『동의보감(東醫寶鑑)』에 활용된 천초(川椒)(촉초(蜀椒), 화초(花椒)) 약대(藥對)에 관한 연구(硏究) (Study on a Herb pair of Pericarpium Zanthoxyli(Zanthoxyli Fructus) in 『Donguibogam』)

  • Objectives : The purpose of this study is to find out herb-pair prototype composed of Pericarpium Zanthoxyli in "Donguibogam", to investigate origin of them. Methods : We researched the usages of Pericarpium Zanthoxyli compatibility as following procedure : 1. Choosing a related words such as Pericarpium Zanthoxyli(蜀椒, 川椒, 花椒) 2. Searching prescriptions: We searched prescriptions containing the related words in "Dongeuibogam" through "wook system", analyzed the selected prescription and found out herb-pair prototype composed of Pericarpium Zanthoxyli. Results : We found 12 fomulas in which Pericarpium Zanthoxyli herb pair was combinated and We made a Table 1. for the description of name of fomula, indication and herb configuration. Conclusions : Pericarpium Zanthoxyli was paired with below herbs; 檳榔(Arecae Semen), 細辛(Asari Radix), 苦楝根(Meliae Radix), 蔥白(Allii Fistulosi Bulbus), 茴香(Foeniculi Fructus), 巴豆(Crotonis Semen), 蓽撥(Piperis Longi Fructus), 露蜂房(Vespae Nidus), 肉桂(Cinnamomi Cortex), 艾葉(Artemisiae Argyi Folium), 細茶(Camellia sinensis), 雄黃(Realgar), 杏仁(Armeniacae Amarum Semen).

랫드와 마우스에서 연생호보액의 급성 경구 독성 시험에 관한 연구 (Acute Toxicity Study of Yan-Sheng Health Liquid in Rats and Mice)

  • The acute toxicity of orally administered Yan-Sheng health liquid (YSHL), water extracts from twelve Chinese drugs (Cervi Cornu, Lonicerae Flos, Foeniculi Fructus, Glycyrrhizae Radix, Liriopis Tuber, Raphani Semen, Bombyx, Ginseng Radix alba, Cinnamomi loureirii Cortex, Epimedii Herba, Zingiberis Rhizoma, Lycii Fructus) was evaluated in male and female Sprague-Dawley rats and ICR mice. Rats and mice aging 5 weeks were gavaged with 0, 2.0, 3.0, 4.4, 6.7, 10.0, 66.7, or 100.0 ml/kg of YSHL. No animal died by oral treatment and no toxic symptom was observed in the treated animals during 5 days. The body weight of the treated animals was not significantly different from the controls. The results of macroscopic examination on the organs of the treated animals revealed no abnormal findings. Therefore, it was concluded that YSHL was practically non-toxic when it was orally administered to rats and mice, and its LD50 was suspected to be greater than 100 ml/kg in rats and mice.

산형과 과실 약재의 형태 및 해부학적 특성 (Morphological and Anatomical Characteristics of Medicinal Fructus in Apiaceae)

  • Background : As the characterization of medicinal plants is an important aspect of traditional Asian herbal medicine, this study examines the morphological and anatomical characteristics in four fructus form medicinally important plants belonging to the family Apiaceae. Methods and Results : Fruit morphology of four genera was examined using microtome sections and light microscopy. The Anethi Fructus, Anethum graveolens (Siraja) has unique wing-like and membranous lateral ribs that are, approximately $500{\mu}m$ wide. The Coriandri Fructus, Coriandrum sativum (Hoyuja) does not have oil ducts at the dorsal region of the mericarp and differs in the development of the dorsal ribs. The ribs appear upon drying and therefore, consist of primary and secondary ribs. The Foeniculi Fructus, Foeniculum vulgare (Hoehyang) characteristically develops three dominant dorsal ribs and has a strong aromatic fragrance. Finally, the Cnidii Fructus, Torilis japonica (Sasangja) has many oil ducts at the dorsal and carpophores regions and develops many hooked trichomes one the fructus surface. Conclusions : We conclude from this study that each plant has several prominent and distinguishing morphological and anatomical characteristics. Therefore, fruit morphology is very useful for identifying these medicinal plants. In addition, the correct use and herbal name need to be standardized for plants used traditionally in Asian medicine within Korea, China, and Japan.

