Serial Block-Face Imaging by Field Emission Scanning Electron Microscopy (전계방사형 주사전자현미경에 의한 연속블록면 이미징)

  • Backscattered electrons (BSE) are generated at the impact of the primary electron beam on the specimen. BSE imaging provides the compositional contrast to resolve chemical features of sectioned block-face. A focused ion beam (FIB) column can be combined with a field emission scanning electron microscope (FESEM) to ensure a dual (or cross)-beam system (FIB-FESEM). Due to the milling of the specimen material by 10 to 100 nm with the gallium ion beam, FIB-FESEM allows the serial block-face (SBF) imaging of plastic-embedded specimens with high z-axis resolution. After contrast inversion, BSE images are similar to transmitted electron images by transmission electron microscopy. As another means of SBF imaging, a specialized ultramirotome has been incorporated into the specimen chamber of FESEM ($3View^{(R)}$). Internal structures of plastic-embedded specimens can be serially revealed and analyzed by $3View^{(R)}$ with a large field of view to facilitate three-dimensional reconstruction. These two SBF approaches by FESEM can be employed to unravel spatial association of (sub)cellular entities for a comprehensive understanding of complex biological systems.

Study on the Thermal Properties and High Impact of Elastic Epoxy Blend System (탄성에폭시 블렌드 시스템의 열적 특성 및 내충격성에 관한 연구)

  • Elastic-factor of elastic epoxy were investigated by TMA (Thermomechanical Analysis), DMTA (Dynamic Mechanical Thermal Analysis), TGA (Thermogravimetric Analysis) and FESEM (Field Emission Scanning Electron Microscope) for structure-images analysis as toughness-investigation to improve brittleness of existing epoxy resin. A range of measurement temperature of the TMA and DMTA was changed from -20($^{\circ}C$) to $200^{\circ}(C)$, and TGA was changed from $0^{\circ}(C)$ to $600^{\circ}(C)$. Glass transition temperature (Tg) of elastic epoxy was measured through thermal analysis devices with the content of 0(phr), 20(phr) and 35(phr). Also, thermal expansion coefficient (a), high temperature, modulus and loss factor were investigated through TMA, TGA, and DMTA. In addition, the structure of specimens was analyzed through FESEM, and then elastic-factor of elastic epoxy was visually showed by FESEM. As thermal analysis results, 20(phr) was more excellent than 30(phr) thermally and mechanically. Specially, thermal expansion coefficient, high temperature, modulus, and damping properties were excellent. By structure-images analysis through FESEM, we found elastic-factor of elastic epoxy that is not existing epoxy, and proved high impact.

  • 본 연구에서는 MILC 및 FALC를 도핑 타입에 따른 온도별 패턴별 인가 전압별로 진행하여 현미경 및 FESEM 관찰을 함으로써 그 메커니즘을 규명하고자 하였다. LPCVD를 이용하여 $1000\;{\AA}$ a-Si 을 glass에 입힌 후 photolithography법 또는 Hard Mask법으로 Ni $200\;{\AA}$ 을 선택적으로 증착하였으며 Pt 전극을 Sputtering법으로 제작하였다. $33\;{\sim}\;200\;V/cm$의 전기장 하에서 MILC 속도가 2배 정도 증가되는 현상이 관찰되었으며 또한 인접패턴에 의해 FALC 속도가 영향을 받는 현상이 관찰되었다. 또한 전자가 움직이는 방향으로 MILC 선단영역 전후에 Void가 발생하는 영역이 존재함을 발견하였다. FESEM 분석을 통하여 FALC 영역 및 Void 영역을 관찰한 결과 도핑 종류에 따라 결정화 양상이 다른 것이 관찰되었으며 Void 분석결과 Charged vacancy가 어닐링시 결집되어 나타나는 것으로 분석할 수 있었다.

Study on PLLA Alloys with Impact Modifier and Talc (충격 보강제와 탈크를 이용한 PLLA 얼로이 연구)

  • In this work, PLLA/EGMA blends were prepared by melt blending of biodegradable Poly-L-lactic acid(PLLA) with Poly(ethylene-co-glycidyl methacrylate)(EGMA) and Engage as impact modifiers by twin screw extruder. Blend compositions of PLLA/Impact modifier blends were 100/0, 75/25, 50/50, 25/75 and 0/100, respectively. Also, Talc was added to 3 PLLA rich phases on PLLA/EGMA blends. The morphology, viscoelastic/mechanical properties were characterized by FESEM, DMA, UTM and Izod impact tester. DMA and Izod impact test data showed that storage modulus at room temperature with increasing EGMA and Engage contents decreased, and impact strength increased. However, storage modulus at room temperature increased by adding talc. From FESEM image, we observed that domain phase was well dispersed into matrix. Although the tensile strength and flexural modulus were decreased with increasing the content of EGMA and Engage in them, they could be supplemented by adding talc.

