BSA 또는 Fatty Acid Free BSA 첨가가 닭 희석 정자와 동결 정자의 생존성에 미치는 영향 (The Effects of Supplementation of BSA or Fatty Acid Free BAS on the Motility of Fresh or Cryopreserved Rooster Spermatozoa)

  • 본 연구에서는 닭 정액의 운동성 분석에 미치는 요인을 분석하기 위하여 희석배율에 따른 생존성을 조사하고, 신선정액의 희석 시에 필요한 조건을 확보하기 위하여 희석제에 단백질원으로서 BSA와 FAF-BSA의 첨가가 닭정자의 생존성에 미치는 영향을 관찰하였다. 최종 희석율이 1/4이 되도록 닭 정액을 희석하여 생존성을 조사하면 신선정액에서 희석율이 1/8, 1/16 및 1/32로 낮아질수록 89.9%, 69.9% 및 53.2%로 생존율이 낮아지는 것을 관찰하였으며, 단백질원으로 BSA나 FAF-BSA를 이용하면 생존율을 유의적으로 증진시킬 수 있음을 증명하였다. 또한 이러한 현상은 동결정액의 희석에도 중요한 요인으로 관찰되었는데, FAF-BSA를 이용하여 희석할 때 단백질이 없는 경우보다 양호한 생존율이 나타난다는 것을 관찰하였다(17.6% 대 34.0%). 그러나 단백질원을 활용하더라도 닭정액의 희석율이 낮아질수록 생존율이 저하하는 현상은 지속적으로 나타나고 있으며, 신선정액에 비하여 동결정액은 장수성이 낮은 것으로 추정되었다. 그러므로 본 연구에서 사용된 정자의 분석방법은 닭 정액동결을 실시하여 운동성을 분석할 때 필요한 기본 정보를 제공할 수 있을 것으로 판단되며, 닭 희석배율을 낮추어 동결정액의 효율성을 높이는 연구에 기여할 것으로 추정된다.

Crystal Structure of p97-N/D1 Hexamer Complexed with FAF1 UBX Domain

  • p97, a universally conserved AAA+ ATPase, holds a central position in the ubiquitin-proteasome system, orchestrating myriad cellular activities with significant therapeutic implications. This protein primarily interacts with a diverse set of adaptor proteins through its N-terminal domain (NTD), which is structurally located at the periphery of the D1 hexamer ring. While there have been numerous structural elucidations of p97 complexed with adaptor proteins, the stoichiometry has remained elusive. In this work, we present the crystal structure of the p97-N/D1 hexamer bound to the FAF1-UBX domain at a resolution of 3.1 Å. Our findings reveal a 6:6 stoichiometry between the p97 hexamer and FAF1-UBX domain, deepening our understanding from preceding structural studies related to p97-NTD and UBX domain-containing proteins. These insights lay the groundwork for potential therapeutic interventions addressing cancer and neurodegenerative diseases.

Developmental Potential of Bovine Nuclear Transfer Embryos Cultured in Serum-free Medium

  • The purpose of this study was to investigate the development of bovine nuclear transfer (NT) embryos cultured in serum-free conditions. Bovine NT embryos cultured in various culture conditions were compared blastocyst development, total cell number and apoptosis using TUNEL assay. In experiment 1, blastocyst rates of NT embryos were significantly higher (P<0.01) in FBS (22.0%) and BSA (26.6%) groups than in PVA (6.3%) group. Total cell number was significantly higher in FBS (78.4$\pm$19.4) and BSA (90.9$\pm$29.1) groups than in PVA group (46.0$\pm$0.0). Apoptotic cell number was significantly fewer in FBS (3.1$\pm$1.4) and BSA (1.7$\pm$1.4) groups than in PVA group (7.0$\pm$20.0) However, all of results were not different between the FBS and BSA group. In experiment 2, blastocyst rates of NT embryos were significantly higher (P<0.05) in fatty acid free-BSA (FAF-BSA) group (26.8%) than in fraction V-BSA group (11.2%). Total cell number were somewhat higher in FAF-BSA group (89.8$\pm$30.7) than in fraction V-BSA group (88.1$\pm$19.3). Apoptotic cell number were somewhat fewer in FAF-BSA (1.7$\pm$1.5) group than in fraction V-BSA group (4.2$\pm$2.9). These findings suggest that serum free condition were effective for the in vitro development of bovine NT embryos. Therefore, we concluded that fatty acid free-BSA has beneficial effect in development bovine NT embryos and can be use as a serum substitute.