  • Cancer is one of the most important cause of death. So recently, investigation of cancer progress prosperously all over the world. Cancer in the present medicine correspond to You-Am, Sin-Am, Young-Soon, Sel-Gyun, Sil-Young, Young-Lyoo, Seg-Je, Seg-Young, Seg-Ha, Jerk-Chui(積聚), Jing-Ha, Oel-Gyek, Ban-Oui, Bi-Gi, Bok-Lyang, Jang-Dan, Hyen-Bek in the oriental medicine. Among these, generally Jerk-Chui(積聚) is expressed to cancer. So to develop of new drugs of cancer in the present medicine, bibliographic investigation of mass-prescriptions was studied in the oriental medicine-books. According to the bibliographic study of Jerk-Chui-prescriptions, the results run as follows. 1. According to the nature and taste in the drugs, warm and heating recipes were used most, heatclearing recipes were used a few times assistantly. 5. The Jerk-Chui-prescription used frequently was Bun-Don-Tang, which was used 13 times ; Bok-Oyang-Hoan 12 times, Bi-Gi-Hoan(肥氣丸) 12 times, Sik-Boon-Hoan 12 times, A-Uie-Hoan 12 times, Bi-Gi-Hoan 12 times, Dai-Cil-Gi-Tang 8 times, San-Cuie-Tang 8 times, Guye-Gyen-Tang 6 times, On-Baig-Won 5 times, So-Jek-Jeng-Ouen-San 5 times, Jin-In-Hoa-Cel-Tang 5 times, Byel-Gab-San 5 times, Sng-Hong-Hoan 5 times, Ji-Sil-San 4 times, So-A-Oie-Hoan 4 times, Hyang-Rng-Hoan 4 times.

Apoptosis Induction Effect of Zingiberis Rhizoma Extract in Microglia BV-2 Cells

  • Microglia have multiple functions in regulating homeostasis of the central nervous system. Microglia cells have been implicated as active contributors to neuron damage in neurodegenerative disorders. In this study, medicinal plant extracts (MPEs) were used to evaluate the cell-death induction effect in microglia BV-2 cells. Among 35 MPEs tested in this study, 4 MPEs showed less than a 30% cell survival after 24 hours of incubation. These were Foeniculi Fructus, Forsythiae Fructus, Zingiberis Rhizoma and Hedera Rhombea. The concentration showed that 50% cell death ($IC_{50}$) occurred with 33, 83, 67 Ed highlight: Please confirm wording, and $81{\mu}/ml$, respectively. For further study, we chose Zingiberis Rhizoma (ZR) which showed a reasonably low $IC_{50}$ value and an induction of cell death in a relatively narrow range. Western blot analysis showed that ZR-treated cells showed activation of caspase-3 and cleavage of PARP Ed highlight: When an acronym is first presented it needs to be spelled out in both dose- and time-dependent manners. However, the level of Bcl-2 and Bax were not changed by ZR-treatment in BV-2 cells. These results suggest that ZR-induced apoptosis in BV-2 cells occured through caspase-3 activation. The results also suggested that ZR may be useful in developing treatments for neurodegenerative diseases.

『醫林改錯』 처방의 현대 질병 범위에 관한 연구 (Study of Clinical Application of Pathology of Blood Stasis, Focused on 33 Prescriptions in 『Yilingaicuo』)

