Rapid and Sensitive Detection of Lettuce Necrotic Yellows Virus and Cucumber Mosaic Virus Infecting Lettuce (Lactuca sativa L.) by Reverse Transcription Loop-Mediated Isothermal Amplification

  • Cucumber mosaic virus (CMV) is damaging to the growth and quality of lettuce crops in Lanzhou, China. Recently, however, for the first time an isolate of lettuce necrotic yellows virus (LNYV) has been detected in lettuce crops in China, and there is concern that this virus may also pose a threat to lettuce production in China. Consequently, there is a need to develop a rapid and efficient detection method to accurately identify LNYV and CMV infections and help limit their spread. Reverse transcription loop-mediated isothermal amplification (RT-LAMP) assays were developed to detect the nucleoprotein (N) and coat protein (CP) genes of LNYV and CMV, respectively. RT-LAMP amplification products were visually assessed in reaction tubes separately using green fluorescence and gel electrophoresis. The assays successfully detected both viruses in infected plants without cross reactivity recorded from either CMV or LNYV or four other related plant viruses. Optimum LAMP reactions were conducted in betaine-free media with 6 mM Mg2+ at 65℃ for LNYV and 60℃ for 60 min for CMV, respectively. The detection limit was 3.5 pg/ml and 20 fg/ml using RT-LAMP for LNYV and CMV plasmids, respectively. Detection sensitivity for both RT-LAMP assays was greater by a factor of 100 compared to the conventional reverse transcription polymerase chain reaction assays. This rapid, specific, and sensitive technique should be more widely applied due to its low cost and minimal equipment requirements.

Molecular Markers for Detecting a Wide Range of Trichoderma spp. that Might Potentially Cause Green Mold in Pleurotus eryngii

  • In Pleurotus sp., green mold, which is considered a major epidemic, is caused by several Trichoderma species. To develop a rapid molecular marker specific for Trichoderma spp. that potentially cause green mold, eleven Trichoderma species were collected from mushroom farms and the Korean Agricultural Culture Collection (KACC). A dominant fungal isolate from a green mold-infected substrate was identified as Trichoderma pleuroticola based on the sequences of its internal transcribed spacer (ITS) and translation elongation factor 1-α (tef1) genes. In artificial inoculation tests, all Trichoderma spp., including T. atroviride, T. cf. virens, T. citrinoviride, T. harzianum, T. koningii, T. longibrachiatum, T. pleurotum, and T. pleuroticola, showed pathogenicity to some extent, and the observed symptoms were soaked mycelia with a red-brown pigment and retarded mycelium regeneration. A molecular marker was developed for the rapid detection of wide range of Trichoderma spp. based on the DNA sequence alignment of the ITS1 and ITS2 regions of Trichoderma spp. The developed primer set detected only Trichoderma spp., and no cross reactivity with edible mushrooms was observed. The detection limits for the PCR assay of T. harzianum (KACC40558), T. pleurotum (KACC44537), and T. pleuroticola (CAF-TP3) were found to be 500, 50, and 5 fg, respectively, and the detection limit for the pathogen-to-host ratio was approximately 1:10,000 (wt/wt).

A Deep Optical Survey of Young Stars in the Carina Nebula. I. UBVRI Photometric Data and Fundamental Parameters

  • We present the deep homogeneous UBV RI photometric data of 135,071 stars down to V ~ 23 mag and I ~ 22 mag toward the Carina Nebula. These stars are cross-matched with those from the previous surveys in the X-ray, near-infrared, and mid-infrared wavelengths as well as the Gaia Early Data Release 3 (EDR3). This master catalog allows us to select reliable members and determine the fundamental parameters distance, size, stellar density of stellar clusters in this star-forming region. We revisit the reddening toward the nebula using the optical and the near-infrared colors of early-type stars. The foreground reddening [E(B-V)fg] is determined to be 0.35 ± 0.02, and it seems to follow the standard reddening law. On the other hand, the total-to-selective extinction ratio of the intracluster medium (RV,cl) decreases from the central region (Trumpler 14 and 16, RV,cl ~ 4.5) to the northern region (Trumpler 15, RV,cl ~ 3.4). It implies that the central region is more dusty than the northern region. We find that the distance modulus of the Carina Nebula to be 11.9 ± 0.3 mag (d = 2.4 ± 0.35 kpc) using a zero-age main-sequence fitting method, which is in good agreement with that derived from the Gaia EDR3 parallaxes. We also present the catalog of 3,331 pre-main-sequence (PMS) members and 14,974 PMS candidates down to V ~ 22 mag based on spectrophotometric properties of young stars at infrared, optical, and X-ray wavelengths. From the spatial distribution of PMS members and PMS candidates, we confirm that the member selection is very reliable down to faint stars. Our data will have a legacy value for follow-up studies with different scientific purposes.