A Maximum Likelihood Estimator Based Tracking Algorithm for GNSS Signals

  • This paper presents a novel signal tracking algorithm for GNSS receivers using a MLE technique. In order to perform a robust signal tracking in severe signal environments, e.g., high dynamics for navigation vehicles or weak signals for indoor positioning, the MLE based signal tracking approach is adopted in the paper. With assuming white Gaussian additive noise, the cost function of MLE is expanded to the cost function of NLSE. Efficient and practical approach for Doppler frequency tracking by the MLE is derived based on the assumption of code-free signals, i.e., the cost function of the MLE for carrier Doppler tracking is used to derive a discriminator function to create error signals from incoming and reference signals. The use of the MLE method for carrier tracking makes it possible to generalize the MLE equation for arbitrary codes and modulation schemes. This is ideally suited for various GNSS signals with same structure of tracking module. This paper proposes two different types of MLE based tracking method, i.e., an iterative batch processing method and a non-iterative feed-forward processing method. The first method is derived without any limitation on time consumption, while the second method is proposed for a time limited case by using a 1st derivative of cost function, which is proportional to error signal from discriminators of conventional tracking methods. The second method can be implemented by a block diagram approach for tracking carrier phase, Doppler frequency and code phase with assuming no correlation of signal parameters. Finally, a state space form of FLL/PLL/DLL is adopted to the designed MLE based tracking algorithm for reducing noise on the estimated signal parameters.

Steroid 물질처리를 받은 Chinese hamster 세포에 있어서의 염색체 이상 (Some Chromosome Alteranations in the Cultured Chinese Hamster Cells Treated by Steroids)

  • This study is concerned with alterations in chromosomes (numbers and morphology) when the culture of Chinese hamster cells (FAF-28 strain) was treated by steroids, testosterone and DOC. 1. In 200 cells of normal untreated cells as control population the chromosome of stemline was decided as which was contained in 158 cells ; that is , in 79 percent of the population. The average chromosome number in above 20 cells observed was calculated as 23.95 with minimum limit at 20 and maximum limit at 70. 2. Many different chromosome numbers, ranging from 19 to 352 were observed in the 200 cells treated by testosterone. The diploid number of 22 showed the peak of variation curve was counted in 71 cells (35.5%) and an average chromosome number of stemline was 22 which was counted in 74 cells (37%). While all of the chromosome number of stemline was 22 which was counted in 74 cells (37%). While all of the chromosome numbers in the 200 cells observed ranged from 20 to 181 , an average chromosome number was also found to be 30.09. 4. The chromosome component in the cultured normal FAF-28 cells with 22 diploid chromosomeswas as follows ; 9a) 2 paris were long and metacentric (LM), (b) 3 pairs were medium length and metacentric (MM), (c) 3 pairs were small and subtelocentric (SS) and (d) 3 pairs were small and metacentric (SM). 5. The twenty cells with 44 chromosomes were selected at random from each cell population treated with testosterone and DOC , so that chromosome idiogram and morphology could be studies. In the twenty cells of the testosterone treated population the average ratio of above four groups, LM ; MM;Ss:SM, was found to be 8.6 : 10.8:13.5:10.7. On the other hand, the average ratio in the same number of cells of the DOC treated one was 7.7 :11.4:12.5:12.7. 6. The five types of the altered chromosomes morphologically in the hundred cells selected at random from each cell population treated by testosterone and DOC were observed (Type I-V). The thirty-one altered chromosomes were found to be in the testosterone treated cell population and the sixteen in DOC treated.

