Effects of Waterborne Iron on Serum Iron Concentration and Iron Binding Capacity of Olive Flounder (Paralichthys olivaceus)

  • Olive flounder (Paralichthys olivaceus) was exposed to waterborne iron (0.1, 0.5, 1, 5 and 10 mg/L) for 50 days. The effects of iron on blood iron status and iron binding capacity were studied. The serum iron concentration was significantly higher than in the group exposed to iron (1, 5 and 10 mg/L) in comparison to the control after 30 days of exposure to iron. A significant decrease in unsaturated iron binding capacity was found between the control and the group exposed to iron (5 and 10 mg/L, respectively) at 40 and 50 days, respectively. The total iron binding capacity of serum in the fish exposed to iron concentrations (5 and 10 mg/L) showed a significant decrease compared to that of the control at 40 days after iron exposure. Serum iron saturation values increased in the flounder exposed to iron concentration (5 and 10 mg/L) at 50 days. Our data suggest that sub-lethal exposure of waterborne iron alters the blood iron concentration and iron binding capacity, and these parameters seems to be valuable factors for screening and diagnosis of iron overload syndromes in fish.

Short-term Toxicity Assay Based on Daphnid Feeding on the Microalga Scenedesmus subspicatus

  • We developed and evaluated a method of short-term acute toxicity testing based on the feeding behavior of Ceriodaphnia dubia. In prior toxicity tests, neonates of C. dubia were hatched and cultivated with the addition of yeast only for the preparation of the transparent daphnid's gut. Scenedesmus subspicatus was supplied as food after 1 to 6 h of exposure to toxicants. The effects of 1-h and 6-h exposure time on test sensitivity did not significantly differ. A comparison of the short-term l-h acute toxicity test developed in this study to the standard 48-h acute toxicity test using heavy metals, cyanide, and pentachlorophenol indicated that the 1-h test provided an acceptable sensitivity level in toxicity testing of C. dubia..

  • Fas-associated death domain (FADD) is an adapter molecule that bridges the interaction between receptor-interacting protein 1 (RIP1) and aspartate-specific cysteine protease-8 (caspase-8). As the primary mediator of apoptotic cell death, caspase-8 has two N-terminal death-effector domains (DEDs) and it interacts with other proteins in the DED subfamily through several conserved residues. In the tumor necrosis receptor-1 (TNFR-1)-dependent signaling pathway, apoptosis is triggered by the caspase-8/FADD complex by stimulating receptor internalization. However, the molecular mechanism of complex formation by the DED proteins remains poorly understood. Here, we found that direct DED-DED interaction between FADD and caspase-8 and the structure-based mutations (Y8D/I128A, E12A/I128A, E12R/I128A, K39A/I128A, K39D/I128A, F122A/I128A, and L123A/I128A) of caspase-8 disrupted formation of the stable DED complex with FADD. Moreover, the monomeric crystal structure of the caspase-8 DEDs (F122A/I128A) was solved at 1.7 Å. This study will provide new insight into the interaction mechanism and structural characteristics between FADD and caspase-8 DED subfamily proteins.

  • The suitability of any fish species for successful aquaculture requires basic information on its reproduction and growth. This study investigated some facets of the reproductive biology of Synodontis shall (Mochokidae) from River Siluko in Nigeria. Fish samples were collected forth-nightly for a duration of fourteen (14) months-March 2015 to April, 2016 with the assistance of artisanal fishermen. Fishes were identified using taxonomic guides and standard techniques were used for determination of sex ratio, gonad maturation and fecundity. Linear regression method was used to define the correlation between fecundity and fish length, body weight and ovary weight. Results showed that sex ratio did not indicate a significant divergence (p > 0.05) from the 1 male to 1 female distribution ratio (1:1.41). Gonad morphology revealed paired gonads. Testes and ovaries were classified into four maturity stages: immature, resting, ripening and ripe. Gonadosomatic index ranged from 0.04 to 5.68 (males) and 0.03 to 20.19 (females). Absolute fecundity ranged from 1,014 to 4,520 eggs (mean = 2,592 eggs) and did not correlate significantly (p > 0.05) to ovary weight. This study has contributed to existing data on the biology of freshwater fish species in Nigeria and provided valuable information for fishery management tools in the conservation and utilization of this valuable freshwater fish species.