Using DQ method for vibration analysis of a laminated trapezoidal structure with functionally graded faces and damaged core

  • This paper has focused on presenting vibration analysis of trapezoidal sandwich plates with a damaged core and FG wavy CNT-reinforced face sheets. A damage model is introduced to provide an analytical description of an irreversible rheological process that causes the decay of the mechanical properties, in terms of engineering constants. An isotropic damage is considered for the core of the sandwich structure. The classical theory concerning the mechanical efficiency of a matrix embedding finite length fibers has been modified by introducing the tube-to-tube random contact, which explicitly accounts for the progressive reduction of the tubes' effective aspect ratio as the filler content increases. The First-order shear deformation theory of plate is utilized to establish governing partial differential equations and boundary conditions for the trapezoidal plate. The governing equations together with related boundary conditions are discretized using a mapping-generalized differential quadrature (GDQ) method in spatial domain. Then natural frequencies of the trapezoidal sandwich plates are obtained using GDQ method. Validity of the current study is evaluated by comparing its numerical results with those available in the literature. After demonstrating the convergence and accuracy of the method, different parametric studies for laminated trapezoidal structure including carbon nanotubes waviness (0≤w≤1), CNT aspect ratio (0≤AR≤4000), face sheet to core thickness ratio (0.1 ≤ ${\frac{h_f}{h_c}}$ ≤ 0.5), trapezoidal side angles (30° ≤ α, β ≤ 90°) and damaged parameter (0 ≤ D < 1) are carried out. It is explicated that the damaged core and weight fraction, carbon nanotubes (CNTs) waviness and CNT aspect ratio can significantly affect the vibrational behavior of the sandwich structure. Results show that by increasing the values of waviness index (w), normalized natural frequency of the structure decreases, and the straight CNT (w=0) gives the highest frequency. For an overall comprehension on vibration of laminated trapezoidal plates, some selected vibration mode shapes were graphically represented in this study.

Free and forced vibration analysis of FG-CNTRC viscoelastic plate using high shear deformation theory

  • This paper investigates the dynamic behavior of a simply supported viscoelastic plate made of functionally graded carbon nanotube reinforced composite under dynamic loading. Carbon nanotubes are distributed in 5 different shapes: U, V, A, O and X, depending on the shape they form through the thickness of the plate. The displacement fields are derived in the Laplace domain using a higher-order shear deformation theory. Equations of motion are obtained through the application of the energy method and Hamilton's principle. The resulting equations of motion are solved using Navier's method. Transforming the Laplace domain displacements into the time domain involves Durbin's modified inverse Laplace transform. To validate the accuracy of the developed algorithm, a free vibration analysis is conducted for simply supported plate made of functionally graded carbon nanotube reinforced composite and compared against existing literature. Subsequently, a parametric forced vibration analysis considers the influence of various parameters: volume fractions of carbon nanotubes, their distributions, and ratios of instantaneous value to retardation time in the relaxation function, using a linear standard viscoelastic model. In the forced vibration analysis, the dynamic distributed load applied to functionally graded carbon nanotube reinforced composite viscoelastic plate is obtained in terms of double trigonometric series. The study culminates in an examination of maximum displacement, exploring the effects of different carbon nanotube distributions, volume fractions, and ratios of instantaneous value to retardation times in the relaxation function on the amplitudes of maximum displacements.