  • In this work, a nano-sized samaria-doped ceria(SDC) was prepared by a urea-based hydrothermal method and characterized by XRD, FESEM and TEM. It was observed that the increase in synthesis time and temperature gave rise to crystallity and particles size. Moreover, the synthesised powders had a excellent ion-conductivity(0.1 S/cm at 600~$800^{\circ}C$) which is suitable for electrolyte of intermediate temperature-solid oxide fuel cell(IT-SOFC). A STUDY ON SURFACE OF VARIOUS ABUTMENT SCREWS

  • Statement of problem: Regardless of any restoration, most of case, we used in screw connection between abutment and implant. For this reason, implant screw loosening has been remained problem in restorative practices. Purpose: The purpose of this study was to compare surface of coated/plated screw with titanium and gold alloy screw and to evaluate physical property of coated/plated material after scratch test in FESEM investigation Material and methods: GoldTite, titanium screw provided by 3i (Implant Innovation, USA) and TorqTite, titanium screw by Steri-Oss (Nobel Biocare, USA) and gold screw, titanium screw by AVANA (Osstem Implant, korea) - were selected for this study. Each abutment screw surface was observed at 100 times, and then screw crest, root, and slope were done more detailed numerical value, at 1000 times with FESEM. A micro-diamond needle was also prepared for the scratch test. Each abutment screw was fixed, micro-diamond scratch the surface of head region was made at constant load and then was observed central region and periphery of fine trace through 1000 times with FESEM. Results: The surface of GoldTite was smoother than that of other kinds of screw and had abundant ductility and malleability compared with titanium and gold screw. The scratch test also showed that teflon particles were exfoliated easily in screw coated with teflon. Titanium screw had a rough surface and low ductility. Conclusion: It was recommended that the clinical use of gold-plated screw would prevent a screw from loosening. CLINICAL IMPLICATIONS Clinical use of gold-plated screw would prevent a screw from loosening because it had abundant ductility and malleability compared with titanium and gold screw.

Ambient Variable Pressure Field Emission Scanning Electron Microscopy for Trichome Profiling of Plectranthus tomentosa by Secondary Electron Imaging

  • Glandular and nonglandular trichomes on the leaf surface of Plectranthus tomentosa were investigated by variable pressure field emission scanning electron microscopy (VP-FESEM). The segments of the plant's leaves were directly mounted without any specimen preparation, and examined at ambient temperature using a variable pressure secondary electron (SE) detector under ca. 15 Pa. Foliar trichomes maintained their shapes and structures without severe surface collapse or charging. The adaxial leaf surface was abundantly covered with different types of trichome. Nonglandular trichomes consisted of a basal cell and a long (up to ca. $300{\mu}m$) stalk. Meanwhile, capitate glandular trichomes had a secretory head and a short or long stalk. Peltate glandular trichomes with globose secretory heads were observed in close contact with the leaf epidermis. Spherical projections on the secretory head showed the secretion process of glandular trichomes. In addition to the trichomes, oval stomata were distributed on the abaxial leaf surface. These results suggest that ambient VP-FESEM can be used to classify the dehydration-sensitive foliar trichomes of succulent plants by SE imaging. At the FESEM resolution, this approach facilitates the rapid and detailed morphological analysis of a variety of trichomes in diverse plant taxa with reduced labor and preparation.

  • A focused ion beam (FIB) system produces a beam of positive ions (usually gallium) which are heavier than electrons and can be focused by electrostatic lenses into a spot on the specimen. With its ability milling of the specimen material by 10 to 100 nm with each pass of the beam, FIB is widely adopted in materials science, semiconductor industry, and ceramics research. Recently, FIB has been increasingly employed in the field of biomedical sciences. Here we provide a brief introduction to FIB and its applications for a wide variety of biomedical research. The surface of specimen can be in situ processed and quasi-real time visualized by two beam combination of FIB and field emission scanning electron microscope (FESEM). Due to its milling process, internal structures can be exposed and analyzed: yeast cells, fungus-inoculated wheat leaf, mannitol particles in inhalation aerosols, and oyster shell. Serial blockface tomography with the system kindles 3-dimensional reconstruction researches in the realm of nervous system and life sciences. Two-beam system of FIB/FESEM is a versatile tool to be utilized in the biomedical sciences, especially in 3-dimensional reconstruction studies.

Thermal and Structural Properties of Epoxy/Elastomer Blend (에폭시/엘라스토머 블렌드의 열적, 구조적 특성)

  • In this paper, the elastic epoxy added elastomer having viscoelasticity to existing epoxy was measured thermal, structural properties by DSC (Differential Scanning Calorimetry) and FESEM (Field Emission Scanning Electron Microscope). Specimens were made of dumbbell forms by the ratio of 5, 10, 15, and 20[phr] by regulation with elastomer contents. The measurement temperature dimensions of DSC were -20[$^{\circ}C$] to 150[$^{\circ}C$] and rising temperature was 4[$^{\circ}C$/min]. Also we observed structure through FESEM at the magnification of 1000 times with the voltage of 15[kV] after breaking by quenching specimens. As experimental results, we could know that thermal and structural properties were improved quantity according to decrease of elastomer contents. Namely, it increased glass transition temperature, high temperature, and matrix structure. In general, thermal, structural properties of 15[phr] was excellent among the specimens.

Surface Modification of Polyacrylonitrile by Low-temperature Plasma (저온플라즈마처리에 의한 폴리아크릴로니트릴의 표면개질)

  • Polyacrylonitrile(PAN) fiber was treated with low-temperature plasmas of argon and oxygen for surface modification, and its surface chemical structure and morphology were examined by a field emission scanning electron microscope(FESEM) and a Fourier-transform infrared microspectroscopy(IMS). The argon-plasma treatment caused the only mechanical effect by sputtering of ion bombardment, whereas the oxygen plasma brought about a chemical effect on the PAN fiber surface. The experimental evidences strongly suggested that cyclization of nitrile group and crosslinking were likely to occur in the oxygen-plasma treatment. On the other hand, with the argon-plasma treatment, numerous my pits resulted in ranging from several tens to hundreds nanometers in radius. The plasma sensitivity of functional groups such as C-H, $C{\equiv}N$, and O-C=O groups in the PAN fiber was dependent on their chemical nature of bonding in the oxygen-plasma, in which the ester group was the most sensitive to the plasma. Vacuum-ultraviolet(VUV) radiation emitted during plasma treatment played no substantial role to alter the surface morphology.