Choline supplementation improves the lipid metabolism of intrauterine-growth-restricted pigs

  • Objective: The objective of this study was to investigate the effects of dietary choline supplementation on hepatic lipid metabolism and gene expression in finishing pigs with intrauterine growth retardation (IUGR). Methods: Using a $2{\times}2$ factorial design, eight normal birth weight (NBW) and eight IUGR weaned pigs were fed either a basal diet (NBW pigs fed a basal diet, NC; IUGR pigs fed a basal diet, IC) or a diet supplemented with two times more choline than the basal diet (NBW pigs fed a high-choline diet, NH; IUGR pigs fed a high-choline diet, IH) until 200 d of age. Results: The results showed that the IUGR pigs had reduced body weight compared with the NBW pigs (p<0.05 from birth to d 120; p = 0.07 from d 120 to 200). Increased (p<0.05) free fatty acid (FFA) and triglyceride levels were observed in the IUGR pigs compared with the NBW pigs. Choline supplementation decreased (p<0.05) the levels of FFAs and triglycerides in the serum of the pigs. The activities of malate dehydrogenase and glucose 6-phosphate dehydrogenase were both increased (p<0.05) in the livers of the IUGR pigs. Choline supplementation decreased (p<0.05) malate dehydrogenase activity in the liver of the pigs. Gene expression of fatty acid synthase (FAS) was higher (p<0.05) in the IC group than in the other groups, and choline supplementation decreased (p<0.05) FAS and acetyl-CoA carboxylase ${\alpha}$ expression in the livers of the IUGR pigs. The expression of carnitine palmitoyl transferase 1A (CPT1A) was lower (p<0.05) in the IC group than in the other groups, and choline supplementation increased (p<0.05) the expression of CPT1A in the liver of the IUGR pigs and decreased (p<0.01) the expression of hormone-sensitive lipase in both types of pigs. The gene expression of phosphatidylethanolamine N-methyltransferase (PEMT) was higher (p<0.05) in the IC group than in the other groups, and choline supplementation significantly reduced (p<0.05) PEMT expression in the liver of the IUGR pigs. Conclusion: In conclusion, the lipid metabolism was abnormal in IUGR pigs, but the IUGR pigs consuming twice the normal level of choline had improved circulating lipid parameters, which could be related to the decreased activity of nicotinamide adenine dinucleotide phosphate-generating enzymes or the altered expressions of lipid metabolism-related genes.

  • Erbium 첨가 광섬유(EDF) 광원은 출력 특성과 온도에 대한 파장 특성이 우수하여 Sagnac 간섭계의 원리를 이용한 광섬유 자이로스코프(이하 줄여 자이로라 함)에 많이 사용되고 있다. 이득매질인 EDF를 광원 겸 광증폭기로 사용하는 광섬유 증폭기형 광원 (Fiber Amplifier/source : FAS) 방식$^{[l-2]}$ 은 기존의 single-pass 방식$^{[3]}$ 에 비해서 구조가 단순하고 검출광 power가 크다는 장점이 있다. 그런데, 검출광 power가 큰 경우에 자이로의 SNR이 광원의 과잉잡음(excess noise)에 의해서 제한되므로 실제로 자이로의 측정감도는 개선되지 않는 문제점이 있다.$^{[4]}$ Single-pass 방식의 광원을 사용하는 경우, 적절한 신호처리를 통해 자이로 출력신호에 포함된 광원의 과잉잡음의 적정주파수 성분을 소거함으로써 자이로 신호의 SNR을 개선시킨 바 있었다.$^{[5]}$ 그러나, 일반적으로 single-pass 방식의 경우에는 검출광 power가 작아서 자이로의 SNR이 광원의 과잉잡음에 의해서 제한되는 경우는 드물다. 반면에 증폭기형 광원 방식은 자이로로부터 되돌아오는 신호광이 다시 광원으로 입사되어 EDF를 반대 방향으로 진행하는 동안 증폭되기 때문에 충분히 큰 검출광 power를 얻을 수 있다. 따라서, 자이로 신호에 포함된 광원의 과잉잡음이 소거된다면 자이로 신호의 SNR은 크게 개선될 것으로 여겨진다. 이 논문에서는 광섬유 증폭기형 광원 방식(FAS)의 자이로에 대해 위와 같은 신호처리를 이용하여 광인의 과잉잡음의 적정주파수 성분을 소거하는 실험을 하였다. Induction of Apoptosis in Human Colon Carcinoma HCT116 Cells Using a Water Extract of Lepidium virginicum L. (콩다닥냉이 추출물에 의한 HCT116 대장암세포의 사멸 유도에 관한 연구)

