips, allowed the researcher to form a better understanding of the daily living area of the Yangban.

Life Cycle-Based Host Range Analysis for Tomato Spotted Wilt Virus in Korea

  • Tomato spotted wilt virus (TSWV) is one of the plant viruses transmitted by thrips and causes severe economic damage to various crops. From 2008 to 2011, to identify natural host species of TSWV in South Korea, weeds and crops were collected from 5 regions (Seosan, Yesan, Yeonggwang, Naju, and Suncheon) where TSWV occurred and were identified as 1,104 samples that belong to 144 species from 40 families. According to reverse transcription-polymerase chain reaction, TSWV was detected from 73 samples from 23 crop species, 5 of which belonged to family Solanaceae. Additionally, 42 weed species were confirmed as natural hosts of TSWV with three different life cycles, indicating that these weed species could play an important role as virus reservoirs during no cultivation periods of crops. This study provides up-to-date comprehensive information for TSWV natural hosts in South Korea.

The Factors Affecting Dining-out Expenses by Generation X and Generation MZ Based on Tobit Model (토빗분석을 적용한 X세대, MZ세대의 외식지출 영향요인)

  • The food service industry has grown larger with changes in the economic and socio-cultural environment. In this saturated food service industry, generation X and MZ are the main consumer forces that demand attention. That is because a generation is the main psychographic factor that reflects personal values and lifestyle based on one's life cycle. From such a perspective, a generation in marketing has been used as a variable to predict a market by supplementing demographic factors. Accordingly, this study classified generations into generation X and generation MZ with the use of the 2019 consumer behavior survey for food by Korea Rural Economic Institute (KREI) and then investigated the factors influencing group and personal dining-out expenses. The analysis was carried out applying the Tobit model using SPSS and R. The positively influential variables on generation X's personal dining-out expenditure were male, single person, high income and simple lifestyle, whereas housewives, personal ethical consciousness, behavioral ethical consciousness, and safe dietary life were negatively influential variables. The positively influential variables on generation MZ's personal dining-out expenditures were male, dual-income, high education level, corporate and governmental ethical consciousness, while the number of family members and safe dietary life were negatively influential variables.

Involvement of TGF-β1 Signaling in Cardiomyocyte Differentiation from P19CL6 Cells

  • Stem cell-based therapy is being considered as an alternative treatment for cardiomyopathy. Hence understanding the basic molecular mechanisms of cardiomyocyte differentiation is important. Besides BMP or Wnt family proteins, $TGF-{\beta}$ family members are thought to play a role in cardiac development and differentiation. Although $TGF-{\beta}$ has been reported to induce cardiac differentiation in embryonic stem cells, the differential role of $TGF-{\beta}$ isoforms has not been elucidated. In this study, employing the DMSO-induced cardiomyocyte differentiation system using P19CL6 mouse embryonic teratocarcinoma stem cells, we investigated the $TGF-{\beta}$-induced signaling pathway in cardiomyocyte differentiation. $TGF-{\beta}1$, but not the other two isoforms of $TGF-{\beta}$, was induced at the mRNA and protein level at an early stage of differentiation, and Smad2 phosphorylation increased in parallel with $TGF-{\beta}1$ induction. Inhibition of $TGF-{\beta}1$ activity with $TGF-{\beta}1$-specific neutralizing antibody reduced cell cycle arrest as well as expression of the CDK inhibitor $p21^{WAF1}$. The antibody also inhibited induction of the cardiac transcription factor Nkx2.5. Taken together, these results suggest that $TGF-{\beta}1$ is involved in cardiomyocyte differentiation by regulating cell cycle progression and cardiac gene expression in an autocrine or paracrine manner.

Expressed Sequence Tag Analysis of Toxic Alexandrium tamarense and Identification of Saxitoxin Biosynthetic Genes (독성 Alexandrium tamarense 의 EST 분석 및 삭시톡신 생합성 유전자의 확인)

  • Expressed sequence tag (EST) library was constructed from A. tamarense. Base sequences of EST clones were analyzed and saxitoxin biosynthesis-related genes were cloned. Sequences of 827 clones were analyzed and 564 EST were functionally clustered using Blast searches against GenBank. Main genes in the EST had functions on cellular organization, cell metabolism, energy, cell cycle and DNA processing, cellular transport and transport, cell rescue, defense, death and aging, and transcription. Moreover, expression of S-adenosylmethionine synthetase and H2A histone family genes were increased in the toxic A. tamarense. These results show that two genes could be a good biomarkers for the detection of saxitoxin biosynthesis in the A. tamarense.