  • This paper researches the features of blood stasis theory of Wangqingren, who wrote 『Yilingaicuo』 that greatly contributed in the development of blood stasis theory at Qing dynasty period. And the disease cause, disease mechanism of blood stasis and scope of modern diseases related with blood stasis are studied by research on clinical papers which used 33 prescriptions in 『Yilingaicuo』 in modern times. Research on the features of blood stasis theory of Wangqingren is proceeded by referring to the annotations of 『Yilingaicuopingyi』 and the papers which related with blood stasis from Korea and China. And clinical papers are searched in China Academic Journals(CAJ) of China National Knowledge Infrastructure(CNKI) to analyse the scope of modern diseases related with blood stasis. The features of blood stasis theory in 『Yilingaicuo』 expanded the range of existing theory. Wangqingren thought that chronic disease, weird disease, the disease of no effect from normal treatments were related with blood stasis. And he attached great importance to qi and blood and thought that the main pathogenesis of blood stasis was qi deficiency. And a lot of Astragalus membranaceus Bunge were combined in many prescriptions to reinforce qi. He also used different herbs according to the location of the disease. Musk and Allium fistulosum were used for the disease located at head or upper part of the patient's trunk. Bupleurum falcatum L., Aurantii Fructus Pericarpium and Platycodon grandiflorum A. De Candolle were used for the disease located at thorax. Cyperus rotundus L., Linderae Radix and Aurantii Fructus Pericarpium were used for the disease located at the stomach or below the costal angle. Foeniculi Fructus and Corydalis remota were used for the disease located at belly or lower part of the patient's trunk. Trogopterorum Faeces, myrrha, Cyperus rotundus L. and Cnidium officinale were used for the disease located at extremity or joint.

5-Lipoxygenase Inhibition of the Fructus of Foeniculum vulgare and Its Constituents

  • The fruits of Foeniculum vulgare (Foeniculi Fructus) have been widely used in Chinese medicine as an antiemetic, ameliorating stomach ailments and as an analgesic. In order to establish its potential for antiallergic use, inhibitory actions of the fruit on 5-lipoxgenase (5-LOX) and ${\beta}$-hexosaminidase release were evaluated. The 70% ethanol extract of this plant material (FR) considerably inhibited 5-LOX-catalyzed leukotriene production from A23187-induced rat basophilic leukemia (RBL)-1 cells. The $IC_{50}$ was $3.2{\mu}g/ml$. From this extract, 12 major compounds including sabinene, fenchone, ${\gamma}$-terpinene, ${\alpha}$-pinene, limonene, p-anisylacetone, panisylaldehyde, estragole (4-allylanisole), trans-anethole, scopoletin, bergapten and umbelliferone were isolated. And it was found that several terpene derivatives including ${\gamma}$-terpinene and fenchone as well as phenylpropanoid, trans-anethole, showed considerable inhibitory action of 5-LOX. In particular, the $IC_{50}$ of trans-anethole was $51.6{\mu}M$. In contrast, FR and the isolated compounds did not show considerable inhibitory activity on the degranulation reaction of ${\beta}$-hexosaminidase release from antigen-treated RBL-2H3 cells. Against arachidonic acid-induced ear edema in mice, FR and trans-anethole showed significant inhibition by oral administration at doses of 100-400 mg/kg. In conclusion, FR and several major constituents are 5-LOX inhibitors and they may have potential for treating 5-LOX-related disorders.

HBX-6의 Sprague-Dawley rat를 이용한 단회경구투여 독성시험 (Single Oral Dose Toxicity Test of HBX-6 in Sprague-Dawley Rat)

  • Objectives : This study was performed to investigate the single oral toxicity of HBX-6 in Sprague-Dawley (SD) rats. Methods : Twenty SD rats were randomly assigned to four groups of 5 rats each and were administrated singly to female and male SD rats, as an oral dose of 2000 mg/kg. HBX-6 is a newly combined Korean herbal medicine formula 30 % Ethanol extract derived from The Dongui Bogam. Now we are developing the prescription for the aim of improving benign prostatic hyperplasia (BPH) without undesirable side effects. HBX-6 is composed of nine medicinal herbs: Aconiti Lateralis Radix Preparata, Corni Fructus, Cistanchis Herba, Psoraleae Semen, Dendrobii Herba, Morindae Radix, Cuscutae Semen, Trigonellae Semen, Foeniculi Fructus. Animals were monitored for the mortality and changes in the body weight, clinical signs, gross observation and necropsy findings for the 14 days according to "Standard for Toxicity Study of Pharmaceuticals" of Korea Food and Drug Administration (KFDA) guideline and "Acute Oral Toxicity - Fixed Dose Procedure" of OECD Test Guideline. Results : We could not find any mortality. Compared with the control group, significant weight change was not observed in the experimental group. After administration, the more common symptoms were not observed. There were no gross abnormalities in all cases. Conclusions : Taken together, these results suggest that the approximate lethal dose of HBX-6 in both female and male SD rats were considered as over 2000 mg/kg.