국내에서 서식하는 쥐똥밀깍지벌레(Ericerus pela)의 분포 및 생태에 관한 연구 (Studies on the Regional Distribution and Some Ecological Characteristics of Chinese White-wax Scale in Korea)

  • 국내에서 서식하고 있는 쥐똥밀까지벌레(Ericerus pela Chavannes)의 지역별 분포 및 월동, 산란 등 몇가지 생태적 특성을 조사한 결과, 본 종의 분포는 증평지역이 조사가지 대비 10.8%의 밀도율을 보여 가장 높았고 영월과 평택지역에서는 각각 1.1%로 가장 낮았다. 암컷성충의 월동후 생종율은 평균 85.3%로 비교적 높은 경향이었고, 월동직후에서 산란직전까지 암컷성충은 약 3.5배의 성장률을 보였다. 암컷 1개체당 평균 산란수는 7,783.5개로서 조사개체의 36.7%가 7,000~10,000개의 범위내에서 산란하였으며 난의 크기는 장경 및 단경이 각각 0.40mm, 0.21mm. 1령유충은 0.52mm, 0.29mm, 2령유충은 0.97mm 0.60mm이었다. 난의 부화율은$ 27^{\circ}C$에서 66.8%로 가장 높았고 용화율은 조사온도 모두 50.0%미만으로 낮았으며 우화율은 $25^{\circ}C$에서 67.3%fg 가장 높았다. 본 종의 총 발육기간은 $25^{\circ}C$, $27^{\circ}C$,$30^{\circ}C$에서 각각 93.27일, 76.83일, 65.42일로서 고온하에서 발육기간이 짧아지는 경향이었다. 한편 난을 일정기간 저온으로 보존한 후 부화율을 조사한 결과 $15^{\circ}C$에서 최대 50일간 보존이 가능함을 알수 있었다.

2009년산 옥수수와 벼에서의 Fusarium 곰팡이독소 자연발생량 조사 (Natural Occurrence of Fusarium Mycotoxins in Field-collected Maize and Rice in Korea in 2009)

  • 2009년 국내산 옥수수 19점과 벼 32점을 대상으로 Fusarium 오염 및 Fusarium 독소 오염을 조사하였다. 옥수수와 벼 시료의 Fusarium 오염률은 각각 54.9%와 8.2%로 확인되었으며, 종 특이 primer를 이용한 PCR 증폭결과 옥수수시료에서 분리된 506균주 중 58균주의 F.graminearum 추정균주(11.5%)와 354균주의 F. verticillioides 추정균주(70.0%)가 동정되었다. 또한 벼의 경우, 분리된 315균주 중 276균주(87.8%)가 F. graminearum으로 추정 되었으며, F. verticillioides 추정균주는 검출되지 않았다. LC 및 LC-MS를 이용한 Fusarium 독소(DON, NIV, ZEA, FB)의 자연발생량 조사 결과, DON과 ZEA이 각각 2개의 옥수수 시료에서만 기준치 이상 검출되었다. FB는 대부분의 옥수수 시료와 한 개의 벼 시료에서 검출 되었으나 모두 기준치 이하였다. 따라서 본 연구에서 사용된 2009년산 옥수수와 벼의 곰팡이독소 오염수준은 대부분 기준치 이하로 심각하지 않았다.

두 복사전달모델 RTTOV와 CRTM으로부터 산출된 밝기온도와 관측된 밝기온도의 비교 (A Comparison of Observed and Simulated Brightness Temperatures from Two Radiative Transfer Models of RTTOV and CRTM)

  • RTTOV와 CRTM은 복사관측자료에 대한 관측연산자로 수치예보에 활용되고 있는 빠른 속도의 복사전달모델이다. 본 연구에서는 두 모델의 기본구조 및 입력자료를 비교했다. 또한, 다양한 파장대를 가진 AMSU-A 마이크로파 센서에 대해 구름에 대한 정보를 포함할 때와 포함하지 않을 때 두 모델로부터 계산된 밝기온도와 관측된 밝기온도를 해양에 대해 비교했다. AMSU-A의 탐측채널(5-14)에 대해서는 두 모델로부터 계산된 밝기온도 값에 큰 차이가 존재하지 않았으나, 대기의 창 채널 및 지표근처의 탐측채널에서는 RTTOV로부터 계산된 밝기온도 값이 관측과 더 가까워 CRTM에 비해 상대적으로 작은 초기추정오차를 보였다. 한편 UM으로부터 제공된 구름물과 얼음의 정보를 추가적으로 활용하였을 때 두 모델로부터 계산된 밝기온도와 관측된 밝기온도의 차이가 감소함을 확인할 수 있었고, 특히 CRTM의 31.4 GHz와 89 GHz 채널에서 모의된 밝기온도와 관측된 밝기온도의 차이가 크게 감소했다.