Ca과 BSA가 소 정자세척액내 수소이온농도에 미치는 영향 (Effect of Ca and BSA on Hydrogen Ion Concentration in Bovine Sperm Washed Solution)

  • This study was carried out to investigate the effects of Ca and BSA on hydrogen ion concentration in sperm washed solution. The results obtained were as follows : 1. The hydrogen concentration in 1st and 2nd sperm washed solutions was signifcinatly(p<0.01) higher when sperm was washed with SHPsolution containing 2mM Ca than when sperm washed with SHP solution or SHP solution containing 10mM Ca. 2. The hydrogen ion concentration in sperm washed solution was significnatly(p<0.05) higher when seprm was washed with SHP solution containing BSA-FAF than when sperm was washed with SHP solution or SHP solution containing BSA-V.

배양대뇌신경세포 저산소증모델에서 반하여 의한 유전자표현의 변화 (Microarray Analysis of Gene Expression Affected by Water-extracts of Pinelliae rhizoma in a Hypoxic Model of Cultured Rat Cortical Cells)

  • 본 연구는 저산소증에서 반하가 대뇌신경세포의 유전자 표현에 미치는 영향을 알아보기 위하여 배양한 $E_{18}$의 흰쥐 대뇌 신경세포를 반하로 처리하고, 저산소증을 유도한 후 microarray 기법으로 유전자 표현 변화를 조사하였다. Microarray 결과 tubb5, tgfa, ptpn11, n-ras, pdgfa 등 세포의 성장 분화에 관여하는 유전자들의 표현이 증가하였으며, 세포 자연사를 억제하는 mcl-1 유전자의 표현 또한 증가하였다. 한편 세포 자연사를 유도하는 tieg 유전자는 표현이 감소하였다(Fig. 3). 반하에 의하여 수많은 유전자의 표현이 변화되었고, 세포사를 촉진하는 유전자의 표현이 크게 증가되는 경우(예, alox12, faf1)도 있어 본 연구결과만으로 일반적인 결론을 유도하기는 어려웠다. 그러나 대략적으로 반하는 저산소증에서 주로 세포의 성장과 분화를 유지하고, 세포 자연사를 방지하는 유전자들의 표현을 증가시켜 신경 세포사를 보호하는 것으로 이해된다.

착상전 생쥐배아의 Glucose에 대한 노출이 체외 배발생에 미치는 영향 (Effect of Glucose Exposure on the Development of the Mouse Preimplantation Embryo In Vitro)

  • 본 연구는 1-세포기배의 glucose에 대한 노출이 상실배기 이후의 배발생에 미치는 영향을 검토하고자 실시되었다. hCG 주사 후 24~25시간 째에, F1hybrid(C57BL/6, ♀ $\times$CBA/N, ♂) 계통 생쥐를 도살하여 1-세포기배를 회수한 후 0.1% hyaluronidase로 처리하여 난구세포를 제거하였다. 1-세포기배는 hCG 주사 후 72시간째 다양한 농도의 glucose(5.5, 16.5, 27.5 및 38.5mM)에 1분 노출 후 glucose가 첨가되어 있거나 혹은 첨가되지 않은 CR1aa배양액에서 계속 배양함으로써 배발생을 유도하였다. 이 실험의 결과를 요약하면 다음과 같다. 1. M2 배양액에서 회수한 후 3mg/ml의 Fatty-acid free BSA가 첨가된 배양액에서 배양한 경우 27.5%의 확장배반포까지의 배발생율과 16.6%의 탈출 배반포까지의 배발생율을 나타낸 반면, TL HEPES 배양액에서 회수한 경우는 전혀 상실배기 이후의 배발생이 나타나지 않았다. 2. hCG 주사 후 72시간째에 단 1분간의 27.5mM glucose에 대한 노출만으로도 68.8% (CR1aa+BSA)와 77.1%(CR1aa+FBS)의 확장배반포까지의 발생을 유도할 수 있었다. 그러나 1분 노출과 이후 계속되는 노출간에는 배발생에 있어서 유의차는 인정되지 않았다. 3. hCG 주사 후 72시간째에 5.5, 16.5, 27.5 및 38.5mM의 glucose 첨가에 따른 확장 배반포까지의 배발생율은 45.7~61.5%로 각 처리군의 유의차는 없었으며, 따라서 고농도의 glucose 첨가에 따른 저해효과는 확인할 수 없었다.