미세아교세포에서 알츠하이머형 치매 치료 처방인 뇌명산(腦明散)의 효능 및 기전연구 (Effects and molecular mechanisms of Noemyeong-san, a novel herbal prescription for treating Alzheimer's disease on microglia)

  • Objectives : Noemyeong-san (NMS) is a novel herbal prescription composed of five oriental medicinal herbs including Prunellae Spica, Betulae Cortex, Foeniculi Fructus, Asiasari Radix, and Clematidis Radix for treating Alzheimer's disease. In the present study, we investigated the effects and molecular mechanisms of NMS on BV2 microglia to evaluate the potential action of this formula for preventing or treating neurodegenerative disease such as Alzheimer's disease. Methods : To determine the cytotoxicity of NMS on BV2 microglia, the MTT assay was performed. The effects of NMS on lipopolysaccharide (LPS)-stimulated BV2 microglia were determined with a nitric oxide (NO) assay and western blots for inflammatory mediator-related proteins, mitogen activated protein kinases (MAPKs), nuclear factor kappa B (NF-${\kappa}B$) pathway-related proteins, and heme oxygenase-1 (HO-1). Result : NMS inhibited induction of iNOS and COX-2 as well as NO production without affecting the cell viability in LPS-stimulated BV2 microglia. NMS also suppressed activation of ERK and p38 MAPK among main kinases of MAPKs as well as NF-${\kappa}B$ by LPS stimulation. Furthermore, NMS dose-dependently induced the expression of HO-1 and the inhibitory effect of NMS on the production of NO were blocked by pretreatment with an HO-1 inhibitor, Snpp. Conclusions : These results demonstrate that NMS has potent anti-neuroinflammatory effect on the LPS-stimulated microglia. These findings provide evidences for NMS to be considered as a new prescription for preventing or treating neurodegenerative disease such as Alzheimer's disease.

수종 산형과 한약재의 혈관이완 및 혈압강하 효능에 대한 문헌고찰 (Vasorelaxant or hypotensive effects of herbal medicines belong to Umbelliferae family: a review)

  • Objectives : The objective of this study was to investigate the possibility for the treatment of hypertension of herbal medicines belong to Umbelliferae family. Methods : Domestic and international articles about Herbology were investigated. A review was performed via the database (DB) search engines such as Pubmed, Korean studies Information Service System (KISS), KoreaScience, and Google Scholar. Hypertension-related terms including "vasorelaxation", "vasorelaxant", "vasodilation", "vasodilatory", "vasodilative", "hypotension", and "hypotensive" were performed as search terms. Results : A list was made about herbal medicines and origin plants belonging to the Umbelliferae family in Korean Pharmacopoeia 12 and Korean Herbal Pharmacopoeia. 14 herbal medicine and 22 origin plants were searched. Ostericum koreanum root and rhizome, Notopterygium incisum root and rhizome, N. forbesii root and rhizome, Ligusticum tenuissimum root and rhizome, L. jeholense root and rhizome, Angelica gigas root, A. dahurica root, A. dahurica var. formosana root, Bupleurum falcatum root, Peucedanum japonicum root, P. praeruptorum root, A. decursiva root, Cnidium officinale rhizome, L. chuanxiong rhizome, Foeniculum vulgare fruit, and Ferula assa-foetida resin and stem showed significant vasorelaxant or hypotensive effects. Conclusion : These review results showed that Osterici seu Notopterygii Radix et Rhizoma, Ligustici Tenuissimi Rhizoma et Radix, Angelicae Gigantis Radix, Angelicae Dahuricae Radix, Bupleuri Radix, Peucedani Japonici Radix, Peucedani Radix, Cnidii Rhizoma, Foeniculi Fructus, and Ferulae Resina had vasorelaxant or hypotensive effects. The results are expected as basic data in clinical trials and experimental researches for the treatment of hypertension of herbal medicines.