감마선을 조사한 생쥐 간에서 발효인삼이 Glutathione 함량에 미치는 영향 (Enact of Glutathione(GSH/GSSG) Contents of Fermented Ginseng on the ${\gamma}$-irradiated Liver of Mice)

  • 목 적: $^{60}Co\;{\gamma}$선을 생쥐에 전신조사로 유도된 생쥐 간의 장해에 대한 발효인삼의 방사선방호효과를 방호제인 glutathione(GSH) 함량 변화를 통하여 실험적 연구를 하였다. 대상 및 방법: ICR종 수컷 생쥐를 발효인삼 투여군, 백삼 투여군, 방사선 조사군, 대조군으로 각각 분류하여 발효인삼 투여군(FG+Rad), 백삼 투여군(WG+Rad)은 $^{60}Co\;{\gamma}$선 조사 전에 백삼과 발효인삼을 7일간(150 mg/kg/day) 경구 투여하였고, 방사선 조사군(Rad)은 $^{60}Co\;{\gamma}$선을 5 Gy(1.01 Gy/min) 선량으로 조사하였으며, 대조군(Co)은 0.1 mL의 생리적 식염수를 투여하였다. 각각의 실험군을 통하여 생쥐 간 조직에서의 환원형 glutathione와 산화형 glutathione(GSSG)의 함량 변화를 조사한 결과 아래와 같다. 결 과: 1. 발효인삼 투여군과 백삼 투여군에서 방사선 조사군보다 유의성 있게 glutathione함량이 증가하였고 산화형 glutathione(GSSG)의 함량은 유의성 있게 감소하였다. 2. 발효인삼투여군과 백삼투여군에서 방사선 조사군보다 산화형 glutathione(GSSG)의 함량과 총 glutathione함량 비율은 유의성 있게 감소하였다. 결 론: 발효인삼투여군이 백삼투여군보다 glutathione함량을 증가시킴으로서 효율적으로 생쥐간의 방사선 방호효과를 증진시킴을 알 수가 있었다.

Quantitative PCR for Etiologic Diagnosis of Methicillin-Resistant Staphylococcus aureus Pneumonia in Intensive Care Unit

  • Background: Ventilator-associated pneumonia (VAP) requires prompt and appropriate treatment. Since methicillin-resistant Staphylococcus aureus (MRSA) is a frequent pathogen in VAP, rapid identification of it, is pivotal. Our aim was to evaluate the utility of quantitative polymerase chain reaction (qPCR) as a useful method for etiologic diagnoses of MRSA pneumonia. Methods: We performed qPCR for mecA, S. aureus-specific femA-SA, and S. epidermidis-specific femA-SE genes from bronchoalveolar lavage or bronchial washing samples obtained from clinically-suspected VAP. Molecular identification of MRSA was based on the presence of the mecA and femA-SA gene, with the absence of the femA-SE gene. To compensate for the experimental and clinical conditions, we spiked an internal control in the course of DNA extraction. We estimated number of colony-forming units per mL (CFU/mL) of MRSA samples through a standard curve of a serially-diluted reference MRSA strain. We compared the threshold cycle (Ct) value with the microbiologic results of MRSA. Results: We obtained the mecA gene standard curve, which showed the detection limit of the mecA gene to be 100 fg, which corresponds to a copy number of 30. We chose cut-off Ct values of 27.94 (equivalent to $1{\times}10^4$ CFU/mL) and 21.78 (equivalent to $1{\times}10^5$ CFU/mL). The sensitivity and specificity of our assay were 88.9% and 88.9% respectively, when compared with quantitative cultures. Conclusion: Our results were valuable for diagnosing and identifying pathogens involved in VAP. We believe our modified qPCR is an appropriate tool for the rapid diagnosis of clinical pathogens regarding patients in the intensive care unit.