  • To examine the anti-cancer effects of Lepidium virginicum L., the anti-proliferative and pro-apoptotic effects of a water extract of L. virginicum leaves (WELVL) and of L. virginicum roots (WELVR) were investigated in HCT116 human colon carcinoma cells. The treatment of HCT116 cells with WELVL and WELVR resulted in the inhibition of growth and morphological changes in a concentration-dependent manner by inducing apoptosis. The growth inhibition and apoptosis induction by WELVR was stronger than that of WELVL thus, we determined that WELVR was the more optimal extract for this study. The increased apoptotic events in HCT116 cells caused by WELVR were associated with an up-regulation of Fas ligand, Bax, and Bad expression, a down-regulation of Bcl-2, Bcl-$_XL$, and Bid expression, and a decrease in the mitochondrial membrane potential (MMP, ${\Delta}{\psi}m$). WELVR treatment induced the proteolytic activation of caspase-3, -8, and -9, and the degradation of caspase-3 substrate proteins, such as poly (ADP-ribose) polymerase (PARP), ${\beta}$-catenin, and phospholipase C-${\gamma}1$ (PLC-${\gamma}1$). In addition, apoptotic cell death induced by WELVR was correlated with a down-regulation of inhibitors of the apoptosis protein (IAP) family, such as the X-linked inhibitor of apoptosis protein (XIAP), cIAP-1, and cIAP-2. These findings suggest that the WELVR-induced inhibition of cell proliferation is associated with the induction of apoptotic cell death. WELVR may be a potential chemotherapeutic agent for the control of HCT116 human colon carcinoma cells.

Activation of Pro-Apoptotic Multidomain Bcl-2 Family Member Bak and Mitochondria-Dependent Caspase Cascade are Involved in p-Coumaric Acid-Induced Apoptosis in Human Jurkat T Cells (p-Coumaric acid에 의해 유도되는 인체 Jurkat T 세포의 에폽토시스 기전)

  • The apoptogenic effect of p-coumaric acid, a phenolic acid found in various edible plants, on human acute leukemia Jurkat T cells was investigated. Exposure of Jurkat T cells to p-coumaric acid (50-$150{\mu}M$) caused cytotoxicity and TdT-mediated dUTP nick-end labeling (TUNEL)-positive apoptotic DNA fragmentation along with Bak activation, ${\Delta}{\psi}m$ loss, activation of caspase-9, -3, -7, and -8, and PARP degradation in a dose-dependent manner. However,these apoptotic events were completely abrogated in Jurkat T cells overexpressing Bcl-2.Under these conditions, necrosis was not accompanied. Pretreatment of the cells with the pan-caspase inhibitor (z-VAD-fmk) could prevent p-coumaric acid-induced sub-$G_1$ peak representing apoptotic cells, whereas it failed to block ${\Delta}{\psi}m$ loss, indicating that the activation of caspase cascade was prerequisite for p-coumaric acid-induced apoptosis as a downstream event of ${\Delta}{\psi}m$ loss. FADD- and caspase-8-positive wild-type Jurkat T cell clone A3, FADD-deficient Jurkat T cell clone I2.1, and caspase-8-deficient Jurkat T cell clone I9.2 exhibited similar susceptibilities to the cytotoxicity of p-coumaric acid, excluding an involvement of Fas/FasL system in triggering the apoptosis. The apoptogenic activity of p-coumaric acid is more potent in malignant Jurkat T cells than in normal human peripheral T cells. Together, these results demonstrated that p-coumaric acid-induced apoptogenic activity in Jurkat T cellswas mediated by Bak activation, ${\Delta}{\psi}m$ loss, and subsequent activation of multiple caspases such as caspase-9, -3, -7, and-8, and PARP degradation, which could be regulated by anti-apoptotic protein Bcl-2.

Delay Bound Analysis of Networks based on Flow Aggregation (통합 플로우 기반 네트워크의 지연시간 최대치 분석)

  • We analyze the flow aggregate (FA) based network delay guarantee framework, with generalized minimal interleaved regulator (IR) initially suggested by IEEE 802.1 time sensitive network (TSN) task group (TG). The framework has multiple networks with minimal IRs attached at their output ports for suppressing the burst cascades, with FAs within a network for alleviating the scheduling complexity. We analyze the framework with various topology and parameter sets with the conclusion that the FA-based framework with low complexity can yield better performance than the integrated services (IntServ) system with high complexity, especially with large network size and large FA size.

Kaurenoic acid, a Diterpene Derived from Aralia continentalis, Alleviates Lipogenesis in HepG2 Cells

  • Objectives: Here we investigated the anti-lipogenic potential of kaurenoic acid (KA), a diterpene derived from Aralia continentalis, in a cellular model of non-alcoholic fatty liver disease. Methods: HepG2 cells were treated with palmitate for 24h to induce intracellular lipid accumulation. To assess the influence of KA on steatotic HepG2 cells, various concentration of KA was co-administered. After palmitate treatment, Intracellular triglyceride content was measured. Expression level of several lipogenic genes, sterol regulatory element-binding transcription factor-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), and stearoyl-CoA desaturase-1 (SCD-1) were measured using Western-blot analyses or RT-PCR. Results: Palmitate markedly increased intracellular triglyceride level and expression of related lipogenic genes in HepG2 cells, and which was relieved by co-administered KA. Conclusions: It is conceivable that that KA may have a pharmacological potential to reduce lipid accumulation in non-alcoholic fatty liver disease.
